During weaning, the transition to solid food diversifies the gut microbiome, triggering a programmed immune response critical for long-lasting mucosal immunity. Previous work showed that the gut microbiome mediates epigenetic development in intestinal stem cells (ISCs) during suckling, but what happens during weaning is unclear. Here, genome-wide profiling revealed that weaning-driven microbiome changes shape the DNA methylome and transcriptome of murine ISCs in an IFNγ-dependent manner. Specifically, we observe demethylation of enhancer elements essential for MHC class II genes, which results in a transcriptional memory that persists through differentiation into adulthood. This paper explores how the recovery of populations of Semibalanus balanoides is affected by climate variation. In intertidal marine ecosystems, larval recruitment is important in shaping all ecological processes. S. balanoides is a cold-water barnacle species that is competitively dominant over Chthamalus montagui and C. stellatus, two warm-water species. This competitive interaction is well studied, though it is not understood how that competition influences the recovery of S. balanoides after a disturbance nor how this process varies over a temperature gradient. 19 sites were studied across the United Kingdom, spanning from northern Scotland to southwestern England. Protein-protein interactions are important targets in cancer biology as they can make historically “undruggable” proteins viable drug targets. However, the current approach to targeting protein-protein interactions relies on the use of small molecule drugs, which have low efficiencies as they must cover a large surface area of the interaction. Further, traditional drug therapies often lead to off-target toxicity, limiting therapeutic specificity. Cannabidiol (CBD) is a non-psychoactive phytocannabinoid derived from Cannabis Sativa with well-documented antioxidant and anti-inflammatory properties, yet its clinical utility is limited by moderate receptor selectivity, rapid hepatic metabolism, and broad pharmacological activity on the central nervous system (CNS). This thesis describes the design and partial execution of a multi-step synthetic route aimed at generating CBD/CBD-derived chalcone intermediates for potential therapeutic applications for neuroinflammation. The steps encompass an O-methylation, O-acetylation, and a Fries rearrangement. The first three steps were completed successfully and reproducibly shown through 1H-NMR and 12C-NMR; however, purification of the fourth step was not successful within the available …

Antibiotic resistance is a rapidly emerging issue within public health. As resistance grows, the effectiveness of traditional antibiotic therapies diminishes while risk of treatment grows. Alongside novel therapies, public knowledge is a strong combatant of the resistance-related risks of antibiotic usage. Through a comprehensive review of current literature, this work serves as an accessible guide to the mechanisms of antibiotics, bacterial resistance, and the challenges related to current antibiotic usage. Hypertension is the leading global risk factor for mortality and is closely associated with vascular inflammation and dysfunction. Damaged mitochondria release cell-free mitochondrial DNA (cf-mtDNA), which activates toll-like receptor 9 (TLR9). The binding of TLR9 to mtDNA activates the NLRP3 inflammasome pathway, promoting the release of pro-inflammatory cytokines via gasdermin D (GSDMD), therefore worsening inflammation. However, the mechanisms linking mitochondrial injury to inflammation-induced vascular dysfunction remain unclear. We hypothesize that cf-mtDNA triggers TLR9-dependent NLRP3 inflammasome activation contributing to vascular and endothelial dysfunction. Small body size and rapid metabolism make annual migrations challenging for North American hummingbirds. Many of these birds make annual journeys of over 10,000 km. During migrations, hummingbirds use stopover sites, where they remain for several days to accumulate body fat. At these stopovers, they often encounter uncertainties in food resources, competition, and weather. To save energy, hummingbirds sometimes enter nocturnal torpor. My dissertation uses respirometry data, lipid measurements from Quantitative Magnetic Resonance, and feather hydrogen stable isotopes to understand how energy levels, migration patterns, torpor use, and water balance interact in four species of migrating hummingbirds. This thesis is concerned with determining the optimal water treatment and harvesting strategy for Lemna minor duckweed plants. The main objective of this thesis is to examine how duckweed plants grow in terms of biomass, photosynthetic activity, leaf morphology, chlorophyll and carotenoid content, and their efficiency in nutrient removal from different water sources. The methodology of the study included assessing duckweed in three water samples including tap water, saline water, and greywater and three harvesting strategies, including low, medium, and high harvesting intensities to assess their growth and nutrient removal efficiency. The One Big Beautiful Bill (OBBB) promises billions in new farm subsidies, but most of that money won’t go to the farmers who need it most. This brief describes how the OBBB’s changes to agricultural subsidy programs will disproportionately benefit large farms and non-farming investors, accelerating farm loss and consolidation across the United States. The authors argue that without reforms, the OBBB will inflate land values, raise barriers for new and small farmers, and further fuel absentee farmland investment. The authors also identify four reforms policymakers should enact to prevent these devastating consequences, including tightening payment limits and closing loopholes, …

In the neuroscience intensive care unit (NSICU), nurses rely on advanced monitoring tools to detect neurological deterioration. Two noninvasive measures, heart rate variability (HRV) and pupillary light reflex (PLR), are used to assess autonomic nervous system function; however, it remains unclear whether these indicators provide overlapping or distinct information. The purpose of this observational prospective study is to evaluate the relationship between HRV and quantitative pupillometry measurements to determine whether these noninvasive indicators provide congruent information. The primary goal of the Arkansas Cotton Variety Tests is to provide unbiased data regarding the agronomic performance of cotton varieties and advanced breeding lines in the major cotton-growing areas of Arkansas. This information helps seed companies establish marketing strategies and assists producers in choosing varieties to plant. These annual evaluations will then facilitate the inclusion of new, improved genetic material in Arkansas cotton production. Purpose: Inhibitors of poly(ADP-ribose) polymerase (PARP), an enzyme with numerous roles in DNA damage response signaling, represent a class of anti-cancer drugs approved for treating solid tumors with defects in the DNA damage response. However, additional methods for identifying patients who would benefit from the use of PARP inhibitors are urgently needed. We evaluated a novel radiotracer, 18F-FluorThanatrace ([18F]-FTT), to noninvasively assess PARP activity with PET/CT before treatment and determine associations between uptake, tumor mutation status, and prior receipt of therapy including PARP inhibitors.

Purpose: To investigate contrast-enhanced T1-weighted MRI and diffusion-weighted magnetic resonance imaging (DWI) for early prediction of tumor response to combined irreversible electroporation (IRE) and anti-PD-1 immunotherapy.
Methods: Murine pancreatic ductal adenocarcinoma (PDAC) cells KRAS* were inoculated into the pancreas of C57BL/6 mice. IRE was performed when tumors became palpable. After IRE, mice received six intraperitoneal injections of anti-PD-1 over 2 weeks. T2-weighted MRI, T1-weighted MRI with macromolecular contrast agent poly(L-glutamic acid)-conjugated gadolinium (PG-Gd), and DWI were performed before and after IRE. Survival was analyzed using Kaplan-Meier curves. Mice surviving at least 100 days without recurrence after IRE were considered complete …

Background & aims: Sorafenib has been a cornerstone of hepatocellular carcinoma (HCC) therapy; however, its efficacy is limited, and identifying patients who will benefit from sorafenib is challenging. We aimed to identify predictive biomarkers of sorafenib benefit in patients with HCC.
Methods: Gene expression data from 33 HCC tumors treated with sorafenib were analyzed to construct a prediction model aimed at identifying patients with greater benefit from sorafenib treatment. The robustness of the predictor was validated using gene expression data from two phase III clinical trials, IMbrave150 and STORM.

BACKGROUND: In patients with hormone receptor-positive (HR
PATIENTS AND METHODS: We performed a retrospective review of patients with clinical stage I-III, HR
RESULTS: A total of 230 patients were included; 120 (52.2%) were clinically node-negative (cN0), while 110 (47.8%) were clinically node-positive (cN+). In the cN+ group, 7.3% (8/110) had a nodal pathologic complete response (pCR). In total, 76.4% (84/110) in the cN+ group underwent axillary lymph node dissection (ALND) as the initial axillary procedure, and 90.9% (100/110) underwent ALND overall. In total, 98.3% (118/120) with cN0 disease underwent sentinel lymph node dissection (SLND), and 28 (23.7%) had positive nodes. …

Background: PD-L1 expression and tumor mutational burden (TMB) are biomarkers for immune checkpoint inhibitor (ICI) therapy in non-small cell lung cancer (NSCLC); however, patients harboring oncogenic alterations have limited benefit from ICIs. The impact of oncogenic alterations on TMB and PD-L1 tumor proportion score in lung cytology specimens is poorly understood. Herein, the association between oncogenic alterations, TMB, and PD-L1 in NSCLC cytology specimens is explored.
Methods: Next-generation sequencing results from 312 NSCLC cytology specimens were retrospectively reviewed that interrogate 610 genes and select immuno-oncology signatures. TMB and PD-L1 immunohistochemical expression across oncogenic alterations were analyzed to explore associations.

Background: Cancer is a systemic disease with most deaths attributed to metastatic burden. Primary and metastatic tumors, albeit at different anatomic locations, are interconnected through multiple biological processes. Pre-clinical and clinical observations of growth acceleration of metastases after surgery, or abscopal effects outside the radiation field are widely reported, yet reliably triggering favorable and avoiding unfavorable systemic responses remains an unmet clinical need. Understanding local and systemic tumor interaction dynamics will help guide future treatments.

Quantifying Nutrient Fluxes Across The Sediment-Water Interface In The Lafayette River Estuary, Adriana E. Amrhein

The Lafayette River, a shallow, eutrophic tributary of the Chesapeake Bay, experiences seasonal harmful algal blooms (HABs). While management efforts focus on reducing external nutrient inputs, internal loading from benthic sediments remains poorly constrained. This study quantifies spatial and seasonal variability in nutrient and radium isotope fluxes across the sediment–water interface at three sites where HABs are known to initiate and represent contrasting sediment textures and hydrodynamic conditions. Sediment cores and porewater samples were collected in summer 2023 and winter 2024 to measure grain size, porosity, dissolved nutrients (NH₄⁺, NOₓ⁻, PO₄³⁻), and radionuclide activities (²²⁴Ra and ²²⁸Th). Listeria monocytogenes is a facultative intracellular foodborne pathogen capable of crossing the placental barrier, leading to miscarriage, stillbirth, and neonatal sepsis, and has a fetal/neonatal fatality rate of ~20–30%. Although the importance of canonical virulence factors such as Internalin A (InlA) and Internalin B (InlB) in L. monocytogenes crossing of the placental barrier has been demonstrated, clinical strains lacking functional canonical internalins have been reported to cause severe neonatal disease, suggesting additional mechanisms of placental translocation.

While several mentorship interventions for STEM students already exist, persistence and retention in STEM fields remain relatively low. Additionally, there is limited research examining the intersections between aspiring (upward) and inspiring (downward) mentorship roles.
This article aims to address these gaps by examining existing literature and mentorship frameworks, with a focus on how mentorship dynamics influence STEM persistence. Two principal findings emerge from the literature: (1) effective mentorship should be conceptualized as an interconnected ecosystem rather than a hierarchical structure, and (2) the intersection of aspiring and inspiring roles contributes to the development of identity, self-efficacy, and a sense of …

Typical antibiotic susceptibility testing (AST) of microbial samples is performed in homogeneous cultures in batch environments, which does not account for the highly heterogeneous and dynamic nature of antibiotic responses. The most common mutation found in P. aeruginosa lineages evolved during chronic cystic fibrosis infections in the human lung, a loss of function of repressor MexZ, increases basal levels of multidrug efflux MexXY, but does not increase resistance by traditional minimal inhibitory concentration (MIC) assays. Here, we use single cell microfluidics to show that P. aeruginosa’s response to aminoglycosides is highly heterogeneous, with only a subpopulation of cells surviving exposure. …

The anadromous Gulf Sturgeon (Acipenser desotoi) is federally listed as “threatened” and is natal to seven rivers from Louisiana to Florida. Only one spawning reach for Gulf Sturgeon natal to the Pascagoula River has been verified via the collection of eggs and larvae; a reach of the lower Bouie River. This reach is anthropogenically modified by four gravel mining formations, hereafter referred to as pits. The greater depths associated with this area have prompted previous researchers to suggest that Gulf Sturgeon may over-summer in pit formations instead of migrating to downstream holding areas in the Pascagoula River after spawning. Stress, especially social stress, has been a leading cause of disruptive emotional behavior in humans, with pregnant and postpartum women being the most vulnerable to these effects. Our study aimed to investigate the effects of gestational social stress on postpartum behavioral outcomes. To accomplish this, we examined the following four key behaviors in female rats after being exposed to chronic witness stress (WS) during pregnancy: burying, freezing, rearing, and grooming. Burying behavior is used to assess a defensive response to threatening situations. Freezing responses are evaluated to show heightened fear or hypervigilance that follows stress. Programming DNA-binding proteins is an effective strategy for developing therapeutics that regulate gene expression. These proteins can directly bind to DNA to activate or repress transcription of specific genes. Zinc finger proteins are a class of DNA-binding proteins commonly found in eukaryotic organisms. Their DNA-binding function is derived from unique amino acid sequences within each domain that recognizes specific nucleotides, allowing each finger to selectively recognize three DNA base pairs. Adjoining of multiple domains in tandem allows recognition of a specific DNA sequence. PURPOSE: The MTXPK.org webtool facilitates model-informed supportive care and glucarpidase use in patients receiving high-dose methotrexate, but its reliance on manual data entry limits workflow integration. We aimed to highlight how translational informatics can advance, automate, and enhance the usability of clinical decision support tools by embedding MTXPK.org within the electronic health record (EHR).
METHODS: A human factors study with 6 clinical providers guided iterative prototype development. MTXPK.org was rebuilt within the EHR using a 3-tier architecture with Fast Healthcare Interoperability Resources-based data retrieval, automated pharmacokinetic modeling, and interactive visualization.

The lack of effective preventive measures for tick-borne diseases represents a major public health challenge. Transmission-blocking vaccines targeting tick molecules essential for feeding or reproduction could provide a sustainable and broad-spectrum solution. In this study, we evaluated two tick-derived proteins as potential vaccine antigens, subolesin, a key regulator of blood-feeding and cellular processes, and vitellogenin, a protein involved in egg development. Using bioinformatic tools, we identified immunogenic peptide sequences from each protein and chemically conjugated them to virus-like particles (VLPs) to enhance antigen presentation. Upon hatching, squid paralarvae must become proficient at locomotion and prey capture before their yolk reserves are depleted, while also contending with the physical constraints associated with low/intermediate Reynolds number (Re) environments. Although some aspects of behavior and jet dynamics have been examined in squids during early ontogeny, little is known about routine behaviors in paralarvae, whether repeatable patterns are employed, or how these behaviors are achieved dynamically.
