Miners Solving for Tomorrow Research Conference

This project investigated the use of engineered bacterial reporter systems to detect cellular stress responses associated with antibiotic activity. While constructs were successfully introduced and tested against known antibiotics, the reporter consistently produced a blue signal across conditions, limiting the ability to distinguish specific stress responses. This suggests issues such as leaky expression, insufficient regulatory control, or lack of specificity in the reporter design. Although results were inconclusive, this work highlights key challenges in developing reliable biosensors and provides a foundation for future optimization. Improving signal specificity and reducing background expression will be critical for enabling accurate characterization of antibiotic …

This thesis is concerned with determining the optimal water treatment and harvesting strategy for Lemna minor duckweed plants. The main objective of this thesis is to examine how duckweed plants grow in terms of biomass, photosynthetic activity, leaf morphology, chlorophyll and carotenoid content, and their efficiency in nutrient removal from different water sources. The methodology of the study included assessing duckweed in three water samples including tap water, saline water, and greywater and three harvesting strategies, including low, medium, and high harvesting intensities to determine their growth and nutrient removal efficiency. Asphalt is a relatively inexpensive material commonly used for road pavements; however, asphalt ages from exposure to oxygen and UV radiation, leading to loss of elasticity, which leads to cracks and potholes. To slow or reverse asphalt aging, pavement preservation treatments are in use. It’s unclear whether these treatments truly rejuvenate the pavement structure or merely offer a short-term benefit. Nuclear Magnetic Resonance (NMR) relaxometry is an analytical technique that uses the recovery of excited nuclear magnetization as a parameter to gain insights into molecular environments. Prader-Willi syndrome (PWS) is a rare progressive metabolic disease characterized by a wide range of behavioral and cognitive defects, most notably severe hyperphagia. The disorder is caused by the complete loss of some paternally inherited genes on chromosome 15q11-q13, resulting in endocrine dysfunction and a loss of control over feeding behaviors. Among the deleted genes within this region is MAGEL2. Our lab recently characterized a paternally deficient MAGEL2 rat model of PWS, discovering a defect in pituitary secretory capacity that is consistent with the impaired vesicular trafficking and secretion observed in PWS patients. Urban ponds are subject to nutrient influxes from stormwater runoff, and blooms of harmful algae can develop in response. One management strategy includes deployment of Floating Treatment Wetlands (FTWs) or floating mats of aquatic plants designed to maximize uptake of nutrients to plant biomass. An understudied component of these artificial systems is their potential to provide microhabitat for aquatic biota. This study focused on the intersection of aquatic insect communities and fish presence between natural and artificial habitat types. Insect communities were found to be weakly dissimilar between habitat types (ANOSIM: R=0.254, p=0.048). Specifically, Anisoptera and Zygoptera were absent or …

Herpetofauna surveys focused on Crayfish Frog, Spring Peeper, Eastern Tiger Salamander, Red-Bellied Snake, and Smooth Earth Snake were conducted concurrently with Bush Honeysuckle removal projects on the Fort Leavenworth Military Reservation.  Survey techniques included coverboard arrays, adaptive hunt drift fences, traditional pit fall traps and visual encounter surveys and were conducted between May and August in 2024 and 2025. The survey effort resulted in the detection of one of the target species, the Smooth Earth Snake. Based on our survey results we are confident that the Crayfish frog and Spring Peeper do not occur on Fort Leavenworth Military Reserve, but …

Fish have a system of sensory organs known as the lateral line system. This system contains numerous mechanoreceptive neuromasts and electroreceptive ampullary organs (Northcutt et al, 2000). Neuromasts are small, hair-like structures located within canals in the lateral line system just under the fish’s skin and are open to the environment through small pores visible on its skin (Parker, 2025). Water flows into these canals through the pores which stimulates the sensitive hair cells, resulting in electrical signals sent to the fish’s brain. Neurospora crassa is a fungus that has been used to study genetics, more specifically, meiotic gene drive through its spore killing abilities. Meiotic drive describes a process in which selfish genetic elements bias Mendelian genetics to increase transmission to their offspring, eliminating any alleles that are sensitive. The spore killing mechanism of Neurospora’s Spore killer-3 (Sk-3) is an example of how meiotic drive works through a poison-antidote model. Neurospora crassa is a useful model organism for genetic research due to its haploid life cycle and its ability to produce large numbers of sexual spores called ascospores. The haploid ascospores form within asci, which typically contain eight ascospores at maturity. However, selfish genetic elements known as meiotic gene drivers have been discovered in Neurospora fungi, and some of these alter the number of viable ascospores that form in each ascus. Western Australia's (WA) sheep industry has undergone substantial structural transformation since the 1990s, with a 67% reduction in the flock, from 38.4 million in 1989-90 to 12.7 million in 2020-21. This transformation process followed the collapse of the Reserve Price Scheme for wool in 1991. This report examines the spatial and temporal changes in flock size, composition, and producer adaptation strategies from 2010-11 to 2020-21, drawing on Australian Bureau of Statistics (ABS) census data and Meat and Livestock Australia (MLA) and Australian Wool Innovation (AWI) producer surveys data.

Introduction: Although heterogeneity of treatment effects (HTEs) is commonly observed in clinical trials, it has received little attention in studies on transcranial direct current stimulation (tDCS).

Objectives: This study identified the presence of HTE in tDCS treatment among participants with symptomatic knee osteoarthritis (KOA) and explored participant characteristics associated with this heterogeneity.

Methods: This secondary analysis of a randomized clinical trial included 120 participants with symptomatic KOA who received 15 daily sessions of home-based 2-mA active or sham tDCS over 3 weeks. Ewing sarcoma is a rare and aggressive pediatric bone cancer with poor survival rates in patients, as low as 30% in advanced or recurrent cases. There are limited treatment options, making molecularly targeted therapy vital to explore. Through CRISPR scans of Ewing sarcoma cell lines, ribosome biogenesis was highlighted as a potential target. As RNA polymerase I is a key component of ribosome biogenesis, the cytotoxic ability of two RNA Polymerase I inhibitors, CX-5461 and BMH-21, were used on patient-derived Ewing sarcoma cells. Clematis socialis (Kral) is a rare, federally endangered perennial species restricted to a small number of fragmented populations in Alabama and Georgia. The species exhibits extensive clonality and limited sexual recruitment, characteristics that complicate expectations regarding genetic diversity and population connectivity. Despite its conservation significance, genome-wide assessments of population structure have been lacking. We used genotyping-by-sequencing (GBS) to generate a genome-wide SNP dataset for 170 individuals sampled from six natural populations. Population structure and genetic differentiation were evaluated using Bayesian clustering, analysis of molecular variance (AMOVA), pairwise FST estimates, and network-based visualization. Analyses revealed strong genetic structuring among populations and …

Stormwater drains and culverts are installed across urban spaces to direct surface water and allow for proper drainage and prevent flooding and erosion. These structures and the water that they carry can provide unintended opportunities to wildlife. Previous research has shown that local ecosystems may utilize and adapt around these culverts in many ways. They can serve as habitats, connect fragmented habitats separated by roads or human traffic, serve as drinking spots, and promote biodiverse plant growth. CIC-DUX4 sarcoma (CDS) is a rare and extremely aggressive soft tissue sarcoma that primarily affects children and young adults. CDS is characterized by an oncogenic chromosomal translocation. Current treatments for CDS are similar to those in other sarcomas like surgery, chemotherapy, and radiation. Treatments, however show that CDS responds poorly compared to other sarcomas, due to relapses of localized disease at high rates and short duration response of treatment, proving the importance of understanding CIC-DUX4’s complex role on how it regulates overall gene expression in CDS cells from a molecular approach.

Within Midwest floodplain forests, there is a lack of long-term studies focusing on native tree recruitment post invasive shrub removal. With interacting threats such as emerald ash borer (Agrilus planipennis), invasive earthworms, and white-tailed deer (Odocoileus virginianus), land managers need particular information about these ecosystems in order to better understand how to protect them. This study aims to explore the efficacy of the removal of Rhamnus cathartica and Lonicera spp. in promoting native tree recruitment within a Minnesota floodplain forest along the Mississippi River. Topographic heterogeneity structures Arctic plant communities, impacting broader ecosystem responses to climate change. The role of topography in shaping leaf physiological, nutritional, and structural traits in the Arctic is important, yet not well characterized across diverse tundra landscapes. To address these controls, we examined plant community composition and measured leaf traits in a few representative species along a heath-slope-valley gradient in Varanger, Norway. Our results indicated that valleys are more biodiverse, driven by forb and graminoid dominance, whereas slopes and heaths had less diversity with high shrub abundance. Leaf physiology and structural traits did not vary across topographic positions, though …

The Developmental Origins of Health and Disease (DOHaD) paradigm posits that early environmental factors may influence a child’s development and long-term health outcomes. Developmental programming (DP) is central to this paradigm, whereby specific early life exposures during critical periods of development are associated with changes to physiological and metabolic pathways, potentially predisposing individuals to disease. However, no standard definition of DP exists, and various terms have been used to describe similar processes. Background

Wrestling, characterized by high-intensity intermittent efforts, demands exceptional anaerobic power and recovery capacity. Nitrate-rich beetroot juice (BRJ), supplemented with vitamin C, has emerged as a potential ergogenic aid through increased nitric oxide bioavailability. However, limited data exists regarding its acute effects on anaerobic performance in combat sport athletes. This study investigated the acute effects of BRJ supplemented with vitamin C on upper- and lower-body anaerobic test performance and selected biochemical markers in collegiate wrestlers.

Background: The purpose of this study was to examine the effects of acute dietary nitrate (NO3−) and caffeine (CAF) supplementation on end power (EP) and work performed above EP (WEP) in trained male cyclists during a 3 min all-out test (3MT) on a cycle ergometer.

Methods: Fifteen healthy, trained male cyclists (28.5 ± 5.3 years, 79.2 ± 9.1 kg, VO2peak 55.2 ± 5.6 mL·kg−1·min−1) completed four exercise trials in a randomized, double-blind, placebo-controlled, crossover study design separated by 3–7 days. Cataract is a leading cause of vision impairment worldwide and are primarily caused by oxidative stress that damages and aggregates lens proteins, leading to lens opacification. However, the eye's anatomical barriers limit the penetration and bioavailability of antioxidant therapies. To address this challenge, a dendrimer-based nanogel with a built-in reactive oxygen species (ROS)-scavenging capability developed by us was employed to deliver the antioxidant N-acetylcysteine (NAC) to the lens. • Microtubules are composed of of a- and b- tubulin heterodimers, vital for cell structure, shape, and intracellular transport • Microtubules are also useful as they separate chromosomes during cell division • Microplastics (< 5 mm) are considered a health threat and have been found throughout the whole body (1) o In several studies, microplastics have been correlated to higher rates cardiorespiratory issues, endocrine disruption, and cancer (1) • Tubulin-binding medications are widely used as chemotherapeutic agents as they are known for disrupting cell division and targeting tubulin (2,3) o In this study, these medications were used to determine whether microtubules play a role in microplastic uptake § Colchicine typically binds to tubulins and arrests polymerization and growth § Nocodazole disrupts polymerization and prevents spindle formation § Vinblastine inhibits microtubule assembly and disruptions miotic spindle formation.

• The purpose of this study was to determine if different concentrations of tubulin-binding medications affect microplastic uptake o C2C12, RAW 264.7, and MOVAS cell lines were used to evaluate whether these changes are cell type specific 

If microtubule dynamics are disrupted, microplastic uptake can be affect in a concentration-dependent and cell type- specific manner.

Aspiration pneumonia is a common mild case of pneumonia, a disease where the lungs are inflamed and filled with fluid, compromising respiration and gas exchange. One common cause of mild pneumonia is bacterial infection via a pathogen known as Mycoplasma pneumoniae. M. pneumoniae causes a variety of respiratory illness- ranging from pharyngitis, tracheobronchitis, mild pneumonia- and in rare cases encephalitis. M. pneumoniae produces the CARDS toxin, which participates in ADP-ribosylation and vacuolization (Kannan et al., 2006)1.
