The Effect Of Fat Content On The Inactivation And Recovery Of Listeria Spp. In Ready-To-Eat Foods After High Pressure Processing, Yhuliana Kattalina Niño Fuerte
Department of Food Science and Technology: Dissertations, Theses, and Student Research
High-pressure processing (HPP) is recognized by the United States Department of Agriculture Food Safety and Inspection Service (USDA FSIS) as a post-lethality treatment to control Listeria monocytogenes in ready-to-eat (RTE) meat and poultry products. A scoping review of 603 journal articles, which ultimately was narrowed down to 16 articles, revealed that the efficacy of HPP varies with fat content. The studies demonstrated that L. monocytogenes inactivation by HPP tends to be lower in RTE meats with greater than 5% fat content, compared to products with lower than 5% fat. The wide range of HPP parameters used in these studies …

Pfos Alters Metabolism In The Liver And Skeletal Muscle Of Cyp2b-Null And Humanized Cyp2b6-Transgenic Mice, Lanie Williams
All Theses
Perfluorooctanesulfonate (PFOS) is a persistent environmental pollutant previously used in industrial products such as fire-fighting foams, furniture, and stain-resistant coatings. PFOS is associated with adverse health outcomes, including hepatosteatosis and diabetes. Previously, humanized CYP2B6-transgenic (hCYP2B6-Tg) mice accumulated more PFOS than Cyp2b-null mice and experienced greater mortality; however, the Cyp2b-null mice experienced greater steatosis. Cyp2b-null and hCYP2B6-Tg mice were treated with 0, 1, or 10 mg/kg/day PFOS for 21 days. To determine how PFOS affected the metabolism of polyunsaturated fatty acids (PUFAs) oxylipin concentrations were measured via LC-MS/MS. Additionally, gene expression changes were measured using RNA sequencing. PFOS treatment increased the …
Venom Complexity Within Garter Snakes And The Role Of Diet In Toxin Diversification, Tucker Heptinstall
Venom Complexity Within Garter Snakes And The Role Of Diet In Toxin Diversification, Tucker Heptinstall
All Theses
Complex traits, such as body size and venom, are often under strong selection by
dietary ecology and can increase efficiency of prey capture and processing. To investigate relationships between ecology and trait evolution in Thamnophis, we amassed nearly 10,000 diet records for 32/35 Thamnophis species and classified each species’ dietary repertoire. We categorized 15/32 species as generalist predators and 17/32 species as specialist predators of varying prey groups and investigated the hypothesis that generalist predators have larger body size. However, we found no significant relationship between body size and dietary specialization. Venom has evolved over 100 times across the Tree …

Survey Of Nematodes Associated With Strawberry In South Carolina And Evaluation Of Short-Term Heat Exposure On Nematode Reproduction Biology, Ivan A. Alarcon Mendoza
All Theses
Nematodes are a growing threat to the multi-billion-dollar strawberry industry in the US due to the lack of safe and effective management methods. Limited information on the distribution of nematodes in fumigated plasticulture strawberry fields further complicates their management. Additionally, climatic anomalies like rising soil temperatures and heat waves are occurring more often, and their impact on crop growth and nematode reproduction is not well understood. The current study was conducted with two main objectives: i) understand distribution and population densities of nematodes associated with plasticulture strawberry production in South Carolina, and ii) understand the impact of short-term high heat …
Effects Of Interrupting The Scotophase Period On Bone Health And Performance Of Laying Hens, Alexis J. Clark
Effects Of Interrupting The Scotophase Period On Bone Health And Performance Of Laying Hens, Alexis J. Clark
All Theses
This thesis aimed to evaluate the effects of interrupting the daily scotophase period on bone health and production in laying hens. Modern commercial laying hens are prone to high rates of bone demineralization due to the constant mobilization of calcium to withstand daily eggshell calcification mechanisms. The industry light standard for laying hens is 16 hours of continuous light (photophase) and 8 hours of continuous darkness (scotophase) daily. During scotophase, physical activity is reduced due to instinctive perching behaviors, and thus, feeding behaviors are halted. However, physiological processes, including digestion, excretion, and reproduction, remain active during scotophase.
Most eggshell formation …

The exponential increase in global population continues to present an ongoing challenge for livestock producers worldwide to consistently provide a safe, high-quality, and affordable source of protein for consumers. In the last 50 years, the poultry industry has spearheaded this effort thanks to focused genetic and genomic selection for feed-efficient, high-yielding broilers. However, such intense selection for productive traits, along with conventional industry farming practices, has also presented the industry with a myriad of serious issues that negatively impacted animal health, welfare, and productivity–such as woody breast and virulent diseases commonly associated with poultry farming. Bacterial chondronecrosis with osteomyelitis (BCO) …
Soil Inversion To Ameliorate Soil Water Repellence, Department Of Primary Industries And Regional Development, Western Australia
Soil Inversion To Ameliorate Soil Water Repellence, Department Of Primary Industries And Regional Development, Western Australia
Natural resources factsheets
One-off soil inversion results in the burial of the water repellent topsoil in a layer that is typically a depth of 15 to 35 cm and brings to the surface a layer of wettable subsoil.
Other benefits include burial of herbicide resistant weed seeds, removal of compaction, improved access to nutrients, reduced disease risk, and incorporation of soil amendments such as lime.
Soil inversion should always be done when the soil is wet to more than the depth of inversion, and a crop or cover can be sown to reduce the risk of wind erosion.
Caution - Deep cultivation can …

Signatures Of Selection, Gene Flow, And Genetic Drift On The Genomes Of A Recent Avian Radiation, Flor Brigitte Hernandez Camacho
Open Access Theses & Dissertations
Speciation is not a discrete event but rather a gradual process that occurs along a continuum. Initially, populations remain genetically and reproductively connected, but they become reproductively isolated as the process of speciation progresses. Advances in DNA sequencing technology has helped our understanding of the processes that contribute to changes in the genetic composition of populations over the progression of speciation, also known as drivers of evolution (i.e. genetic drift, mutation, selection, and gene flow). Among them, hybridization, or the interbreeding between distinct lineages, is increasingly recognized as a natural process that can have significant consequences to the speciation process. …

Establishment Of A Novel Prairie Vole (Microtus Ochrogaster) Sars-Cov-2 Infection And Transmission Model, Stephanie Rebecca Kehl
Open Access Theses & Dissertations
Considering that the number of SARS-CoV-2 cases and deaths continues to increase three years after the onset of the pandemic, the need for animal models to study disease pathology and test antiviral effectiveness remains paramount. Prairie voles offer unique advantages as an infection model, such as mirroring human social behavior, including pair bonding and biparental offspring rearing, having inducible estrus and ovulation cycles, and not being overly domesticated. Based on the sequence homology of the vole ACE2 receptor to that of other species that are susceptible to SARS-CoV-2, such as the ferret and hamster, we hypothesized that the vole ACE2 …

This study addresses the prevalent issue of missing data in patient-reported outcome datasets, particularly focusing on head and neck cancer patient symptom ratings sourced from the MD Anderson Symptom Inventory. Given that many data mining and machine learning algorithms necessitate complete datasets, the accurate imputation of missing data as an initial step becomes crucial. In this study we propose, for the first time, the use of collaborative filtering for imputing missing head and neck cancer patient symptom ratings. Two configurations of collaborative filtering, namely patient-based and symptom-based, leverage known ratings to infer the missing ones. Additionally, this study compares the …

Extending and safeguarding tropical forest ecosystems is critical for combating climate change and biodiversity loss. One of its constituents, lianas, is spreading and increasing in abundance on a global scale. This is particularly concerning as lianas negatively impact forests’ carbon fluxes, dynamics, and overall resilience, potentially exacerbating both crises. While possibly linked to climate-change-induced atmospheric CO2 elevation and drought intensification, the reasons behind their increasing abundance remain elusive. Prior research shows distinct physiological differences between lianas and trees, but it is unclear whether these differences confer a demographic advantage to lianas with climate change. Guided by extensive datasets collected …

Elexacaftor/tezacaftor/ivacaftor (ETI) therapy has revolutionized the treatment of cystic fibrosis (CF) for most affected individuals but the effects of treatment on sinus microbiota are still unknown. Changes to the airway microbiota in CF are associated with disease state and alterations to the bacterial community after ETI initiation may require changes to clinical management regimens. We collected sinus swab samples from the middle meatus in an observational study of 38 adults with CF and chronic rhinosinusitis (CRS) from 2017 to 2021 and captured the initiation of ETI therapy. We performed 16S and custom amplicon sequencing to characterize the sinus microbiota pre- …

Identifying New Potential Breeding Pond Sites For The Dusky Gopher Frog In Southern Mississippi, Shelbie Kellum
Master's Theses
The main objective of this thesis was to identify potential new pond sites across southern Mississippi for the critically endangered dusky gopher frog (DGF). Ephemeral ponds are the preferred breeding habitat for the species, but they are quite rare in the region due to the diminishment of the longleaf pine ecosystem. This research addresses gaps in the current literature by providing a broad overview of dusky gopher frog breeding habitat identification. LiDAR derived datasets as well as tools within ArcGIS Pro were utilized in order to perform spatial analysis on sites of interest in south Mississippi. By using this methodology …

Effects Of Increasing Cycling Cadence On Post-Cycling Gait Velocity: An Experimental Study, Nitu Lama
Master's Theses
Previous research has shown that increasing cycling cadence, rather than the workrate, can result in improved post-cycling gait velocity. However, the specific threshold of cycling cadence required to bring about clinically meaningful changes in gait velocity remains unknown. To address this knowledge gap, our study aimed to determine the minimum incremental increase in cycling cadence that would lead to a significant improvement in post-cycling gait velocity. A total of 42 young adults participated in our study and were randomly assigned to one of three groups: TEN, TWENTY, and THIRTY. Each group was assigned to cycle at a cadence at the …

Investigating The Response Of The Eastern Oyster (Crassostrea Virginica) To Low Dissolved Oxygen Conditions, Abiola Obafemi
Master's Theses
The Eastern oyster (Crassostrea virginica) requires dissolved oxygen (DO) of about 4 mg/L to support proper physiological function. Hypoxia and/or microxia occur when DO decreases to ≤ 2 mg/L. Salinity and temperature changes resulting from large-scale freshwater influx have been identified as key factors influencing the oyster populations in the Mississippi Sound. It is important to investigate the impacts of DO conditions on oyster mortality, biomass, size, and condition during these freshening events. Under controlled laboratory experiments, this thesis investigated 1) the response of oysters to microxic, hypoxic, and normoxic conditions 2) the response of oysters to combined hypoxia and …
Projected Range Shift Of The Atlantic Surfclam, Spisula Solidissima, And The Ocean Quahog, Arctica Islandica, Under Climate-Induced Benthic Warming: Forecasting Fishery Influence In The Mid-Atlantic Bight Throughout The 21st Century, Molly Mae Spencer
Master's Theses
The Atlantic surfclam, Spisula solidissima, and ocean quahog, Arctica islandica, are biomass dominant bivalve species on the eastern North American continental shelf, both supporting lucrative commercial fisheries in the Mid-Atlantic with a combined value of $53.6 million in ex-vessel revenue per year. Rapid climate change is expected to modify the geographic range of these bivalve populations across the Mid-Atlantic Bight (MAB), suggesting transformative consequences for the surfclam fishery. This study forecasted future distributions of the two species from years 2016 – 2095, based on projected bottom water temperatures and a temperature-dependent population dynamics model, to project subsequent fishery-based indicators …

Objective
Livestock disease management is crucial for producers. To control and eradicate disease, the US government has a duty to depopulate infected or potentially infected animals, and current indemnity policy dictates that producers must receive fair market compensation for depopulated animals. This study surveys producers’ preferences regarding livestock indemnity policy to better understand ordered preferences for any changes in the future.Materials and Methods
Through a ranked-ordered probit model, we analyzed producers’ rankings of 4 different types of indemnity from an online producer survey. Results and Discussion
Based on the responding producers, the most preferred method of livestock indemnity is …The Relationship Between Phosphorus Retention And Fibroblast Growth Factor 23 In Broiler Breeders, A. D. Magnuson, N. Boonsinchai, J. Caldas, J. England, C. Coon

Previous studies with broiler breeders indicate a P retention threshold when fed daily dietary levels of non-phytate P (NPP) exceeding 320 mg. Fibroblast growth factor 23 (FGF23) is a hormone secreted by osteocytes which modulates P retention and could be the biological agent which controls the P threshold in breeders. To evaluate the relationship between FGF23 and the P retention threshold, a 4 -wk study with 32 -wk -old breeders was conducted with 6 dietary treatments with daily NPP intake of 216 to 576 mg/d/h with increments of 80 mg/kg diet. The goals were 1) to …

Bacterial chondronecrosis with osteomyelitis (BCO) lameness is a major welfare issue for broiler production worldwide affecting approximately 1.5% of broilers over 42 days old. Excessive body weight gain causes mechanical stress on long bones, leading to micro-fractures. This condition induces a bacterial infection of fractures, resulting in bone necrosis and eventual BCO lameness. Increasing gut integrity and supporting Calcium metabolism contribute to the optimal bone structure and subsequently reduce BCO lameness. Probiotics thus provide an excellent strategy for alleviating BCO due to the improvement of intestinal integrity and barrier function. Accordingly, the present study investigated the lameness reduction through the …

Sustainability in agriculture starts with the health of soil-- from its productivity and fertility, to its ability to retain water during droughts and allow for drainage during rain. One approach of improving soil health is the use of cover crops, a practice associated with improved nutrient availability in soils, the promotion of soil biodiversity and organic matter to improve density and structure, effective weed management, and ultimately improved yields of subsequent cash crops. Assessing the cumulative impacts of cover crops is difficult, with various metrics and indicators. Crop dry weight may be a way to predict the effects cover crops …

Researchers are often interested in estimating the effect of sustained use of a treatment on a health outcome. However, adherence to strict treatment protocols can be challenging for individuals in practice and, when non-adherence is expected, estimates of the effect of sustained use may not be useful for decision making. As an alternative, more relaxed treatment protocols which allow for periods of time off treatment (i.e. grace periods) have been considered in pragmatic randomized trials and observational studies. In this article, we consider the interpretation, identification, and estimation of treatment strategies which include grace periods. We contrast natural grace period …

Renal cell carcinoma (RCC) bone metastatis progression is driven by crosstalk between tumor cells and the bone microenvironment, which includes osteoblasts, osteoclasts, and osteocytes. RCC bone metastases (RCCBM) are predominantly osteolytic and resistant to antiresorptive therapy. The molecular mechanisms underlying pathologic osteolysis and disruption of bone homeostasis remain incompletely understood. We previously reported that BIGH3/TGFBI (transforming growth factor-beta-induced protein ig-h3, shortened to BIGH3 henceforth) secreted by colonizing RCC cells drives osteolysis by inhibiting osteoblast differentiation, impairing healing of osteolytic lesions, which is reversible with osteoanabolic agents. Here, we report that BIGH3 induces osteocyte apoptosis in both human RCCBM tissue specimens …

The T2-Fluid-Attenuated Inversion Recovery (T2-FLAIR) mismatch sign is a radiogenomic marker that is easily discernible on preoperative conventional MR imaging. Application of strict criteria (adult population, cerebral hemisphere location, and classic imaging morphology) permits the noninvasive preoperative diagnosis of isocitrate dehydrogenase (IDH)-mutant 1p/19q-non-codeleted diffuse astrocytoma with near-perfect specificity, albeit with variably low sensitivity. This leads to improved preoperative planning and patient counseling. More recent research has shown that the application of less strict criteria compromises the near-perfect specificity of the sign but remains adequate for ruling out IDH-wildtype (glioblastoma) phenotype, which bears a far grimmer prognosis compared to IDH-mutant diffuse …

Objective: We provide evidence-based recommendations regarding the treatment of interstitial lung disease (ILD) in adults with systemic autoimmune rheumatic diseases (SARDs).
Methods: We developed clinically relevant population, intervention, comparator, and outcomes questions. A systematic literature review was then performed, and the available evidence was rated using the Grading of Recommendations, Assessment, Development, and Evaluation methodology. A panel of clinicians and patients reached consensus on the direction and strength of the recommendations.
Results: Thirty-five recommendations were generated (including two strong recommendations) for first-line SARD-ILD treatment, treatment of SARD-ILD progression despite first-line ILD therapy, and treatment of rapidly progressive ILD. The strong …

Neutrophil extracellular traps (NETs) function to control infectious agents as well as to propagate inflammatory response in a variety of disease conditions. DNA damage associated with chromatin decondensation and NACHT domain-leucine-rich repeat-and pyrin domain-containing protein 3 (NLRP3) inflammasome activation have emerged as crucial events in NET formation, but the link between the two processes is unknown. In this study, we demonstrate that poly(ADP-ribose) polymerase-1 (PARP-1), a key DNA repair enzyme, regulates NET formation triggered by NLRP3 inflammasome activation in neutrophils. Activation of mouse neutrophils with canonical NLRP3 stimulants LPS and nigericin induced NET formation, which was significantly abrogated by pharmacological …

Objectives: The type I interferon pathway is a promising target for treatment of patients with systemic sclerosis (SSc). Here, we describe the design of a multinational, randomised phase 3 study to Determine the effectiveness of the type I interferon receptor antibody, Anifrolumab, In SYstemic sclerosis (DAISY).
Methods: DAISY includes a 52-week double-blind, placebo-controlled treatment period, a 52-week open-label active treatment period, and a 12-week safety follow-up period. The patient population includes a planned 306 adults with limited or diffuse cutaneous active SSc who satisfied American College of Rheumatology/European Alliance of Associations for Rheumatology 2013 SSc criteria. Use of standard immunosuppressants, …

Study aims: To evaluate the outcomes of patients with 3q26.2/MECOM-rearranged chronic myeloid leukemia (CML).
Methods: We reviewed consecutive adult patients with 3q26.2/MECOM-rearranged CML between January 1, 1998 and February 16, 2023. Rearrangements of 3q26.2/MECOM were confirmed by conventional cytogenetics, and fluorescence in situ hybridization starting in 2015.
Results: We identified 55 patients with MECOM-rearranged CML, including 23 in chronic phase (CP) or accelerated phase (AP) and 32 in blast phase (BP). Nine patients (16%) achieved a major cytogenetic response (MCyR) or deeper. At a median follow-up of 89 months, median survival was 14 months. The 5-year survival rate was 19% …
