Discovery Of Small Molecules To Alleviate Heart Disease-Induced Proteinopathy And Proteotoxicity Characteristic Of Alzheimer’S Disease, Nirjal Mainali
Theses and Dissertations
Cardiovascular disease (CVD) is the leading cause of death in the world and in the United States. Reduced cardiac output due to CVD has been previously associated with decreases in systemic and cerebral perfusion. Cerebral hypoperfusion increases the risk of Alzheimer’s disease (AD), accompanied by accumulation of aggregates that feature Aβ amyloid and hyperphosphorylated tau. Protein aggregation is a shared hallmark of aging and neurodegenerative diseases. We previously reported elevated aggregation in hearts of hypertensive and aged mice and now find that aggregation is induced in hearts and brains of male C57BL/6 mice after transient ligation of the left coronary …

The spatial extent of semi-arid hot regions is forecasted to grow through the twenty-first century, complicating restoration and reforestation plans. In arid and semi-arid climates, seedlings are more susceptible to transplant shock due to lower soil moisture throughout the year. Determining strategies to reduce seedling stress and improve survival post-planting will be paramount to continued reforestation efforts in a changing climate. We quantified seedling physiological, morphological, and field performance (mortality and growth) response for five species native to the semi-arid region of South Texas (Erythrina herbacea L., Celtis pallida Torr., Fraxinus berlandieriana DC, Malpighia glabra L., and Citharexylum berlandieri B.L …

While poly (ADP-ribose) polymerase (PARP) inhibitors (PARPi) have improved the prognosis of ovarian high-grade serous carcinoma (HGSC) tumors that are homologous recombination (HR) deficient (HRD), new therapeutic strategies are needed for tumors that are HR proficient (HRP) because they demonstrate greater resistance to current treatments and thus have poorer clinical outcomes. Additionally, clinical precautionary statements regarding potential risks associated with PARPi, such as myelodysplastic syndrome, highlight the need for combinatorial approaches that can lessen the dose and duration of PARPi treatment to reduce toxicities. Here, we evaluated DNA double-strand damage repair pathways in HRD and HRP ovarian cancer cell lines …

Pancreatic ductal adenocarcinoma (PDAC) presents a fatal clinical challenge characterized by a dismal 5-year overall survival rate, primarily due to the lack of early diagnosis and limited therapeutic efficacy. Immunotherapy, a proven success in multiple cancers, has yet to demonstrate significant benefits in PDAC. Recent studies have revealed the immunosuppressive characteristics of the PDAC tumor microenvironment (TME), including immune cells with suppressive properties, desmoplastic stroma, microbiome influences, and PDAC-specific signaling pathways. In this article, we review recent advances in understanding the immunosuppressive TME of PDAC, TME differences among various mouse models of pancreatic cancer, and the mechanisms underlying resistance to …

CRISPR base editors can introduce point mutations into DNA precisely, and cytosine base editors (CBEs) catalyze C to T transitions. While CBEs have been thoroughly explored in cell culture and organisms such as mice, little is known about DNA base editing in insects. In this study, we evaluated germline editing rates of three different CBEs expressed under actin (ubiquitous) or nanos (germline) promoters utilizing Drosophila melanogaster. The original Rattus norvegicus-derived cytosine deaminase APOBEC1 (rAPO-1) displayed high base editing rates (~99%) with undetectable indel formation. Additionally, we show that base editors can be used for generating male sterility and female lethality. …

Mesenchymal stem/stromal cells (MSCs) are an attractive platform for cell therapy due to their safety profile and unique ability to secrete broad arrays of immunomodulatory and regenerative molecules. Yet, MSCs are well known to require preconditioning or priming to boost their therapeutic efficacy. Current priming methods offer limited control over MSC activation, yield transient effects, and often induce the expression of pro-inflammatory effectors that can potentiate immunogenicity. Here, we describe a genetic priming method that can both selectively and sustainably boost MSC potency via the controlled expression of the inflammatory-stimulus-responsive transcription factor interferon response factor 1 (IRF1). MSCs engineered to …

Internal combustion engine fueled by carbon-free hydrogen (H2-ICE) offers a promising alternative for sustainable transportation. Herein, we report a facile and universal strategy through the physical mixing of Pt catalyst with zeolites to significantly improve the catalytic performance in the selective catalytic reduction of nitrogen oxides (NOx) with H2 (H2-SCR), a process aiming at NOx removal from H2-ICE. Via the physical mixing of Pt/TiO2 with Y zeolite (Pt/TiO2 + Y), a remarkable enhancement of NOx reduction activity and N2 selectivity was simultaneously achieved. The incorporation of Y zeolite effectively captured the in-situ generated water, fostering a water-rich environment surrounding the …

Evaluation Of Satellite-Based Remote Sensing Methods For Monitoring Of Winter-Flooding Of Agricultural Fields For Migratory Birds, Thomas Patrick Dill
Evaluation Of Satellite-Based Remote Sensing Methods For Monitoring Of Winter-Flooding Of Agricultural Fields For Migratory Birds, Thomas Patrick Dill
Student Theses and Dissertations
Agricultural land conversion in Arkansas has caused the loss of 72% of wetland habitat, reducing wintering ground for migratory birds. Winter-flooded agriculture fields resemble seasonal wetlands. Federal agencies and private organizations have recognized the importance of this and have invested millions of dollars in subsidies. While remote sensing has been used in wildlife management, there is currently no method to remotely track adoption or monitor fields. To create this ability, we examined four spectral indices: normalized difference vegetation index (NDVI), normalized difference water index (NDWI), and two different versions of the modified normalized difference water index (MNDWI). We also compared …

Spatial processes across both broad and fine scales are important for understanding ecosystem function. Broad-scale spatial processes can be defined as elevational, latitudinal, or longitudinal processes whereas fine-scale spatial processes represent movement and dispersal among habitat patches. Historically, studies on freshwater snail distributions have focused on fine-scale processes with secondary consideration given to spatial sampling. Species inventories for freshwater snails are outdated because historically snail species descriptions were based on shell morphology, which is highly plastic. In Arkansas, it is estimated that there are 36 freshwater snail species, but this estimate does not account for shell plasticity and does not …

An Integral Approach To Philosophy Of Nature, Carol Richardson
An Integral Approach To Philosophy Of Nature, Carol Richardson
Journal of Conscious Evolution
Abstract
Quantum physicists and quantum biologists enjoy a mathematically-inspired romance with the wonders of nature at the quantum level. Recognizing the romance of mathematics and the quantum-to-cosmic level relational energetics of Nature can lead to a more integral, romantic, and even embodied phenomenology of the intelligence inherent within Nature’s relational and embodied energies. Approaching this quantum nature of Nature from the more romantic, wholistic views of the right hemisphere of the brain, rather than from the fragmenting views of the left hemisphere of the brain, one can begin to discover the self-organizing abilities of the energies of Nature at all …

Mexico enacted a decree to ban the sale of genetically modified (GM) maize seed and maize for human consumption. Maize is particularly important to the average diet in Mexico as it is the main feed for the primary source of protein (poultry) and the main ingredient for the primary source of calories (corn tortillas). This study aimed to assess consumer awareness of the decree, support for the decree, and sensitivity of support given possible economic outcomes related to the decree. Additionally, we estimate the premiums consumers were willing to pay (WTP) for non-GM products relative to GM products (i.e., chicken …

Acute lower gastrointestinal GVHD (aLGI-GVHD) is a serious complication of allogeneic hematopoietic stem cell transplantation. Although the intestinal microbiota is associated with the incidence of aLGI-GVHD, how the intestinal microbiota impacts treatment responses in aLGI-GVHD has not been thoroughly studied. In a cohort of patients with aLGI-GVHD (n = 37), we found that non-response to standard therapy with corticosteroids was associated with prior treatment with carbapenem antibiotics and a disrupted fecal microbiome characterized by reduced abundances of Bacteroides ovatus. In a murine GVHD model aggravated by carbapenem antibiotics, introducing B. ovatus reduced GVHD severity and improved survival. These beneficial effects …

Myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) belong to a continuous disease spectrum of myeloid malignancies with poor prognosis in the relapsed/refractory setting necessitating novel therapies. Natural killer (NK) cells from patients with myeloid malignancies display global dysfunction with impaired killing capacity, altered metabolism and exhausted phenotype at the single cell transcriptomic and proteomic levels. In this study we identified that this dysfunction was mediated through a crosstalk between NK cells and myeloid blasts necessitating cell-cell contact. NK cell dysfunction could be prevented by targeting the αvβ integrin/TGF-β/SMAD pathway but, once established, was persistent due to profound epigenetic reprogramming. …

Genomic Analysis Of Enterobacter Species Isolated From Patients In United States Hospitals, Fred C. Tenover, Isabella A. Tickler
Guidelines, Recommendations, and Other Resources on Antimicrobial Resistance
Abstract: We analyzed the whole genome sequences (WGS) and antibiograms of 35 Enterobacter isolates, including E. hormaechei and E. asburiae, and the recently described E. bugandensis, E. kobei, E. ludwigii, and E. roggenkampii species. Isolates were obtained from human blood and urinary tract infections in patients in the United States. Our goal was to understand the genetic diversity of antimicrobial resistance genes and virulence factors among the various species. Thirty-four of 35 isolates contained an AmpC class blaACT allele; however, the E. roggenkampii isolate contained blaMIR-5. Of the six Enterobacter isolates resistant to ertapenem, imipenem, and meropenem, …

Low-Emission Beef Production In The Southern Rangelands Of Western Australia: An Analysis Of Herd Structure And Stocking Rate Experiencing Droughts, Christophe D'Abbadie
Animal production and livestock research articles
Reconciling profitable cattle production with rangeland health and reduced emissions is a key challenge in the southern rangelands of Western Australia (WA). Stocking rate and herd structure selection are crucial decisions to achieve this balance. This study assessed the emission profiles of three contrasting herd structures (weaner production, live export, and slaughter production), and three stocking rates within a herd–carbon accounting modelling framework. The analysis considers the impact of varying drought frequencies on these cattle production systems. Herd models were developed for the semiarid southern WA rangelands. Stocking rates were set at 100%, 80% and 66% of the government recommended …

Dissecting The Interplay Of Protein Synthesis And Degradation Pathways In Cellular Adaptation To Stress, Brittany Friedson
Theses and Dissertations
Adaptation to stress requires cells to reprogram transcription, translation, and proteolytic pathways. Although much is known about the response of each program, it remains unclear how they coordinate following stress. My studies in S. cerevisiae identified the Cdk8 kinase module (CKM) of the Mediator complex as a new player in coordinating these processes. It is well established that the CKM consists of four highly conserved proteins (cyclin C, its cognate kinase Cdk8, and two structural proteins Med12 and Med13) and predominantly represses a subset of stress responsive genes in yeast. We demonstrated for the first time that the CKM also …

Importin β-superfamily nuclear import receptors (NIRs) mitigate mislocalization and aggregation of RNA-binding proteins (RBPs), like FUS and TDP-43, which are implicated in neurodegenerative diseases. NIRs potently disaggregate RBPs by recognizing their nuclear localization signal (NLS). However, disease-causing mutations in NLS compromise NIR binding and activity. Here, we define features that characterize the anti-aggregation activity of NIR and NLS. We find that high binding affinity between NIR and NLS, and optimal NLS location relative to the aggregating domain plays a role in determining NIR disaggregation activity. A designed FUS chimera (FUSIBB), carrying the importin β binding (IBB) domain, is …

MicroRNAs (miRNAs) are regulators of gene expression, and their dysregulation is linked to cancer and other diseases, making them important therapeutic targets. Several strategies for targeting and modulating miRNA activity are being explored. For example, steric-blocking antisense oligonucleotides (ASOs) can reduce miRNA activity by either blocking binding sites on specific mRNAs or base-pairing to the miRNA itself to prevent its interaction with the target mRNAs. ASOs have been less explored as a tool to elevate miRNA levels, which could also be beneficial for treating disease. In this study, using the PKD1/miR-1225 gene locus as an example, where miR-1225 is located …

An ongoing canine sterilisation programme in rural and remote Western Australia provided an opportunity to assess the dingo ancestry of camp dogs and opportunities for hybridisation. Blood samples were collected from 345 individual community dogs at 21 locations. Dogs were screened using 23 microsatellite loci and ancestry percentage assigned using an iterative Bayesian assignment algorithm. A single individual was a dingo, 96% were domestic dogs and 3.5% were hybrids. Camp dog and dingo hybridisation in these areas is of little concern in terms of conserving …

The role of CD4+ T cells in the induction of protective CD8+ T cells by mRNA lipid nanoparticle (LNP) vaccines is unknown. We used B6 or Tlr9−/− mice depleted or not of CD4+ T cells and LNP vaccines loaded with mRNAs encoding the ectromelia virus (ECTV) MHC class I H-2 Kb-restricted immunodominant CD8+ T cell epitope TSYKFESV (TSYKFESV mRNA-LNPs) or the ECTV EVM158 protein, which contains TSYKFESV (EVM-158 mRNA-LNPs). Following prime and boost with 10 μg of either vaccine, Kb-TSYKFESV-specific CD8+ T cells fully protected male and female mice from …

Eukaryotic gene expression is regulated at the transcriptional and post-transcriptional levels, with disruption of regulation contributing significantly to human diseases. The 5' m7G mRNA cap is a central node in post-transcriptional regulation, participating in both mRNA stabilization and translation efficiency. In mammals, DCP1a and DCP1b are paralogous cofactor proteins of the mRNA cap hydrolase DCP2. As lower eukaryotes have a single DCP1 cofactor, the functional advantages gained by this evolutionary divergence remain unclear. We report the first functional dissection of DCP1a and DCP1b, demonstrating that they are non-redundant cofactors of DCP2 with unique roles in decapping complex integrity and specificity. …

Purpose: : Polyvinyl alcohol-capped silver nanostructures (cAgNSs) were investigated in order to enhance the cytotoxicity, proapoptotic, and oxidant patterns of in human laryngeal carcinoma Hep-2 cells by employing a 50 mT electromagnetic field (LEMF) for 30 min. Methods: Wet chemical reduction was used to synthesize the cAgNSs, and after they had been capped with polyvinyl alcohol, they were specifically examined for particle size analysis and structural morphology. To visualize how the silver may attach to the protein targets, a molecular docking study was conducted. Estimation of cytotoxicity, cell cycle progression supported by mRNA expression of three apoptotic-promoting genes and one …
