The growing global population and increasingly unpredictable weather patterns have intensified the demand for sustainable food production methods. Hydroponics, a form of Controlled Environment Agriculture (CEA), has gained attention as a viable solution due to its efficient use of resources such as land and water. Although hydroponic systems are perceived as safer alternatives to conventional farming, recent foodborne outbreaks associated with these systems have raised significant public health concerns. As the human population grows, demand for agriculture and renewable energy rises. Landuse competition is caused by the expansion of area-intensive solar photovoltaic (PV) facilities. While solar energy is crucial to mitigate climate change, it presents challenges related to land availability and effects on habitats for essential pollinators. In Chapter II, a spatial analysis was conducted to examine the relationship between solar panel density and pollinator diversity, exploring regions with the least interaction between these two factors. Drug development research has long focused on synthesizing novel therapeutics while overlooking isolation from naturally available sources. Recently, the discovery of the therapeutic effects of highly bioavailable animal venom caused the field of venom research to soar in popularity. Animal venom serves as a natural, sophisticated tool optimized over millions of years by evolution with the ability to bind to ion channels such as nAChRs, non-specific ligand-gated ion channels distributed throughout the human nervous system, with high selectivity and specificity. Urbanization can negatively impact birds of prey, which occupy higher trophic levels. The Gray Hawk (Buteo plagiatus) is a neotropical raptor inhabiting the Lower Rio Grande Valley (LRGV) of Texas, where it is threatened. The species’ ecology has been little studied and it is unclear how urbanization may affect population viability. I studied Gray Hawk reproductive success, diet, and thermal ecology at nests in urban and natural habitats in the LRGV. Camera trap data showed adults at urban nests delivered mostly avian prey, while mammalian prey was most frequently brought to natural nests. Data from temperature loggers showed …

Nutrition plays a vital role in early childhood, where rapid growth and development occur. This study investigates Early Interventionists’ (EIs) nutrition knowledge and perceptions of nutrition-related concerns in children receiving services through the Tennessee Early Intervention System (TEIS). An electronic survey was distributed to EIs in Tennessee to assess their nutrition knowledge and perceptions and to identify nutrition concerns. Findings revealed that most EIs have never received any nutrition education, despite being asked for nutrition guidance by families and being open to nutrition-related training. The EIs reported seeing nutrition concerns such as growth delays, food refusal, and picky eating, but …

Cholesterol is crucial in modulating the behavior and characteristics of biological membranes, liposomes, and proteoliposomes, for instance in drug delivery applications. Cholesterol regulates membrane stability by influencing lipid density, phase separation, and protein-membrane interactions. In planar bilayers, cholesterol enhances membrane thickness, reduces permeability, and promotes lipid ordering, adding rigidity and fluidity. Cholesterol also modulates membrane curvature in curved bilayers and vesicles, stabilizing low-curvature regions. In this work, all-atom molecular dynamics (MD) simulations were employed to examine how varying cholesterol concentrations influence the structure and dynamics of membranes, focusing on planar and curved geometries. We investigated the real-time metabolic rates of activated macrophage cell lines derived from mouse, chicken and rainbow trout. Each species showed a different kinetic signature with metabolic rates accelerating under fever conditions specific to each organism. Upon activation mouse macrophages increased their rate of glycolysis early and then a later reprogramming phase occurred with a dramatic decrease in oxidative phosphorylation (OXPHOS) accompanied by a reciprocal second increase in glycolytic rates. Chicken macrophages showed an initial increase in both glycolytic and OXPHOS rates. The late drop OXPHOS rates also occurred in chicken however there was no reprogramming that occurred as glycolytic …

The relative failure of immune checkpoint inhibitors in pancreatic ductal adenocarcinoma (PDAC) despite having a dense, immunosuppressive tumor microenvironment highlights the need to target alternate/escape pathways. We have previously examined C–C chemokine receptor type 9 (CCR9) as a candidate immune checkpoint and developed a targeted, humanized monoclonal antibody (SRB2). Cytotoxicity of SRB2 was evaluated in vitro and in vivo. CCR9 expression on PDAC cells/tissues, immune components of patient-derived organoids (PDOs), and antibody-dependent cell-mediated cytotoxicity were examined. In PANC-1 and MIA PaCa-2 cell lines, we demonstrated highest CCR9 expression; however, no direct cytotoxic effect was observed with SRB2 treatment. In PANC-1 …

Uncoupling protein 2 (UCP2) is a mitochondrial protein that regulates the flow of protons down the gradient established by the electron transport chain (ETC) without generating ATP, thus uncoupling the proton gradient from ATP generation. In tumors, specifically pancreatic cancer (PDAC), the ETC is highly stimulated to generate the copious ATP needed by the tumor to grow. However, extended ETC use increases reactive oxygen species (ROS), and if left unchecked, results in cell death. To evade this, PDAC employs antioxidant strategies, including upregulating UCP2 in tumor cells. In addition to its uncoupling function, UCP2 indirectly controls ROS levels by maintaining …

Measuring soil water content is crucial for various applications, such as agriculture and environmental monitoring. Scientists use sensors that rely on electromagnetic waves to estimate soil water content by measuring the dielectric of media. However, factors, including the frequency of the sensor's measurements, soil particle size and distribution, and the soil's electrical conductivity, can affect the dielectric measurements and, consequently, the accuracy of the water content estimation. Anthropogenic methane emissions contribute to the greenhouse gas profile which is the chief cause of climate change. One significant source of methane emissions is biogas produced from bacteria in wastewater facilities and landfills. Methane rich biogas has the potential to be valorized into high value bioproducts by using methanotrophs (microbes that use methane as their primary carbon source). Microalgae are incredible organisms that have a variety of uses such as wastewater treatment, fertilizer, livestock feed, and bio-compound production. Microalgae require a light source and minimal nutrients space to proliferate, making them an environmentally friendly and sustainable option for such processes. The Sustainable Wastes-to-Bioproducts Engineering Center at Utah State University is developing and implementing a Rotating Algae Biofilm Reactor (RABR) to remove total nitrogen and phosphorous from the Central Valley Wastewater Reclamation Facility wastewater effluent. In chocolate-coated products, oil migration in fats can lead to chocolate softening, filling hardening, discoloration, and lower consumer acceptance. This study examined three different fats (soy- palm-, and palm-kernel (PK) based), crystallized under different conditions (cooling rate and use of high intensity ultrasound (HIU)) to examine how the crystallization conditions impact the physical properties of confectionery products. These conditions produced fats with high and low oil binding capacities (OBCs); the ability of crystal networks to entrap liquid oil. Pathogens that infect commercially reared bee species, such as honey bees and bumble bees, have been extensively studied within the last several years, and studies that explore pathogen impacts on solitary bee health are significantly fewer. Consistent pesticide exposure methods are limited to honey bees and do not outline exposure routes to solitary bees, including how to test chemical impacts on immature stages. Dryland crop production in the Intermountain West is largely limited by the amount of precipitation received. Increasing dryland safflower (Carthamus tinctorius L.) production yields in a semi-arid climate is possible with a dormant planting but is not well understood. A two-year (2020-22) project was designed to gain an understanding of the effects of variety, seeding rate, and planting date, on dormant seeded safflower. Chemical weed control options for safflower were also evaluated. Food insecurity is a critical issue globally and in the United States. The USDA’s Special Supplemental Nutrition Program for Women, Infants, and Children (WIC) aims to address this by improving the nutrition of low-income pregnant women and children. However, immigrants and refugees, who are particularly vulnerable to food insecurity, often encounter barriers to accessing WIC benefits. This study aimed to: i) evaluate how immigrant and refugee families in Utah utilize WIC benefits, and ii) explore their experiences, challenges, and suggestions for improving the program. Aquatic invasive plants are non-native plants that invade a water body and displace native aquatic plants. Invasion depletes the native plant community's ability to provide ecosystem services, including improved water quality, decreased problematic algae, and increased animal habitat. The loss of these ecosystem services reduces the value of fishing, boating, and swimming opportunities in aquatic ecosystems. Encroachment of human infrastructure fragments wild landscapes and introduces a suite of negative impacts for humans and wildlife. In Utah, roadways threaten migratory ungulates by creating a barrier to movements and preventing access to seasonal migration routes. To mitigate these conflicts, state transportation agencies often install wildlife crossing structures to allow animals to safely cross over or under roadways. Current food packaging materials are often non-biodegradable and harmful for ingestion, which poses a threat to the environment and public health. Thus far, the dangers of microbial contamination of food on human health have outweighed the risks associated with using these conventional packaging plastics. North American rangelands are vast, open landscapes that support diverse ecosystems and provide many ecosystem services. Many wildlife call sagebrush rangelands home, including the Greater sage-grouse (sage-grouse; Centrocercus urophasianus). Sage-grouse habitat encompasses 165 million acres across western North America, and this bird has been of conservation concern as their populations drastically declined since the 1960s. Rangelands are also grazed by livestock, which has the potential to negatively affect sagebrush ecosystems when overgrazed or overstocked, which could be detrimental to sage-grouse nesting habitat. Grass-dominated rangelands in the Western United States often lack sufficient protein for pregnant cows after grasses mature in mid- summer, leading ranchers to rely on expensive feed supplements such as protein cubes or alfalfa hay. The goal of this study was to screen 27 perennial legume and forb species with the potential to remain green into late summer and provide sufficient protein to supplement cows on grass-dominated rangeland. Three experiments were carried out to understand germination of species, plant establishment and persistence at five Utah locations, and agronomic characteristics. The escalating impact of climate change on agricultural production underscores the need to investigate sustainable crop enhancement methods. This research explores the use of plant biostimulants to grow onion (Allium cepa L.) under varying environmental conditions. In the initial germination and greenhouse screening, seven biostimulants were tested on ‘Vaquero’ onion seeds: three bacterial products (Continuum, Spectrum DS, and Tribus Original), two mycorrhizal products (Mighty Mycorrhizae and Myco Apply), one seaweed extract product (Seaweed), and one product containing humic acid (Huma Pro 16). River metabolism, the measure of energy flow in a river, is driven by hydrological and physicochemical regimes (e.g., temperature, turbidity, and nutrients). However, these regimes are increasingly altered by human water demand and climate change-driven drought. These stressors alter the magnitude, timing, and duration of critical spring-snowmelt floods with poorly understood consequences on ecosystem function, particularly in desert rivers. The Cheddar cheese industry has recently observed variation in the pH of their cheese during the early stage of storage. This variation may be due to the addition of the faster acid producing thermophilic starter culture Streptococcus thermophilus which was recently introduced into the Cheddar cheese making process. This starter culture may impact the calcium buffering properties of the cheese due to more retention of the insoluble calcium (INSOL Ca) within the cheese. To investigate this problem, we studied how insoluble calcium affects cheeses with pH ranging from 4.8 to 6.6 and found that INSOL Ca was positively correlated with …

Due to harsh and variable environmental conditions, successful plant establishment in "dryland" ecosystems – areas with scarce precipitation – is a major challenge for restoration. One of the most important decisions influencing restoration outcomes is choosing which species to plant (i.e., species selection). Here I explored two opportunities to improve restoration success related to species selection. Glioblastoma (GBM) is a highly aggressive brain tumor that remains a significant challenge in neuro-oncology today. Approximately 90% of GBM patients experience tumor recurrence within one to two years of diagnosis. Therefore, due to its high resistance to conventional therapies, new treatment strategies are urgently needed. The current standard of care for GBM is the chemotherapeutic Temozolomide (TMZ). TMZ is an oral alkylating agent, it functions by delivering a methyl group to purine bases. The consequences of climate change and anthropogenic activity have decreased wildlife biodiversity. Plants are particularly vulnerable to climate change due to their large biomass on the planet and sessile nature. A particularly useful method for plants to cope with environmental changes is phenotypic plasticity. Adjusting morphological traits through plasticity can help plants cope with environmental changes. Furthermore, plastic responses can be passed down through methods such as changes in DNA sequence or epigenetic modifications.
