Innate immune system plays an important role in individual's protection against pathogens and in activation of adaptive immune system. Utilizing RAW 264.7 murine macrophages as an innate immune response representative in this study, we analyzed the effect of invasive pathogen's components (e.g. flagellin) on the arrangement of macrophage's cytoskeleton, on viability of immune cells and on secretion of pro-inflammatory and anti-inflammatory cytokines and on fluorescence intensity of cytoskeleton after rearrangement. Additionally, we studied the similarity and differences between bacterial (Salmonella typhimurium) and synthetic TLR4 agonist (synthetic lipid-A) on viability, fluorescence intensity, cytokine secretion, and cytoskeleton rearrangements. Similarly, we studied the …

Amyotrophic Lateral Sclerosis (ALS) is a fatal, adult-onset progressive degenerative motor neuron disease that is characterized by muscle atrophy and weakness due to the loss of upper and lower motor neurons. Average survival time for individuals diagnosed with the disease is three to five years; currently there is no cure and only one drug approved by the Food and Administration (FDA). Scientists have proposed various theories in order to solve the mystery which surrounds ALS. One of these theories hypothesizes how hyperexcitability and excitotoxicity leads to the death of motor neurons. In this study, we will address ways of combatting …

The removal of mRNA transcript poly(A) tails by 3'-5' exonucleases is the rate-limiting step for controlled mRNA decay in eukaryotes. Poly(A)-specific ribonuclease (PARN) is one such exonuclease that degrades poly(A) tails, and although its in vitro activity is well-characterized, PARN’s patho-physiological roles in the cell are not well understood. Prior studies have found a possible role for PARN in cancer, in that PARN expression levels in human breast cancer tissues are often decreased compared to normal control tissues. Indeed, data mined from the ONCOMINE cancer array database showed that PARN is downregulated in patient invasive breast carcinoma samples compared to …

Silver nanoparticles (AgNPs) and ionic silver (Ag+) are known to be broad-spectrum antimicrobial agents. Recent studies show these agents may be an alternative to the most widely used drinking water disinfectant, chlorine. Chlorine is a toxic industrial chemical with a lethal concentration of 430 ppm after 30 minutes. Additionally, chlorine can react with naturally occurring materials to produce a number of disinfection byproducts such as chloroform and trihalomethanes. Some of these byproducts pose cancer risks in addition to other negative impacts to human health. These would be eliminated with the use of Ag+ or AgNPs. The main goal of this …

Translation of falls prevention evidence into practice is problematic. Understanding older adults’ views about falls prevention information could enhance delivery of falls prevention, resulting in better engagement and uptake of recommended activities. The aim of this study was to examine the views and preferences of community-dwelling older adults about seeking and receiving falls prevention information. A community forum using a modified World Café approach was conducted. Participants discussed five topic areas in small groups, under the guidance of table facilitators. Perspectives were captured on paper. Thematic analysis was conducted to identify factors that influence participants’ engagement and uptake of information. …
The Neonatal Anti-Viral Response Fails To Control Measles Virus Spread In Neurons Despite Interferon-Gamma Expression And A Th1-Like Cytokine Profile, Priya Ganesan
Electronic Theses and Dissertations
Neonates are highly susceptible to infections in the central nervous system (CNS) and have a greater risk of viral infections and encephalopathies. Neurotropic viral infections can lead to blindness, hearing loss and neurological deficiencies such as cognitive impairment, epilepsy, and even death in the neonatal and pediatric populations. Viral infections also are hypothesized to indirectly contribute to neurodegenerative and neuropsychiatric diseases such as Schizophrenia and Parkinson’s disease later in life due to early neuronal damage or stress. Many diverse viruses are capable of invading the neonatal CNS including Borna Disease Virus, Coxsackievirus (CV), Herpes simplex viruses (HSV), and measles virus. …

Neuronal Gq Structures In Neurodegeneration, Damian S. Mcaninch
Electronic Theses and Dissertations
This study investigates protein nucleic acid interactions between various proteins and G quadruplex (GQ) forming messenger RNAs (mRNAs) in human neurological disorders. GQ structures are formed in DNA/RNA, when four guanine residues form planar tetrads stabilized by Hoogsteen base pairing, that stack forming a GQ structure stabilized by potassium ions. These GQ structures are targeted by the arginine-glycine-glycine (RGG) repeat domain containing RNA-binding domain.
Three RGG domain containing RNA-binding proteins, all of which have been implicated in neurological disorders, and their interactions with GQ forming mRNAs, were investigated in this study: fused in sarcoma (FUS), fragile X mental retardation protein …
Distribution Changes, Genetic Population Structure, And A Novel Environmental Dna (Edna) Detection Method For Darters (Subgenus Nothonotus) In The Upper Ohio River Watershed, Anthony S. Honick
Electronic Theses and Dissertations
In the upper Ohio River watershed three species of small-bodied benthic fish the Bluebreast Darter, Etheostoma (Nothonotus) camurum (Cope), the Tippecanoe Darter, Etheostoma (Nothonotus) tippecanoe Jordan and Evermann, and the Spotted Darter, Etheostoma (Nothonotus) maculatum Kirtland previously existed in disjunct distributions due to poor water quality and habitat degradation. Signs of recovery indicated that these species were moving from areas of refugia into the deeper mainstem waters of the Allegheny and Ohio rivers and expanding their distributions. To provide information for the proper conservation management of these species this dissertation was divided into three stages: 1) distribution records were updated …

Accuracy And Repeatability Of Single-Pose Calibration Of Inertial Measurement Units For Whole-Body Motion Analysis, Xavier Robert-Lachaine, Hakim Mecheri, Christian Larue, André Plamondon
Études primaires
Portable inertial measurement units (IMUs) are suitable for motion analysis outside the laboratory. However, IMUs depend on the calibration of each body segment to measure human movement. Different calibration approaches have been developed for simplicity of use or similarity to laboratory motion analysis, but they have not been extensively examined. The main objective of the study was to determine the accuracy and repeatability of two common single-pose calibrations (N-pose and T-pose) under different conditions of placement (self-placement and passive placement), as well as their similarity to laboratory analysis based on anatomical landmarks. A further aim of the study was to …

Underlying mechanisms of obesity-related back pain remain unexplored. Thus, we aim to determine the effect of obesity and its shapes on the spinal loads and the associated risks of injury. Obesity shapes were initially constructed by principal component analysis based on datasets on 5852 obese individuals. Spinal loads, cycles to vertebral failure and trunk stability margin were estimated in a subject-specific trunk model taking account of personalized musculature, passive ligamentous spine, obesity shapes, segmental weights, spine kinematics and bone mineral density. Three obesity shapes (mean and extreme abdominal circumferences) at three body weights (BWs) of 86, 98 and 109 kg …

Background
Lumbar stabilization exercise programs should normalize the aberrant movements patterns often observed in patients with low back pain. This study aimed to determine the effect of an 8-week lumbar stabilization program on EMG/kinematics measures of the aberrant movement patterns in such patients. A secondary goal was to assess the 8-week test-retest reliability of these measures.
Methods
The patients followed an 8-week lumbar stabilization program while no intervention was carried out on the controls. Before and after this period, kinematics of the spine along with the EMG of paraspinal muscles were recorded during trunk maximal flexion-extension. ANOVAs tested the effect …

Applying Theory Of Constraints To Timber Harvesting: A Case Study From The Northeast Usa, Matthew Kelly, René H. Germain
Applying Theory Of Constraints To Timber Harvesting: A Case Study From The Northeast Usa, Matthew Kelly, René H. Germain
Michigan Tech Publications, Part 1
Logging firms are a critical link in wood supply chains, connecting forest landowners with markets for wood products. Improving operational planning can benefit individual logging firms as well as the larger wood supply chain in which they operate. Applying concepts from Theory of Constraints (TOC) to timber harvesting may help achieve greater predictability and efficiency when planning harvest operations. However, examples that demonstrate how TOC can improve logging operations are lacking. This study focuses on the analysis of production and activity data collected during the harvest of a temperate mixed hardwood forest in the Northeast United States using a chainsaw-forwarder …

Multi-scale models can facilitate whole plant simulations by linking gene networks, protein synthesis, metabolic pathways, physiology, and growth. Whole plant models can be further integrated with ecosystem, weather, and climate models to predict how various interactions respond to environmental perturbations. These models have the potential to fill in missing mechanistic details and generate new hypotheses to prioritize directed engineering efforts. Outcomes will potentially accelerate improvement of crop yield, sustainability, and increase future food security. It is time for a paradigm shift in plant modeling, from largely isolated efforts to a connected community that takes advantage of advances in high performance …

The methylation status of O-6-methylguanine-DNA methyltransferase (MGMT) is associated with the prognosis in gliomas and in other cancers. Recent studies showed that rs16906252, an SNP in the MGMT promoter, is associated with promoter methylation and is a predictor of the overall survival time (OST) and the response to temozolomide (TMZ) treatment. However, these findings haven't been systematically investigated in the Han-Chinese population. We analyzed the relevance between rs16906252 polymorphisms, the MGMT methylation status, and the OST in 72 Han-Chinese gliomas patients. The MGMT promoter methylation was measured by bisulfite conversion followed by pyro-sequencing, while rs16906252 was measured by restriction endonuclease …

Articles, Abstracts, and Reports
We have established proof of principle for the Indicator Cell Assay Platform™ (iCAP™), a broadly applicable tool for blood-based diagnostics that uses specifically-selected, standardized cells as biosensors, relying on their innate ability to integrate and respond to diverse signals present in patients' blood. To develop an assay, indicator cells are exposed in vitro to serum from case or control subjects and their global differential response patterns are used to train reliable, disease classifiers based on a small number of features. In a feasibility study, the iCAP detected pre-symptomatic disease in a murine model of amyotrophic lateral sclerosis (ALS) with 94% …

Differences Between The Genomes Of Lymphoblastoid Cell Lines And Blood-Derived Samples., Lena M Joesch-Cohen, Gustavo Glusman
Articles, Abstracts, and Reports
Lymphoblastoid cell lines (LCLs) represent a convenient research tool for expanding the amount of biologic material available from an individual. LCLs are commonly used as reference materials, most notably from the Genome in a Bottle Consortium. However, the question remains how faithfully LCL-derived genome assemblies represent the germline genome of the donor individual as compared to the genome assemblies derived from peripheral blood mononuclear cells. We present an in-depth comparison of a large collection of LCL- and peripheral blood mononuclear cell-derived genomes in terms of distributions of coverage and copy number alterations. We found significant differences in the depth of …

Distribution Of The North American Porcupine (Erethizon Dorsatum) In Northern California, Cara L. Appel, William J. Zielinski, Frederick V. Schlexer, Richard Callas, Tim Bean
Open Educational Resources
Western Wildlife 4:17–28, 2017: The North American Porcupine (Erethizon dorsatum) is one of the most widely distributed mammals in North America, but recent reports have suggested declines in parts of its range in the West. In California, little is known about the historical or current status of the porcupine, and maps of its distribution conflict considerably. Nevertheless, the species is of interest to natural resource managers. For much of the 1900s, foresters and others primarily treated porcupines as pests because of the undesirable damage they inflict feeding on trees and gnawing on manmade items in search of salt. …
Glaucous-Winged Gull Predation On Chicken Eggs Treated With A Uv-Blocking Agent, Athena T. Smith
Glaucous-Winged Gull Predation On Chicken Eggs Treated With A Uv-Blocking Agent, Athena T. Smith
Master's Theses
Egg cannibalism plays a major role in the life histories of Glaucous-winged Gulls (Larus glaucescens) breeding at Protection Island, Washington. Gulls, along with other birds, visualize reflected ultraviolet (UV) light and I wondered if reflected UV light plays a role in determining which eggs are selected for cannibalization. Motivated by studies showing that tree-nesting bird eggs coated with a UV-blocking agent were less subject to predation than control eggs, I tested whether ground-nesting Glaucous-winged Gulls would preferentially predate control chicken eggs over those coated with a UV-blocking agent. Early during five mornings I formed artificial nests at randomly-determined …
