[Objective ] A simple and sensitive detection method for Staphylococcus aureus is established for the efficient detection of this pathogen in food.[Method ] Targeting Staphylococcus aureus,a simple and sensitive fluorescent detection method for Staphylococcus aureus,EXPAR-P-AgNCs,was developed based on exponential amplification reaction (EXPAR ) combined with DNA silver nanoclusters (P-AgNCs ).A recognition probe was designed to initiate EXPAR in the presence of Staphylococcus aureus,generating a large amount of target DNA(T).The binding of T to hairpin P displaced the G-rich strand from P-AgNCs,resulting in a decrease in the relative fluorescence intensity of P-AgNCs and enabling the detection of Staphylococc us aureus.[Result ] …

[Objective ] This study aims to explore the physicochemical indicators,microbial diversity,and volatile substances in the Qupi (surface ) and Quxin (core) of high-temperature Daqu.[Method ] High-throughput sequencing and headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS ) were employed to compare the microbial diversity,volatile substances,and physicochemical indicators between Qupi and Quxin of three types of Daqu.[Result ] The moisture mass fraction in Qupi of the three types of Daqu was lower than that in Quxin (P<0.05),and the moisture mass fraction was the lowest in the Qupi of white Daqu (WP) and the highest in the Quxin of yellow Daqu (YX).The acidity of Qupi and Quxin were in the order of Quxin of black Daqu (BX) > YX > Qupi of black Daqu (BP) > Quxin of white Daqu (WX) > Qupi of yellow Daqu (YP) > WP.High-throughput sequencing results showed that Firmicutes and …0.05),and>

The genus Aspergillus comprises over 250 species of fungi. Within this genus, only a handful of species cause disease in humans. One species is Aspergillus fumigatus (A.f.), an environmentally ubiquitous, saprophytic fungal pathogen. Lung infections due to A.f. range from invasive infection to allergic forms of the disease. Allergic bronchopulmonary aspergillosis (ABPA) is one of the most severe forms and presents as a hypersensitivity that often results in off-target damage to the surrounding airways. A novel nanocomposite of polyvinyl alcohol formaldehyde (PVF) sponge loaded with green zinc oxide nanoparticles (ZnONP) and referred to as (NP-PVF) was fabricated as a potential adsorbent of ciprofloxacin from water. A simple mechanical foaming process was used to synthesize the adsorbent. Rice husk extract was used to synthesize zinc oxide nanoparticles (ZnONP) for its phytochemicals as reducing agents. A facile and low-energy-dependent extraction method was used that did not require the use of high-temperature annealing. The synthesized nanoparticles showed a maximum UV absorption at 347 nm, which corresponds to a 3.58 eV band gap energy. FTIR confirmed the successful …

Mitochondrial health relies on the membrane fission mediated by dynamin-related protein 1 (Drp1). Previous structural studies of Drp1 on remodeled membranes were hampered by heterogeneity, leaving a critical gap in the understanding of the mitochondrial fission mechanisms. Here we present a cryo-electron microscopy structure of full-length human Drp1 decorated on membrane tubules. Using the reconstruction of average subtracted tubular regions (RASTR) technique, we report that Drp1 forms a locally ordered lattice along the tubule without global helical symmetry. The filaments in the lattice are similar to dynamin rungs with conserved stalk interactions. Adjacent filaments are connected by GTPase domain interactions …

Macrophages are critical custodians of the lung, orchestrating immune surveillance, maintaining tissue homeostasis, and coordinating both innate and adaptive responses. This dissertation examines their heterogeneity and specialized functions, focusing on IMs, which reside in the lung interstitium, and AMs, which occupy the airspace. Through comprehensive single-cell transcriptomic profiling of murine and human samples, we identify multiple IM and AM subsets defined by distinct chemokine, cytokine, receptor, and innate immune gene signatures. These subsets display clear divisions of labor, contributing to immune cell recruitment, tertiary lymphoid structure formation, and alveolar homeostasis.

Most vertebrates need a constant supply of oxygen to sustain life, with disruption to O2 supply or delivery quickly leading to cell and tissue damage and death. Many aquatic species are able to withstand long periods without O2 due to evolving to survive the regular periods of hypoxia and anoxia in their natural habitat. The Austrofundulus limnaeus, or the annual killifish, in particular have shown the highest anoxia tolerance in all vertebrates and is a promising model for developing effective treatments for anoxia-related damage, such as heart attacks and strokes. Type I interferons (IFNs-I), a group of pleiotropic cytokines, critically modulate host response in various inflammatory diseases. However, the role of the IFN-I pathway in periodontitis remains largely unknown. In this report, we describe that the IFN-β levels in the gingival crevicular fluid of human subjects were negatively associated with periodontitis and clinical gingival inflammation. Disruption of IFN-I signaling worsened alveolar bone resorption in a ligature-induced periodontitis murine model. Deficiency of the IFN-I pathway resulted in an exaggerated inflammatory response in myeloid cells and drastically increased the interleukin-17 (IL-17)-mediated neutrophil recruitment in the gingiva. Coral reefs are vulnerable to disruptive human activities such as pollution, overfishing, and coastal development—degrading water quality and reef structures. Active reef restoration entails temporarily relocating several corals to aquaculture facilities to achieve successful rehabilitation; however, current coral transport methods pose certain risks, such as stress-induced mortality, which can result from physical damage during transportation. Existing studies on coral transport have explored various techniques for collecting, shipping, and maintaining live coral specimens. Bacterial chondronecrosis with osteomyelitis (BCO) is a major cause of lameness in broiler chickens. The anterior pituitary is comprised of five cell types which secrete regulatory hormones into the bloodstream. Specifically, gonadotrophs synthesize and release sex hormones follicle-stimulating hormone (FSH) and luteinizing hormone (LH) in response to gonadotropin-releasing hormone (GnRH). GnRH signals from the hypothalamus regulate the differential secretion of LH and FSH from the anterior pituitary. Exocytosis of FSH and LH from dense core vesicles (DCVs) occurs in response to an intracellular calcium signal, sensed by the synaptotagmins proteins. Conservation professionals expect increased attempts to weaken the US Endangered Species Act (ESA) during the second Trump administration. As such, it is important to understand Americans’ level of support for the ESA. Prior research indicates that support for the ESA remained consistently strong across four studies conducted over a two-decade period, 1996–2015. The research presented here extends those observations to six studies conducted over a three-decade period, 1996–2025. We find that support of the ESA over that period has remained consistently high, at about 84%, and opposition has remained consistently low, at about 12%. We also report on other trends …

The natural world abounds with examples of complex behavior in humans and many other species. Evolutionary game theory is a powerful mathematical framework to understand the origins of many such behaviors like cooperation. Since these behaviors are often selected against initially, understanding why they are so widespread has been a longstanding question. Rather than assuming agents' rationality, like in traditional game theory, this approach studies the mutation and selection of strategies themselves. Purpose of review: Virotherapy has emerged as a promising approach to cancer treatment. Over the past two decades, early-phase clinical trials have demonstrated the safety and promising efficacy of virotherapy in subsets of cancer patients. However, a significant knowledge gap needs to be filled to propel the field further and achieve substantial anti-cancer benefits for more than the current 10-20% of treated patients. Following the observed significant improvements in overall survival and progression-free survival in clinical trials, the combination of atezolizumab and bevacizumab has been recommended as a first-line therapy for patients with unresectable hepatocellular carcinoma. Despite its clinical benefits, the high cost associated with this treatment poses a substantial challenge in routine practice in Taiwan. The Ping-Chong-Jiang-Ni Formula (PCJNF), a compound of Traditional Chinese Medicine (TCM), has demonstrated remarkable clinical effectiveness and is widely utilized in the treatment of endometriosis (EMs). Previous studies have shown that PCJNF inhibits the proliferation and induces the apoptosis of ectopic endometrial stromal cells (EESCs) by modulating the JNK signaling pathway. Synthetic cathinones are a widely abused class of new psychoactive substances that pose significant challenges in forensic toxicology owing to their complex metabolic pathways and lack of verified reference standards. This study investigated the in vitro metabolism of nine methylenedioxy-substituted synthetic cathinones, including methylone and eight structural isomers, which were categorized into three groups: Group 1 (dimethylone, ethylone, and butylone), Group 2 (dibutylone, pentylone, and eutylone), and Group 3 (N-ethylpentylone and dipentylone). Chronic atrophic gastritis (CAG) is precancerosis with limited treatment options. This study investigated the therapeutic effects of velvet antler (VA), a traditional natural medicine, on CAG, with a focus on ferroptosis. In vivo experiments demonstrated the efficacy of VA in alleviating CAG, and network pharmacology analysis revealed its involvement in related terms, including inflammation, oxidative stress, and cell proliferation. Analyses further confirmed the modulation of ferroptosis by VA, particularly via the Keap1-Nrf2/HO-1/GPX4 pathway. In vitro assays supported these findings, demonstrating the ability of VA to reduce methylnitronitrosoguanidine-induced inflammation, oxidative stress, and ferroptosis in GES-1. Jiao Sanxian (JSX) consists of three traditional Chinese medicine including charred hawthorn, malt and Liu Shen Qu, they are also from Chinese medicine homologous food. It is commonly used to relieve functional dyspepsia (FD). The present study aims to explore the mechanism of JSX (granule), with the addition of prebiotics for the treatment of FD using mice model. The network pharmacology and molecular docking were used to forecast therapeutic targets for JSX. Based on the protein-protein interaction (PPI) results, thirteen targets exhibited strong interactions with each other. BACKGROUND: Acute myeloid leukemia (AML) is an aggressive hematologic cancer with a notoriously bleak prognosis; for non-M3 AML, the overall 5-year survival rate is ∼30%. While 60-70% of newly diagnosed AML patients will achieve complete remission (CR), half of these patients will experience relapse (secondary resistance) by three years from their diagnosis. Moreover, 30-40% of AML patients present with refractory disease (primary resistance) and cannot respond to frontline treatments. Leukemia stem cells (LSCs) are implicated in both primary and secondary resistance, and their eradication is necessary to maintain CR. Quinones are ubiquitous species that can be produced from the photochemical aging of combustion-derived particles (CDPs). Polycyclic aromatic hydrocarbons (PAHs) are a major component of CDPs and are precursors to quinones and other oxidized products (OPAHs). My work first expanded on the PAH and OPAH photochemistry research of Dr. John Haynes, who showed that anthracene (ANT) oxidizes into 1,4-naphthoquinone (1,4-NAPQ), 1,4-anthraquinone (1,4-ANTQ), and 9,10-anthraquinone (9,10-ANTQ), and of Dr. Heather Runberg, who demonstrated the ability of ANT and these quinones to generate reactive oxygen species (ROS). Clathrin-mediated endocytosis (CME) is a cellular process for internalizing extracellular materials and recycling parts of the plasma membrane into intracellular vesicles. When a threshold of membrane-bound receptors is met at the cell surface, endocytic proteins organize into a conformation that can invaginate the membrane and separate a vesicle into the cytosol. Our current model for understanding CME has been derived from studies in Saccharomyces cerevisiae. Previous research has led to a model for how Las17, a homolog of Wiskott Aldrich Syndrome Protein (WASP), contributes to CME. The goal of this project is to study the protein CowN in the diazotroph Azotobacter vinelandii (Av) in comparison to CowN in Gluconacetobacter diazotrophicus (Gd). Nitrogenase is an enzyme that catalyzes the reduction of nitrogen gas into ammonia in diazotrophic bacteria. In many diazotrophs, the protein CowN prevents nitrogenase inhibition from the environmental gas carbon monoxide (CO). Previous work studied CowN in the diazotroph G. diazotrophicus. In G. diazotrophicus, CowN binds to nitrogenase and weakens CO binding to its active site. The mechanism of CowN has only been investigated in G. diazotrophicus.
