Opening of long-lived pores in the cell membrane is the principal primary effect of intense, nanosecond pulsed electric field (nsPEF). Here we demonstrate that the evolution of pores, cell survival, the time and the mode of cell death (necrotic or apoptotic) are determined by the level of external Ca2+ after nsPEF. We also introduce a novel, minimally disruptive technique for nsEP exposure of adherent cells on indium tin oxide (ITO)-coated glass coverslips, which does not require cell detachment and enables fast exchanges of bath media. This fourth special electroporation-based technologies and treatments issue of the Journal of Membrane Biology contains reports on recent developments in the field of electroporation by participants in the 7th International Workshop and Postgraduate Course on electroporation based technologies and treatments (EBTT 2013) held in Ljubljana, November 17–23, 2013. The 65 participants included faculty members, invited lecturers, special guests, and young scientists, and students from 16 countries. In addition to lectures on the fundamentals, this year’s sessions included talks on microbial inactivation by pulsed electric fields, modeling of intracellular electroporation, electroporation in food processing, and electrotransfer-facilitated DNA vaccination.

The atmospheric pressure dielectric barrier discharge in helium is a pulsed discharge in nature and the moment of maximum species densities is almost consistent with peak discharge current density. In this paper, a one-dimensional fluid model is used to investigate the temporal structure of plasma species in an atmospheric He-N2 dielectric barrier discharge (DBD). It is demonstrated that there exist microsecond delays of the moments of the maximum electron and ion densities from the peak of discharge current density. Nuclei/Cell detection is usually a prerequisite procedure in many computer-aided biomedical image analysis tasks. In this thesis we propose two automatic nuclei/cell detection frameworks. One is for nuclei detection in skeletal muscle fiber images and the other is for brain tumor histopathological images.
For skeletal muscle fiber images, the major challenges include: i) shape and size variations of the nuclei, ii) overlapping nuclear clumps, and iii) a series of z-stack images with out-of-focus regions. Although brain-computer interfaces (BCIs) can be used in several different ways to restore communication, communicative BCI has not approached the rate or efficiency of natural human speech. Electrocorticography (ECoG) has precise spatiotemporal resolution that enables recording of brain activity distributed over a wide area of cortex, such as during speech production. In this study, we investigated words that span the entire set of phonemes in the General American accent using ECoG with 4 subjects. We classified phonemes with up to 36% accuracy when classifying all phonemes and up to 63% accuracy for a single phoneme. Further, misclassified phonemes follow articulation …

Noble metal nanoparticles supporting localized surface plasmon resonances (LSPR) have been extensively investigated for label free detection of various biological and chemical interactions. When compared to other optical sensing techniques, LSPR sensors offer label-free detection of biomolecular interactions in localized sensing volume solutions. However, these sensors also suffer from a major disadvantage – LSPR sensors remain highly susceptible to interference because they respond to both solution refractive index change and non-specific binding as well as specific binding of the target analyte. Array of atmospheric pressure surface discharges confined by a two-dimensional hexagon electrode mesh is studied for its discharge modes and temporal evolution so as to a theoretical underpinning to their growing applications in medicine, aerodynamic control, and environmental remediation. Helium plasma surface-confined by one hexagon-shaped rim electrode is shown to evolve from a Townsend mode to a normal and abnormal glow mode, and its evolution develops from the rim electrodes as six individual microdischarges merging in the middle of the hexagon mesh element. The dynamical translocation of lipids from one leaflet to another due to membrane permeabilization driven by nanosecond, high-intensity (>100 kV/cm) electrical pulses has been probed. Our simulations show that lipid molecules can translocate by diffusion through water-filled nanopores which form following high voltage application. Our focus is on multiple pulsing, and such simulations are relevant to gauge the time duration over which nanopores might remain open, and facilitate continued lipid translocations and membrane transport. Our results are indicative of a N1/2 scaling with pulse number for the pore radius. These results bode well for the use of pulse …

Particle-based vaccine delivery systems are under exploration to enhance antigen-specific immunity against safe but poorly immunogenic polypeptide antigens. Chitosan is a promising biomaterial for antigen encapsulation and delivery due to its ability to form nano- and microparticles in mild aqueous conditions thus preserving the antigenicity of loaded polypeptides. Despite tremendous scientific achievements, cancer remains the second leading cause of death in the United States. Metabolic reprogramming has been increasingly recognized as a core hallmark of cancer. My dissertation work identified novel diagnostic markers and therapeutic targets for human cancers through the study of cholesterol in cancer cells.
Enabled by label-free Raman spectromicroscopy, we performed the first quantitative analysis of lipogenesis at single cell level in human patient cancerous tissues. The successful clinical use of implantable intracortical microelectrodes (ICMs) to treat certain types of deafness, blindness, and paralysis is limited by a reactive tissue response (RTR) of the brain. This RTR culminates in the formation of a tight glial scar and a loss of neuronal density around implanted ICMs, and is accompanied by a decrease in signal to noise ratio and an increase in impedance. The noninvasive measurement of finite displacements and strains in biomaterials by magnetic resonance imaging (MRI) may be shown to enable mathematical estimates of stress distributions and material properties within structures of the body such as articular cartilage or the intervertebral disc. Such methods will allow for non-contact and patient-specific modeling in a manner not currently possible with traditional mechanical testing or finite element techniques. Therefore, the objective of this thesis was to develop computational methods incorporating imaging-based measures of deformation, composition, and local microstructure to permit nondestructive analysis of a range of complex biomechanical systems.

Hearing aids are able to restore some hearing abilities for people with auditory impairments, but background noise remains a significant problem. Unfortunately, we know very little about how speech is encoded in the auditory system, particularly in impaired systems with prosthetic amplifiers. There is growing evidence that relative timing in the neural signals (known as spatiotemporal coding) is important for speech perception, but there is little research that relates spatiotemporal coding and hearing aid amplification.

Computational fluid dynamics (CFD) simulations are becoming a reliable tool in understanding disease progression, investigating blood flow patterns and evaluating medical device performance such as stent grafts and mechanical heart valves. Previous studies indicate the presence of highly disturbed, transitional and mildly turbulent flow in healthy and pathological arteries. Accurate simulation of the transitional flow requires high order numerics together with a scale resolving turbulence model such as large eddy simulation (LES). Natural Family Planning (NFP) is a method to help couples determine the fertile and infertile times of a woman’s menstrual cycle with natural indicators of fertility. NFP methods have advantages over other methods of family planning. Proper use of NFP methods also ensures high effectiveness (close to 98%) in helping couples avoid pregnancy. However, very few physicians prescribe NFP to their patients due to lack of credibility to the fertility methods and lack of access to NFP knowledge. Antennas designed to link footwear sensors within body centric networks are introduced with two small UWB antennas, one directional and another quasi-omnidirectional. The radiating characteristics are evaluated for three positions on a sample sports shoe using a detailed simulation model and measurements with a homogenous foot phantom. Antenna performance is assessed for resilience to close proximity loading by the footwear materials and the phantom foot.

Modern implantable and body worn medical devices leverage wireless telemetry to improve patient experience and expand therapeutic options. Wireless medical devices are subject to a unique set of regulations in which monitoring of the available frequency spectrum is a requirement. To this end, implants use software protocols to assess the in-band activity to determine which channel should be used. An estimated 0.5% of world’s population has been diagnosed with epilepsy. Of these patients 20-30% will be unable to achieve seizure control with anti-epileptic drugs. Vagus nerve stimulation (VNS) may be an appropriate treatment option for some patients with pharmaceutically refractory, partial-onset seizures.
VNS therapy uses a helical electrode to interface between the implantable pulse generator and the vagus nerve. Microcontrollers, the brains of embedded systems, have found their way into every aspect of our lives including medical devices such as pacemakers. Pacemakers provide life supporting functions to people therefore it is critical for these devices to meet their timing constraints. This thesis examines the use of hardware co-processing to accelerate the calculation time associated with the critical tasks of a pacemaker. In particular, we use an FPGA to accelerate a microcontroller’s calculation time of the Kendall Tau Rank Correlation Coefficient algorithm. Throughout the years, sensors have been an integral part of automation, alert, and medical systems. Many of these systems measure physiological characteristics of the human body to alert themselves of their current conditions. Drowsy driver systems, for instance, measure the eyes and facial movements with a camera to determine if the driver is falling asleep at the wheel. Electroencephalography (EEG), electrooculography (EOG), and electromyography (EMG) employ electrodes on the human body to measure electrical activity of a patient’s REM sleep cycle patterns. Pulse oximeters use optical light through a process called photoplethysmography (PPG) to measure heart rate.
