This project developed and evaluated an optical sensing system for detecting changes associated with glucose concentration. The system combined a laser-diode, cuvette sample holder, photodiode, resistive-feedback transimpedance amplifier, high-resolution analog-to-digital converter, and microcontroller. Parametric testing evaluated the effects of input current and feedback resistance on transimpedance gain, output range, and linearity. Firmware was developed to configure the ADC, average repeated conversions, monitor measurement variation, convert raw digital counts into voltage using a source-meter calibration equation, and compare sample measurements with a water reference. A cuvette enclosure maintained alignment between the laser-diode, sample, and photodiode while reducing external optical interference. Testing …

This thesis presents a comprehensive framework for the automated tracking and visualization of articulatory movements based on magnetic resonance imaging (MRI) data. A well-known data analysis tool for markerless pose estimation, known as DeepLabCut, is investigated for this purpose. The performance of this tool is enhanced through the design and implementation of a pre-processor. DeepLabCut is a markerless pose estimation toolbox based on deep learning, which overcomes the issue of making manual annotations frame-by-frame. Limitations from manually marking the MRI images are addressed by implementing transfer learning with convolutional neural networks to achieve accurate, user-defined articulator tracking without markers. With the continued growth of the biopharmaceutical industry, the demand for scalable, robust, and resource-efficient platforms for large-scale mammalian cell culture is amplified. Recent developments in microfluidic technology, such as precise control of the microenvironment, showed the potential to improve the performance of cell culture systems. However, constrained by scalability and operational efficiency, applying such approaches to large-scale cell culture and biopharmaceutical production presents challenges. This dissertation addresses these challenges through three independent but conceptually related technological developments. Every second, human skin processes over one million sensory signals while maintaining properties such as electrical conductivity, mechanical adaptability, and regenerative capability that surpass all synthetic materials. Contemporary bioelectronic devices prove inadequate when contacting skin surfaces due to poor adhesion and electrical contact issues that prevent effective sensing. Despite advancements in wearable and bioelectronic technologies, current devices face major drawbacks when interfacing with human skin, particularly in maintaining firm adhesion, conformability, and low-noise electrical signal acquisition.

Properly designed prosthetics hands can enhance the quality of life for those suffering from limb loss. Recently, 3D-printed prosthetic hands are becoming common. In these prostheses, fingers flex through the tendons that are activated by motion of the wrist. To provide spring action, thermoplastic polyurethane (TPU) hyperelastic joints are used to connect digits to each other as well as to the wrist. While these designs are common, no model for the relationship between tendon tension and joint flexion is available. Point-of-care (POC) electric field-based biosensors have emerged as a promising tool to detect early cancer biomarkers such as circulating tumor DNA (ctDNA), microRNA (miRNA), and proteins. To be effective in screening in clinical settings, these biosensors must be simple and easy to use. In this study, we have studied the manipulation of short DNA molecules suspended in a sessile drop to achieve this goal. Evaluating the effectiveness of transcranial direct current stimulation (tDCS) is essential for guiding its integration into therapeutic and performance-enhancement applications. In our laboratory, we investigate the efficacy of tDCS across multiple experimental models, including both animal and human studies. I have contributed significantly to the execution and analysis of these experiments, which include studies in rats and healthy human participants aimed at evaluating whether electrical stimulation of the motor cortex can enhance motor learning. These studies assess improvements in fine motor performance resulting from tDCS. In stroke patients, I contribute to our investigation of tDCS as a rehabilitative intervention, particularly …

Cochlear implant surgery is a delicate procedure performed by Otolaryngologists (ENTs) to implant an electronic device into the inner ear to provide a sense of sound for people who are profoundly deaf or hard of hearing. The current practices of training involve cadavers and 3D-printed models. Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. Alzheimer’s disease (AD) is a progressive neurodegenerative disease with the clinical symptoms of gradual cognitive decline and memory loss. Biologically, abnormal accumulation of Aβ (Aβ) plaque and tau tangles in the brain are the core pathological hallmarks in AD. Although inflammatory response is not specific to AD, emerging evidence suggests that the disrupted innate immune system within the central nervous system, particularly microglial activation, might act as the bridge linking Aβ aggregation to pathological phosphorylation and aggregation of tau. Genetics plays a critical role in understanding the molecular mechanisms underlying neurodegenerative disorders and pathogen evolution in infectious diseases. For example, identifying genetic variants associated with a disease phenotype uncovers functional pathways that could lead to potential drug targets and therapeutic interventions. In addition, tracking the genetic evolution of pathogens enables early detection and warning of infectious disease outbreaks. The Microphysiological Systems Laboratory aims to develop colorectal cancer tumor models under a hypoxic environment to assess model response to pharmaceutical compounds in vitro. To perform relevant studies, researchers have attempted to use different hypoxic inducing strategies such as a nitrogen pod and hypoxic incubator to recreate in vivo physiological responses to hypoxia. This work describes the design of an EEG-based neurofeedback system which provides users with real-time feedback on their level of focus or relaxation. By analyzing the spectral content of the brain activity measured via scalp electrodes, focus and relaxation levels can be quantified. Based on these measurements, live feedback in the form of moving bar graphs is provided to users, allowing them to gain awareness of their mental-state and more efficiently learn how to consciously relax or focus. However, many standardized assessment tools rely on human raters for performance assessment, which is resource-intensive and subjective. Cellular senescence, a process of irreversible cell cycle arrest, is associated with various changes in cellular morphology and function. While senescence will occur naturally in a time-dependent manner, it can be prematurely induced by various extrinsic and intrinsic factors. Recent advancements in bioimpedance technology have demonstrated significant promise in the application of cardiac health monitoring. This research explores the design and development of a forearm-based wearable bioimpedance device for non-invasive measurement of heart rate and respiratory rate at an accuracy level comparable to medical-grade monitors. Dental implantation is an increasingly common procedure used to treat missing teeth. However, surgical drilling to place the implant poses a high risk of injury to critical anatomy, such as inferior alveolar nerve injury or maxillary sinus perforation. A real-time surgical feedback system sensing proximity of these critical anatomy could reduce injury risks. Breast Cancer Related Lymphedema (BCRL) is a common co-morbidity in cancer survivors following neoadjuvant therapies such as chemotherapy, radiation, and/or surgery. It is brought about by the disruption in the lymphatic system (think lymph node biopsy) that leads to a buildup of lymphatic fluid in the arm. Current diagnostic strategies for this condition are merely retroactive, and fairly limited in the parameters that are examined to ensure patient well-being long term. Dangers associated with high radiation and microgravity exposure in space are critical challenges inhibiting us from exploring deep space and pursuing long-duration missions, as current medical systems are unable to monitor, diagnose, or treat tissue injury within physical spacecraft constraints and communication limits. This research aims to transform cancer treatment through the optimization of brachytherapy, with a focus on reducing treatment duration, setup complexities, and financial burdens, all while emphasizing patient safety. Patients living at a distance from radiation clinics, particularly those undergoing extended Low Dose Radiation brachytherapy, often struggle with the formidable financial challenges associated with securing nearby accommodations. In response to these issues, the research introduces a radiation capsule designed to condense the conventional six-month treatment period to approximately just one week, thereby significantly reducing the duration of required accommodations. Traditional scales utilized for recording pain are known to be highly subjective and biased due to inaccuracies in recollecting actual pain intensities. As a result, machine learning (ML) models that are trained using these scores as ground truth are reported to have low performance for objective pain classification because of the huge disparity between what was felt in moments of pain and the scores recorded afterward.

Piezoelectric materials show great promise for low-power wearable and implantable sensing, but their rigidity makes it challenging to integrate them with biological tissue. To address this, researchers have started exploring polymer-based functional materials that offer flexibility and are suitable for interfacing with the human body. However, these materials are still in their early stages, and a framework is necessary to illustrate how these materials, in conjunction with novel fabrication techniques and device designs, can enable the development of multi-functional sensing and energy harvesting devices.
