Effective nanopulse generators have become critical in recent decades concerning the study of subcellular affects in response to nanosecond pulsed electric fields. It has been observed that nanosecond duration electric pulses can target intracellular organelles, ultimately leading to cell apoptosis, suggesting the possibility of a new, minimally invasive, low risk cancer therapy methodology. The standard topology for developing a medical nanopulser is the Blumlein “transmission line” approach. This approach relies on the nearly infinitesimal, yet finite amount of time required for an electromagnetic field to propagate down a short transmission line. Mesenchymal stem cells are derived from a variety of human tissues and are being bioengineered and studied for possible uses in the advancement of medicine. Recent efforts are being focused on Dental Pulp Stem Cells (DPSC's) due to the accessibility of this tissue. Many factors influence DPSC quality and quantity, including the specific methods used to isolate, collect, concentrate, and store these isolates once they are removed. Ancillary factors, such as the choice of media, the selection of early versus late passage cells, and cryopreservation techniques may also influence the differentiation potential and proliferative capacity of DPSC isolates.

Drosophila melanogaster is a dominant model organism for studying the function of animal genes in initial stages of embryogenesis. Usually, images containing Drosophila gene expression patterns are captured at different developmental stages to study the interconnection of animal genes. To achieve most biologically meaningful results, gene expression images from a similar stage should be compared. Currently, biologists manually classify embryos in images into different stages, which is time intensive and infeasible for current massively produced gene expression images. Novel prevention and therapeutic treatment for cartilage degradation is urgently called for, as cartilage degradation is a hallmark for arthritic diseases including osteoarthritis (OA) and rheumatoid arthritis (RA), and there is a high incidence of arthritis-related disability and high medical costs. While both underuse (e.g. physically inactive lifestyle) and overuse (e.g. high impact or intense repetitive joint use as seen in certain sports) are risk factors for cartilage degradation, recent studies highlight that dynamic moderate loading is associated with reduced incidence of developing OA. Parkinson Disease (PD) is a debilitating and progressive movement disorder that is estimated to affect over six million worldwide. One of the most characteristic symptoms of PD is resting tremor, which involves unintentional and rhythmic muscle oscillations of an afflicted extremity while the muscles of said extremity are relaxed. When compared to traditional surgery, laparoscopic procedures result in better patient outcomes: shorter recovery, reduced post-operative pain, and less trauma to incisioned tissue. Unfortunately, laparoscopic procedures require specialized training for surgeons, as these minimally-invasive procedures provide an operating environment that has limited dexterity and limited vision. Advanced surgical robotics platforms can make minimally-invasive techniques safer and easier for the surgeon to complete successfully. For this project, the design, implementation, characterization, calibration and possible applications of a multiple transducer stethoscope apparatus were investigated. The multi-transducer sensor array design consists of five standard stethoscope diaphragms mounted to a rigid frame for a-priori knowledge of their relative spatial locations in the x-y plane, with compliant z-direction positioning to ensure good contact and pressure against the subject’s skin for reliable acoustic coupling. In this thesis, a machine learning based method is proposed to predict the putt outcomes of golfers based on their electroencephalogram (EEG) data. The method can be used as a core building block of a brain-computer interface, which is designed to provide guidance to golf players based on their EEG patterns. The proposed method includes three steps. First, multi-channel 1-second EEG trials were extracted during golfers' preparation of putting. A silent speech interface (SSI) maps articulatory movement data to speech output. Although still in experimental stages, silent speech interfaces hold significant potential for facilitating oral communication in persons after laryngectomy or with other severe voice impairments. Despite the recent efforts on silent speech recognition algorithm development using offline data analysis, online test of SSIs have rarely been conducted. In this paper, we present a preliminary, online test of a real-time, interactive SSI based on electromagnetic motion tracking. This project details the design development of an alternative computer input system that allows a person with quadriplegia to move a computer's cursor and activate left and right click button inputs. After researching and analyzing possible solutions, an end design was chosen that most appropriately satisfied all user requirements and engineering specifications. There are currently no inexpensive assistive devices that allow the visually impaired detect low hanging objects that are above waist height and may cause injury, such as a rope or tree branch. The challenge presented to the team by the Quality of Life Plus Lab (QL+) was to develop a Sonar Cane device that attaches to a traditional white cane to address this problem. This device must be lightweight, inexpensive to manufacture, have a long battery life, and accurately detect obstacles to prevent user injury.

Bipolar pulse generation has been studied using transmission line technology, which was mainly used for monopolar pulse generation. In this thesis, low impedance, high current, nanosecond pulse generators were designed, constructed and tested. Five transmission lines were connected in parallel, so the impedance was 10 n. The transmission lines were switched by a spark gap switch operated in air. Opening of long-lived pores in the cell membrane is the principal primary effect of intense, nanosecond pulsed electric field (nsPEF). Here we demonstrate that the evolution of pores, cell survival, the time and the mode of cell death (necrotic or apoptotic) are determined by the level of external Ca2+ after nsPEF. We also introduce a novel, minimally disruptive technique for nsEP exposure of adherent cells on indium tin oxide (ITO)-coated glass coverslips, which does not require cell detachment and enables fast exchanges of bath media. This fourth special electroporation-based technologies and treatments issue of the Journal of Membrane Biology contains reports on recent developments in the field of electroporation by participants in the 7th International Workshop and Postgraduate Course on electroporation based technologies and treatments (EBTT 2013) held in Ljubljana, November 17–23, 2013. The 65 participants included faculty members, invited lecturers, special guests, and young scientists, and students from 16 countries. In addition to lectures on the fundamentals, this year’s sessions included talks on microbial inactivation by pulsed electric fields, modeling of intracellular electroporation, electroporation in food processing, and electrotransfer-facilitated DNA vaccination.

Nuclei/Cell detection is usually a prerequisite procedure in many computer-aided biomedical image analysis tasks. In this thesis we propose two automatic nuclei/cell detection frameworks. One is for nuclei detection in skeletal muscle fiber images and the other is for brain tumor histopathological images.
For skeletal muscle fiber images, the major challenges include: i) shape and size variations of the nuclei, ii) overlapping nuclear clumps, and iii) a series of z-stack images with out-of-focus regions. The atmospheric pressure dielectric barrier discharge in helium is a pulsed discharge in nature and the moment of maximum species densities is almost consistent with peak discharge current density. In this paper, a one-dimensional fluid model is used to investigate the temporal structure of plasma species in an atmospheric He-N2 dielectric barrier discharge (DBD). It is demonstrated that there exist microsecond delays of the moments of the maximum electron and ion densities from the peak of discharge current density.
