This project developed and evaluated an optical sensing system for detecting changes associated with glucose concentration. The system combined a laser-diode, cuvette sample holder, photodiode, resistive-feedback transimpedance amplifier, high-resolution analog-to-digital converter, and microcontroller. Parametric testing evaluated the effects of input current and feedback resistance on transimpedance gain, output range, and linearity. Firmware was developed to configure the ADC, average repeated conversions, monitor measurement variation, convert raw digital counts into voltage using a source-meter calibration equation, and compare sample measurements with a water reference. A cuvette enclosure maintained alignment between the laser-diode, sample, and photodiode while reducing external optical interference. Testing …

This thesis presents a comprehensive framework for the automated tracking and visualization of articulatory movements based on magnetic resonance imaging (MRI) data. A well-known data analysis tool for markerless pose estimation, known as DeepLabCut, is investigated for this purpose. The performance of this tool is enhanced through the design and implementation of a pre-processor. DeepLabCut is a markerless pose estimation toolbox based on deep learning, which overcomes the issue of making manual annotations frame-by-frame. Limitations from manually marking the MRI images are addressed by implementing transfer learning with convolutional neural networks to achieve accurate, user-defined articulator tracking without markers. With the continued growth of the biopharmaceutical industry, the demand for scalable, robust, and resource-efficient platforms for large-scale mammalian cell culture is amplified. Recent developments in microfluidic technology, such as precise control of the microenvironment, showed the potential to improve the performance of cell culture systems. However, constrained by scalability and operational efficiency, applying such approaches to large-scale cell culture and biopharmaceutical production presents challenges. This dissertation addresses these challenges through three independent but conceptually related technological developments. This study investigates the relationship between electrical stimulus characteristics of shape and charge on evoked sensations and electroencephalogram (EEG) data during multi-waveform transcutaneous electrical nerve stimulation (TENS) of the median nerve. Neuromodulation methods have traditionally had little control over the location and quality of their associated evoked sensation (e.g., electric, vibration, touch). This experiment utilized five unique stimulus waveforms during TENS stimulation. EEG data were collected concurrently to provide an introductory objective measure of the neural responses underlying these sensory changes. Every second, human skin processes over one million sensory signals while maintaining properties such as electrical conductivity, mechanical adaptability, and regenerative capability that surpass all synthetic materials. Contemporary bioelectronic devices prove inadequate when contacting skin surfaces due to poor adhesion and electrical contact issues that prevent effective sensing. Despite advancements in wearable and bioelectronic technologies, current devices face major drawbacks when interfacing with human skin, particularly in maintaining firm adhesion, conformability, and low-noise electrical signal acquisition.

Properly designed prosthetics hands can enhance the quality of life for those suffering from limb loss. Recently, 3D-printed prosthetic hands are becoming common. In these prostheses, fingers flex through the tendons that are activated by motion of the wrist. To provide spring action, thermoplastic polyurethane (TPU) hyperelastic joints are used to connect digits to each other as well as to the wrist. Point-of-care (POC) electric field-based biosensors have emerged as a promising tool to detect early cancer biomarkers such as circulating tumor DNA (ctDNA), microRNA (miRNA), and proteins. To be effective in screening in clinical settings, these biosensors must be simple and easy to use. In this study, we have studied the manipulation of short DNA molecules suspended in a sessile drop to achieve this goal. Concerted binocular coordination evoking oculomotor and refractive responses to visual stimuli are essential to daily function. Oculomotor dysfunctions can inhibit binocular responses to visually-near stimuli and have high comorbidities to accommodative dysfunctions. Three visual cues for inward (convergent) and outward (divergent) oculomotor movements, when presented concertedly create natural-viewing conditions: disparity- the binocular difference in light cast onto the fovea due to differing ocular perspectives, blur- the acuity of a visual target which stimulates accommodation, and proximal- the perceived distance of a visual stimuli based on size.

Background: Dopamine plays a critical role in various essential functions, including motor control, hormone regulation, cognition, learning, and the reward system. In healthy individuals, dopamine levels are extremely low, with concentrations ranging from 0 to 0.25 nM in blood and 0.3 to 3.13 µM in urine. Abnormal levels are linked to disorders like Parkinson’s, schizophrenia, Alzheimer’s, epilepsy, hypertension, and arrhythmia. Abnormal dopamine levels can indicate the presence of certain cancers. Conversational Voice User Interfaces (CVUIs) are widely used in commercial applications such as personal assistants. CVUIs are beneficial for most users as they enable interaction through speech and natural language. However, recent studies indicate that underrepresented user groups, such as individuals with speech impairments and specifically those with Down syndrome, face challenges in using voice commands to control CVUIs. Evaluating the effectiveness of transcranial direct current stimulation (tDCS) is essential for guiding its integration into therapeutic and performance-enhancement applications. In our laboratory, we investigate the efficacy of tDCS across multiple experimental models, including both animal and human studies. I have contributed significantly to the execution and analysis of these experiments, which include studies in rats and healthy human participants aimed at evaluating whether electrical stimulation of the motor cortex can enhance motor learning. Traumatic brain injury (TBI) is a major cause of neurological impairment, often leading to variable recovery and uncertain prognosis in the neurocritical care setting. There is a pressing clinical need for robust, physiologically grounded biomarkers to inform prognosis and therapeutic decision-making in acute TBI. This thesis investigates neurovascular coupling (NVC), the physiological coordination between neuronal activity and cerebral blood flow, as a candidate biomarker for brain function and recovery after injury.

Cochlear implant surgery is a delicate procedure performed by Otolaryngologists (ENTs) to implant an electronic device into the inner ear to provide a sense of sound for people who are profoundly deaf or hard of hearing. The current practices of training involve cadavers and 3D-printed models. Seismocardiography (SCG), the measurement of cardiac-induced vibrations at the chest surface, has shown potential clinical utility. Improving the reliability of detecting fiducial points of electrocardiography (ECG) and SCG, which collectively capture the electro-mechanical cardiac activities, could expand ECG/SCG utility as a low-cost, accessible tool for clinical assessment. Biofeedback training for box breathing is becoming increasingly accessible due to advancements in consumer-grade breathing sensors. However, there is limited research on their design and applications for specialized populations. This study evaluates a novel biofeedback holographic game, EtherealBreathing, designed to support autistic children. In EtherealBreathing, children practice box breathing to collect virtual elements to maintain the Earth's balance, using a wearable sensor to measure chest expansion for breath detection. A deployment study with 20 autistic children revealed that EtherealBreathing effectively promotes box breathing, leading to better health-related outcomes, such as lowering participants’ heart and respiratory rates than traditional practices. Humans, as bipedal locomotors, are effective at reducing the mechanical cost of transport (CoTmech) by adopting movement strategies and gaits that minimize energy expenditure for a given distance. By using different gaits at different speeds, leveraging their long spring-like tendons and muscle elasticity which store and release energy during movement, humans reduce the mechanical effort required for locomotion. Current locomotion solutions offered in bipedal robots, based on legacy walking and running gait models, are not great at energy efficiency unless walking at very low speeds. Alzheimer’s disease (AD) is a progressive neurodegenerative disease with the clinical symptoms of gradual cognitive decline and memory loss. Biologically, abnormal accumulation of Aβ (Aβ) plaque and tau tangles in the brain are the core pathological hallmarks in AD. Although inflammatory response is not specific to AD, emerging evidence suggests that the disrupted innate immune system within the central nervous system, particularly microglial activation, might act as the bridge linking Aβ aggregation to pathological phosphorylation and aggregation of tau. The Microphysiological Systems Laboratory aims to develop colorectal cancer tumor models under a hypoxic environment to assess model response to pharmaceutical compounds in vitro. To perform relevant studies, researchers have attempted to use different hypoxic inducing strategies such as a nitrogen pod and hypoxic incubator to recreate in vivo physiological responses to hypoxia. This dissertation aimed to (1) quantify morphological variations in the pediatric hip joint and (2) evaluate the sensitivity of an infant musculoskeletal model (MSM) to these variations, considering hip joint center estimation errors. A shape statistical model (SSM) of decedent infant femurs from the Ortolani collection was created using ShapeWorks, capturing key morphological features, such as variations in the femoral neck-shaft and anteversion angles. Seven synthetic femurs were generated from the SSM to create SSM-informed MSMs, which were systematically evaluated through kinematics and kinetics analyses in OpenSim. In electrocardiograms (ECGs), multiple forms of encryption and preservation formats create difficulties for data sharing and retrospective disease analysis. Additionally, photography and storage using mobile devices are convenient, but the images acquired contain different noise interferences. To address this problem, a suite of novel methodologies was proposed for converting paper-recorded ECGs into digital data. Firstly, this study ingeniously removed gridlines by utilizing the Hue Saturation Value (HSV) spatial properties of ECGs.
