This study investigates the relationship between electrical stimulus characteristics of shape and charge on evoked sensations and electroencephalogram (EEG) data during multi-waveform transcutaneous electrical nerve stimulation (TENS) of the median nerve. Neuromodulation methods have traditionally had little control over the location and quality of their associated evoked sensation (e.g., electric, vibration, touch). This experiment utilized five unique stimulus waveforms during TENS stimulation. EEG data were collected concurrently to provide an introductory objective measure of the neural responses underlying these sensory changes. Eleven participants completed three tasks (thresholding, super-threshold stimulation, two-alternative forced choice) using a two-electrode TENS approach. Stimulus waveforms were …

Evaluating the effectiveness of transcranial direct current stimulation (tDCS) is essential for guiding its integration into therapeutic and performance-enhancement applications. In our laboratory, we investigate the efficacy of tDCS across multiple experimental models, including both animal and human studies. I have contributed significantly to the execution and analysis of these experiments, which include studies in rats and healthy human participants aimed at evaluating whether electrical stimulation of the motor cortex can enhance motor learning. These studies assess improvements in fine motor performance resulting from tDCS. In stroke patients, I contribute to our investigation of tDCS as a rehabilitative intervention, particularly …

Traumatic brain injury (TBI) is a major cause of neurological impairment, often leading to variable recovery and uncertain prognosis in the neurocritical care setting. There is a pressing clinical need for robust, physiologically grounded biomarkers to inform prognosis and therapeutic decision-making in acute TBI. This thesis investigates neurovascular coupling (NVC), the physiological coordination between neuronal activity and cerebral blood flow, as a candidate biomarker for brain function and recovery after injury.

Fast Scan Cyclic Voltammetry (FSCV) is a widely used electrochemical technique for real-time measurement of the brain’s chemical messengers, including the molecule dopamine, with high temporal resolution. Currently the financial burden of performing FSCV is quite high, ranging from $8,000 to $20,000+ making the barrier to entry nearly insurmountable for laboratories and classrooms at small institutions. However, many standardized assessment tools rely on human raters for performance assessment, which is resource-intensive and subjective. Dental implantation is an increasingly common procedure used to treat missing teeth. However, surgical drilling to place the implant poses a high risk of injury to critical anatomy, such as inferior alveolar nerve injury or maxillary sinus perforation. A real-time surgical feedback system sensing proximity of these critical anatomy could reduce injury risks. This dissertation investigates the development of such a system that incorporates an electrical impedance sensor into the tip of a surgical drill.

Significant progress has been made toward model-based prediction of neral tissue activation in response to extracellular electrical stimulation, but challenges remain in the accurate and efficient estimation of distributed local field potentials (LFP). Analytical methods of estimating electric fields are a first-order approximation that may be suitable for model validation, but they are computationally expensive and cannot accurately capture boundary conditions in heterogeneous tissue. Nanosecond pulsed electric fields (nsPEFs) offer a plethora of opportunities for developing integrative technologies as complements or alternatives to traditional medicine. Studies on the biological effects of nsPEFs in vitro and in vivo have revealed unique characteristics that suggest the potential for minimized risk of complications in patients, such as the ability of unipolar nsEPs to create permanent or transient pores in cell membranes that trigger localized lethal or non-lethal outcomes without consequential heating. My research focuses on electrokinetic transport. Particularly, in this dissertation, we focus on fabrication and testing of micro electrodes with nanostructured surfaces to minimize the electrode polarization (EP) effects for biosensor applications. In the first study, electrochemical deposition of gold nanoparticles on to planar gold electrodes was used to generate rough surfaces. Dendritic nanostructures that reduced EP up to two orders of magnitude was obtained by optimizing the deposition conditions. These structures also enhanced dielectrophoresis (DEP) response of our bio-chips, making them usable in physiological buffers. Studies have shown that the activity of muscarinic receptors and their affinity to agonists are sensitive to membrane potential. It was reported that in airway smooth muscle (ASM) depolarization evoked by high K+ solution increases contractility through direct effects on M3 muscarinic receptors. In this study, we assessed the physiological relevance of voltage sensitivity of muscarinic receptors on ASM contractility. Our findings reveal that depolarization by high K+ solution induces contraction in intact mouse trachea predominantly through activation of acetylcholine release from embedded nerves, and to a lesser extent by direct effects on M3 receptors. Electrophysiological measurements have been used in recent history to classify instantaneous physiological configurations, e.g., hand gestures. This work investigates the feasibility of working with changes in physiological configurations over time (i.e., longitudinally) using a variety of algorithms from the machine learning domain. We demonstrate a high degree of classification accuracy for a binary classification problem derived from electromyography measurements before and after a 35-day bedrest. The problem difficulty is increased with a more dynamic experiment testing for changes in astronaut sensorimotor performance by taking electromyography and force plate measurements before, during, and after a jump from a small platform. The Anterior Cruciate Ligament (ACL) is the primary source of rotational stability in the knee by preventing the tibia from sliding in front of the femur. When the ACL is torn, it typically must be repaired through reconstructive surgery which results in proprioceptive deficiencies in the knee. Proprioception plays an important role in understanding where one’s knee is in space, sensing movement and reacting accordingly. This study examines an alternative method of measuring proprioceptive responses to a stimulus (motion) by using electromyogram (EMG) and electroencephalogram (EEG) signals to observe muscle and brain activity. Alzheimer’s disease (AD) is a chronic and progressive, irreversible syndrome that deteriorates the cognitive functions. Official death certificates of 2013 reported 84,767 deaths from Alzheimer’s disease, making it the 6th leading cause of death in the United States. The rate of AD is estimated to double by 2050. The neurodegeneration of AD occurs decades before symptoms of dementia are evident. Therefore, having an efficient methodology for the early and proper diagnosis can lead to more effective treatments.

Functional magnetic resonance imaging (fMRI) and electroencephalography (EEG) are useful tools to analyze brain activities given active stimulation. However, the electromagnetic noise from the MRI distorts the brain signal recording and damages the subject with excessive heat generated on the electrodes attached to the skin. MRI-compatible recording and stimulation systems previously developed at LIBI lab were capable of removing the electromagnetic noise during the imaging process. Previously, the hardware systems had required the integrative software that could control both circuits simultaneously and enable users to easily change recording and stimulation parameters. Electroencephalogram (EEG) has been used in a wide array of applications to study mental disorders. Due to its non-invasive and low-cost features, EEG has become a viable instrument in Brain-Computer Interfaces (BCI). These BCI systems integrate user's neural features with robotic machines to perform tasks. Hydrocephalus is a neurological disorder that typically requires a long-term implantation of a shunt system to manage its symptoms. These shunt systems are notorious for their extremely high failure rate. More than 40% of all implanted shunt systems fail within the first year of implantation. On average, 85% of all hydrocephalus patients with shunt systems undergo at least two shunt-revision surgeries within 10 years of implantation. A large portion of this high failure rate can be attributed to biofouling-related obstructions and infections. Previously, we developed flexible polyimide-based magnetic microactuators to remove obstructions formed on hydrocephalus shunts. To test the long-term …
