[Objective ] This study aims to employ synthetic biology and metabolic engineering techniques to engineer Pichia pastoris (P.pastoris ) for the efficient heterologous synthesis of the UV resistant compound gadusol.[Method ] On the basis of a bioinformatic analysis and codon optimization of the genes involved in the gadusol biosynthetic pathway in Danio rerio,the gadusol fermentation pathway was constructed in P.pastoris via strategies such as promoter engineering,metabolic engineering,and fermentation optimization.[Result ] A high-purity gadusol standard was prepared through optimized extraction and chromatographic separation.The gadusol yield in the recombinant strain P.pastoris HME reached 105 mg/L.By screening of gene expression promoters and optimization …

[Objective ] This study aims to improve the utilization of maize resources and address the shortage of feed resources.[Method ] The intestinal colonization capacity,tolerance,and antibacterial properties of three Lactobacillus strains,Lactobacillus paracasei ZB- 1 (ZB-1),L.paracasei DY- 2 (DY-2),and L.rhamnosus DY- 4 (DY-4) were evaluated.The most suitable strain for fermenting cornhusks was identified,and its fermentation process was optimized.[Result ] ZB-1 exhibited higher intestinal colonization ability and antibacterial properties than DY- 2 and DY- 4.The antibacterial mechanism of ZB- 1 was likely driven by the action of organic acids in an acidic environment and certain protein-based antimicrobial substances.The fermentation conditions for ZB- 1 …

[Objective ] This study aims to compare the effects of hesperetin and eriocitrin on the gut microbiota in different enterotypes.[Method ] An in vitro gut fermentation model was established,which was combined with 16S rRNA high-throughput sequencing and gas chromatography-mass spectrometry (GC-MS ) technique to evaluate the regulatory effects of the two flavonoids on the gut microbiota in different enterotypes.[Result ] After fermentation,both flavonoids significantly reduced the Firmicutes-to-Bacteroidota ratio (F/B ratio ) in terms of relative abundance at the phylum level in enterotype Bacteroides and enterotype Prevotella.At the genus level,pro-inflammatory bacteria including Paeniclostridium and Sutterella showed decreased relative abundance in treatment …

[Objective ] The effects of different extraction methods on the yield,purity,structural characteristics,and physicochemical properties of tortoise shell collagen were investigated.[Method ] The ultrasound-assisted enzymatic method,acid method,and enzymatic method were used to extract collagen from tortoise shell powder.[Result ] The yield and purity of tortoise shell collagen extracted by ultrasound-assisted enzymatic method were 2.29% and 85.65%,respectively,significantly higher than those extracted by acid method (0.78% and 73.15%) and enzymatic method (1.77% and 75.63%).Ultraviolet absorption spectroscopy,infrared absorption spectroscopy,circular dichroism spectroscopy,and electrophoresis profiles showed that collagens extracted from tortoise shell by three methods were all type I collagen and retained intact triple helix structures.X-ray …

Black youth in the United States disproportionately experience fatal drowning at rates up to five times higher than their white peers. Eliminating these inequities requires new approaches that address historical and structural barriers of learning to swim. The objective of this study was to evaluate the three-week long swimming component of a community-based discovery program to empower Black youth in Evanston, IL. Participants were Black youth ages 8-10 years enrolled in summer 2023 and 2024. Swim skills were assessed by instructors using the Pictorial Scale of Perceived Water Competence (PSPWC). The silent epidemic of childhood drowning, and the racial disparities associated with it require scalable and innovative approaches that both break cycles of exclusion and circumvent our societal tendency to overlook drowning as a matter in need of urgent resolution. No More Under, a Seattle-based water safety organization whose mission is to eradicate childhood drowning, has launched the Swim Safely Series. The series is a national program uniquely poised to overcome the various barriers associated with racial and socioeconomic drowning disparities by tapping into underutilized collegiate aquatic spaces and merging water safety with mentorship and social intervention.

Plant transformation efficiency is highly dependent on species, individual genotypes, and tissue types. In maize, immature embryos are regularly used for transformation. The process relies heavily on callus development, as it is intricately associated with somatic embryogenesis and subsequent plant regeneration, both of which directly affect transformation efficiency. Immature embryos of the segregation progeny derived from the two inbred parents, a transformation-amenable line A188 and a recalcitrant line B73, can be cultured to form two primary callus types: Type I and Type II. Northern hardwood forests of the Lake Superior region face a series of novel disturbance pressures including canopy dieback. Previous studies have linked regional sugar-maple (Acer saccharum) canopy dieback to introduced earthworms, which may have coinciding impacts on the ground-layer plant community. Dieback–earthworm interactions may lead to important longer-term changes in forest structure and function, but these relationships but have not been characterized. We sampled ground-layer plant communities in five national forest units in Michigan, Wisconsin, and Minnesota in 2010, and again just over a decade later in 2021. Background: The vast amount of natural language clinical notes about patients with cancer presents a challenge for efficient information extraction, standardization, and structuring. Traditional NLP methods require extensive annotation by domain experts for each type of named entity and necessitate model training, highlighting the need for an efficient and accurate extraction method.

Methods: This study introduces a tool based on the Large Language Model (LLM) for zero-shot information extraction from cancer-related clinical notes into structured data aligned with the minimal Common Oncology Data Elements (mCODE™) structure. Human speech perception is multisensory, integrating auditory information from the talker's voice with visual information from the talker's face. BOLD fMRI studies have implicated the superior temporal gyrus (STG) in processing auditory speech and the superior temporal sulcus (STS) in integrating auditory and visual speech, but as an indirect hemodynamic measure, fMRI is limited in its ability to track the rapid neural computations underlying speech perception. Purpose: Purine metabolism is a promising therapeutic target in cancer; however, how cancer cells respond to purine shortage, particularly their adaptation and vulnerabilities, remains unclear.

Experimental design: Using the recently developed purine shortage-inducing prodrug DRP-104 and genetic approaches, we investigated the responses in prostate, lung, and glioma cancer models.

Results: We demonstrate that when de novo purine biosynthesis is compromised, cancer cells employ microtubules to assemble purinosomes, multiprotein complexes of de novo purine biosynthesis enzymes that enhance purine biosynthesis efficiency. Patients with non–small cell lung cancer (NSCLC) with loss of the tumor suppressor gene STK11 are resistant to immune checkpoint therapies like anti–PD-1. In this study, we conducted an in vivo CRISPR screen that identified histone deacetylase 1 as a target to reverse anti–PD-1 resistance driven by loss of STK11 and developed TNG260, a potent small-molecule inhibitor of the CoREST complex with selectivity exceeding previously generated inhibitors in this class in preclinical studies. Treatment with TNG260 led to increased expression of immunomodulatory genes in STK11-deficient cancer cells. When operating on gliomas near critical language regions, surgeons risk either leaving residual tumor or inducing permanent postoperative language deficits (PLDs). Despite the advent of intraoperative mapping techniques, subjective judgments frequently determine important surgical decisions. We aim to inform data-driven surgery by constructing a non-invasive mapping approach that quantitatively predicts the impact of individual surgical decisions on long-term language function. This study included 79 consecutive patients undergoing resection of language-eloquent gliomas. Phytophthora species are of global importance (Sinclair and Lyon, 2005). For most Phytophthora species, the natural mechanism of how these pathogens travel and spread long distances is not fully understood. They are have been shown to travel by air, water, nursery stock, and via inadvertent human transport. To combat the growing issue of petroleum plastic waste, alternative bio-based polymers are being developed. Many of these biopolymers are made from bio-derived materials, or are biodegradable, but the most promising polymers fall in both categories. Polyhydroxyalkanoates (PHAs) are one such class of polymers, and poly-3-hydroxybutyrate (P3HB), the most popular PHA, has shown great potential. This study utilized two types of cotton-derived glucose, alongside commercial glucose, as a feedstock for the biosynthesis of P3HB by Cupriavidus necator (also known as Ralstonia eutropha). Despite the incremental increase in the number of Black people who are learning to swim, the number of Black divers has not kept pace. In the NCAA Database, swimmers and divers were not segmented; thus, they all appear in the category of “swimming” by race and sex. When viewing the 2024 Division I NCAA Demographics Database, 89 (.02%) of 4307 student-athletes participating in swimming were Black women. For men, the data is equally dismal, revealing that 81 (.03%) of 2,608 student-athletes who were identified as swimmers were Black. The public is paying increased attention to the environmental impacts associated with cattle production systems in the US. Thus, cattle producers are increasingly exploring ways to communicate their efforts to reduce those environmental and climate impacts. This study uses online and social media data from 2018 to 2022 to examine public interest in and sentiment about beef and dairy cattle production when discussed within the context of greenhouse gas (GHG) emissions. Neoadjuvant chemotherapy (NAC) is a critical component of breast cancer treatment, but the molecular mechanisms underlying resistance remain poorly understood. This study aimed to identify transcriptomic changes associated with NAC resistance across four breast cancer subtypes: Luminal A, Luminal B/HER2-positive, Luminal B/HER2-negative, and Triple-Negative Breast Cancer (TNBC). RNA-seq analysis was performed on paired pre- and post-NAC breast cancer samples from 32 nonresponders. Differentially expressed genes (DEGs) were identified, and functional enrichment analyses were conducted. Protein-protein interaction (PPI) networks were constructed to identify hub genes. Objectives: To evaluate the antibacterial efficacy of Ginger, Resin Gum, and Nigella Sativa, against Enterococcus Faecalis.

Methods: Thirty-eight extracted single-rooted premolars were inoculated with E.faecalis, except the negative control, and incubated for 4 weeks. After instrumentation samples were divided into 4 groups (n=7) according to final irrigation: NaOCl+EDTA (Group A), Nigella sativa (B), Resin Gum (C), Ginger (D), plus 2 control groups (n=5): Saline (E), and Saline without bacterial inoculation (F). Cultures were collected at baseline and after irrigation.

RAS pathway (RASp) mutations induce proliferative features, and promote transformation in chronic myelomonocytic leukemia (CMML). However, the unique clonal landscape and hierarchy of distinct RASp mutations remain unexplored. To characterize the landscape, architecture, and implications of unique RASp mutations in CMML, we evaluated a cohort of 814 patients with CMML. We identified 461 RASp mutations among 342 patients (42%). N/KRAS and CBL mutations were the most common, frequently involved the P-loop or RING domains, respectively, and frequently appeared as dominant events (63% and 65%, respectively). Nongenetic transcription evolution has been increasingly explored and recognized to drive tumor cell progression and therapeutic resistance. As the regulation hub of transcription machinery, cyclin-dependent kinase 9 (CDK9) is the gatekeeper of RNA polymerase II transcription, and CDK9 dysfunction results in transcriptomic reprogramming and tumor cell progression. We recently reported that the heat shock protein 90 (HSP90)-MYC-CDK9 network drives therapeutic resistance in mantle cell lymphoma (MCL) through transcriptomic reprogramming. Background: Genetic ancestry is an important factor to account for in DNA methylation studies because genetic variation influences DNA methylation patterns. One approach uses principal components (PCs) calculated from CpG sites that overlap with common SNPs to adjust for ancestry when genotyping data is not available. However, this method does not remove technical and biological variations, such as sex and age, prior to calculating the PCs. This study experimentally investigates the dynamic damping performance of passive tuned liquid column vibration absorbers (TLCVAs), focusing on the influence of vertical column inclination angle and orifice disc configuration. Free vibration tests were conducted on a primary system equipped with a TLCVA using a custom-designed test rig.
