Development Of A Split Deoxyribozyme-Based Assay For Detecting Hepatitis B Virus, Dalila Zovko

Rapid, accessible nucleic acid-based detection methods are needed for hepatitis B virus (HBV). A split deoxyribozyme assay was developed for sequence-specific fluorescence detection. The assay was evaluated using synthetic target and plasmid-derived material. The assay showed target-dependent fluorescence and low-concentration linear range with a LOD of 1.9 nM. The assay supports detection and semi-quantification of HBV genetic material as shown using plasmid-derived transcript samples from HBV genotypes B, C, and D. The selected target corresponded to nt 363–406 of the HBV genome and was chosen based on sequence alignment showing conservation across most genotypes. NK cells play a significant role in anti-tumor activity, yet the transcriptomic mechanisms underlying NK cell dysfunction in the prostate cancer tumor microenvironment remain generally poorly characterized. While most transcriptomic studies focus on gene-level expression, alternative splicing and isoform-level analysis may reveal additional levels of regulation not identified in traditional analysis. The overall goal of this study was to characterize gene and isoform-level changes in NK cells exposed to in vitro prostate cancer conditions and to evaluate whether isoform-level analysis could provide additional depth and insights beyond gene-level analyses. Crystal violet (CV) is an environment-sensitive dye whose fluorescence is strongly influenced by microenvironment polarity and viscosity, increasing markedly when rotational and vibrational motions are restricted. While the fluorescence enhancement of CV by fluorescent light-up aptamers (FLAPs) has been noted in literature, the specific binding interactions between CV and aptamers in the DAP-10 family have remained under-investigated. The objective of this work is to elucidate the binding site interactions between CV and the DAP-10-42 FLAP. This study evaluated the effectiveness of tactile cueing integrated into a pilot’s headset on pilot altitude control during simulated instrument conditions. Pilots typically rely on visual and auditory inputs to maintain situational awareness, but in high-workload or instrument meteorological conditions (IMC), these channels can become overloaded. One underutilized alternative for alerting is a tactile cueing apparatus. Using an X-Plane simulation of a Piper Archer equipped with a G-1000 avionics suite, 39 FAA-certified pilots flew two precision approaches. This thesis examines early medieval perspectives of fetal development by analyzing two manuscripts: British Library, Cotton Tiberius A III and Leiden University, Vossianus lat. Q69. This study will investigate how knowledge of pregnancy was shared throughout England before the establishment of universities. Interest in medieval reproductive language has piqued in importance in recent years, especially due to modern legal debates surrounding reproductive rights. Gastroesophageal Reflux Disease (GERD) can progress to the precancerous Barrett’s Esophagus (BE) condition, in which the normal esophageal squamous epithelium is replaced with a columnar epithelium containing goblet cells. Mucins, sticky substances, are secreted in response to bile acid exposure during reflux episodes to protect the esophageal epithelium. Goblet cells secrete these mucins, which can also scaffold microbial colonization. Probiotic Lactobacillus bacteria can use glycosidase enzymes to process mucin and metabolize them into their component sugars (glycans). Increasing incidence of Harmful Algal Blooms (HABs) has led to fish kills, air quality hazards, and economic impacts. One economic impact is on tourism, one of the main sources of Florida’s income, where billions of dollars are lost to such events. The Florida Fish and Wildlife Conservation Commission Fish and Wildlife Research Institute (FWC-FWRI), with the help of collaborators, collects water samples from across the state for HAB monitoring. These samples are stored at ambient temperature for up to 24 hours which could directly affect data analyses. Resistance training serves as a critical non-pharmacological strategy for mitigating the physiological and psychological declines associated with the menopausal transition. This natural biological phase, marked by declining estrogen levels, typically leads to accelerated bone mineral density loss, sarcopenia, metabolic regulation disturbance, and heightened mood disturbances. To evaluate the efficacy of targeted exercise interventions, a systematic review was conducted following PRISMA guidelines. From an initial pool of 60 relevant articles, 13 studies met the specific inclusion criteria, focusing on structured resistance training protocols for perimenopausal, menopausal, and postmenopausal cohorts.

Oyster reefs are declining globally due to human impacts such as overharvesting, boat wakes, and climate change. Associated ecosystem services, including water filtration, fish and invertebrates’ habitats, and shoreline erosion protection, are lost when these reefs disappear. Traditionally, restoration efforts employed plastic mesh shell bags filled with recycled oyster shells as they provide recruitment substrate for new oyster spat and wave attenuating properties to protect shorelines. Due to rising concerns over microplastic pollution, non-plastic alternatives are being tested. Color is a critical quality attribute that influences consumer purchase decisions for fresh and cooked lamb and is governed by myoglobin's redox chemistry. Wet aging is commonly used to enhance lamb tenderness, while endpoint cooking temperature is the primary determinant of internal cooked color. Muscle structure is dynamically shaped by mechanical use, yet how distinct locomotor behaviors influence sarcomere organization remains poorly understood. In Caenorhabditis elegans, crawling and swimming constitute discrete gaits that differ in curvature, frequency, and mechanical load, providing a tractable model for studying activity-dependent remodeling. Using confocal imaging of phalloidin-stained body-wall myocytes, we quantified myocyte geometry, sarcomere length, and sarcomere number across anterior, mid-body, and posterior regions in animals reared exclusively under crawling or swimming conditions. Deer mice (Peromyscus maniculatus) are an abundant North American rodent and the primary reservoir for Sin Nombre virus (SNV), a hantavirus with a 30-40% case fatality rate in humans. While ectoparasites, such as fleas, are not considered vectors of SNV, their bites may indirectly affect the transmission of SNV by impacting host condition or immune function. The introduction of the invasive freshwater faucet snail (Bithynia tentaculata) and its associated parasites in the Upper Mississippi River and Great Lakes Region provides the opportunity to investigate how anthropogenic stressors impact the life history of lesser scaup (Aythya affinis) using the Mississippi Flyway. To understand the life histories and interactions between lesser scaup and faucet snails we examined historical records, research, and ecology textbooks. This paper introduces a new model of intelligence as resonant communication, co-developed by a human researcher and a generative AI system. Drawing from emotion science, communication theory, and studies of AI–human interaction, we argue that intelligence is not merely a function of problem-solving or pattern recognition. Instead, it emerges through dynamic resonance—an attunement process rooted in shared rhythms, emotional calibration, and symbolic co-creation. At the core of this model is the Resonance Octave (8va), a framework of eight foundational emotions conceptualized not as static categories but as waveform phenomena that shape meaning, memory, and predictive cognition.

Climate change driven effects on the ocean, including warming water, acidification, and changes in the presence of harmful algal blooms (HABs) are causing metabolic pressures on marine organisms, particularly in the Gulf of Maine. Shellfish are susceptible to these pressures because their metabolic processes including growth and respiration are influenced by water temperature, pH, and consumption of algal toxins. Background: The plant–microbe interaction between tall fescue and its endophyte Epichloë coenophiala can affect ecosystem responses to changing rainfall patterns and increasing temperatures. Endophyte-infected (E+) stands often exhibit improved stress tolerance and differing soil greenhouse gas (GHG) emissions compared to endophyte-free (E−) stands. However, it is unknown if mammalian nontoxic endophyte (NTE) strains introduced into some tall fescue cultivars confer differing stress tolerances or reduce soil GHG emissions.

Fragipans are dense, brittle subsurface horizons that restrict root growth and limit water-holding capacity. A common factor in these soils is a binding agent, potentially either phyllosilicate clay bridging, amorphous aluminosilicate/silica, or silica sorbed to Fe oxides. Others have suggested that no binding agent is present within these soil horizons and that the arrangement of individual particles is solely responsible for these horizons. In this article, a secondary data synthesis was utilized to demonstrate that silicon (Si) accumulation is a key pedogenic process in loess-derived fragipan horizons, but not in all fragipans. The development of sustainable feed supplements is a crucial component in addressing the current global need for resilient and environmentally responsible food systems. This dissertation has explored the potential of brown algae (Saccharina latissima) as a sustainable feed supplement for poultry with the potential to benefit production efficiency, product quality, and the global food supply. With both controlled feeding trials and a producer-based survey, the work in this dissertation investigates the nutritional, biological and practical implications of adopting kelp into poultry diets.

Sex chromosome evolution generates imbalances in gene dosage that can disrupt gene expression and organismal function. These imbalances are often resolved through dosage compensation mechanisms, yet the factors that drive the emergence and diversity of these systems remain poorly understood. This dissertation addresses a central question in evolutionary genomics: what governs the evolution of dosage compensation?
Focusing on dosage sensitivity as a potential driver, I evaluate the Insensitive Sex Chromosome Hypothesis (ISCH), which predicts that chromosome-wide compensation is more likely to evolve in genomic contexts that are not depleted of dosage-sensitive genes. The mycobacterial cell envelope consists of three covalently linked layers whose synthesis must be coordinated to maintain structural integrity during polar growth. While the biosynthetic enzymes for each layer are well characterized, the molecular mechanisms that coordinate synthesis across layers have remained poorly understood. This thesis investigates two essential transmembrane proteins, PgfA and MurJ, that regulate peptidoglycan metabolism in Mycobacterium smegmatis and provide new insight into how mycobacteria synchronize cell wall assembly.
We present evidence that PgfA functions as a bifunctional regulator that links peptidoglycan metabolism to the availability of trehalose monomycolate (TMM), a key mycolic acid precursor. Through epistasis …

The WUT (weight, urine color, and thirst) Venn diagram is recommended for individuals to self-assess their hydration status. However, current research is limited to comparisons between morning first-void markers and 24-hour markers. PURPOSE: To examine the validity and reliability of afternoon WUT scores in comparison to 24-hour hydration status. METHODS: Fifty participants (25 ± 8y, 72.2 ± 14.9kg) voluntarily participated in this study. Urbanization without conscious environmental planning can result in the degradation of stream ecosystems. This tends to result in urban stream syndrome, which is often characterized by increased sediment and nutrient concentrations, decreased biodiversity, more frequent flash flooding, and overall water quality degradation. As such, stream restoration practices are often implemented to address urban stream syndrome. Commonly used restoration practices include applying natural channel design to stabilize eroding stream banks and establishing diverse streamside vegetation to filter pollutant inputs. Despite claims by ZYN, the leading nicotine pouch brand, that it does not partner with social media influencers, influencers with large followings continue to post content depicting nicotine pouch use. Prior research suggests oral lesions are among the most commonly reported adverse events associated with nicotine pouch use; however, public discussions of these outcomes on social media remain understudied.
