Journal of Arid Land

Studies on the ecosystem service value (ESV) of gardens are critical for informing evidence- based land management practices based on an understanding of the local ecosystem. By analyzing equivalent value factors (EVFs), this paper evaluated the values of 11 ecosystem services of gardens in the Yellow River Basin of China in 2019. High-precision land use survey data were used to improve the accuracy of the land use classification, garden areas, and spatial distribution of the ESVs of gardens. The results showed that garden ecosystem generally had high ESVs, especially in terms of the ESV of food production, which is worthy …

Neural Substrates Of Homing Pigeon Spatial Navigation: Results From Electrophysiology Studies, Gerald E. Hough Apr 2022

Neural Substrates Of Homing Pigeon Spatial Navigation: Results From Electrophysiology Studies, Gerald E. Hough

College of Science & Mathematics Departmental Research

Over many centuries, the homing pigeon has been selectively bred for returning home from a distant location. As a result of this strong selective pressure, homing pigeons have developed an excellent spatial navigation system. This system passes through the hippocampal formation (HF), which shares many striking similarities to the mammalian hippocampus; there are a host of shared neuropeptides, interconnections, and its role in the storage and manipulation of spatial maps. There are some notable differences as well: there are unique connectivity patterns and spatial encoding strategies. This review summarizes the comparisons between the avian and mammalian hippocampal systems, and the …

Crispr-Mediated Multiplexed Live Cell Imaging Of Nonrepetitive Genomic Loci With One Guide Rna Per Locus., Patricia A Clow, Menghan Du, Nathaniel L. Jillette, Aziz Taghbalout, Jacqueline J Zhu, Albert Cheng

Faculty Research 2022

Three-dimensional (3D) structures of the genome are dynamic, heterogeneous and functionally important. Live cell imaging has become the leading method for chromatin dynamics tracking. However, existing CRISPR- and TALE-based genomic labeling techniques have been hampered by laborious protocols and are ineffective in labeling non-repetitive sequences. Here, we report a versatile CRISPR/Casilio-based imaging method that allows for a nonrepetitive genomic locus to be labeled using one guide RNA. We construct Casilio dual-color probes to visualize the dynamic interactions of DNA elements in single live cells in the presence or absence of the cohesin subunit RAD21. Using a three-color palette, we track …

Feasibility Of Tubulin As A Control For Gene Expression Following Transfection In Mouse Monocyte/Macrophage-Like Cells, Ankita Chabra

Honors Program Theses and Research Projects

Transfection, which is the ability to modify host cells’ genetic content, has broad application in studying normal cellular processes, molecular mechanism of disease and gene therapy. There are several transfection techniques, and all require either a control or a reference gene. Commonly used controls for transfection experiments are housekeeping genes, which maintain expression for a given cell/tissue, experimental conditions, and treatment. However, recent research has uncovered that expression levels of housekeeping genes may vary depending on the gene, cell type and experimental conditions. A growing body of evidence demonstrates that housekeeping genes are inadequate internal standards for measuring gene expression …

Farm Succession Planning: Why And How?, Allan Vyhnalek Apr 2022

Farm Succession Planning: Why And How?, Allan Vyhnalek

Cornhusker Economics

If you visit with an ag professional and ask them “What percentage of farmers and ranchers have an estate plan, or an up-to-date estate/succession plan?” the answer will be half, or just less than half. This article will examine why that happens and give some simple steps for anyone to follow to create an up-to-date estate or succession plan. Farmers and ranchers don’t have a plan for a variety of reasons. These might include not ever planning to retire, not wanting to give up control, or not wanting to admit that they will ever pass away. Too often, we don’t …

Lis1 And Ndel1 Regulate Axonal Trafficking Of Mitochondria In Mature Neurons, Jai P. Pandey, Liang Shi, Remi A. Brebion, Deanna S. Smith

Faculty Publications

Defective mitochondrial dynamics in axons have been linked to both developmental and late-onset neurological disorders. Axonal trafficking is in large part governed by the microtubule motors kinesin-1 and cytoplasmic dynein 1 (dynein). Dynein is the primary retrograde transport motor in axons, and mutations in dynein and many of its regulators also cause neurological diseases. Depletion of LIS1, famous for linking dynein deregulation to lissencephaly (smooth brain), in adult mice leads to severe neurological phenotypes, demonstrating post-developmental roles. LIS1 stimulates retrograde transport of acidic organelles in cultured adult rat dorsal root ganglion (DRG) axons but findings on its role in mitochondrial …

Rising obesity rates have become a major public health concern within the United States. Understanding the systemic and neural effects of obesity is crucial in designing preventive and therapeutic measures. In previous studies, administration of a high fat diet has induced significant weight gain for mouse models of obesity. Interestingly, sex differences in high-fat diet-induced weight gain have been observed, with female mice gaining significantly less weight compared to male mice on the same high-fat diet. It has also been observed that consumption of a high-fat diet can increase neurogliosis, but the mechanism by which this occurs is still not …

Adapt Lake Mead Releases To Inflow To Give Managers More Flexibility To Slow Reservoir Draw Down, David E. Rosenberg

Publications

A 20-year Colorado River drought continues and Lake Mead draws down. As Lake Mead falls through 8 elevation tiers to 1,020 feet (5.7 million acre-feet [maf]), releases drop and mandatory water conservation targets for California, Arizona, Nevada, and Mexico grow to 1.375 maf per year (USBR, 2019). How will different reservoir inflows, releases, and additional water conservation efforts beyond mandatory targets speed or slow Lake Mead draw down, stabilization, and recovery?

Lake Urmia, Iran Collaborations

Public concern over environmental issues such as ecosystem degradation is high. However, restoring coupled human-natural systems requires integration across many science, technology, engineering, management, and governance topics that are presently fragmented. Here, we synthesized 544 peer-reviewed articles published through September 2020 on the desiccation and nascent recovery of Lake Urmia in northwest Iran. We answered nine questions of scientific and popular interest about causes, impacts, stabilization, recovery, and next steps. We find: (1) Expansion of irrigated agriculture, dam construction, and mismanagement impacted the lake more than temperature increases and precipitation decreases. (2) Aerosols from Lake Urmia's exposed lakebed are negatively …

Genetic Characterization Of Resistance To Bacterial Panicle Blight And Sheath Blight In Rice Using Qtl Linkage Analysis And Qtl-Seq, John Christian E. Ontoy

LSU Master's Theses

Bacterial panicle blight (BPB) and sheath blight (SB) are major diseases of rice causing significant yield reduction in conducive conditions. Few cultivars have shown reliable resistance to these diseases. In this study, a recombinant inbred line (RIL) population derived from the U.S. cultivars Jupiter (moderately resistant) and Trenasse (susceptible) was investigated to identify genomic regions associated with BPB and SB resistance. Phenotypes in BPB and SB, as well as days to heading (DH), of the RIL population were evaluated in the field. DH was found to be correlated to BPB and SB diseases. BPB was also positively correlated to SB. …

Sex And Starvation Influences Latrotoxin Expression In The Brown Widow Spider, Mattie Harris Apr 2022

Sex And Starvation Influences Latrotoxin Expression In The Brown Widow Spider, Mattie Harris

Honors College Theses

Widow spiders (genus Latrodectus) are well-known for their potent venom. Seven latrotoxin proteins constitute the main components of widow spider venom. The vertebrate specific (α-latrotoxin) and insect specific (α-latroinsectotoxin) latrotoxins have been well-characterized with respect to structure and function. Regulation of latrotoxin gene expression is not well understood but sex and feeding could be factors influencing production. In this study, I used quantitative qPCR to (1) characterize the expression patterns of both the insect and vertebrate specific latrotoxins in male and female brown widow spiders (Latrodectus geometricus) to characterize sex-biased expression and to (2) study expression patterns …

The FGFR3-TACC3 (F3-T3) fusion gene was discovered as an oncogenic molecule in glioblastoma and bladder cancers, and has subsequently been found in many cancer types. Notably, F3-T3 was found to be highly expressed in both untreated and matched recurrence glioblastoma under the concurrent radiotherapy and temozolomide (TMZ) treatment, suggesting that targeting F3-T3 is a valid strategy for treatment. Here, we show that the F3-T3 protein is a client of heat shock protein 90 (HSP90), forming a ternary complex with the cell division cycle 37 (CDC37). Deprivation of HSP90 or CDC37 disrupts the formation of the ternary complex, which destabilizes glycosylated …

Over the past few decades, neuroimaging has become a ubiquitous tool in basic research and clinical studies of the human brain. However, no reference standards currently exist to quantify individual differences in neuroimaging metrics over time, in contrast to growth charts for anthropometric traits such as height and weight1. Here we assemble an interactive open resource to benchmark brain morphology derived from any current or future sample of MRI data (http://www.brainchart.io/). With the goal of basing these reference charts on the largest and most inclusive dataset available, acknowledging limitations due to known biases of MRI studies …

In order to facilitate hydrological restoration, initiatives have been conducted to promote tree growth in degraded and rewetted peatlands in Indonesia. For these initiatives to be successful, tree seedlings need to be able to survive flooding episodes, with or without shade. We investigated the survival rates and the formation of adventitious roots in the case of four tree species exposed to combinations of different shading and water levels under controlled conditions in a nursery, with artificial rainwater and with peat soil as the medium. The research focused on the following questions (i) whether trees can grow on flooded peat soils; …

Assessing The Influence Of Urban Greening On Urban Arthropods, Brendon Lawrence Apr 2022

Assessing The Influence Of Urban Greening On Urban Arthropods, Brendon Lawrence

Scholars Week

Green Heart is an urban greening experiment in Louisville, KY seeking to create new urban green spaces to ultimately increase air quality and improve human health. Changes in biodiversity in response to urban greening may influence human health via multiple hypothesized pathways including reducing harm (e.g. medicine provisioning), restoring capacities (e.g. reductions in stress), building capacities (e.g. increasing activity outdoors), and causing harm (e.g. zoonotic diseases). Arthropods are one component of diversity that may influence human health via each of these pathways. However, few studies have assessed the influence of Arthropod diversity on human health. Thus, the objective of our …

Movement Patterns Of The Shovelnose Guitarfish (Pseudobatos Productus) And California Bat Ray (Myliobatis Californica) In The Southern California Bight, Annabel Gong

Theses

While the movement patterns of large elasmobranch species have been studied extensively, those of smaller, mesopredatory species remain understudied. The shovelnose guitarfish (Pseudobatos productus) and the California bat ray (Myliobatis californica) are among the least studied elasmobranchs in the Southern California Bight. This study quantified the broad- and fine-scale movement patterns of these species using passive acoustic telemetry. Twelve guitarfish were surgically implanted with coded acoustic transmitters at a known aggregation site off La Jolla (San Diego County), California, USA and tracked for 849.5 days each, on average. Six bat rays were also implanted here and tracked for …

Results: When tested on the baseline of isolated tissues, Tfg.Cr was devoid of any activity (stimulant or relaxant) till 10 mg/ml. This is an interesting finding, keeping in mind that the fenugreek seeds are used to alleviate constipation and diarrhoea. When Tfg.Cr was tried for any potential AChE inhibitory activity, it did show an inhibitory effect in increasing concentrations (47-380 μg/ml). This inhibitory effect was comparable to the effect produced by a standard AChE inhibitor physostigmine. One of the known fenugreek constituents, diosgenin, was also tested, and it also showed an AChE inhibitory effect in a concentration-dependent manner (11-190 …

Alcohol is a risk factor for hepatocellular carcinoma (HCC). However, the molecular mechanism by which chronic alcohol consumption contributes to HCC is not well understood. The purpose of the study was to demonstrate the effects of chronic ethanol exposure on the damage of human normal hepatocytes. Our data showed that chronic exposure of hepatocytes with ethanol induced changes similar to transformed hepatocytes that is, exhibited colonies and anchorage-independent growth. These damaged hepatocytes contained high levels of reactive oxygen species (ROS) and showed induction of the SATB2 gene. Furthermore, damaged hepatocytes gained the phenotypes of CSCs which expressed stem cell markers …

Healthcare Simulation: A Key To The Future Of Medical Education - A Review, Omair Ayaz, Faisal Wasim Ismail

Medical College Documents

Aim: Simulation originates from its application in the military and aviation. It is implemented at various levels of healthcare education and certification today. However, its use remains unevenly distributed across the globe due to misconception regarding its cost and complexity and to lack of evidence for its consistency and validity. Implementation may also be hindered by an array of factors unique to the locale and its norms. Resource-poor settings may benefit from diverting external funds for short-term simulation projects towards collaboration with local experts and local material sourcing to reduce the overall cost and achieve long-term benefits. The recent shift …

Fruits Of Nandina Domestica Are (Sometimes) Cyanogenic And (Sometimes) Hazardous To Birds, Scott Zona

Poisonous Plant Research (PPR)

A 12-month survey of cyanogenesis in the fruits and leaves of Nandina domestica (Berberidaceae) revealed that the leaves are strongly cyanogenic throughout the year, as measured by the Feigl-Anger test. Fruits vary in their cyanogenic potential depending on the degree of ripeness and clone. Green fruits are strongly and rapidly cyanogenic, but most ripe fruits are weakly and slowly cyanogenic. Some fruits tested negative for cyanide. Fruits tend to become less cyanogenic the longer they remain on the plant, so birds feeding on older fruits are at a lower risk of poisoning. More than other frugivores, cedar waxwings (Bombycilla …

Theses and Dissertations

Penstemon is one of North America's largest endemic genera with over 280 described species. These species are distributed throughout most of North America from the Arctic northern latitudes to tropics of Central America. The genus has historically been divided into six subgenera, but has recently been reorganized into two subgenera following some recent phylogenetic studies. I made a comprehensive assessment of the Penstemon genus' geographic distribution utilizing herbaria databases by ecoregion to discuss the general ecologic adaptations of each historic subgenera. I also assessed the Penstemon genus' bee pollinator diversity utilizing online databases of bee specimen collections associated with Penstemon …
