The Salinity Of The Great Salt Lake And Its Deep Brine Layer, Madeline F. Merck, David G. Tarboton Apr 2023

The Salinity Of The Great Salt Lake And Its Deep Brine Layer, Madeline F. Merck, David G. Tarboton

Publications

The Great Salt Lake is a highly saline terminal lake with considerable fluctuations in water surface elevation and salinity. The lake is divided into two arms by a railroad causeway. River inflows enter the larger south arm, while the north arm only receives minimal surface runoff. Evaporation from both arms and limited exchange of water and salt through causeway openings result in complex water and salinity processes in the lake. The north arm is typically homogeneous and close to saturation. The south arm is typically stratified with periodic occurrences of a deep brine layer. This paper analyzes the lake’s long-term …

Kimmel Cancer Center Faculty Papers

BACKGROUND: Malignant perivascular epithelioid cell tumors (PEComas) are exceedingly rare malignant mesenchymal neoplasms with characteristic morphological and immunohistochemical (IHC) patterns. However, some malignant PEComas are poorly differentiated with atypical histopathological features, making a definitive diagnosis difficult. PEComas are most commonly found in females and often show either TSC1 or TSC2 alterations, which result in the activation of the mTOR pathway, or TFE3 fusions. Given these molecular characteristics, mTOR inhibitors have recently been approved by the FDA in the treatment of malignant PEComas, particularly in those with TSC1/2 alterations. Therefore, molecular analyses may be helpful for both the diagnostic workup of …

Autotaxin (ATX), the key enzyme that generates lysophosphatidic acid (LPA) from lysophosphatidylcholine (LPC), is involved in tumorigenesis through the ATX-LPA axis and is regarded as a valuable target in tumor therapy. Hypoxia is a major feature of solid tumors and contributes to tumor development with striking alterations in the gene expression profile. Here, we show that hypoxia induces ATX expression in a hypoxia-inducible factor (HIF) 2α-dependent fashion in human colon cancer SW480 cells. HIF-2α is directly bound to specific hypoxia response elements (HREs) in the ATX promoter. Under hypoxic conditions, knockout or inhibition of ATX suppressed the migration of SW480 …

Molecular Mechanisms Of Ferroptosis And Updates Of Ferroptosis Studies In Cancers And Leukemia, Hiroki Akiyama, Bing Z Carter, Michael Andreeff, Jo Ishizawa

Faculty, Staff and Student Publications

Ferroptosis is a mode of cell death regulated by iron-dependent lipid peroxidation. Growing evidence suggests ferroptosis induction as a novel anti-cancer modality that could potentially overcome therapy resistance in cancers. The molecular mechanisms involved in the regulation of ferroptosis are complex and highly dependent on context. Therefore, a comprehensive understanding of its execution and protection machinery in each tumor type is necessary for the implementation of this unique cell death mode to target individual cancers. Since most of the current evidence for ferroptosis regulation mechanisms is based on solid cancer studies, the knowledge of ferroptosis with regard to leukemia is …

BACKGROUND: We sought to discover serum biomarkers of ankylosing spondylitis (AS) for diagnosis and monitoring disease activity.

METHODS: We studied biologic-treatment-naïve AS and healthy control (HC) patients' sera. Eighty samples matched by age, gender, and race (1:1:1 ratio) for AS patients with active disease, inactive disease, and HC were analyzed with SOMAscan™, an aptamer-based discovery platform. T-tests tests were performed for high/low-disease activity AS patients versus HCs (diagnosis) and high versus low disease activity (Monitoring) in a 2:1 and 1:1 ratio, respectively, to identify differentially expressed proteins (DEPs). We used the Cytoscape Molecular Complex Detection (MCODE) plugin to find clusters …

Habitat Effects On Scorpion Densities And Microhabitat Use By Centruroides Vittatus And Vaejovis Waueri In South Texas, Jonathan Beltran Apr 2023

Habitat Effects On Scorpion Densities And Microhabitat Use By Centruroides Vittatus And Vaejovis Waueri In South Texas, Jonathan Beltran

Theses and Dissertations

Co-existence between Centruroides vittatus and Vaejovis waueri is influenced by environmental factors that shape their habitats. Differences in surface activity frequency between the two species suggest differential microhabitat use. The observed frequency of scorpions using microhabitats in the Martinez Ranch and La Union Ranch were not significantly different from the expected frequencies. Frequency of scorpions observed using microhabitats in La Union South was significantly different from the expected frequency (P<0.001). Microhabitat use of catclaw was lower than expected and higher than expected for other vegetation. Conditional comparisons of microhabitats were significantly different among habitats (P<0.001), size classes were significantly different among microhabitats (P<0.001), and size classes were significantly different among habitats (P<0.01). Microhabitat use by scorpions on the ground was higher among all habitats. Scorpion use of ground and grass were different among size classes. Size class I-II scorpions had a higher frequency than size class III. Scorpion density for C. vittatus was significantly different among habitats (P<0.0001) and scorpion density for V. waueri was significantly different among habitats (P<0.0001). Centruroides vittatus was significantly higher in the Martinez Ranch and in La Union South. Vaejovis waueri was significantly higher in La iv Union South. Associations between plant densities and scorpion size classes were tested. Plant densities had a significant association with C. vittatus size classes I and II in the Martinez Ranch. Association between plants had a significant correlation with C. vittatus size classes I and IV in La Union Ranch. Association between plants had a significant correlation with V. waueri’s total density. These associations may suggest plant densities may have a synergistic effect on scorpion densities. Soil type composition for the different habitats was hypothesized to influence scorpion densities. Maps for each site were created to show soil type compositions. Soil type findings suggest that La Union South had the soil types with the lowest soil strength. Vaejovis waueri was shown to have a higher density in La Union South where soils are sandier. Results indicate that co-existence between C. vittatus and V. waueri is possible because competition is low through minimal overlap in microhabitat use and intraguild predation avoidance.

Aspen Bibliography

Background Wood represents the majority of the biomass on land and constitutes a renewable source of biofuels and other bioproducts. However, wood is recalcitrant to bioconversion, raising a need for feedstock improvement in production of, for instance, biofuels. We investigated the properties of wood that affect bioconversion, as well as the underlying genetics, to help identify superior tree feedstocks for biorefining.

Results We recorded 65 wood-related and growth traits in a population of 113 natural aspen genotypes from Sweden (https://doi.org/10.5061/dryad.gtht76hrd). These traits included three growth and field performance traits, 20 traits for wood chemical composition, 17 traits for wood anatomy …

The severe consequences of the Zika virus (ZIKV) infections resulting in congenital Zika syndrome in infants and the autoimmune Guillain–Barre syndrome in adults warrant the development of safe and efficacious vaccines and therapeutics. Currently, there are no approved treatment options for ZIKV infection. Herein, we describe the development of a bacterial ferritin-based nanoparticle vaccine candidate for ZIKV. The viral envelope (E) protein domain III (DIII) was fused in-frame at the amino-terminus of ferritin. The resulting nanoparticle displaying the DIII was examined for its ability to induce immune responses and protect vaccinated animals upon lethal virus challenge. Our results show that …

MUSC Theses and Dissertations

Neuropsychiatric disorders have a significant impact on modern society. These disorders affect a large percentage of the population: schizophrenia has a world-wide prevalence of 1% and autism spectrum disorders (ASD) affects 1 in 59 school-aged children in the US. There is substantial evidence that most neuropsychiatric disorders have a genetic component. Thus, with the advent of high throughput sequencing much effort has gone into identifying genetic variants associated with these disorders. The emerging picture from these studies is a complex one where hundreds of genes with small effects interact with a varied landscape of common variants to result in disease. …

Halophilic Genes That Impact Plant Growth In Saline Soils, Mckay A. Meinzer Apr 2023

Halophilic Genes That Impact Plant Growth In Saline Soils, Mckay A. Meinzer

Theses and Dissertations

Many plants are highly sensitive to salt in the soil, and their growth and yield can be greatly hindered by as little as less than 1% salt concentration in the soil. Additionally, soil salinity is a growing issue globally and affects significant areas in Utah. Halophytes are plants that are adapted to grow in saline soils and have been widely studied for their physiological and molecular characteristics, but little is known about their associated microbiomes. Bacteria were isolated from the rhizosphere and as root endophytes of Salicornia rubra, Sarcocornia utahensis, and Allenrolfea occidentalis, three native Utah halophytes. Several strains of …

Utilizing Mass Spectrometry Imaging To Correlate N-Glycosylation Of Hepatocellular Carcinoma With Tumor Subtypes For Biomarker Discovery, Andrew Delacourt

MUSC Theses and Dissertations

Hepatocellular carcinoma (HCC) is a leading cause of cancer deaths globally and is a growing clinical problem with poor survival outcomes beyond early-stage disease. Surveillance for HCC has primarily relied on ultrasound and serum α-fetoprotein (AFP), but combined they only have a sensitivity of 63% for early-stage HCC tumors, suggesting a need for improved diagnostic strategies. Alterations to N-glycan expression are relevant to the progression of cancer, and there a multitude of N-glycan-based cancer biomarkers that have been identified with sensitivity for various cancer types including HCC. Spatial HCC tissue profiling of N-linked glycosylation by matrix-assisted laser desorption ionization imaging …

Multispecies Use Of Various Habitats In The Mississippi Sound With An Emphasis On Future Oyster Reef Construction, Austin Draper Apr 2023

Multispecies Use Of Various Habitats In The Mississippi Sound With An Emphasis On Future Oyster Reef Construction, Austin Draper

Master's Theses

Transient fish have high economic value and effective restoration projects must consider the variable impacts they may have on these fish in addition to the target organism or habitat being restored. Globally, oyster reef construction is common and will continue but data on their impacts to transient fish is lacking. This study used acoustic telemetry to provide baseline data on the potential for interaction between five transient fish species, and oyster reef construction activities. Overall, I noted that habitat preferences are hierarchical with sandy habitats nearest to bay or river mouths having high transient fish use. Oyster reef construction in …

Utilizing Isothermal Titration Calorimetry To Measure Β-Galactosidase Activity In Dairy Products, Tyler Ronald Jarrard

Theses and Dissertations

The dairy industry uses enzymes to make cheese, alter product flavor, and eliminate lactose. The activities of these enzymes have been measured in clear buffered solutions, but because of the limitations of spectrophotometric methods, enzyme activities have not been measured in opaque or colored dairy products where they are used. Isothermal titration calorimetry (ITC) can be used to determine reaction kinetics in opaque and colored solutions by measuring the heat rate from enzyme-catalyzed reactions as a function of time. This study used ITC to measure β-galactosidase activity in opaque solutions of milk, sweet whey, sweet whey permeate, acid whey, and …

Consciousness, Evolution, And The Self-Organizing Brain, Karen Seymour Apr 2023

Consciousness, Evolution, And The Self-Organizing Brain, Karen Seymour

Journal of Conscious Evolution

While evolution is guided by natural selection, it is internally driven by self-organizing processes. The brain encompasses these complementary forces and dynamics of evolution in both its structure and dynamics by embodying a historical record of the factors that have shaped it throughout its evolutionary past, as well as by being shaped by selective parameters in real time. Self-organization is evident in not only the brain’s structure and form, but also in the processes that support consciousness. From the convergence of complex structure and the novelty-generating dynamics of chaos that both characterize the brain arises the experience of explicit consciousness, …

What Can Plant Science Learn From Animal Nervous Systems?, Luiz Pessoa

Animal Sentience

I welcome Segundo-Ortin & Calvo’s (2023) call for a rigorous science of plant behavior and physiology. My commentary addresses three points drawn from the literature on animal brains that could help elucidate the possibility of cognition and sentience in plants: (1) the presumed requirement of a centralized brain; (2) centralization of control versus heterarchical organization; and (3) connecting plant research with research on animal nervous systems.

ARID1A, encoding a subunit of the SWI/SNF complex, is mutated in ∼50% of clear cell ovarian carcinoma (OCCC) cases. Here we show that inhibition of the mevalonate pathway synergizes with immune checkpoint blockade (ICB) by driving inflammasome-regulated immunomodulating pyroptosis in ARID1A-inactivated OCCCs. SWI/SNF inactivation downregulates the rate-limiting enzymes in the mevalonate pathway such as HMGCR and HMGCS1, which creates a dependence on the residual activity of the pathway in ARID1A-inactivated cells. Inhibitors of the mevalonate pathway such as simvastatin suppresses the growth of ARID1A mutant, but not wild-type, OCCCs. In addition, simvastatin synergizes with anti-PD-L1 antibody in a genetic OCCC …

Diabetes is associated with increased risk for kidney disease, heart failure, and mortality. Sodium-glucose cotransporter 2 inhibitors (SGLT2i) prevent these adverse outcomes; however, the mechanisms involved are not clear. We generated a roadmap of the metabolic alterations that occur in different organs in diabetes and in response to SGLT2i. In vivo metabolic labeling with 13C-glucose in normoglycemic and diabetic mice treated with or without dapagliflozin, followed by metabolomics and metabolic flux analyses, showed that, in diabetes, glycolysis and glucose oxidation are impaired in the kidney, liver, and heart. Treatment with dapagliflozin failed to rescue glycolysis. SGLT2 inhibition increased glucose oxidation …

Switchgrass (SG) is considered a model bioenergy crop and a warm-season perennial grass (WSPG) that traditionally served as forage feedstock in the United States. To avoid the sole dependence on SG for bioenergy production, evaluation of other crops to diversify the pool of feedstock is needed. We conducted a 3-year field experiment evaluating eastern gamagrass (GG), another WSPG, as complementary feedstock to SG in one- and two-cut systems, with or without intercropping with crimson clover or hairy vetch, and under different nitrogen (N) application rates. Our results showed that GG generally produced lower biomass (by 29.5%), theoretical ethanol potential (TEP, …
