Is Vdac1 A Novel Bcl2 Family Member That Binds Bax?, Claire Pearson
Honors Theses
Apoptosis is a type of regulated cell death important for normal embryonic development and maintenance of adult tissues by removing excess or dysfunctional cells to ensure proper functioning of organs. The Bcl-2 family of proteins determines whether apoptosis remains suppressed or becomes activated through the balance of interactions among pro-survival and pro-death members. A defining feature of the Bcl-2 family is a BH3 domain that drives interactions between the family members. Isoform 1 of the voltage dependent anion channel (VDAC1) has an important role in metabolism, but was recently found to have high homology with known BH3 domains. This study …

Mitochondria are signaling organelles implicated in cancer, but the mechanisms are elusive. Here, we show that Parkin, an E3 ubiquitination (Ub) ligase altered in Parkinson's disease, forms a complex with the regulator of cell motility, Kindlin-2 (K2), at mitochondria of tumor cells. In turn, Parkin ubiquitinates Lys581 and Lys582 using Lys48 linkages, resulting in proteasomal degradation of K2 and shortened half-life from ∼5 h to ∼1.5 h. Loss of K2 inhibits focal adhesion turnover and β1 integrin activation, impairs membrane lamellipodia size and frequency, and inhibits mitochondrial dynamics, altogether suppressing tumor cell-extracellular matrix interactions, migration, and invasion. Conversely, Parkin does …

Transcriptome Profile In Drosophila Kc And S2 Embryonic Cell Lines, Daniel Klonaros, Jacqueline M. Dresch, Robert A. Drewell
Biology
Drosophila melanogaster cell lines are an important resource for a range of studies spanning genomics, molecular genetics, and cell biology. Amongst these valuable lines are Kc167 (Kc) and Schneider 2 (S2) cells, which were originally isolated in the late 1960s from embryonic sources and have been used extensively to investigate a broad spectrum of biological activities including cell-cell signaling and immune system function. Whole-genome tiling microarray analysis of total RNA from these two cell types was performed as part of the modENCODE project over a decade ago and revealed that they share a number of gene expression features. Here, we …

Mitochondrial outer membrane permeabilization (MOMP) by Bax oligomerization triggers apoptosis. BCl-2 family proteins, classified as BH3 only proteins, pro-survival proteins, or pro-apoptotic proteins, control apoptosis partly through their agonist or antagonistic effects on Bax, which are mediated by their conserved BH3 domains. All BH3 domains form an alpha helix containing 5-7 conserved hydrophobic residues, designated H0-H5, and one conserved aspartic acid that drive interaction with Bax and other ‘multi-domain’ BCl-2 members. BH3 agonists induce Bax oligomerization, while BH3 antagonists sequester Bax to prevent MOMP. We discovered that voltage dependent anion channels (VDACs) in the MOM contain a putative BH3-like domain …

BACKGROUND AND OBJECTIVES: Previous studies suggest that lower mitochondrial DNA (mtDNA) copy number (CN) is associated with neurodegenerative diseases. However, whether mtDNA CN in whole blood is related to endophenotypes of Alzheimer disease (AD) and AD-related dementia (AD/ADRD) needs further investigation. We assessed the association of mtDNA CN with cognitive function and MRI measures in community-based samples of middle-aged to older adults.
METHODS: We included dementia-free participants from 9 diverse community-based cohorts with whole-genome sequencing in the Trans-Omics for Precision Medicine (TOPMed) program. Circulating mtDNA CN was estimated as twice the ratio of the average coverage of mtDNA to nuclear …

Advanced high-grade serous ovarian cancer (HGSC) is an aggressive disease that accounts for 70% of all ovarian cancer deaths. Nevertheless, 15% of patients diagnosed with advanced HGSC survive more than 10 years. The elucidation of predictive markers of these long-term survivors (LTS) could help identify therapeutic targets for the disease, and thus improve patient survival rates. To investigate the stromal heterogeneity of the tumor microenvironment (TME) in ovarian cancer, we used spatial transcriptomics to generate spatially resolved transcript profiles in treatment-naïve advanced HGSC from LTS and short-term survivors (STS) and determined the association between cancer-associated fibroblasts (CAF) heterogeneity and survival …

The global threat of antimicrobial resistance (AMR) varies regionally. This study explores whether geospatial analysis and data visualization methods detect both clinically and statistically significant variations in antibiotic susceptibility rates at a neighborhood level. This observational multicenter geospatial study collected 10 years of patient-level antibiotic susceptibility data and patient addresses from three regionally distinct Wisconsin health systems (UW Health, Fort HealthCare, Marshfield Clinic Health System [MCHS]). We included the initial Escherichia coli isolate per patient per year per sample source with a patient address in Wisconsin (N = 100,176). Isolates from U.S. Census Block Groups with less than 30 isolates …

Dynamic Transcriptomic Responses Of Circulating Immune Cells In Response To Subsequent Days Of Exercise Heat Stress, Soohyun Oh
University Scholar Projects
Acute exercise and chronic training have diverse effects on immune function that are still not well understood. There are remaining questions about how an acute bout of exercise, competition, or repeated training over a period affects an individual’s immune defense system. In a related study, there is evidence that acute exercise may serve as an adjuvant to enhance response to immune function as well as contradicting evidence, including our preliminary data (unpublished). Understanding the fundamental function and gene expression of circulating immune cells in response to acute exercise and in various contexts such as heat stress is critical in understanding …

An Analysis Of Sediment Collected By Pogonomyrmex Salinus From The Jurassic Sundance Formation, Bighorn Basin, Wyoming, Josephine H. Laprad
Graduate Dissertations and Theses
Pogonomyrmex salinus is a species of harvester ant found in the Bighorn Basin area of Wyoming. Harvester ants are aptly named as they “harvest” seeds for food, but this harvesting behavior is also seen in their collection of sediment particles to build their mounds. For decades, paleontologists have looked to harvester ants for microfossils because the small size of the fossils makes them difficult for human eyes to find. But there are unresolved questions about the data that can be gleaned from the fossils collected by the ants. The mounds collected for this study include material from the Stockade Beaver …

The Role Of Fto, A Human Rna Demethylase In Perennial Grass Development And Abiotic Stress Responses, Andrew Fiorentino, Qian Hu, Xiaotong Chen, Zhaohui Chen
Honors College Theses
The integration of the human fat mass and obesity associated (FTO) gene into turfgrass is a novel approach at improving cell proliferation and abiotic stress resistance. The FTO protein is an RNA demethylase responsible for epigenetic regulation of the genome. In related rice, the gene is associated with increased crop yield, tiller number, and aerial biomass. It is proposed to work via demethylation of repeat RNA associated with chromatin remodeling, causing widespread transcriptional activation. In this study, the feasibility of using FTO for plant trait modification in perennial grasses is being investigated. Potentially transformed embryogenic calli of creeping bentgrass with …

Temporal, Phenotypic, And Quantitative Characterization Of Thyroid Infiltrating Mononuclear Cells During Development Of Spontaneous Autoimmune Thyroiditis In Obese Strain Chickens, Katelyn M. Clark
Poultry Science Undergraduate Honors Theses
The Obese strain (OS) of chickens spontaneously develops autoimmune thyroiditis and is a well-established biomedical model for Hashimoto’s thyroiditis in humans. Both conditions are characterized by the infiltration of the thyroid glands with mononuclear cells resulting in destruction of the thyroid tissue and impairment of the thyroid’s endocrinological functions. Past studies have described mononuclear cell infiltration in thyroids of the OS chickens, but the time-course, cell composition, and relative amounts of the various mononuclear cells infiltrating the thyroids has not been well defined. In this project, the mononuclear cell infiltrate was characterized phenotypically and quantitatively in OS chickens over the …

An Investigation Of Resistance Training Behavior, Non-Specific Psychological Distress, And Perceived Barriers To Resistance Training In Self-Identifying Female Undergraduate Students, Jody Sobel
Master's Theses
The prevalence and severity of depression symptoms amongst college students, specifically female identifying undergraduate students, is worrisome. There is a great deal of research that suggests a relationship between exercise, specifically resistance training (RT), and mental health. That said, current literature suggests that many female identifying students are not engaging due to perceived barriers. This, perhaps because of the plethora of barriers one can perceive. The purpose of this study was multifold 1) to examine the prevalence and severity of non-specific psychological distress; 2) to determine if there were differences between Kessler 6 Non-Specific Psychological Distress Scale (K6) categories (i.e., …

Hawk Mountain Raptor Migration Phenology’S Relation To Weather, Eric Burgos
Computer Science and Information Technology Faculty
We have been studying year-round raptor migration phenology across the United States and North America for multiple decades now. Hawk Mountain Sanctuary’s Autumn migration hawk count began in 1934 and is the longest running raptor migration count in the world. A decline in total raptor counts passing through Hawk Mountain’s North Lookout is well documented and much research has already been done in what could be the main causes for this decrease in counts year-over-year. We know that cold front passages have long been associated with autumnal migration in northeastern North America. Using updated analysis techniques, we examined 60 years’ …
