Confocal Z-series .nd2 files (Nikon NIS) showing GFP fluorescence from dissected egg chambers. These files were used to create the maximum intensity projections displayed in figure 1 (UAS-GFPnls drd-gal4 stage 10B and 14 10.27.16.nd2) and figure S3 (UAS-GFPnls drd-gal4 stage 14 7.16.21.nd2 and UAS-GFPnls control stage 14 7.16.21.nd2). Files with “UAS-GFPnls drd-gal4” were experimental samples containing both the drd-GAL4 and UAS-GFP.nls transgenes, while files with “UAS-GFPnls control” were controls lacking the drd-GAL4 transgene. The “10.27.16” and “7.16.21” file sets were imaged on different days with different settings, so image intensities should not be compared between the two file sets.

Factors such as soil type and precipitation vary across rangeland landscapes, and these factors affect restoration outcomes and ultimately mean that “one size fits all” management strategies are not effective across large, complex landscapes. Big sagebrush (Artemisia tridentata) is a foundational rangeland species that is important to wildlife habitat across the western U.S. On the Colorado Plateau, sagebrush is important browse for ungulates, such as mule deer and pronghorn, which motivates a great deal of restoration effort. However, most scientific knowledge of big sagebrush comes from the Great Basin, and we know much less about how to restore sagebrush on …

Progression of disease within 24 months (POD24) from diagnosis in marginal zone lymphoma (MZL) was shown to portend poor outcomes in prior studies. However, many patients with MZL do not require immediate therapy, and the time from diagnosis-to-treatment interval can be highly variable with no universal criteria to initiate systemic therapy. Hence, we sought to evaluate the prognostic relevance of early relapse or progression within 24 months from systemic therapy initiation in a large US cohort. The primary objective was to evaluate the overall survival (OS) in the two groups. The secondary objective included the evaluation of factors predictive of …

Involvement Of Trpm7 In Alcohol-Induced Damage Of The Blood-Brain Barrier In The Presence Of Hiv Viral Proteins, Michelle L. Mack

Seton Hall University Dissertations and Theses (ETDs)

Ethanol (EtOH) exerts its effects through various protein targets, including transient receptor potential melastatin 7 (TRPM7) channels, which play an essential role in cellular homeostasis. We demonstrated that TRPM7 was expressed in rat brain microvascular endothelial cells (rBMVECs), the major cellular component of the blood–brain barrier (BBB). Heavy alcohol drinking is often associated with HIV infection, however mechanisms underlying alcohol-induced BBB damage and HIV proteins, are not fully understood. We utilized the HIV-1 transgenic (HIV-1Tg) rat to mimic HIV-1 patients on combination anti-retroviral therapy (cART) and demonstrated TRPM7 expression in rBMVECs was lower in adolescent HIV-1Tg rats compared to control …

Dendroctonus Ponderosae (Coleoptera: Curculionidae) Observations From Wheeler Peak Campground, Great Basin National Park, Justin P. Williams, Danielle Malesky

Western North American Naturalist

Mountain pine beetle (Dendroctonus ponderosae Hopkins, 1902; MPB) has been causing limber pine (Pinus flexilis E. James) mortality at Great Basin National Park, Nevada, including at Wheeler Peak Campground, for many years. Research has shown that MPB voltinism and phenology are strongly influenced by climate and can vary by location and elevation, indicating the need for localized observations to inform treatment recommendations in some cases. We used emergence cages on infested tree boles, baited Lindgren funnel traps, and ambient air temperatures to describe MPB adult flight timing over 2 years. Adult MPB flight at this high-elevation site occurred …

Background: Genetic variation in regulatory sequences that alter transcription factor (TF) binding is a major cause of phenotypic diversity. Brassinosteroid is a growth hormone that has major effects on plant phenotypes. Genetic variation in brassinosteroidresponsive cis-elements likely contributes to trait variation. Pinpointing such regulatory variations and quantitative genomic analysis of the variation in TF-target binding, however, remains challenging. How variation in transcriptional targets of signaling pathways such as the brassinosteroid pathway contributes to phenotypic variation is an important question to be investigated with innovative approaches.

Results: Here, we use a hybrid allele-specific chromatin binding sequencing (HASChseq) approach and …

Cancer remains a major global health challenge. Traditional chemotherapy often results in side effects and drug resistance, necessitating the development of alternative treatment strategies such as gene therapy. Mesoporous silica nanoparticles (MSNs) offer many advantages as a gene delivery carrier, including high loading capacity, controlled drug release, and easy surface functionalization. MSNs are biodegradable and biocompatible, making them promising candidates for drug delivery applications. Recent studies demonstrating the use of MSNs for the delivery of therapeutic nucleic acids to cancer cells have been reviewed, along with their potential as a tool for cancer therapy. The major challenges and future interventions …

BACKGROUND: The role of ovulation in epithelial ovarian cancer (EOC) is supported by the consistent protective effects of parity and oral contraceptive use. Whether these factors protect through anovulation alone remains unclear. We explored the association between lifetime ovulatory years (LOY) and EOC.

METHODS: LOY was calculated using 12 algorithms. Odds ratios (ORs) and 95% confidence intervals (CIs) estimated the association between LOY or LOY components and EOC among 26 204 control participants and 21 267 case patients from 25 studies. To assess whether LOY components act through ovulation suppression alone, we compared beta coefficients obtained from regression models with …

The G-Signaling Protein Rcp Controls The Polarized Basement Membrane Deposition In Epithelial Cells, Rebecca M. Brnot May 2023

The G-Signaling Protein Rcp Controls The Polarized Basement Membrane Deposition In Epithelial Cells, Rebecca M. Brnot

Honors Capstones

Epithelial tissues are the most common type of tissue in the human body, forming the outer layer of the skin and most organs. They are composed of epithelial cells and rely heavily on their cellular architecture, which is organized by an apical-basal polarity, for their function. One critical component for the establishment and maintenance of the epithelial cell architecture is the proper placement of the basement membrane. The basement membrane is a specialized sheet within the extracellular matrix that lines the basal side of epithelial cells, and there is a biological pathway that is responsible for the proper placement of …

An In-Depth Study Of Border Cell Migration Within Drosophila Melanogaster, Leif R. Verace May 2023

An In-Depth Study Of Border Cell Migration Within Drosophila Melanogaster, Leif R. Verace

Honors Capstones

Cell migration plays a key role in many biological processes including embryonic development, tissue morphogenesis, and the metastasis of cancer cells; thus, understanding how this behavior works is highly important. To study cell migration, we use the process of border cell (BC) migration in Drosophila melanogaster, which sees a cluster of cells migrate through a multicellular structure known as the egg chamber found in the Drosophila ovary. Here, I will describe my results in studying the role of a particular kinesin-like gene, CG14535, in BC migration. Through analysis of a mutant for this gene (labelled KC53), RNAi …

The Impacts Of Seasonality And Nutrient Loading On Microcystis Bloom Development In Wall Pond, Alyssa Antolak May 2023

The Impacts Of Seasonality And Nutrient Loading On Microcystis Bloom Development In Wall Pond, Alyssa Antolak

Honors Theses

The harmful cyanobacteria Microcystis globally dominates eutrophic freshwater systems. Eutrophication leading to nitrogen and phosphorus loading into aquatic systems is increasing bloom propagation and shifting diatom/dinoflagellate dominated systems to cyanobacteria dominated systems. Understanding seasonal variability and environmental parameters combined with nutrient loading will allow for a better understanding of what factors are influencing Microcystis blooms. Biweekly plankton samples and environmental parameters were collected from Wall Pond from spring 2022-spring 2023. Results show that yearly plankton samples shifted from Microcystis dominated in the early summer to diatom dominated in the late summer-early fall then back to Microcystis dominated in late fall-early …

Elucidating The Impact Of Sos-Response Timing In On Escherichia Coli Survival Following Treatment With Fluoroquinolone Topoisomerase Inhibitors, Stephanie Schofield May 2023

Elucidating The Impact Of Sos-Response Timing In On Escherichia Coli Survival Following Treatment With Fluoroquinolone Topoisomerase Inhibitors, Stephanie Schofield

Honors Scholar Theses

Antibiotic treatment failure is a public health crisis, with a 2019 report stating that roughly 35,000 deaths occur in the United States yearly due to bacterial infections that are unresponsive to antibiotics (1). One complication in the treatment of bacterial infection is antibiotic persistence which further compromises our battle to effectively treat infection. Bacterial persisters can exist in clonal bacterial cultures and can tolerate antibiotic treatment by undergoing reversible phenotypic changes. They can survive drug concentrations that their genetically identical kin cannot. Some persisters remain in a slow growing state and are difficult to target with current antibiotics. A specific …

Lessons From Chloroviruses: The Complex And Diverse Roles Of Viruses In Food Webs, John Delong, James L. Van Etten, David Dunigan

School of Biological Sciences: Faculty Publications

Viruses can have large effects on the ecological communities in which they occur. Much of this impact comes from the mortality of host cells, which simultaneously alters microbial community composition and causes the release of matter that can be used by other organisms. However, recent studies indicate that viruses may be even more deeply integrated into the functioning of ecological communities than their effect on nutrient cycling suggests. In particular, chloroviruses, which infect chlorella-like green algae that typically occur as endosymbionts, participate in three types of interactions with other species. Chlororviruses (i) can lure ciliates from a distance, using them …

Targeting The Cxcr1/2 Axis In Pancreatic Ductal Adenocarcinoma, Caitlin Molczyk May 2023

Targeting The Cxcr1/2 Axis In Pancreatic Ductal Adenocarcinoma, Caitlin Molczyk

Theses & Dissertations

Pancreatic ductal adenocarcinoma (PDAC) is notoriously challenging to diagnose and treat. This tumor has an aggressive phenotype and benign clinical features until severe disease dissemination, which makes it too late to eradicate. Although it is only the 10th most common cancer in the United States, it is the fourth leading cause of cancer-related deaths. The treatment options are complicated further due to tumor heterogeneity, meaning not all tumor cells are in the same state phenotypically or epigenetically. They also express cytokines, chemokines, and receptors differently. Here we explore the usage of CXCR1 and CXCR2 and their ligands in PDAC tumor …

Synaptic Transmission At The Photoreceptor Ribbon Synapse, Chris Mesnard May 2023

Synaptic Transmission At The Photoreceptor Ribbon Synapse, Chris Mesnard

Theses & Dissertations

Synaptic transmission relies on the Ca2+-dependent release of neurotransmitters packaged into synaptic vesicles. Ribbon synapses have the capability to release multiple vesicles at a time in a highly coordinated and synchronized manner. Glutamate release from rod and cone photoreceptor cells involves presynaptic ribbons composed largely of the protein RIBEYE. To examine roles of ribbons in rods and cones, we studied mice in which GCamP3 replaced the B-domain of RIBEYE. We discovered that ribbons were absent from rods and cones of both knock-in mice possessing GCamP3 and conditional RIBEYE knockout mice. Mice lacking ribbons showed reduced temporal resolution and …

The Relationship Between Cerebrovascular Impairment And Behavioral Abnormalities In Rats Exposed To Alcohol In Utero, Tiffany M. Knecht

Honors Thesis

Maternal consumption of alcohol during pregnancy can result in fetal alcohol spectrum disorders (FASD), which describes a range of developmental disorders. The goal of this study was to examine the association between impaired cerebrovascular function and behavioral abnormalities associated with FASD. We fed Sprague-Dawley dams a liquid diet with or without alcohol (3% ethanol) for the duration of their pregnancy. We then performed short-term memory and motor coordination assessments on the offspring during adolescence or adulthood. Next, we examined cerebral arteriole reactivity to the vasodilators nitroglycerin, adenosine diphosphate, N-methyl-D-aspartate, and iloprost. We found impairment in eNOS-, nNOS-, and K-channel-dependent dilation …

Cognition Is Not Evidence Of Sentience, Tom Bennett May 2023

Cognition Is Not Evidence Of Sentience, Tom Bennett

Animal Sentience

The ability of plants to detect their environment, including other organisms within that environment, is unquestioned. The ability of plants to differentially process, integrate and respond to complex combinations of environmental information could perhaps be described as cognition. But no amount of evidence for cognitive abilities in plants equates to evidence for plant sentience. Nor is plant sentience required to understand or interpret the behaviour of plants in their environment.

Investigating Host Cell Transcriptional Heterogeneity And Metabolism In Staphylococcus Aureus Biofilm Infections, Christopher M. Horn May 2023

Investigating Host Cell Transcriptional Heterogeneity And Metabolism In Staphylococcus Aureus Biofilm Infections, Christopher M. Horn

Theses & Dissertations

Staphylococcus aureus (S. aureus) is a leading cause of prosthetic joint infection (PJI). These infections are often intractable due to biofilm formation, which are complex bacterial communities that adhere to biotic and abiotic surfaces. Within a biofilm, there is considerable heterogeneity in bacterial metabolism and gene expression that contributes to the establishment an anti-inflammatory milieu at the site of infection, which favors bacterial persistence even in the context of an immunocompetent host. This is due, in part, to the preferential recruitment of granulocytic myeloid-derived suppressor cells (G-MDSCs), which are a population of pathologically activated immature leukocytes that develop …

Investigating Plasmodial Proteostasis Mechanisms To Counter Artemisinin Resistance, Melissa Rosenthal May 2023

Investigating Plasmodial Proteostasis Mechanisms To Counter Artemisinin Resistance, Melissa Rosenthal

Theses & Dissertations

Malaria remains a significant global health threat, responsible for 691,000 deaths in 2021. Artemisinin-based combination therapies have contributed significantly to decline of malaria disease burden. Upon activation by heme, artemisinins are believed to kill parasites by non-specifically alkylating proteins and causing widespread damage to the parasite. Artemisinin resistance is now prevalent in regions of Southeast Asia and resistance has recently emerged in Africa, where over 90% of all malaria deaths occur. Mutations in Kelch13 (K13) are believed to mediate artemisinin resistance by either reducing parasite hemoglobin digestion, and consequently artemisinin activation, or by increasing parasite stress response mechanisms. We hypothesize …

The Interface Between Host And Bacterial Metabolism During Staphylococcus Aureus Prosthetic Joint Infection, Blake P. Bertrand May 2023

The Interface Between Host And Bacterial Metabolism During Staphylococcus Aureus Prosthetic Joint Infection, Blake P. Bertrand

Theses & Dissertations

Staphylococcus aureus (S. aureus) is a versatile pathogen that can occupy a wide range of niches within the human body, including implanted medical devices such as in prosthetic joint infection (PJI). S. aureus adheres to these medical devices and forms a biofilm, which is a community of bacteria contained within an extracellular matrix that leads to gradients in nutrient and oxygen availability. During PJI, the biofilm is recalcitrant to antibiotics and immune-mediated clearance, leading to treatment failure, mortality, and a high healthcare burden. This is partially due to skewing of the host immune system to an anti-inflammatory state, …

A Critical Assessment Of The Role Of Clpxp-Mediated Proteolysis In The Development And Differentiation Of Chlamydia Trachomatis, Nicholas A. Wood May 2023

A Critical Assessment Of The Role Of Clpxp-Mediated Proteolysis In The Development And Differentiation Of Chlamydia Trachomatis, Nicholas A. Wood

Theses & Dissertations

Chlamydia trachomatis, an obligate intracellular human pathogen, is the etiological agent of the most commonly reported bacterial sexually transmitted infection and the leading cause of preventable infectious blindness worldwide. In adapting to an intracellular niche, Chlamydia has evolved to eliminate superfluous genes from its chromosome. Despite its highly reduced genome, Chlamydia undergoes a complex developmental cycle in which the bacteria differentiate between two functionally and morphologically distinct forms: the infectious, non-replicative elementary body (EB) and the non-infectious, replicative reticulate body (RB). An EB initiates infection by binding to and inducing uptake into a host cell. Within the cell, the EB …
