Mycobacterium avium subsp. paratuberculosis (MAP) is the etiological agent of Johne’s disease, a severe gastroenteritis of ruminants. This study developed a model cell culture system to rapidly screen MAP mutants with vaccine potential for apoptosis. Two wild-type strains, a transposon mutant, and two deletion mutant MAP strains (MOI of 10 with 1.2 × 106 CFU) were tested in murine RAW 264.7 macrophages to determine if they induce apoptosis and/or necrosis. Both deletion mutants were previously shown to be attenuated and immunogenic in primary bovine macrophages. All strains had similar growth rates, but cell morphology indicated that both deletion mutants …
Complement System In Multiple Sclerosis: Its Role In Disease Course And Potential As A Therapeutic Target, Michael R. Linzey
Complement System In Multiple Sclerosis: Its Role In Disease Course And Potential As A Therapeutic Target, Michael R. Linzey
Dartmouth College Ph.D Dissertations
Multiple sclerosis (MS) is a clinically heterogeneous neurological condition characterized by neuroinflammation and neurodegeneration. Relapsing-remitting MS, defined by inflammatory attacks, is the most common initial form of MS and there are currently 23 FDA-approved treatments for these patients. These therapies work primarily by reducing inflammation in the CNS; they do not work well in progressive disease. Therefore, an unmet medical need exists for effective therapeutic options to treat progressive MS (PMS).
In MS, intrathecal immunoglobulins synthesis (IIgS) correlates with disease progression. My goals for this dissertation were to establish the pathological role of IIgS and identify new potential therapeutic …

Paleoamerican Exploitation Of Extinct Megafauna Revealed Through Immunological Blood Residue And Microwear Analysis, North And South Carolina, Usa, Christopher R Moore, Larry R Kimball, Albert C Goodyear, Mark J Brooks, I Randolph Daniel, Sean G Taylor, John L Fagan, Cam M Walker
Faculty, Staff and Student Publications
Previous immunological studies in the eastern USA have failed to establish a direct connection between Paleoamericans and extinct megafauna species. The lack of physical evidence for the presence of extinct megafauna begs the question, did early Paleoamericans regularly hunt or scavenge these animals, or were some megafauna already extinct? In this study of 120 Paleoamerican stone tools from across North and South Carolina, we investigate this question using crossover immunoelectrophoresis (CIEP). We find immunological support for the exploitation of extant and extinct megafauna, including Proboscidea, Equidae, and Bovidae (possibly Bison antiquus), on Clovis points and scrapers, as well as possible …

Vertebrate Animal Behaviors And Abundances On Estuarine Shorelines Stabilized With Biodegradable Materials Utilizing Wildlife Cameras, Katherine Guise
The Pegasus Review: UCF Undergraduate Research Journal
Living shoreline stabilization is a restoration technique that utilizes natural materials as breakwaters, plus vegetation landward of the breakwaters, to protect coastlines. This research does not provide information about how new, biodegradable restoration materials affect vertebrates that utilize these shorelines. For this project, I monitored 18 restoration sites along Canaveral National Seashore with wildlife trail cameras: 3 made with cement-infused jute breakwaters, 3 with metal gabion oyster shell breakwaters, and 4 with previously used breakwaters manufactured from plastic mesh oyster shell bags. This project used 4 sites as positive controls (intact vegetation) and 4 as negative controls (highly eroded, no …

Faculty Publications
The Lake Tana Labeobarbus species flock represents one of the world’s most famous examples of lacustrine species radiations. Previous studies of this group have resulted in the description of at least 15 species based on their differences in functional morphology and definition of two clades (lacustrine and riverine spawning clades) based on life history traits. A total of 166 fish representing 14 Labeobarbus species were genotyped using 10 lineage-specific hexaploid microsatellite loci. Six of these loci were developed for this study based on DNA sequence contigs derived from a microsatellite-enriched genomic library of Labeobarbus intermedius from Lake Tana; the remaining …

Phylogeography Of The Pacific Sardine, Sardinops Sagax, In The Northeastern Pacific, Ella Adams
Theses
The Pacific Sardine, (Sardinops sagax), is a small, coastal pelagic species in the family Clupeidae. Sardine are an ecologically important forage fish for many animals, including larger, economically and ecologically important fishes, and have historically supported an important commercial fishery. When the fishery declined in the 1940s, a massive effort in understanding population structure for the Pacific Sardine resulted in a wealth of literature. Initially, these studies agreed on a large panmictic population with high annual variation, but the general consensus has since been that there are multiple subpopulations of the Pacific Sardine along the West Coast of …

Effects Of Interleukin-22 (Il-22) On Necroptosis, Inflammatory Responses, And Metabolism Of Mycobacterium Tuberculosis Stimulated Type 2 Diabetes Mellitus Mouse Macrophages, Bismark Owusu-Afriyie
Biotechnology Theses
Mycobacterium tuberculosis (Mtb) infects one-third of the world’s population and causes nearly 1.3 million deaths per year. Limited information is available about the immune responses during Mtb infection in type 2 diabetic hosts. Our laboratory developed an experimentally induced type 2 diabetes (T2DM) model in wild-type C57BL/6 mice and found that IL-22 and type 3 innate lymphoid cells (ILC3s) reduce inflammation and mortality of Mtb-infected T2DM mice. Our laboratory also found that Mtb-infected alveolar macrophages (AMs) from T2DM mice undergo necroptosis compared to Mtb-infected AMs of non-T2DM mice. In the current study, we determined whether …

Viral Dna Accumulation Regulates Replication Efficiency Of Chlorovirus Osy-Ne5 In Two Closely Related Chlorella Variabilis Strains, Ahmed Esmael, Irina V. Agarkova, David D. Dunigan, You Zhou, James L. Van Etten
Nebraska Center for Virology: Faculty Publications
Many chloroviruses replicate in Chlorella variabilis algal strains that are ex-endosymbionts isolated from the protozoan Paramecium bursaria, including the NC64A and Syngen 2-3 strains. We noticed that indigenous water samples produced a higher number of plaque-forming viruses on C. variabilis Syngen 2-3 lawns than on C. variabilis NC64A lawns. These observed differences led to the discovery of viruses that replicate exclusively in Syngen 2-3 cells, named Only Syngen (OSy) viruses. Here, we demonstrate that OSy viruses initiate infection in the restricted host NC64A by synthesizing some early virus gene products and that approximately 20% of the cells produce a …

Alternariol (AOH), alternariol monomethyl ether (AME), alternuene (ALT), tenuazonic acid (TeA) and tenoxin (TEN) are the most studied at present. Alternaria mycotoxins was reported in apples, orange, tomatoes, corns and other agricultural products. Eating these foods contaminated by Alternaria mycotoxins will cause serious harm to human health. In this paper, the pollution status and the detection technology of Alternaria mycotoxins were summarized, and the future research direction of Alternaria mycotoxins was prospected. The purpose was to provide …

Context
One in 8 women will develop breast cancer in their lifetime. Yet, the burden of disease is greater in Black women. Black women have a 40% higher mortality rate than White women, and a higher incidence of breast cancer at age 40 and younger. While the underlying cause of this disparity is multifactorial, exposure to endocrine disrupting chemicals (EDCs) in hair and other personal care products has been associated with an increased risk of breast cancer. Parabens are known EDCs that are commonly used as preservatives in hair and other personal care products, and Black women are disproportionately exposed …
