The histone acetyltransferase p300/CBP is composed of several conserved domains, among which, the TAZ2 domain is known as a protein-protein interaction domain that binds to E1A and various transcription factors. Here we show that TAZ2 has a HAT autoinhibitory function. Truncating p300/CBP at TAZ2 leads to hyperactive HAT and elevated histone H3K27 and H3K18 acetylation in cells. Mechanistically, TAZ2 cooperates with other HAT neighboring domains to maintain the HAT active site in a 'closed' state. Truncating TAZ2 or binding of transcription factors to TAZ2 induces a conformational change that 'opens' the active site for substrate acetylation. Importantly, genetic mutations that …

The Effects Of Different Training Types On Phase Angle In Men - An Exploratory Study, Trevor Short, Paulette Yamada

Topics in Exercise Science and Kinesiology

Topics in Exercise Science and Kinesiology Volume 4: Issue 1, Article 12, 2023. Phase Angle (PhA) has emerged as a valuable measure in clinical and sports settings due to its predictive relationship with health and sports performance. Fitness is related to PhA, but the type of exercise training that is most influential over PhA has yet to be determined. The purpose was to explore the effects of endurance training (ET), strength-based resistance training (RT), and sprint-interval training (SIT) on PhA, resistance (R), and reactance (Xc) in men. Thirty-five recreationally-trained healthy young adult males (24±4 years) were randomly assigned to one …

Comparative Investigations On Microextraction And Conventional Air Sampling Techniques: Challenges And Future Directions, Firoz Ahmed, Mehedi Hasan Roni, Ashiqur Rahman, Sayed M A Salam

Al-Bahir

Microextraction technique (e.g., solid phase microextraction, thin film microextraction, in-tube extraction) brings a revolutionary change in air sampling techniques over the recent few years. This advanced technique exhibits a high pollutant extraction rate, a low retention time, and a lower error margin compared to conventional air sampling techniques. The accuracy range of microextraction technique (MET) was recorded ~90-95% to isolate the volatile organic components, oxygenated and halogenated carbon particles from the air. However, the efficiency of MET increases additional >3-5% when employed by coupled with gas chromatography or gas chromatography-mass spectrometry. The conventional sampling techniques (e.g., bag sampling, grab sampling) …

Agronomy, Horticulture and Plant Science Faculty Publications

Plant biostimulants include beneficial fungi and bacteria, and are often applied to foliage to improve crop growth, yield, and/or crop quality. Crop improvements due to biostimulant addition may be modest; therefore, solo applications may not be economical or climate smart. However, biostimulants combined with other postemergence treatments, such as herbicides, may provide an alternative application method, if mixtures do not harm the living organism(s). The growth of Bradyrhizobium japonicum, as a biostimulant surrogate, was assessed in solutions of glyphosate [N-(phosphonomethyl)glycine] and dicamba (3,6-dichloro-2-methoxybenzoic acid), with and without common spray additives (ammonium sulfate [AMS] and nonionic surfactant) in laboratory studies over …

Prevalence Of Adulteration In Dietary Supplements And Recommendations For Safe Supplement Practices In Sport, Andrew R. Jagim, Patrick Harty, Jacob L. Erickson, Grant M. Tinsley, Dan Garner, Andrew J. Galpin

Faculty Scholarship

The prevalence of dietary supplement use among athletes continues to rise with 60–80% of athletes often reporting current or previous use of dietary supplements. While select dietary ingredients have been shown to improve acute performance and enhance training adaptations over time, it is important to still consider the risk vs. reward for athletes before opting to consume a dietary supplement. Previous work has indicated that certain dietary supplements may pose risks for inadvertent doping, may be susceptible to mislabelling, could be banned by certain governing bodies of sport, or pose health risks for certain populations. The purpose of the current …

Fire Effects On Soil Organic Matter In The Creek Fire, Gracie E. Doolin Sep 2023

Fire Effects On Soil Organic Matter In The Creek Fire, Gracie E. Doolin

Master of Science in Environmental Sciences and Management Projects

Wildfires have increased in frequency and severity over the past few decades due to the increased concertation of CO2 emissions from anthropogenic influence. Soil carbon (C) sequestration has been identified as a climate change mitigation strategy; however, the influx of large-scale wildfires has accelerated landscape processes such as erosion, reducing soil aggradation, and soil C and nitrogen (N) protection. This trend is highlighted by the Creek Fire that occurred in September 2020 and burned 379,895 acres in the Sierra National Forest. This research is designed to close the knowledge gap regarding the impact of burn severity on soil organic matter …

Adipocyte-Derived Pxr Signaling Is Dispensable For Diet-Induced Obesity And Metabolic Disorders In Mices, Fang Wang, Jinwei Liu, Rebecca Hernandez, Se-Hyung Park, Ying-Jing Lai, Shuxia Wang, Bruce Blumberg, Changcheng Zhou

UK CARES Faculty Publications

Pregnane X receptor (PXR) is a xenobiotic receptor that can be activated by numerous chemicals including endogenous hormones, dietary steroids, pharmaceutical agents, and environmental chemicals. PXR has been established to function as a xenobiotic sensor to coordinately regulate xenobiotic metabolism by regulating the expression of many enzymes and transporters required for xenobiotic metabolism. Recent studies have implicated a potentially important role for PXR in obesity and metabolic disease beyond xenobiotic metabolism, but how PXR action in different tissues or cell types contributes to obesity and metabolic disorders remains elusive. To investigate the role of adipocyte PXR in obesity, we generated …

Evidence That Bivalve Burrowing Is Mediated By Serotonin Receptors: Activation Of Foot Inflation And Protrusion By Serotonin, Serotonergic Ligands And Ssri-Type Antidepressants In Three Species Of Freshwater Bivalve, Peter P. Fong, Rebecca Blaszczyk, Mary Butler, Jake Stergio Sep 2023

Fong, Rebecca Blaszczyk, Mary Butler, Jake Stergio

Biology Faculty Publications

Foot inflation and protrusion in bivalve mollusks is part of a sequence of events necessary for burrowing into the substrate. Although this process been observed after exogenous exposure to the neurotransmitter serotonin [5-hydroxytryptamine (5-HT)] and by some selective serotonin reuptake inhibitor (SSRI) type antidepressants, no study has shown that foot inflation and protrusion as a precursor to burrowing is under serotonergic control. In order to elucidate the physiological mechanism mediating this response, we tested over 2,000 freshwater bivalves in three species by exposing them to 5-HT, two 5-HT receptor agonists, two receptor antagonists and five antidepressants. Fingernail clams (Sphaerium striatinum), …

Selected Fermented Indigenous Vegetables And Fruits From Malaysia As Potential Sources Of Natural Probiotics For Improving Gut Health, Olaide Olawunmi Ajibola, Raymond Thomas, Babatunde Femi Bakare

Biochemistry Publications

In the Peninsular Malaysia and Northern Borneo island of Malaysia, various rich indigenous leafy vegetables and fruits grow and contribute to the nutritional and dietary values of the population. They have high water contents, thus, naturally vulnerable to rapid food spoilage. Food preservation and processing play a vital role in the inhibition of food pathogens in fruits and vegetables that are prevalent in Malaysia. Lactic acid fermentation is generally a local-based bioprocess, among the oldest form and well-known for food-processing techniques among indigenous people there. The long shelf life of fermented vegetables and fruits improves their nutritional values and antioxidant …

Corticosterone Alters Body Weight, But Not Metabolites, During Chronic Stress, Carolyn M. Bauer, M. A. Oranges, G. Firempong, L. M. Romero Sep 2023

Corticosterone Alters Body Weight, But Not Metabolites, During Chronic Stress, Carolyn M. Bauer, M. A. Oranges, G. Firempong, L. M. Romero

Biology Faculty Works

The stress response is partially mediated by increased levels of circulating glucocorticoids. While the stress response may be adaptive in the short term, chronically elevated levels of glucocorticoids can be pathological. We aimed to verify that chronic stress causes metabolic dysregulation via increased corticosterone (Cort) exposure by monitoring free fatty acid (FFA) concentrations (evidence of fat breakdown), uric acid concentrations (evidence of protein breakdown), and organ weights (furcular fat, abdominal fat, liver, and pectoralis muscle) in chronically stressed juvenile house sparrows (Passer domesticus). The sparrows were chronically stressed for 3 wk by applying a series of rotating mild psychological stressors. …

Western Australian Sheep Producer Survey 2022, Janet Conte, Katherine Davies Sep 2023

Western Australian Sheep Producer Survey 2022, Janet Conte, Katherine Davies

Animal production and livestock published reports

The Western Australian Sheep Producer Survey is conducted to gather information on sheep enterprises and producer practices. It has now been conducted four times, in 2011, 2014, 2018 and 2022 with large random samples of sheep producers in the Medium Rainfall Zone (MRZ) and Cereal Sheep Zone (CSZ).

Some questions were included in all four surveys providing a chance to examine trends over time. Many of the results have remained stable over time giving some confidence that significant changes in survey responses are reflecting trends in the broader population of WA sheep producers.

Once You See It, You Can’T Unsee It?: Racial Justice Activism And Articulations Of Whiteness Among White Collegiate Athlete Activists, Yannick Kluch, Emma Calow, Eric M. Martin, Travis R. Scheadler, Andrew Mac Intosh, Shannon Jolly

Kinesiology Faculty Publications and Presentations

The goal of this study was to examine how athletes holding privileged racial identities understand their whiteness as they engage in racial justice activism. Drawing from 12 semistructured interviews with white collegiate athletes who have engaged in activism for racial justice, we identified four higher order themes which we situate within a broader discussion of how each theme either reinforces or disrupts racial power: articulations of (a) racial consciousness, (b) white privilege, (c) white empathy, and (d) white accountability. While the white accountability theme has the potential to disrupt racial power due to its relying on rigorous self-critique, the remaining …

Biological Sciences Faculty Publications

Phylogenetic relationships within the oestroid subclades Rhinophorinae (Calliphoridae) and Polleniidae were reconstructed for the first time, applying a Sanger sequencing approach using the two protein-coding nuclear markers CAD (carbamoyl-phosphate synthetase 2, aspartate transcarbamylase, and dihydroorotase; 1794 bp) and MCS (molybdenum cofactor sulfurase; 2078 bp). Three genera of Polleniidae and nineteen genera of Rhinophorinae were analyzed together with a selection of taxa representing the major lineages of Oestroidea (non-rhinophorine Calliphoridae, Oestridae, Sarcophagidae, Tachinidae). The selected markers provide good resolution and moderate to strong support of the distal branches, but weak support for several deeper nodes. Polleniidae (cluster flies) emerge as monophyletic …

Dousing The Flame: Reviewing The Mechanisms Of Inflammatory Programming During Stress-Induced Intrauterine Growth Restriction And The Potential For Ω-3 Polyunsaturated Fatty Acid Intervention, Melanie White, Dustin T. Yates

Department of Animal Science: Faculty Publications

Intrauterine growth restriction (IUGR) arises when maternal stressors coincide with peak placental development, leading to placental insufficiency. When the expanding nutrient demands of the growing fetus subsequently exceed the capacity of the stunted placenta, fetal hypoxemia and hypoglycemia result. Poor fetal nutrient status stimulates greater release of inflammatory cytokines and catecholamines, which in turn lead to thrifty growth and metabolic programming that benefits fetal survival but is maladaptive after birth. Specifically, some IUGR fetal tissues develop enriched expression of inflammatory cytokine receptors and other signaling cascade components, which increases inflammatory sensitivity even when circulating inflammatory cytokines are no longer elevated …

The Philippine Agricultural Scientist

A rare, nodule-forming, Gram-positive bacterium (Sub4) was isolated from the forage and cover crop Pueraria phaseoloides, which aligned at 99.26% sequence identity to a partial 16S rDNA sequence of Staphylococcus cohnii sp. This was the first record of a strain/species of Staphylococcus capable of independent, unassisted nodule formation in a legume host. When a nodA gene was sought by PCR using a pair of gene-specific primers synthesized using a related (Firmicutes) Paenibacillus sp. nodA gene, the reaction yielded a PCR product of similar size but a distinct identity. The resulting ~400bp PCR product coded for a likely N-acetylmuramoyl-L-alanine amidase …
