Many organisms rely upon the internal and/or external microbial communities they harbor. Microbial communities which depend upon other organisms for survival are known as host-associated (HA) microbial communities. HA microbial communities differ in diversity and composition from surrounding environmental microbiota and are generally less diverse than surrounding soil microbiomes. Although distinct bacterial profiles exist between host and environment, microbial scale dispersal often links the two. For this reason, host and environmental microbial communities can be correlated (e.g., dominant taxa in HA microbiota are dominant in the host microenvironment or vice versa). The sugarcane aphid (SCA, Melanaphis sorghi), first discovered on grain sorghum in late 2013, has gradually become a major insect pest of sorghum. It inflicts significant damage to the crop year after year, causing millions of dollars in losses throughout the sorghum-producing regions of the United States. Similarly, Armillaria root rot—also referred to as oak root rot—is a major cause of decline in peach (Prunus persica) production in the southeastern United States. This disease is caused by Armillaria mellea and Armillaria tabescens. Current management practices for controlling M. sorghi include the use of synthetic pesticides, resistant varieties, and regular scouting. …

The buildup of protein in tissues and organs plays a major role in unfunctional several serious diseases, including Alzheimer’s disease, Parkinson’s disease, and Type 2 diabetes, each tied to specific proteins that form harmful amyloid deposits. Amyloid aggregation typically follows a sigmoidal kinetic curve, beginning with a slow lag phase, followed by rapid fibril growth, and eventually reaching a plateau where mature, stable fibrils are formed. However, the early stage of aggregation remains particularly difficult to capture experimentally due to the highly dynamic nature and low abundance of small oligomeric species. P. radiata trees are invading Southern Hemisphere Eucalyptus forests within New Zealand and southeastern Australia, resulting in a novel codominant forest system. This codominant invasion is unique relative to other pine invasions throughout the Southern Hemisphere. Ectomycorrhizal fungi (EMF) play an important role in all pine invasions and are found within each instance of invasion. The impact of ectomycorrhizal fungi and Pinus trees upon the properties of the soil within Eucalyptus forests is not very well studied nor understood. The confinement of horses to stalls with limited or no turnout time restricts their natural roaming behaviors, which potentially increases stress and behavioral stereotypies. This study investigated the impact of turnout duration on behavioral and physiological stress indicators in healthy horses. Adult horses (n = 11) were enrolled in a 3 treatment (0-, 15-, and 60-minute turnout/day) Latin square design, including 3 12 d experimental periods over 6 weeks. Horses were acclimated to turnout (24.38 x 64.01m indoor arena) and stalls (2.74 x 4.87m - 3.66 x 3.66m) stalls prior to data collection on day 1. Experienced handlers managed …

Snakes in human care often lack diversity in the diet. Rodents account for the majority of meals, while wild snakes are consuming a varied diet composed of multiple different whole prey species. By feeding only one species of prey, the chance of nutrient deficiencies and other health concerns increases. Two Sweet Bran products were evaluated at 13%, 18%, and 23% inclusion of the diet on total tract digestibility and rumen fermentation parameters. Seven ruminally cannulated, heifers were used in a 6 x 7 row-column design with 21-d periods consisting of 16-d adaptation period followed by a 5-d collection. Dietary treatments were designed in a 2 x 3 factorial arrangement with 6 periods. Factors included two Sweet Bran products that differed in particle size (Sweet Bran-SP; GMD = 1.79 mm or Sweet Bran-LP; GMD = 2.51 mm) and three dietary inclusion (13%, 18%, or 23% of diet dry matter). Improved methane (CH4) mitigation strategies are needed to achieve the greenhouse gas emission goals of the dairy industry. The first experiment was conducted to evaluate the effects of feeding black cumin (Nigella sativa) seed oil and acetate (BCA) on CH4 production and whole-animal energy utilization. Twelve dry Jersey cows were used in a 2-period crossover design study with 28d periods. The BCA treatment was compared to a control diet with corn oil. This study evaluated the amount of detectable DNA accumulated on clean plastic fidget blasters, used to simulate firearms, over a seven-day storage period inside a vehicle. Twelve volunteers were asked to place one fidget blaster each in the glove box and underneath the driver’s seat from behind. After the seven-day period, the volunteers returned the fidget blasters, donated a reference sample, and filled out a questionnaire. The grip, slide, and trigger were swabbed for DNA and extracted. The extracts were quantified to determine the concentration of human DNA, and if the samples met the PCR threshold to be genotyped. The …

Melanaphis sacchari (Zehntner;Hemiptera: Aphididae), sugarcane aphid (SCA), is an invasive phloem-feeder found worldwide with a wide host range of economically important plants including sorghum and sugarcane. Given its high reproductive capacity and ability to rapidly spread over long distances, SCA presents challenges for effective control, leading to substantial economic losses. Peripheral artery occlusive disease (PAOD) is a progressive vascular condition that affects over 200 million people worldwide and approximately 8 million in the United States. It is characterized by atherosclerotic narrowing of peripheral arteries, leading to reduced blood flow, ischemia, and, in severe cases, tissue loss and limb amputation. Although surgical revascularization is the gold standard, many patients—particularly those with comorbidities like diabetes— continue to experience poor outcomes, emphasizing the need for alternative therapies. The stress response of Saccharomyces cerevisiae is a valuable area of study for understanding cellular adaptation mechanisms that govern survival in fluctuating environments. This dissertation investigates stress response regulation in yeast through the roles of Hog1 mitogen-activated protein kinase (MAPK) and the post-transcriptional regulator Mkt1, a key post-transcriptional regulator, in diverse wild yeast isolates. Hog1 is traditionally known for its central role in osmotic stress regulation in the high-osmolarity glycerol (HOG) pathway. This study expands the functional scope of Hog1 to include oxidative and ethanol-induced stress responses.

Fish are a rich source of EPA and DHA, which are well-known long-chain polyunsaturated fatty acids (LC-PUFAs). The sustainable production of these essential fatty acids poses significant challenges, mainly due to our dependence on marine fish and certain nuts. CRISPR-Cas system was utilized to insert the elongation of long-chain fatty acids-like2 (Elovl2) gene from Salmo salar into Danio rerio. Elovl2 was introduced at 10-15-minute post-fertilization, and successful integration confirmed by PCR genotyping, sequencing and expression of EGFP. Melanoma is a highly aggressive form of skin cancer frequently driven by mutations in the BRAF gene, particularly the BRAFⱽ⁶⁰⁰ᴱ variant, which leads to sustained activation of the MAPK signaling cascade. Recent studies implicate the cyclic AMP (cAMP) signaling axis in melanoma progression and therapeutic resistance, highlighting the role of the Inducible cAMP Early Repressor (ICER), a transcriptional antagonist of CREB and CREM. ICER is downregulated during melanomagenesis and subject to post-translational regulation via phosphorylation and ubiquitin-mediated degradation. This dissertation examines decision-making under risk in grain marketing, focusing on how information influences behavior, attitudes toward accessible information, and methods for forecasting commodity basis. Through three essays, it explores the impact of information on hedging decisions, the determinants of information-seeking behavior, the extent to which students can serve as proxies for farmers in risk experiments, and the effectiveness of different forecasting approaches for commodity basis.

Urban forests are valued as green infrastructure for the variety of benefits they provide across ecological, social, public health, and economic domains. To understand the distribution of trees, maps are required. Many methods of mapping tree cover use expensive and data-intensive datasets such as hyperspectral and LiDAR imagery, making these methods inaccessible to municipal forest managers. Pigs are the only mammalian livestock species encoding functional proteins for the second form of gonadotropin-releasing hormone (GnRH-II) and its receptor (GnRHR-II). In the first study, GnRHR-II localized to developing germ cells and the connecting piece of ejaculated spermatozoa. Concurrently, GnRH-II was most abundantly produced in the testis compared with other reproductive tract organs and was present within seminal plasma. Moreover, treatment of extended semen with GnRHR-II analogues altered sperm motility, morphology, and kinematics. Recent advances in computational biology, artificial intelligence, and sequencing technologies have enabled new perspectives for addressing longstanding questions in evolutionary biology. Increased computational power has made large-scale simulations feasible for investigating diverse aspects of evolutionary and phylogenetic modeling. Heir’s property arises when the original owner still holds the title despite their passing, and ownership is distributed among their heirs. This was driven by barriers to retaining land ownership and access to legal support in predominantly African American populations in the early 20th century. This approach allowed land to remain in the original owner's name while granting continued ownership to future generations without formal paperwork. Additionally, allowing limited land access, land management, and disparities in capital access compared to property with direct ownership rights. Our research investigates the structure disorder and magnetic behavior of Li(Mn,Fe)PO₄, an olivine-type material relevant for lithium-ion battery applications. Using single-crystal neutron diffraction at Oak Ridge National Laboratory, we precisely determined Mn and Fe occupancy, revealing a 56% Fe and 44% Mn distribution at the atomic 4c site within the Pnma space group. Additionally, we observed significant lithium site vacancies (78% occupied), influencing the material's electrochemical and magnetic properties. The varying Mn/Fe ratio affected spin reorientation transitions, shifting from antiferromagnetic alignment along the a-axis to the b-axis. Crocodilians inhabit a wide variety of aquatic, marine, and estuarine habitats on 4 continents. Saltwater-adapted species and populations face considerable osmoregulatory challenges in maintaining homeostasis with respect to plasma salt [NaCl] concentration. In addition to the kidneys and cloaca found in all crocodilians, crocodiles possess a salt-secreting lingual gland whose presence and function is less understood in other crocodilians.
