Vaccines have been used to prevent infectious disease for over 200 years. While they are an incredible way to prevent loss of life, they can be improved upon. Researchers around the globe are working to improve the immune response and ultimately the protection provided by existing and new vaccines. In this dissertation, I investigate three ways to improve vaccine immune responses: using an adjuvant, using a prime-boost intramammary delivery route in cows, and using microneedle delivery route. Collagen α1(XI) is a minor fibrillar collagen involved in the critical regulation of collagen fibrils, such as nucleation, assembly, and regulation of fibril diameter. The amino-terminal domain consists of an amino-propeptide (Npp) domain and a variable region, which is the result of alternative splicing from exon 6A, 6B, 7, and 9. The spliced variants involving Npp, with the combinations of exon 6A, 6B, 7, and 9 gives rise to different isoforms with very distinctive chemical nature. Despite major advancements in the management of chronic lymphocytic leukemia (CLL), drug resistance remains a major challenge. CLL resistant to venetoclax (VEN) have limited available therapeutic options, especially when the leukemic cells have dysfunctional p53 and have relapsed on Bruton’s tyrosine kinase inhibitors (BTKi). The broad hypothesis of this thesis is that VEN-relapsed CLL exhibits altered dependence on BCL2 family members, which can be exploited for therapeutic benefit. Colorectal cancer (CRC) is the second leading cause of cancer related deaths with nearly a quarter of patients presenting with metastatic disease at the time of their diagnosis. Therapeutic management of metastatic CRC continues to be a major challenge due to therapy resistance and disease heterogeneity. Due to its central role in tumorigenesis, CRC is commonly treated with MAPK pathway inhibitors (MAPKi). However, clinical trials repeatedly demonstrates that durability of benefit is short-lived in many patients. The introduction of non-native fish species in the freshwater ecosystems of the Philippines presents a clear threat to native ichthyofauna, making verified reports of such introductions all the more critical. Through the examination of a collected specimen and anecdotes from fisherfolk and social media, we present here the first records of the occurrence of the tinfoil barb, Barbonymus schwanefeldii (Bleeker, 1854), in Laguna de Bay, Luzon Island, Philippines. This represents the addition of another non-native fish species in this lake, which is increasingly becoming dominated by non-native and invasive fishes that have been intentionally or accidentally introduced. Williams syndrome (WS) is caused by a rare microdeletion of 25 – 27 genes on chromosome 7q11.23, characterized by a distinctive cognitive profile with considerable phenotypic heterogeneity. However, the genetic factors underlying this phenotypic variability remain unclear. I explored two genetic mechanisms—parent-of-origin (PoO) effects and genome-wide polygenic variation, as measured by polygenic scores (PGSs)—to elucidate possible influences on cognitive variability in WS. The first study examined PoO effects on cognitive abilities in two overlapping samples of children with WS (N = 211, N = 251). Agroecology incorporates ecology and agronomy to inform crop management. Industrial hemp (Cannabis sativa L.) is a prime system to investigate agroecological questions because it has only recently been authorized for commercial cultivation and its sensitivity to biotic and abiotic stressors is unresolved. Here, I investigate how some conventional management choices, including the application of fungicide, plastic mulch, and irrigation, impact the physiology, chemistry, and microbial ecology of industrial hemp. Overall, I found that industrial hemp can be both resilient and resistant to these stressors, an important development for the industry. Cardiovascular disease (CVD) is a multifaceted pathology that affects almost half of Americans ≥20 years old. One modifiable risk factor associated with CVD is air pollution exposure. The intricacy of air pollution exposure and CVD can make it difficult to elucidate on the multitude of mechanisms underpinning these associations. In this dissertation, we selected a relevant exposure (i.e., volatile organic compounds (VOC)) and an intermediate outcome (i.e., autonomic balance) that may represent one of these pertinent mechanisms. In evaluating the potential effects of nitrogen cycles and pH on plant diversity and richness after the removal of Lonicera maackii in a temperate urban forest in Louisville, Kentucky, data was collected including nitrogen, pH, canopy, stem count, and plant surveys. The cutting and treatment with chemicals to remove L. maackii were successful one year after the treatments, but legacy effects ensued about five years thereafter. Native diversity waned as monoculture proliferated in some areas, including second invasions of the invasive. OBJECTIVES: School food can have a profound effect on the population’s health. Thirty million children receive food at school and consume half their daily calorie intake at school. Competitive foods are foods sold outside of school breakfast and lunch, such as those sold in vending machines. A new ruling for Smart Snacks Standards, a set of competitive food guidelines, started in the school year 2014-2015. It is known that competitive foods undermine school lunch participation, and a decrease in school lunch consumption is associated with poorer student health. Lung cancer remains the leading cause of cancer death, often linked to smoking. However, smoking alone does not account for all cases. Exposure to environmental metals like hexavalent chromium [Cr(VI)] also contributes to lung cancer. Cr(VI) causes lung cancer by inducing chromosome instability (CIN), an early hallmark of its carcinogenic mechanism. While it has been established that Cr(VI) induces chromosome instability, the precise mechanisms underlying the chromosome instability are poorly understood. This dissertation investigates the mechanisms of carcinogenesis associated with zinc chromate, a particularly potent particulate form of hexavalent chromium. Arbuscular mycorrhizal fungi (AMF) are obligate biotrophs that form symbiotic associations with the roots of over 70% of terrestrial plants. These symbioses are crucial in plant nutrient acquisition, stress tolerance, and soil health. AMF inoculum could be valuable for ecological restoration, but challenges in culturing and multiplying AMF limit their use in natural habitats. In this project, I investigated methods to characterize and culture AMF native to the sagebrush steppe. For comparison, I also grew a non-native strain of the AMF fungus Rhizophagus clarus. Introduction: As individuals age, they are often confronted with age-related physical declines that can impede their ability to perform activities of daily living (ADLs) and elevate the risk of experiencing falls. While multiple factors may contribute to these physical decrements, muscular power diminishes more rapidly than strength across the lifespan, and serves as a more reliable predictor of fall risk among community-dwelling older adults. Population growth has only amplified the demand, resulting in 430 million tons of plastic produced annually, with 11 million tons ending up in our oceans. The typical modern diet is brimming with food additives used in food processing. Additives such as antibiotic residues, GMOs, pesticide residues, anti-caking agents, emulsifiers, artificial sweeteners, colorants, flavor enhancers, and preservatives. Most of these additives seem to shift the gut microbiome towards more pro-inflammatory signals. When all these additives are considered as an amalgam, such as with the modern processed diet, the ramifications on the gut microbiome are concerning. Obesity, which is characterized by low-level chronic inflammation, is likely exacerbated by these food additives. Consequently, most of these additives should be avoided or banned for optimal health.

The parasite Toxoplasma gondii is a highly ubiquitous parasite that latently infects an estimated 33% percent of the global population. Toxoplasmosis is of obvious clinical significance due to the extremely high risks to the unborn who contract it congenitally. Less attention has been given to a more subtle aspect of latent infection, which is the behavioral alterations that are commonly experienced by those who contract the parasite. The purpose of this study was to assess the relationship between school-based agricultural education (SBAE) teacher Technological Content Knowledge, Pedagogical Content Knowledge, and Technological Pedagogical Knowledge and teachers’ technology integration self-efficacy through the use of TPACK and the Intrapersonal Technology Integration Scale (ITIS). In-service SBAE teachers from four upper-Midwest states were surveyed to assess their current self-efficacy related to the integration and utilization of the educational technology available in their classroom and curriculum. Kansas was historically home to 50 species of freshwater mussels, of which 42 remain extant. The Spring River of southeastern Kansas is home to 34 of the remaining 42 mussels, many of which are imperiled. Elevated metal concentrations that resulted from past lead and zinc mining in the Tri-State Mining District historically imperiled Spring River mussels, but this threat has abated over the last 20 years. Ammonia pollution may have replaced the threat of metals, however, with potentially severe consequences for Spring River mussels. Background/Objectives: Shiga-Toxin producing Escherichia coli (STEC) is a contaminant found in multiple food types and causes multistate outbreaks due to the industrialization of food production. In this report I examine the differences in outbreaks categorized by the regulating agency responsible in outbreak prevention and compare outbreak case counts when organized by positive outbreak investigation and response outcomes.

Background: The fecal immunochemical test for hemoglobin (FIT) is now a widely used non-invasive test in population-based organized screening programs for colorectal neoplasia. The positivity thresholds of tests currently in use are based on the fecal hemoglobin concentration (f-Hb), but the rationale for the adopted thresholds are not well documented. To understand current global usage of FIT in screening programs we conducted an international survey of the brands of FIT used, the f-Hb positivity threshold applied and the rationale for the choice.

Background: Utility analysis is well-established for comparing treatment options but challenging to assess with patients in clinical care. Preference assessment may be more feasible, but it is not yet known whether it correlates with utilities. The aims of this study were to (1) assess women's rankings, ratings, and utilities for tissue-based, implant-based, and no reconstruction after total mastectomy; and (2) explore assessment methods and correlations with clinical and psychosocial factors.

Targeted cancer therapies aim to effectively treat patients with specific biomarker profiles. Nevertheless, these therapies may not always precisely hit their intended targets, leading to uncertainty about the specific subset of patients who will benefit. To address this uncertainty, the identification of sensitive patient subsets in clinical trials becomes crucial. Our proposed phase IIB/III clinical trial design seeks to pinpoint a biomarker signature with precision, ensuring the accurate identification of patients who will respond to a specific treatment. While mostly de novo truncating variants in SCAF4 were recently identified in 18 individuals with variable neurodevelopmental phenotypes, knowledge on the molecular and clinical spectrum is still limited. We assembled data on 50 novel individuals with SCAF4 variants ascertained via GeneMatcher and personal communication. With detailed evaluation of clinical data, in silico predictions and structural modeling, we further characterized the molecular and clinical spectrum of the autosomal dominant SCAF4-associated neurodevelopmental disorder. The molecular spectrum comprises 25 truncating, eight splice-site and five missense variants. Detecting the efficacy signal and determining the optimal dose are critical steps to increase the probability of success and expedite the drug development in cancer treatment. After identifying a safe dose range through phase I studies, conducting a multidose randomized trial becomes an effective approach to achieve this objective. However, there have been limited formal statistical designs for such multidose trials, and dose selection in practice is often ad hoc, relying on descriptive statistics. Despite advancements in catheter technology, intraoperative assessment of radiofrequency ablation (RFA) lesions remains elusive. Prior ex vivo data suggest photoacoustic imaging (PAI) can provide RFA lesion characteristics, but in vivo data are lacking. PAI involves delivering near-infrared light to tissue, leading to transient acoustic waves that can be detected by clinically available ultrasound transducers, providing optically based tissue characterization at depth. Three epicardial RFA lesions were delivered in an open-chest porcine model. In vivo PAI-based measurements of RFA lesion dimensions matched (< 0.7-mm error) gross pathologic assessment, yielding in vivo feasibility data of PAI to provide intraoperative RFA lesion dimensions.

Introduction: Rare solid tumors account for one-quarter of cancers among adults in the United States, but few resources have been devoted to their treatment. We evaluated the efficacy of pembrolizumab, a programmed cell death-1 inhibitor, in patients with rare solid tumors.

Methods: We conducted a phase 2 basket trial that included patients with rare, advanced tumors. Patients were enrolled in the study in nine tumor-specific and a 10th cohort of miscellaneous rare histologies. Patients received pembrolizumab 200 mg intravenously every 21 days. Antibody-drug conjugates (ADCs) are a potent class of antineoplastic drugs that deliver cytotoxic agents via targeting with monoclonal antibodies; however, ADCs are associated with high rates of lung toxicities, limiting the therapeutic potential in many patients. We report a case of a patient with metastatic non-small cell lung cancer taking the ADC telisotuzumab vedotin (Teliso V) plus osimtertinib who developed ADC-induced pneumonitis despite clinical benefit. This patient was able to tolerate treatment for a prolonged period on a novel steroid regimen, using steroids when pneumonitis was asymptomatic but radiologically evident and as a daily suppressant.
