A Peptoid-Based Targeted Drug Delivery System For The Treatment Of Metastatic Cancer, Hugh M. Purdy
A Peptoid-Based Targeted Drug Delivery System For The Treatment Of Metastatic Cancer, Hugh M. Purdy
Chemical Engineering Undergraduate Honors Theses
Development of a targeted drug delivery system is a critical step in the effort to improve cancer treatments. Such a system would greatly reduce the harmful side effects of chemotherapy by delivering toxic drugs directly to cancerous cells. Peptoids—synthetic compounds that can be easily produced from readily available amine monomers—have great potential for use in targeted drug delivery. This project aimed to develop peptoids that would bind to specific proteins expressed on the surface of cancer cells. These peptoids could be combined into a complex that would bind to the proteins with an even greater affinity than the individual compounds. …
Classification And Quantification Of Phenotypic Characteristics Of Normal And Dysplastic Oral Cells, Jackson W. Boice
Classification And Quantification Of Phenotypic Characteristics Of Normal And Dysplastic Oral Cells, Jackson W. Boice
Biomedical Engineering Undergraduate Honors Theses
Introduction Oral cancer accounts for 2.5% of all cancer cases in the United States. The 5 year survival rate or stage 1 or localized oral cancer is 82%, and the survival rate of unstaged oral cancer is only 50%; therefore, early diagnosis is key. Current clinical practices for determining malignancy of tissue include physical examination and surgical biopsy, which can be plagued by high variability between observers and are invasive for the patient. Proflavine intercalates between base pairs of nucleic acids and has excitation and emission peaks at 455 (+/- 20) nm and 515 nm respectively; therefore, it can be …
Optimizing In Vitro Extracellular Matrix Production Using Polymer Scaffolds With Targeted Pore Size, Addison M. Walker
Optimizing In Vitro Extracellular Matrix Production Using Polymer Scaffolds With Targeted Pore Size, Addison M. Walker
Biomedical Engineering Undergraduate Honors Theses
Extracellular matrix (ECM) has been shown to provide the framework needed for healthy tissue to grow after experiencing an injury. The optimization of growing ECM is a pressing concern in the tissue regeneration field. From prior research, an in vitro approach for harvesting ECM has been created using polyurethane (PU) foams seeded with cells. Our approach aimed at discovering if the pore size of the foams increased the amount of recovered ECM. Scaffolds of targeted pore size were created by separating sugar granules using a sieve, packing the sugar into a cylindrical mold, then pouring PU over the sugar. Three …
How Imbalanced Excitation And Inhibition May Cause Low Entropy Brain Dynamics, Qusay Redwan Alfaori
How Imbalanced Excitation And Inhibition May Cause Low Entropy Brain Dynamics, Qusay Redwan Alfaori
Biomedical Engineering Undergraduate Honors Theses
To study the effect of the imbalance of excitation and inhibition neurons on brain entropy using a computer model of a brain. The brain model will be composed of a neural network and entropy of the neural network will be measured quantitatively. Entropy in this context quantifies how many different patterns of activated neurons the brain can create. Imbalance between excitation and inhibition neurons causes the brain to malfunction, which can be an underlying cause of autism.
5ht And The 5ht Receptors- Relevance To Cardiac Valve Disease, Christopher Ryan Martindale
5ht And The 5ht Receptors- Relevance To Cardiac Valve Disease, Christopher Ryan Martindale
Biomedical Engineering Undergraduate Honors Theses
Recent studies have revealed that elevated levels of serotonin or 5HT lead to valve dysfunction. A mouse model was successfully established to investigate the effects on the aortic valve in subjects with elevated 5HT levels along with cardiovascular hypertension. For each two-week trial, eight C57BL/6J mice were divided into the following expermental groups: control, 5HT, angiotensin II, and 5HT+angiotensin II. The drug was delivered to the mice via osmotic pumps (Ang II: 0.4ng/g/min, 5HT:0.0025ng/g/min) or direct injections(Ang II: 0.1 mg, 5HT: 2.12 mg, every other day) and the blood pressure of the mice was continually monitored via the CODA tail …
Modeling Of An Oxygenation-Aided 3d Culture For Functional Beta-Cell Expansion, John Lance Mcreynolds
Modeling Of An Oxygenation-Aided 3d Culture For Functional Beta-Cell Expansion, John Lance Mcreynolds
Biomedical Engineering Undergraduate Honors Theses
Introduction: Diabetes affects millions of people and its prevalence is increasing by 2-5% per year. A promising method for the control of diabetes is the transplant of pancreatic β-cell mass. Unfortunately, there is a shortage of the donor tissue that is needed to replace β-cell mass. A potential source of β-cells is the production of β-cells in vitro, but the proliferation rate of β-cells is very slow and elevating the proliferation rate can result in loss of cell function. We observed that β-cells benefit from being cultured in a 3D environment, but in the 3D culture the β-cells often die …
Beginning Of Malignant Tumor Cells, Emily K. Shields
Beginning Of Malignant Tumor Cells, Emily K. Shields
Biomedical Engineering Undergraduate Honors Theses
Despite advances in cancer treatments, cancer is still ranked the second leading cause of death. Cancers are started by cancer stem cells (CSCs). CSCs, also called tumor-initiating cells, are found in tumors and are responsible for the growth of tumors and the spread of cancer to other parts of the body. CSCs are very resistant, and cannot be killed by chemotherapy or radiation treatments. These cells can come from healthy stem cells. The environment surrounding the healthy stem cells and mature cells can cause mutations in the cell’s genetics, resulting in development of CSCs. The environment can also increase tumor …
Comparing Virus Ultrafiltration Of Bacteriophages With Filtration Of Minute Virus Of Mice, Kent Smith
Comparing Virus Ultrafiltration Of Bacteriophages With Filtration Of Minute Virus Of Mice, Kent Smith
Graduate Theses and Dissertations
Industrial production of protein therapeutics demand rigorous testing and clearance of viruses. The U.S. Food and Drug Administration dictate the purity of pharmaceuticals with regards to viral contamination. As this testing is time consuming and expensive using mammalian cells and viruses, bacteriophages may provide a faster and cheaper alternative for membrane filtration processes. We used ultrafiltration membranes to filter protein solutions with viruses. Two bacteriophages were tested against membranes with two different pore sizes. These membranes were then tested by inverting the membrane's orientation. Flux measurements and log virus removal data were taken. Flux and log virus removal were seen …

In the future, the availability of reliable alternative fuels will be crucial for any country to become energy independent. One such alternative is ethanol as it can be used both as a fuel and as a fuel additive. Most of the ethanol produced in the world today is derived from biomass. The biomass feedstocks and fermentation broths used in ethanol production both contain high amounts of water and therefore, the energy efficiency of the process is lessened by product separation processes (azeotropic separation of water and ethanol) that are non-trivial and highly inefficient (due to the evaporation of water). An …

Synthesis And Characterization Of Extractive Scintillating Resin For Ultra-Trace-Level Quantification Of Uranium In Aqueous Media, C Duval, V Bliznyuk, A Seliman, T Devol, S Husson
Graduate Research and Discovery Symposium (GRADS)
The detection and quantification of radionuclides in aqueous media is of interest to both the Environmental Protection Agency, for matters of public health, and international organizations like the Comprehensive Test Ban Treaty Organization, that use environmental samples to investigate claims of illicit nuclear activities. The current method of quantification involves batch style analytical techniques which utilize bulky instruments. Portable, flow cell detectors utilizing extractive scintillating resin with physically sorbed extractants and fluorophores have been developed; however, this resin shows poor long term stability as the active components leach from the resin over time. There is a need to develop chemically …

Effect Of Surface Parameters On Interfacial Water Film Behavior, Brittany Glatz, Luke Rhym, Sapna Sarupria
Graduate Research and Discovery Symposium (GRADS)
Vapor-to-liquid and liquid-to-solid transitions on mineral surfaces are the primary pathways for phase transitions in atmospheric water. These phase transitions affect the microphysics of clouds and have significant effects on the weather and climate. Our overall goal is to elucidate the mechanisms through which surfaces affect these transitions, and develop predictive abilities to correlate surface properties to the thermodynamics and kinetics of the phase transitions. In this work, we use molecular dynamics simulations to study the structure, and dynamics of water near kaolinite-like surfaces. Kaolinite is the most abundant mineral dust in the atmosphere. We specifically investigate the effect of …

Development And Utilization Of A Pair Of Sol-Gel Entrapped Lipases For Biodiesel Production From High Free Fatty Acid Oils, Cory M. Schwartz
Department of Chemical and Biomolecular Engineering: Dissertations, Theses, and Student Research
Biodiesel, which consists of fatty acid alkyl esters, is one of the most widely adopted and successful renewable fuels. Traditional physiochemical biodiesel production methods require high cost refined feedstocks, and so alternative methods of catalysis and feedstocks have been explored. This research investigated the use of polysiloxane entrapped lipases to catalyze the production of biodiesel from low cost feedstocks.
In this work, lipase from Burkholderia cepacia (lipase PS) and lipase B from Candida antarctica (CalB) were separately entrapped using sol-gel chemistry. Optimal reaction conditions for the esterification of free fatty acids by immobilized CalB with methanol were determined. Immobilized CalB …

Chemical and Biochemical Engineering Faculty Research & Creative Works
Highly porous titania particles were prepared by depositing thin films of titania, using alternating reactions of TiCl4 and hydrogen peroxide, on poly(styrene-divinylbenzene) (PS-DVB) template particles via atomic layer deposition (ALD) at 77 C. The composition of the titania films was verified by XPS analysis, and the titania films were directly observed by TEM. TGA/DSC was used to study the thermal decomposition of the polymer template. Porous titania particles with uniform wall thicknesses were successfully obtained after the template PS-DVB was removed by oxidation in air at 400 C for 24 h. Verification of the resulting porous structure of the titania …

While ultrasound has been used in many medical and industrial applications, only recently has research been done on phase transformations induced by ultrasound. This paper presents a numerical model and the predicted results of the phase transformation of a spherical nanosized droplet of perfluorocarbon in water. Such a model has applications in acoustic droplet vaporization, the generation of gas bubbles for medical imaging, therapeutic delivery and other biomedical applications. The formation of a gas phase and the subsequent bubble dynamics were studied as a function of acoustic parameters, such as frequency and amplitude, and of the physical aspects of the …

Catalytic Hydrogen Transfer Of Ketones Over Atomic Layer Deposited Highly-Dispersed Platinum Nanoparticles Supported On Multi-Walled Carbon Nanotubes, Chengjun Jiang, Xinhua Liang
Chemical and Biochemical Engineering Faculty Research & Creative Works
Hydrogen transfer of ketones, catalyzed by highly dispersed platinum nanoparticles supported on multi-walled carbon nanotubes (MWCNTs), was studied. Pt nanoparticles were deposited on gram quantities of non-functionalized MWCNTs by atomic layer deposition (ALD) at 300 C, using (methylcyclopentadienyl) trimethylplatinum and oxygen as precursors. TEM analysis showed that ~ 1.4 nm Pt nanoparticles were highly dispersed on MWCNTs. The heterogeneous hydrogen-transfer reactions of acetophenone indicated that an acetophenone conversion of 100% and a 1-phenylethanol selectivity of 99.0% could be obtained with a ketone to Pt mass ratio of 24,690 and a ketone to KOH mass ratio of 22 at 150 C …

Environmental Stress Cracking Of Glassy Polymers, Parthasakha Neogi, Gholamreza Zahedi
Chemical and Biochemical Engineering Faculty Research & Creative Works
When a glassy polymer is exposed to an organic solvent, it can crack spontaneously or fail more readily under extension. This is environmental stress cracking. When the solvent diffuses into the polymer, stresses develop because of swelling of the polymer. In addition, fluctuations give rise to stresses, because of the variation in surface tension from changes in the surface concentration of the solute, which is the Marangoni effect. A linear stability analysis is performed to show that such a system is unstable to infinitesimal disturbances. Large tensile stresses arise locally, leading to failures, which will be observed in the interfacial …

Process For The Purification Of Benzphetamine Hydrochloride, Keith Tomazi
Chemical and Biochemical Engineering Faculty Research & Creative Works
The present invention relates to the economical and separation of benzphetamine hydrochloride and methamphetamine by liquid extraction. An extraction process employing a suitable organic solvent and water at a pH in the range of from about 6 to about 8 provides excellent removal of the methamphetamine by dissolution in the water phase while the benzphetamine dissolves in the organic phase. Simple separation of the two phases results in separation of the two amines.

The effects of increasing bioactive glass additions,
SiO2-TiO2-CaO-Na2O-ZnO up to 25 wt% in increments of 5 wt%, on the physical and mechanical properties of hydroxyapatite (HA) sintered at 900, 1000, 1100 and 1200°C for 2 h was investigated. Increasing both the glass content and the temperature resulted in increased HA decomposition. This resulted in the formation of a number of bioactive phases. However the presence of the liquidus glass phase did not result in increased densification levels. At 1000 and 1100°C the additions of 5 wt% glass resulted in a decrease in density which …
