Currently, cryovials on dry ice or liquid nitrogen vapor transportation vessels are primary ways utilized to transport mammalian cells from one location to another. Due to the safety concerns of dry ice and liquid nitrogen, associated costly shipping charges, and limitations of proper packaging and labeling of materials, these conventional shipping techniques are unacceptable. Analysis Of The Relationship Between Flow Patterns In The Post-Stenotic Region Of Model Arteries And Stenosis Severity., Michael Laroche

In this study, contour fields of mean age and turbulence kinetic energy (TKE) in the post-stenotic region of model arteries were analyzed to explore their correlation with FFR. While both high and low FFR cases exhibited similarities with an inner low-age region and an outer high-age region, low FFR cases displayed asymmetry in their low-age regions due to eccentric stenosis. Moreover, gradients of age across the artery diameter were more pronounced for low FFR cases. Additionally, low FFR cases demonstrated significantly higher TKE in the post-stenotic blood jet stream compared to high FFR cases that are reflected in low-age boundary …

This dissertation explores the application of novel molecular dynamics simulation methods to better understand complex systems, with a particular focus on silica materials and protein interactions. Silica materials, known for their versatile surface and solution chemistries, play critical roles in catalysis, chromatography, and wastewater treatment. Despite their widespread use, the molecular-level interactions between silica surfaces and their environments remain poorly understood. To address this knowledge gap, we employed λ-dynamics enabled constant pH molecular dynamics to dynamically model protonation and deprotonation events on silica surfaces. Electroporation is a technique that induces the formation of open pores in cell membranes by the application of an electric field. Electroporation is widely practiced in research and clinical work for transfection of genetic sequences and drug molecule transport through the membrane barrier. However, a full theoretical explanation of the molecular mechanisms and thermodynamics responsible for pore formation, structure, and longevity does not yet exist. Advances in molecular dynamics simulations have enabled theoretical studies of electroporation with previously unobtainable fidelity spanning biologically relevant timescales. With the growth of the distillery and brewery industries and the potential jobs created for chemical engineers, it is imperative to equip the chemical engineering graduates from the University of Louisville with knowledge of bioreaction kinetics and experience in performing calculations to solve for concentration profiles and reaction rates. Unassisted, the lifetime of many proteins outside their natural environment is very short. Scientists generally need to extract the sample, often in remote locations, and then transport, isolate, and store such biospecimens relatively far from the point of origin in order to study them. Currently, the most commonly used methods for storage of biospecimens are cryopreservation or refrigeration which, though effective, have their fair share of flaws. Cryopreservation and refrigeration-based storage of biospecimens requires a great deal of space and other resources to be effective. Over one million invasive coronary angiography procedures are performed annually in patients who experience chest pain or are known to have coronary artery disease. The procedure is carried out to ascertain the degree of arterial blockage (stenosis) that hinders blood flow to the heart. A cardiologist performing the procedure determines the physiological degree of a stenosis by either visual estimation, which is routine practice, or by invasively measuring fractional flow reserve (FFR), which is the current gold standard that has been demonstrated to improve patient outcomes and temper the cost of healthcare. Wall shear stress (WSS) is commonly accepted as the primary influence affecting characteristics of anchored endothelial cells when subjected to fluid flow. Orbital shakers are commonly used to study cellular responses due to their ease of use, ability to run several experiments simultaneously, and since they exert physiologically relevant oscillatory shear. These studies require comprehensive resolution of WSS, however the fluid dynamics inside orbiting culture dishes has not yet been well described since the flow is complex and difficult to quantify analytically.
