Cardiac fibrosis (CF) is a major contributor to the high mortality rate of heart failure which affects over 23 million people worldwide. CF involves cardiac fibroblasts depositing extracellular matrix, made primarily of collagen type I, which stiffens the heart walls over time. As testing new drugs for CF is deterred by the high cost and time required for animal and clinical trials, this research helps to solve the problem by providing an in vitro platform to study fibrosis.

This thesis explores the use of electrochemically produced aqueous bicarbonate as a novel liquid-phase carbon source for enhancing algal biomass productivity. Targeting the freshwater microalgae Chlorella Saccharophila, the study investigates its growth response to varying bicarbonate concentrations and compares the efficacy of liquid bicarbonate delivery with traditional gaseous CO₂ bubbling methods. Results demonstrate that Chlorella Saccharophila can sustain growth solely on bicarbonate, and that bicarbonate-fed cultures yielded higher biomass than those supplied with gaseous CO₂ or hybrid systems. Cardiac fibrosis, a large contributor to heart failure, is the excessive accumulation of extracellular matrix in response to stress or injury. There are no approved treatments for cardiac fibrosis, and targeting specific species involved creates complex problems for drug development, so a computational model of the cardiac fibroblast signaling network can be used to observe the interactions involved in the progression of cardiac fibrosis. Liquid crystals (LCs) constitute a subset of soft matter possessing hybrid characteristics from both liquid and crystalline phases. In this state, the center of masses remains uncorrelated similar to an isotropic phase, while the long axis of molecules aligns in parallel, resulting in molecules exhibiting orientational and positional order, either individually or concurrently. The directional alignment inherent in LCs arises from the asymmetrical shape and structure of the constituent molecules, causing a spontaneous self-assembly into ordered phases. In pursuit of sustainability, we explored replacing conventional dissolved air floatation (DAF) in poultry processing wastewater (PPW) treatment with a precisely tuned 0.02 µm stainless-steel ultrafiltration (SSUF) membrane. SSUF is a robust, homogenously porous membrane with strong chemical resistance, ease of cleaning, and exceptional resistance to organic fouling. Unlike polymeric membranes, it can be regenerated multiple times, making it a cost-effective choice due to its compatibility with harsh chemical cleaning. The PPW used for the study was untreated wastewater from all processing units and post-initial screening. Our study revealed the SSUF membrane’s exceptional efficiency at eliminating contaminants. The microneedle patches are composed of polyvinyl alcohol (PVA), type I collagen (COL), and chitosan (CHI) as base material that carries NSAIDs to achieve therapeutic purposes. Clinical studies have shown that patients undergoing renal replacement therapy are more susceptible to developing hemolysis, or the rupturing of red blood cells. Rapid hemolysis can cause symptoms such as anorexia, vomiting, and even death in severe cases. In this research project, induced mechanical stress on the cell nucleus via synthetic liquid crystals (LCs) was explored. Points of interest were the shape alterations and distortions of the nuclear envelope under strain, providing insight into the mechanics and stiffness of the cell nuclei. LCs are substances with structure and fluidity, and LCs in the nematic phase are aligned in a single direction without layers, inducing shear stress on inclusions. Dissipative particle dynamics (DPD), a coarse grain simulation method, was applied to the membrane formation process of non-solvent induced phase separation (NIPS) to gain further insight on the mechanism of certain variables and how they affect the final morphology. NIPS involves two solutions, an organic polymer dissolved in an organic solvent colloquially called the dope and an aqueous coagulation bath, brought into contact with one another. This study was designed to test the hypothesis that in addition to repairing the architectural and cellular cues via regenerative medicine, the delivery of immune cues (immunotherapy) may be needed to enhance regeneration following volumetric muscle loss (VML) injury. We identified IL-10 signaling as a promising immunotherapeutic target. To explore the impact of targeting IL-10 signaling, tibialis anterior (TA) VML injuries were created and then treated in rats using autologous minced muscle (MM). When a peripheral nerve injury (PNI) occurs, the gold standard for tissue regeneration is the use of autografts. However, due to the secondary effects produced by multiple surgeries involved in the removal and implantation of autografts for very small lesions, it is possible to replace them with the use of Nerve Guide Conduits (NGCs). However, NGCs are limited to short lesions (less than 1 cm). This limitation is caused by the absence of compounds in the extracellular matrix (ECM) that autografts can provide. First, to help survivors of Drug-Facilitated Sexual Assault (DFSA) process their experience by compiling the toxicological, pharmacological, and distribution of the three most used date-rape drugs. Second, to gauge the knowledge and interest of University of Arkansas students regarding drug impairments, sexual assault education, and bystander intervention training. A survey was conducted for the latter and revealed that 91.6% of students believe the University’s existing sexual assault prevention education and bystander intervention training have room for improvement. People with diabetes have trouble regulating their blood glucose levels which typically leads to hyperglycemia. Under normal physiological conditions, erythrocytes can undergo deformations in response to shear stress when passing through capillaries with a smaller diameter. Poorly managed hyperglycemia can lead to the glycosylation of erythrocyte membrane proteins and hemoglobin. In this work, we have made and characterized a pair of immunobiosensors for detecting the green fluorescent protein (GFP) in an aqueous matrix. An anti-GFP antibody-based biosensor was assembled to detect GFP, while a novel peptoid (N-substituted oligomers of glycine designated as IOS-1) biosensor was also assembled for GFP detection. A quartz crystal microbalance (QCM) gold sensor was used as the supporting substrate for self-assembly of the immunobiosensors. Gravimetric measurements of the QCM gold sensor during immunobiosensor construction and operation were available in real-time using a QCM instrument. Microdialysis (µD) sampling is a diffusion-limited sampling method that has been widely used in different biomedical fields for greater than 35 years. Device calibration for in vivo studies is difficult for current non-steady state analytes of interest correlated with both inflammatory response and microbial signaling molecules (QS); which exist in low ng/mL to pg/mL with molecular weights over a wide range of 170 Da to 70 kDa. The primary performance metric, relative recovery (RR), relating the collected sample to the extracellular space concentration varies from 10% to 60% per analyte even under controlled bench-top conditions.
