Legionella pneumophila is a persistent opportunistic pathogen that poses a significant threat to domestic water systems. Previous studies suggest that copper (Cu) is an effective antimicrobial in water systems. A rapid and sensitive quantification method is desired to optimize the conditions of L. pneumophila treatment by Cu and to better understand the interaction mechanisms between Cu and cells. In this study, we developed a highly sensitive single cell (SC)-ICP-MS method to monitor L. pneumophila cell concentration and track their uptake of Cu. The SC-ICP-MS method showed excellent sensitivity (with a cell concentration detection limit of 1000 cells/mL), accuracy (good agreement …

The most common scale-up approach for gas–solids fluidized beds is based on matching the governing dimensionless parameters. In the literature, this approach has been validated only by means of measuring global parameters between different sizes of fluidized beds. However, such global measurements are not sufficient to depict all the interplaying hydrodynamic phenomena and hence verify the scale-up relationships. Therefore, to assess this approach, an advanced gas–solids optical probe and pressure transducer measurement techniques have been applied to quantify local hydrodynamic parameters in two different sized fluidized beds. Four different sets of experimental conditions were designed and conducted to examine the …

Knowledge and proper safety analyses of the gas coolant and heat transport mechanism in the dynamic core of packed pebble bed nuclear reactors pose challenges to the reliable design and efficient operation of these reactors. Therefore, this paper carefully reviews most of the gas coolant mixing and heat transport studies performed for the fluid flow and heat transfer processes in packed pebble bed reactors (PBRs). It begins with a brief introduction and description of nuclear PBRs. The second section summarizes the physical characteristics of packed bed reactors in terms of the bed structure (porosity) and its radial and axial distributions. …

Boron-containing materials are efficient catalysts for the oxidative dehydrogenation of propane to propylene, proceeding via radical intermediates. The radical mechanism is initiated by the solid surface and propagated in the gas phase. It has been hypothesized that the propylene selectivity could be increased by enhancing the gas-phase contributions by favoring the formation of iso- over n-propyl radical intermediates. Indeed, whereas n-propyl radicals can be converted to both propylene and ethylene, iso-propyl radicals yield exclusively propylene. In this contribution, we explore 3D printing to structure the hexagonal boron nitride (hBN) heterogeneous catalyst with high void space. 3D-printed hBN monoliths were found …

Metal-organic frameworks (MOFs) often exhibit an exceptional adsorption-based separation performance for a variety of gases, ions, and liquids. While most radioactive iodine removal studies focus on the capture of radioactive iodine from off-gas streams, few studies have systematically investigated the effect of structure-property relationships of MOFs on iodine removal performance in the presence of interfering ions in liquid solutions. Herein, we investigated the iodide ion (I-) adsorption performance of two model MOFs (e.g., Ni-MOF-74 and Zr-UiO-66) in liquid phase as a function of iodine concentration (e.g., 0.125 to 0.25 and 0.50 mmol/L) and adsorption temperature (e.g., 25 to …

Using microwave irradiation, PtCo alloy nanoparticles were deposited within a few minutes on COOH-functionalized MWCNT supports. The obtained catalysts were used for selective hydrogenation of cinnamaldehyde, a reaction whose products are widely used in various fields. In the selective cinnamaldehyde hydrogenation to cinnamyl alcohol, microwave-prepared catalysts (generically, PtxCoy-MW) outperformed a catalyst prepared by the conventional method (Pt1Co2-con). The highest selective hydrogenation to cinnamyl alcohol, 89%, was obtained using Pt1Co2-MW, while Pt1Co2-con showed a selectivity of 76%. Characterization results confirmed that the microwave prepared samples …

Computer Automated Radioactive Particle Tracking (CARPT) technique has been successfully utilized to measure the velocity profiles and mixing parameters in different multiphase flow systems where a single radioactive tracer is used to track the tagged phase. However, many industrial processes use a wide range of particles with different physical properties where solid particles could vary in size, shape and density. For application in such systems, the capability of current single tracer CARPT can be advanced to track more than one particle simultaneously. Tracking multiple particles will thus enable to track the motion of particles of different size shape and density, …

Current sorbents investigated for light olefin/paraffin separation usually suffer from low selectivity. Besides, multicomponent analysis of this important separation is usually overlooked in the literature. To enhance the separation efficiency of zeolite 13X, we developed a series of Fe2O3/13X composite sorbents and assessed their separation performance using binary, ternary, and multicomponent gas mixtures of C2H4, C2H6, CH4, and H2. In these composites, nano- and micro Fe2O3 particles (NPs and MPs) with varied loadings were used, while Fe was also ion-exchanged into …

Gas drying by adsorption on molecular sieves is one of the first unit operations in natural gas processing. Nevertheless, the thermal swings during regeneration combined with the presence of a complex gas mixture may lead to early adsorbent fouling. The shaping of adsorbent materials is crucial not only to reduce pressure drop in columns, but also for enhanced thermal and mechanical properties. In this work, synthesized sodium-based zeolite Linde Type A (Na-LTA) in powder was shaped by extrusion into cylindrical pellets using a clay binder followed by calcination at 673 K for 24 h. The produced 2-mm pellets had binder …

Surfactants could react with adsorbates on carbonate rock surface to alter wettability from oil-wetness to water-wetness, which is effective to enhance oil recovery. Surfactant huff-puff treatment is mostly applied for this purpose and the resulting surfactant performance is the outcome of complex interfacial processes. Currently, the effect of important parameters on surfactant performance is not completely reported and the contribution of each parameter to surfactant performance is hard to be quantified. Traditional methods to optimize surfactant performance are time-consuming and show strong dependency on extensive experiments. in this paper, we address these problems from machine learning (ML) perspectives. Several ML …

Transcription factor, STAT3, is inappropriately expressed in cancer cells, and has contrasting activation in 2D versus 3D microenvironments. 2D plates are often used for drug screening and do not always recapitulate in vivo responses. To combat inaccurate 2D drug studies, a 3D hydrogel was created to support the growth of cancer cells into a tumor-like environment. The hydrogel consists of a biocompatible dextran homopolysaccharide, cell adhesion RGD sequences, and crosslinker MMP labile peptides. A pH dependent reaction couples the RGD sequences to dextran then the polymers are crosslinked into a gel. Crosslinking is accomplished using terminal cysteine peptide sequences, allowing …

High viscosity friction reducers (HVFRs) have been gaining more attention and increasing in use as proppant carriers. Reusing of produced water has also been driven by both environmental and economic benefits. However, high total dissolved solids (TDS) of produced water and high reservoir temperature significantly decrease the viscoelastic properties of HVFRs even for the cationic ones, which further reduce their proppant transport capabilities. Increasing loadings of HVFRs also has very limited effects on improving their proppant transport performances under high-TDS and high-Temperature conditions. The ultimate objective of this experimental study is to investigate the injection rate effects on the proppant …

Bubble and slurry bubble column reactors (BCRs/SBCRs) are used for various chemical, biochemical, and petrochemical applications. They have several operational and maintenance advantages, including excellent heat and mass transfer rates, simplicity, and low operating and maintenance cost. Typically, a catalyst is present in addition to biochemical processes where microorganisms are used to produce industrially valuable bio-products. Since most applications involve complicated gas-liquid, gas-liquid-solid, and exothermic processes, the BCR/SBCR must be equipped with heat-exchanging tubes to dissipate heat and control the reactor's overall performance. In this review, past and very recent experimental and numerical investigations on such systems are critically discussed. …

The effects of various material compositions and reservoir environments on the ultimate strength and swelling kinetics of a commercial preformed particle gel (PPG) have been investigated. This study used different ratios of acrylamide and acrylic acid copolymers with a specific crosslinker concentration. Results have indicated that increasing the acrylic acid proportion enhances the PPGs' ability to swell but weakens their network structure. In contrast, increasing the crosslinker content decreases the swelling ratio and increases the gel strength. The highest equilibrium swelling capacity among the six preformed particle gel samples was obtained for PPG2, which has the highest acrylic acid amount …

Preformed particle gels (PPG) have been widely applied in oilfields to control excessive water production. However, PPG has limited success in treating opening features because the particles can be flushed readily during post-water flooding. We have developed a novel micro-sized Re-crosslinkable PPG (micro-RPPG) to solve the problem. The microgel can re-crosslink to form a bulk gel, avoiding being washed out easily. This paper evaluates the novel microgels' transport and plugging performance through super-permeable channels. Micro-RPPG was synthesized and evaluated for this study. Its storage moduli after fully swelling are approximately 82 Pa. The microgel characterization, self-healing process, transportation behavior, and …

Cross-Seeding Of Protegin-1 And Amyloid Beta To Inhibit Amyloid Beta Aggregation, Sarah Robinson
Williams Honors College, Honors Research Projects
Amyloid beta (Aβ) is considered a major component in the progression of degenerative disease like Alzheimer’s disease. In an attempt to cure Alzheimer’s before it has presented, this project proposes a study on the effects of protegrin-1 (PG-1) on the aggregation of Aβ. By cross-seeding to form a PG-1- Aβ heterocomplex, it is believed that aggregation of Aβ on the human brain can be inhibited, thus helping to stop the neurodegeneration caused by Alzheimer’s disease. To complete this project, an initial literature review of previous work done by the graduate student leading the project will be completed. After this, experiments …

Background: Hepatitis B virus (HBV) infection develops as an acute or chronic liver disease, which progresses from steatosis, hepatitis, and fibrosis to end-stage liver diseases such as cirrhosis and hepatocellular carcinoma (HCC). An increased stromal stiffness accompanies fibrosis in chronic liver diseases and is considered a strong predictor for disease progression. The goal of this study was to establish the mechanisms by which enhanced liver stiffness regulates HBV infectivity in the fibrotic liver tissue. Methods: For in vitro studies, HBV-transfected HepG2.2.15 cells were cultured on polydimethylsiloxane gels coated by polyelectrolyte multilayer films of 2 kPa (soft) or 24 kPa (stiff) …

Rationale: Trauma, surgery, and infection can cause severe inflammation. Both dysregulated inflammation intensity and duration can lead to significant tissue injuries, organ dysfunction, mortality, and morbidity. Anti-inflammatory drugs such as steroids and immunosuppressants can dampen inflammation intensity, but they derail inflammation resolution, compromise normal immunity, and have significant adverse effects. The natural inflammation regulator mesenchymal stromal cells (MSCs) have high therapeutic potential because of their unique capabilities to mitigate inflammation intensity, enhance normal immunity, and accelerate inflammation resolution and tissue healing. Furthermore, clinical studies have shown that MSCs are safe and effective. However, they are not potent enough, alone, to …

The settling velocity of the proppant is one of the key outputs to describe their proppant transport capability. However, it is influenced by many factors such as fluid properties, proppant properties, and fracture properties. Many empirical/physics-based models and correlations to predict particle settling velocity have been developed. However, they are usually based on certain assumptions and have applicable limits. In contrast, machine learning models can be considered as a black box. The objective of this …

Volumetric muscle loss (VML) remains one of the few skeletal muscle injuries without a reliable and repeatable treatment. In large volume muscle injuries, muscle fibers as well as the surrounding connective tissue are damaged, preventing therapeutic muscle stem cells, called myogenic progenitor cells (MPCs), from reaching the injury site and initiating repair. There is a clinical need to rapidly fabricate in vitro muscle tissue constructs that mimic the native tissue organization, with aligned myotubes, for insertion and integration at the patient’s injury site. In this dissertation, we utilize the MPC’s natural propensity to close gaps across an injury site to …

Optical 3D printing is a branch of Additive manufacturing (AM) that utilizes low-waste production leading to efficient and rapid prototyping. Currently, this method of AM is associated with mainly petroleum-based resins. Bio-resin polymerization is an emerging research area for finding replacements for petroleum-based resins, but curing the resin with laser energy is not normally a practical use case. Current optical 3D printing uses lasers to cure liquid resins to form solid 3D objects in a layer-by-layer process which is time consuming, defect prone, and a leading limiting factor for high volume production; so, finding a way to incorporate bio-resin into …

Renewable fuels from lignocellulosic biomass are an appealing option because they can seamlessly integrate into the existing fuel distribution infrastructure. Lignocellulosic biomass constitutes nonedible plant material obtained from plant cell walls. The natural recalcitrance of lignocellulosic biomass poses a challenge in accessing the cell wall carbohydrates during biochemical conversion. Despite various approaches, enzymatic hydrolysis of lignocellulosic biomass remains economically impractical due to incomplete knowledge about biomass recalcitrance and the influence of environmental factors on biomass quality.
The first goal of this dissertation was to construct a microfluidic imaging reactor to better understand the tissue-specific deconstruction of plant materials. Confocal laser …
