A Small-Angle Neutron Scattering Study Of Γ-Crystallins Near Their Isoelectric Point, Paul Petitt, Michael Eugene Edwards, Daniel Forciniti

In this paper, a small-angle neutron scattering study of γII-crystallins near their isoelectric point is presented. The experiments were carried out using protein concentrations of 5.7-85.7 mg/ml at temperatures in the range 11-60°C. The experimental data were analyzed using an ellipsoidal model for intraparticle correlations and the mean spherical approximation for interparticle correlations. Our studies revealed that γII-crystallins have a thick hydration layer, which is possibly due to the special arrangement of polar and ionic groups on their surface. Recombinant human protein C (rhPC) secreted in the milk of transgenic pigs was studied. 'Ikansgenes having different regulatory elements of the murine milk protein, whey acidic protein, were used with cDNA and genomic human protein C (hPC) DNA sequences to obtain lower and higher expressing animals. The cDNA pigs had a range of expression of about 0.1-0.5 g/l milk. Two different genomic hPC pig lines have expressed 0.3 and 1-2 g/l, respectively. The rhPC was first purified at yields greater than 60 per cent using a monoclonal antibody (mAb) to the activation site on the heavy chain of hPC. Subsequent …

The impact of antibody orientation on immunosorbent efficiency was quantitatively assessed. A pH-dependent murine monoclonal antibody (Mab) against human protein C (hPC), recombinant hPC (rhPC) and two different immobilization chemistries and matrices were used as model systems. The lysyl groups of the rhPC were covalently modified with an acetic acid ester of N-hydroxysuccinimide and this modified rhPC was used as a Fab masking agent (FMA). The FMA was used to mask the antigen bind-ing regions (Fab) of the Mab prior to and during covalent immobilization. Thereafter, the residual active sites of the support were in-activated and the FMA was removed. …

Multicomponent adsorption equilibrium data are essential for the reliable design of processes and equipment for gas separation by adsorption. We discuss techniques for the measurement and analysis of multicomponent adsorption equilibrium data, and present a comprehensive set of equilibrium data for the adsorption of oxygen and nitrogen on 5A-zeolite.

A mathematical model describing dynamic adsorption in columns with spherical bidisperse perfusive or spherical bidisperse purely diffusive adsorbent particles is used to study the effects of the structural parameters of the adsorbent particles and of the parameters of the adsorption mechanism on the performance of chromatographic systems involving the adsorption of bovine serum albumin onto anion-exchange porous adsorbent particles. Four structural parameters are studied: the particle diameter, d(p), the macropore void fraction, ε(p), the microparticle diameter, d(m) and the parameter φ which affects the value of the permeability of the adsorbent particle. Purple membrane (PM, λ" role="presentation">λmax" role="presentation">max 570 nm) of H. halobium on treatment with sulphuric acid changes its colour to blue (λ" role="presentation">λmax" role="presentation">max 608 nm). The purple chromophore can be regenerated from the blue chromophore by exogeneous addition of anions such as CI−" role="presentation">− and HPO42−" role="presentation">2−4. Chloride ion is found to be more effective than the dibasic phosphate ion in regenerating the purple chromophore. Nevertheless, one thing common to the anion regeneration is that both CI−" role="presentation">− and HPO42−" role="presentation">2−4 show marked pH effect. At pH 1.0 the efficiency of …

A mathematical model of multi-component dynamic adsorption in columns with spherical perfusive and spherical purely diffusive adsorbent particles with a bidisperse porous structure is constructed, solved, and used to study the performance of a binary adsorption system in a single-column fixed-bed process and in a two-column periodic countercurrent process in which the adsorbent is equally distributed over two beds. A mathematical model describing the dynamic elution (elution stage) in columns with spherical bidisperse perfusive or spherical bidisperse purely diffusive adsorbent particles is presented and its solution is obtained numerically. The desorption of bovine serum albumin (BSA) from spherical anion-exchange porous particles was studied for different values of the intraparticle Peclet numbers of the adsorbate, (Peintra)a, and eluent, (Peintra)e, and of the microsphere diameter, dm. A sub grid turbulence model for the lattice Boltzmann method is proposed for high Reynolds number fluid flow applications. The method, based on the standard Smagorinsky sub grid model and a single-time relaxation lattice Boltzmann method, incorporates the advantages of the lattice Boltzmann method for handling arbitrary boundaries and is easily implemented on parallel machines. The method is applied to a two-dimensional driven cavity flow for studying dynamics and the Reynolds number dependence of the flow structures. The substitution of other sub grid models, such as the dynamic sub grid model, in the framework of the LB method is discussed.

The Isothermal Decomposition of Hydrogen Peroxide on a CuCr Catalyst in a Laboratory Scale Trickle Bed Reactor Was Used to Test Model Predictions of the Dependence of Liquid Reactant Conversion on Space Time for Different Operating Conditions. It is Assured that the Decomposition of Hydrogen Peroxide is a First Order Liquid-Limited Reaction. Comparison of Model Predictions and Experimental Data Indicates that Both External Mass Transfer Effects and Incomplete External Catalyst Wetting Need to Be Accounted For. Dudukovic's (1977) Approximate Model for the Catalyst Effectiveness Factor Adequately Simulates Both Effects.

Unsteady conjugate mass transfer between a spherical bubble, droplet, or particle and a surrounding fluid flow with chemical reaction in the continuous phase has been analyzed. An algorithm based on spectral methods has been developed and implemented for the simulation of the interphase mass transfer. The influence of Pe (convection) and Da (rate of external reaction), but in particular of D (diffusivity ratio) and H (distribution coefficient), which characterize the coupling features of the conjugate mass transfer, is studied in a wider range of parameters than earlier investigations. In this paper we analyze the reflection of non-reacting shock waves as oblique detonation waves. The 2D, steady, inviscid analysis makes use of detonation polar diagrams to identify incident shock angles that lead to reflected detonations, with and without detonation Mach stem formation. The implications of the results for design and operation of superdetonative ram accelerators are explored. The influence of the projectile forebody geometry, the propellant composition, and the takeover velocity is described. We conclude with a discussion of the significance of these parameters on superdetonative ram accelerator unstart phenomena.

In this paper, we discuss the cyclic operation of an enhanced combustion pulsejet engine (ECPE) applicable in the Mach number range of 0 to 3. The geometry of this engine naturally provides for integration into a new combined cycle engine with ramjets. As in the operation of conventional pulsejets, an ECPE provides engine aspiration, charge compression, combustion and expansion without the use of turbomachinery. The use of modern electronics, fuel injection, and feedback control offers the potential for performance improvements that could make the ECPE a cost-effective competitor for the turbojet engine. Conventional path tracking algorithms used in homotopy continuation systems sometimes miss roots owing to jumping from one segment of the homotopy path to another even if there exists homotopy paths to the roots. A robust path tracking algorithm is proposed which loses significant efficiency only on those portions of the path where segment jumping is likely to occur. The method presented here basically performs the predictor-corrector procedure using the Euler predictor and the Newton corrector. Any available algorithm can be used to control the step size. In this article we report steady, two-dimensional, inviscid solutions for the near field and far field of a supersonic reactive flow over a variable-double-ramp geometry. The incident shock wave compresses and heats the reactants that will combust after flowing some induction length. Upon reaction, a detonation wave forms and intersects the leading wave at some distance from the ramp surface. In this article, reaction-polar diagrams are developed, and the detonation branch solutions are used to investigate the wave interaction processes that may lead to a steady three-wave structure. Interphase mass transfer has been analyzed for a reactant being transferred from a sphere into a quiescent, infinite continuous phase where it undergoes a chemical reaction. Ranges of the diffusivity ratio D, the distribution coefficient H, and the Damköhler number Da have been delineated, within which the overall resistance to mass transfer may be reasonably ascribed to the sum of the individual resistances in each of the two phases. Experimental total pressure measurements of propyne mixed with propane, propene, and propadiene are presented for five isotherms from 253 to 353 K. Thermodynamically consistent vapor compositions and saturated phase densities are calculated using a cubic equation of state with three isothermal pure component parameters and two binary mixture parameters. The precision vapor—liquid equilibrium cell and experimental procedure which are particularly useful for measurements involving close boiling light hydrocarbons are described, and the experimental and modeled results are presented. © 1995, American Chemical Society. The sorption kinetics of benzene in silicalite have been measured gravimetrically using large silicalite crystals of sizes 350 µm × 105 µm × 105 µm and 270 µm × 70 µm × 70 µm in the temperature range 283–343 K. Experiments were performed under conditions that ensured isothermal operation with intracrystalline diffusion control. This paper presents a parallel solution method of large sparse systems of linear equations arising in the context of a chemical process flowsheeting application on a message passing multicomputer. To maximize the performance, the algorithm uses a novel matrix decomposition and solution method, called parallel two-phased LU decomposition, which schedules the concurrent tasks in a maximally overlapping manner, and at the same time, tries to minimize the interprocessor data dependencies and obtain optimal load balancing. ABSTRACT The serious shortage of human organs available for transplantation has engendered a heightened interest in the use of animal organs (xenografts) for transplantation. However, the major barrier to successful discordant xenogeneic organ transplantation is the phenomenon of hyperacute rejection. Hyperacute rejection results from the deposition of high-titer preformed antibodies that activate serum complement on the luminal surface of the vascular endothelium, leading to vessel occlusion and graft failure within minutes to hours. Although endogenous membrane-associated complement inhibitors normally protect endothelial cells from autologous complement, they are species restricted and thus confer limited resistance to activated xenogeneic complement. Projectile angular scattering is investigated following electron capture by protons from circular Rydberg atoms oriented along a quantization axis perpendicular to the incident projectile direction. We find strong left-right scattering asymmetry as a function of the orientation angle at collision speeds comparable to the electron orbital speed. The capture cross section is dramatically enhanced due to velocity matching when the angular momenta of the proton and the electron are aligned. The scattering asymmetry exhibits characteristic impact parameter dependencies suitable for studying three-body dynamics influenced by both long and short range forces of comparable strength. © 1994 IOP Publishing Ltd.
