Background: Wilson disease (WD) is an autosomal recessive disorder that results in excessive hepatic copper, causing hepatic steatosis, inflammation, fibrosis, cirrhosis, and liver failure. Previous studies have revealed dysregulation of many farnesoid X receptor (FXR) metabolic target genes in WD, including the bile salt exporter pump, the major determinant of bile flow.
Methods: We tested the hypothesis that the FXR-cistrome is decreased in Atp7b-/- mice in accord with dysregulated bile acid homeostasis.
Results: FXR binding within Atp7b-/- mouse livers displayed surprising complexity: FXR binding was increased in distal intergenic regions but decreased in promoter regions in Atp7b-/- versus wild-type mice. …

Eating disorders can cause a disturbance in how the health of food is viewed (Cerniglia et al., 2017). If a person has disturbances in their eating behaviors that can cause harm to their health, but the behaviors do not fit into the criteria of a specific eating disorder, they fall into the category of a disordered eating pattern (Academy of Nutrition and Dietetics, 2018). Because of the stress the college environment can cause students, some college students may cling to anything they can control, which may be reflected in their dietary habits. Orthorexia Nervosa (ON), a disordered eating pattern, is …

Background/objectives: Diet quality and sleep regularity both influence cardiovascular disease (CVD) risk and may influence each other, but there is scarce evidence for their joint or interacting associations in relation to CVD. We assessed these associations in the Multi-Ethnic Study of Atherosclerosis (MESA).
Methods: Participants free of CVD with valid diet and sleep measures in 2010-2013 were included and followed through 2020 for detection of incident CVD (188 events detected over 8.8 years among 1782 participants; 55% women). Sleep timing and duration regularity were assessed via the intra-individual SD of sleep onset time and duration across 5- to 7-day actigraphy. …

Biliary atresia (BA) is a pediatric liver disease that often necessitates parenteral nutrition (PN) to support growth due to impaired liver function. While soy-based lipid emulsions (SLE) are commonly used in PN, they may contribute to cholestatic liver injury. In contrast, mixed lipid emulsions (MLE) show promise in preventing cholestasis in infants without BA, potentially by restoring bile flow. However, their effectiveness in patients of complete bile duct obstruction, as seen in BA, remains uncertain. To explore the potential benefits of MLE in BA, we utilized a neonatal pig model of bile duct ligation (BDL). Pigs underwent either BDL or …

The extent of genetic variation and its influence on gene expression across multiple tissue and cellular contexts is still being characterized, with germline Structural Variants (SVs) being historically understudied. DNA methylation also represents a component of normal germline variation across individuals. Here, we combine germline SVs (by short-read sequencing) with tumor DNA methylation across 1292 pediatric brain tumor patients. For thousands of methylation probes for CpG Islands (CGIs) or enhancers, rare and common SV breakpoints upstream or downstream associate with differential methylation in tumors spanning various histologic types, a significant subset involving genes with SV-associated differential expression. Cancer predisposition genes …

Assessment Of The Risk Factors Affecting Diabetes Outcome In Kumasi Metropolis Of Ghana, Audrey Jacobs
Honors College Theses
This paper examines Kumasi Metropolis, Ghana risk factors for diabetes outcomes, considering the prominent determinants such as geographic location, income, and educational level. Such determinants play a role when it comes to shaping health outcomes, with research indicating positive correlation between per capita expenditure on healthcare and life expectancy. In spite of that, inequalities do occur, even in developing nations such as Ghana, where minimal health care investment impacts poor health outcomes. Socioeconomic factors, including low living conditions, poor access to healthcare, and illiteracy, reinforce health disparities, especially in rural areas. Informed by a review of past research, we hypothesized …

Exploring The Relationship Between Diet And Mental Health, Emory Layne
Honors Theses
Dietary patterns in Western societies have shifted over the past century due to the rise of food manufacturing and technological advancements. Ultra-processed foods now account for approximately half of the daily energy intake in high-income countries. Similarly, the national mental health crisis emerged in the 21st century and worsened after the COVID-19 pandemic. The changes in diet have impacted the gut microbiome, which interacts with the brain through the bidirectional gut-brain axis. Research suggests the concept of the gut-brain axis is a principal indicator of diet’s impact on mental health. However, this area of research is still emerging and requires …

Background/Objectives: Disorders of gut-brain interaction (DGBI), characterized by chronic abdominal pain and significant disability, affect 15-20% of children and adults and continue into adulthood in ~60% of cases. Costs for adults reach USD 30 billion per year, yet effective management strategies are elusive. Studies support using cognitive behavioral therapy (CBT), but abdominal pain only improves in ~40% of patients. Dietary management (low FODMAP diet; LFD) has also shown promise but it is effective in only a similar percentage of patients. Studies suggest that biologic factors (biomarkers) contribute to CBT response. Similarly, gut microbiome composition appears to influence abdominal pain …

OBJECTIVES: School food can have a profound effect on the population’s health. Thirty million children receive food at school and consume half their daily calorie intake at school. Competitive foods are foods sold outside of school breakfast and lunch, such as those sold in vending machines. A new ruling for Smart Snacks Standards, a set of competitive food guidelines, started in the school year 2014-2015. It is known that competitive foods undermine school lunch participation, and a decrease in school lunch consumption is associated with poorer student health. Additionally, an increase in competitive foods tends to be of nutrient-poor …
