Malignant melanoma arising in the oral cavity is particularly rare. The clinical presentations of malignant melanoma can range from a benign-looking pigmented macule or nodule to a non-pigmented neoplasm which can be categorized into primary or metastatic lesions. Less than 2% of all melanomas show no pigmentation and could lead to misdiagnosis. Objective: To present a benign looking but life-threatening malignancy of melanotic melanoma. Case report: A 59-year-old female patient was referred to a regional hospital for further treatment of a localized gingival swelling with no ulceration at the anterior maxilla. Lactobacillus reuteri is known as probiotics that effective to improve oral health. Objective: To analyze the effect of L. reuteri Indonesian strain, towards Enterococcus faecalis and Candida albicans biofilm growth. Methods: This study was conducted using biofilm assay. The purpose of this investigation was to evaluate the efficacy of a glass ionomer varnish in preventing demineralization of enamel adjacent to and some distance away from an orthodontic bracket. Forty extracted human molars were divided into four different treatment groups (varnish in distilled water, varnish in acetic acid, enamel in acetic acid, and enamel in distilled water). Two 3 x 3 mm graphite windows were drawn on the tooth, one adjacent to the bracket and one 3 mm away from the first window, indicating the scanning area. Background: Gingivo-buccal oral squamous cell carcinoma (OSCC-GB) is the most common cancer among men in India and is associated with high mortality. Although OSCC-GB is known to be quite different from tongue cancer in its genomic presentation and its clinical behavior, it is treated identically as tongue cancer. Oral Squamous Cell Carcinoma (OSCC) affects 53,000 Americans each year making it the sixth most common cancer in the United States. The incidence rate may be even higher since current screening methods are underutilized and physical examinations can misdiagnose malignant oral tumors as benign inflammatory lesions. Recent advances in genetic, protein, and extracellular vesicle biomarker detection techniques can monitor the transformation of healthy cells into tumors before terminal stages arise. Biomarkers can be easily extracted from biofluids such as saliva making them less invasive than traditional tissue biopsies. Dental Pulp Stem Cells (DPSCs) are non-embryonic, mesenchymal stem cells that may have significant potential for therapeutic and regenerative biomedical applications. Studies of DPSC differentiation have demonstrated the potential to form many tissue types, including neural, osteogenic and vascular precursors using cytokines and growth factors, such as Vascular Endothelial Growth Factor (VEGF). Eight previously isolated Dental Pulp Stem Cell (DPSC) isolates were grown in culture and treated with VEGF to evaluate any effects on growth, viability or biomarker expression. Background: Porphyromonas gingivalis, a major pathogen associated with chronic periodontitis, secretes variety of proteins, majority of which begins with glutamine. Several of these proteins were found with pyroglutamate (pGlu) at N-terminus suggesting the presence of this posttranslational modification pathway in P.gingivalis. The observation that N-terminal glutamine is over-represented as the first amino acid after signal peptide cleavage, and subsequent confirmation of pGlu formation on the nascent protein via mass spectrometry, led us to conclude that an enzyme must be present as the executor of this reaction. Objective: Extracellular vesicles derived from oral cancer cells, which include Exosomes and Oncosomes, are membranous vesicles secreted into the surrounding extracellular environment. These extracellular vesicles can regulate and modulate oral squamous cell carcinoma (OSCC) progression through the horizontal transfer of bioactive molecules including proteins, lipids and microRNA (miRNA). The primary objective of this study was to examine the potential to isolate and evaluate extracellular vesicles (including exosomes) from various oral cancer cell lines and to explore potential differences in miRNA content. Since periodontitis is a chronic, progressive condition, uninterrupted disease progression leads to irreversible oral damage; therefore, periodontitis often reduces the oral health-related quality of life. Streptococcus sanguinis, an abundant and benign inhabitant of the oral cavity, is an important etiologic agent of infective endocarditis, particularly in people with pre-disposing cardiac valvular damage. Although commonly isolated from patients with IE, little is known about the factors that make any particular S. sanguinis isolate more virulent than another or, indeed, whether significant differences in virulence exist among isolates. To investigate the virulence of multiple isolates, a variation of the Bar-seq (barcode sequencing) method was employed. A recent study demonstrated an association between the Porphyromonas gingivalis (Pg)infection and the progression of ESCC (Gao et al., 2016). However, how Pg infection affects the nature of ESCC remains unknown. We examined the effects of Pg on the proliferation and chemotherapy drug-induced apoptosis of ESCC cells and elucidated the signaling molecules involved in these processes. Streptococcus mutans (S. mutans) is the main etiological bacteria present in the oral cavity that leads to dental caries. All of the S. mutans in the oral cavity form biofilms that adhere to the surfaces of teeth. Dental caries are infections facilitated by the development of biofilm. An esterified derivative of epigallocatechin-3-gallate (EGCG), epigallocatechin-3-gallate-stearate (EGCG-S), was used in this study to assess its ability to inhibit the growth and biofilm formation of S. mutans. Background: Porphyromonas gingivalis is an important etiologic agent of chronic periodontitis, an infectious disease defined by destruction of supporting tissues of the teeth. Marijuana is a risk factor for chronic periodontitis, although underlying mechanisms are poorly understood. As phytocannabinoids have been ascribed antiinflammatory properties, we hypothesized that cannabidiol exposure would lead to altered immune response to P. gingivalis infection facilitating bacterial persistence and, subsequently, increased alveolar bone loss in cannabidiol (CBD)-exposed mice. While Streptococcus sanguinis plays a beneficial role in the oral cavity as a competitor of Streptococcus mutans and Streptococcus sobrinus, the bacteria that produce dental caries, it can cause deadly infective endocarditis if given the opportunity to colonize the vegetations that form over damaged endocardial tissue. Pre-existing heart conditions, surgery, and intravenous drug use predispose individuals to endocarditis. S. sanguinis growth and consequential virulence is significantly impeded by restriction to manganese. The oral cavity harbors hundreds of microbial species that are present either as planktonic cells or incorporated into biofilms. The majority of the oral microbes are commensal organisms. Those that are pathogenic microbes can result in oral infections, and at times can initiate systemic diseases. Biofilms that contain pathogens are challenging to control. Many conventional antimicrobials have proven to be ineffective. Recent advances in science and technology are providing new approaches for pathogen control and containment and methods to characterize biofilms. Drug abuse, currently a national epidemic affecting millions of Americans, causes numerous health issues including increased dental disease. There are several factors which can contribute to an increase in caries and missing teeth. One of the most discussed mechanisms is xerostomia. Drug abusers tend to have large sugar intakes which exacerbates the problems caused by xerostomia. Drug abusers are also at risk for oral infections associated with dental decay due to their altered saliva composition. Objectives: This study aimed to i) determine the kinetic changes of immune cell phenotypes, including an exhaustion marker PD-1, in spleens of murine sepsis ii) investigate the susceptibility against secondary fungal infection after sepsis, and iii) investigate the efficacy of an anti-PD-1 treatment in post-sepsis fungal infection. Methods: Cecal ligation and puncture (CLP) was used as a sepsis model and splenocytes post-CLP were assessed by flow cytometry. Periodontium is a specialized tissue laying between tooth and surrounding bone. It functions as force cushion. In particular stimulation including mechanical force, periodontal ligament (PDL) cells which are mechanosensitive cells have the ability to secrete specific cytokines and proteins. PDL cells also have the potential to differentiate into osteoblast-like cells or fibroblast-like cells. Periodontal diseases result from the interplay between the dysregulation of the host inflammatory response and the actions of a dysbiotic bacterial community. These chronic inflammatory conditions affect persons on a worldwide scale. P. gingivalis is strongly implicated as a key periodontal pathogen in the more severe manifestations of periodontal diseases such as periodontitis. A core element of P. gingivalis pathogenicity is its dysregulation of innate immunity, including suppression of IL-8 production by gingival epithelial cells. The focus of this project is to understand the role of Retinoic Acid (RA) produced by RDH10-mediated metabolism of Vitamin A in palate development. For 80 years now, we have known that dietary restriction of RA/Vitamin A can cause cleft palate but we have failed to understand how RA deficiency causes cleft palate.
