Impaired Neutrophil Extracellular Trap-Forming Capacity Contributes To Susceptibility To Chronic Vaginitis In A Mouse Model Of Vulvovaginal Candidiasis, Junko Yano, Paul L. Fidel

School of Dentistry Faculty Publications

Vulvovaginal candidiasis (VVC), caused by is characterized by aberrant inflammation by polymorphonuclear neutrophils (PMNs) in the vaginal lumen. Data from the established murine model shows that despite potent antifungal properties, PMNs fail to clear due to local heparan sulfate that inhibits the interaction between PMNs and , resulting in chronic vaginal immunopathology. To understand the role of neutrophil extracellular traps (NETs) in defense against at the vaginal mucosa, we investigated the NET-forming capacity of PMNs in chronic VVC-susceptible (CVVC-S/C3H) and -resistant (CVVC-R/CD-1) mouse strains. Immunofluorescence revealed the formation of NETs (release of DNA with PMN-derived antimicrobial proteins) in PMN cocultures …

The Effect Of Parabiotic Reuterin On The Expression Of Genes Involved In Candida Albicans Biofilm Formation: An Ex Vivo Study, Anastasya Muna Riad, Armelia Sari Widyarman

Journal of Dentistry Indonesia

Candida albicans has a number of properties, including resistance to various antimicrobial agents, which allow it to survive in the root canals. Lactobacillus reuteri plays a role in maintaining oral health through interactions with the oral microbiome. L. reuteri has potential as a preventive and therapeutic agent against inflammatory diseases. Objective: The aim of this study was to investigate the effect of irrigation with a reuterin-containing solution on BCR1, ACE2, EFG1, and TEC1 gene expression in C. albicans root canal biofilms. Methods: L. reuteri was cultured in MRS broth and incubated anaerobically for 24 hours at 37°C. C. albicans was …

Persistent Endodontics Pathogens Biofilm Inhibited By Lactobacillus Reuteri Indonesian Strain, Armelia Sari Widyarman, Njoo Kiky Elysia Lazaroni

Journal of Dentistry Indonesia

Lactobacillus reuteri is known as probiotics that effective to improve oral health. Objective: To analyze the effect of L. reuteri Indonesian strain, towards Enterococcus faecalis and Candida albicans biofilm growth. Methods: This study was conducted using biofilm assay. Each E. faecalis ATCC-29212 and C. albicans ATCC-10231 were cultured in Brain Heart Infusion Broth, distributed into 96 well-microplate and incubated for 24h, 37°C, anaerobic condition for E. faecalis and aerobic condition for C. albicans. L. reuteri LC382415 (Indonesian strain) was cultured in deMann Rogosa Shar pe Broth and diluted into different concentrations (108,106, and 104 …

Mass Spectrometric Analyses Of Surface Proteins From Candida Albicans After Treatment With Histatin 5, Shirley Fan

Electronic Thesis and Dissertation Repository

Current drug therapies have lost their effectiveness in controlling the fungal species known as C. albicans; therefore, the need for newer drugs becomes crucial. Histatin 5 (Hst 5), a naturally occurring peptide found in the oral cavity, has demonstrated strong effects on controlling candidiasis. Here, we have proposed that the adhesion of cells through surface proteins is an important process. To allow the analysis of surface proteins, the concept of on-whole cell tryptic digestions were developed. This on-target approach allows for isolation of the peptides of interest, while greatly reducing digestion times.

To study the effects of Histatin 5 …

Candida albicans is an opportunistic pathogen that is present in the normal flora in a majority of individuals. One key factor in C. albicans virulence is the ability to change its morphology from yeast to an elongated or hyphal form. The regulation of this morphogenesis relies in part upon quorum sensing (QS) molecules. C. albicans often exists as part of a mixed culture alongside other microbes and is influenced by their presence as well as the presence of QS molecules that they produce. In this study, a library of diploid homozygous transcriptional regulator knockout (TRKO) mutants were screened to identify …
