Background: Atherosclerosis is an inflammatory cardiovascular disease that progresses with ageing. Glycosylation plays a significant role in inflammation and contributes to the pathogenesis of ageing-related diseases, including atherosclerosis. Cluster of differentiation 147 (CD147), a glycosylated agonist of matrix metallopeptidase 9 (MMP9), serves as an inducer of atherosclerosis and atherothrombosis. However, the effect of glycosylated CD147 (GlyCD147) on atherosclerosis remains unclear. This research aims to identify whether GlyCD147 is associated with atherosclerosis. The pathogenesis of acute rheumatic fever (ARF) is poorly understood, limiting the development of immune-modulating therapies to treat disease and prevent progressive heart damage. Here, participants with definite ARF were compared to other severe acute paediatric conditions and matched healthy controls by profiling circulating immune molecules and cells to inform disease mechanisms and potential druggable pathways. ARF shared immunological similarities with other inflammatory conditions, including elevated serum IL-6 and an increased frequency of circulating CD4+ T cells. Rheumatoid arthritis (RA) is a chronic autoimmune disease characterised by persistent inflammation, joint destruction, and notable differences in prevalence and severity between sexes. Elevated levels of pro-inflammatory cytokines, such as interleukin (IL)-1, IL-6, and tumour necrosis factor alpha (TNF-α), are well established mediators of RA pathology. Although extensive research has elucidated their pathological roles, the influence of sex on cytokine profiles and RA outcomes remains undefined.

This review examines preclinical and clinical studies analysing sex-specific cytokine responses in RA. Findings reveal that female patients and experimental models exhibit higher concentrations of pro-inflammatory mediators, with oestrogen and androgen signalling playing relevant …

Objectives: This study aimed to investigate the impact of Obstructive Sleep Apnea (OSA) on immune cell profiles by utilizing both the National Health and Nutrition Examination Survey (NHANES) dataset and a two-sample Mendelian Randomization (MR) method. Methods: We included 20,732 participants from the NHANES to explore the relationship between symptom defined OSA and peripheral immune cells. Additionally, we conducted a two-sample MR analysis to investigate the effect of OSA on 35 summarized peripheral immune cell properties and 731 immunophenotypes. Introduction. Systemic lupus erythematosus (SLE) is a complex chronic autoimmune disease with diverse clinical manifestations. Treatment stabilization is crucial for optimal disease control, but the consistency of immunosuppressive therapy may be affected by non-ideal conditions, including drug unavailability or adverse effects. This study aimed to assess the association between unplanned immunosuppressant switching and increased disease activity in SLE patients.

Methods. A retrospective cohort study of SLE patients meeting the inclusion criteria, whose disease activity was evaluated using the MEX-SLEDAI score during immunosuppressant substitution. Medical case management (MCM) plays a critical role in supporting engagement in HIV care; however, differences in implementation across agencies may influence service delivery and outcomes. Existing research has focused primarily on outcomes such as retention in care and viral suppression, while less is known about how the MCM intervention is implemented in practice. Studies characterising the immunoglobulin (Ig)-bound microbiota apply varying methodologies, making comparisons difficult. This scoping review synthesised evidence on Ig–microbiota binding patterns in maternal and infant contexts, identified recurrent Ig-bound and -unbound bacteria across studies, and highlighted knowledge gaps for further study. Nine articles investigating Ig–microbiota binding patterns in stool or breastmilk samples in mothers or infants were included. Ig–microbiota associations were influenced by sample type, Ig-subclass, genetics, and diet. The most important antibody was IgA, with partial functional redundancy with IgM, while IgG appeared more selective for pathobionts. Toll-like receptors are a class of innate immune pattern recognition receptors that detect pathogen-associated molecular patterns and regulate downstream inflammatory responses. Toll-like receptor 10 remains the only human TLR whose ligand specificity, cellular localization, and immunological function are not fully defined. Emerging evidence suggests that TLR10 may play an anti-inflammatory role and can heterodimerize with TLR2 to modulate immune signaling. Previous studies from our laboratory demonstrated that stimulation with heat-killed Mycobacterium tuberculosis promotes TLR2-TLR10 heterodimerization and trafficking to the cell surface. Type 1 diabetes (T1D) is an autoimmune disease targeting the insulin-producing beta cells of the pancreas. The discovery of insulin revolutionized the treatment of T1D, but current research into T1D has progressed from disease management to possible prevention or delayed onset of diagnosis. One drug currently in use is teplizumab, an anti-CD3 monoclonal antibody that has been shown to delay the onset of T1D in some, but not all, at-risk individuals. Introduction: Previous research has found that the calcineurin-mediated immunosuppressants cyclosporine A (CsA) and tacrolimus inhibit binge alcohol drinking in mice. Further research has shown that this effect is mediated directly in brain, as intracerebroventricular administration also significantly decreases drinking. Introduction: The infant gut microbiome plays a critical role in immune system development during the early-life “window of immune education.”1 Microbial colonization influences immune tolerance through mechanisms such as regulatory T-cell (Treg) induction, short-chain fatty acid (SCFA) production, and mucosal immune regulation. Cold agglutinin disease is a rare subtype of autoimmune hemolytic anemia (AIHA) characterized by complement-mediated hemolysis triggered by cold temperatures. Because of its rarity and variable presentation, cold agglutinin disease is frequently underrecognized, often leading to extensive and sometimes unnecessary diagnostic evaluations. By recognizing cold agglutinin disease as a differential diagnosis in patients with recurrent admissions for hemolysis, clinicians can avoid unnecessary workups, consults, and distress for their patients.

Background: Food allergy negatively impacts numerous children in the United States. Oral immunotherapy (OIT) and biologic therapy (omalizumab) are emerging promising strategies for desensitization. This literature review aims to compare the effectiveness of omalizumab and oral immunotherapy in pediatric populations.

Hypothesis: If the use of omalizumab therapy is used to treat children with food allergies compared to OIT, then there would be higher desensitization, lower post-treatment IgE levels, higher IgG4 levels, and decreased chance of a positive skin prick test.

Sepsis is a leading cause of morbidity and mortality worldwide, driven by a dynamic immune continuum that progresses from early hyperinflammation to profound immunosuppression. This review examines the three overlapping phases of sepsis: the inflammation-mediated cytokine storm driven by dysregulated PRR activation, NF-κB signaling, and NETosis; apoptosis-driven lymphopenia involving caspase-mediated lymphocyte death, dendritic cell dysfunction, and metabolic exhaustion; and prolonged immunoparalysis culminating in persistent inflammation, immunosuppression, and catabolism syndrome (PICS). Key mechanisms linking these phases, including the co-existence of hyper- and hypo-inflammatory states, are synthesized alongside relevant clinical biomarkers such as IL-6, HLA-DR, and single-cell immune profiles. Identifying Major Sources Of Leptospira Interrogans In San Juan Bay And Río Grande De Loíza Estuaries Of Puerto Rico, Logan Mccullers, Dave Bachoon

This study evaluated the presence of pathogenic Leptospira interrogans, potential carriers, and environmental factors influencing their occurrence in the San Juan Bay Estuary (SJBE) and Río Grande de Loíza (RGDL) watersheds of Puerto Rico from 2024–2025. Detection of this pathogen from water samples was performed using the lipL32 PCR assay. The likely hosts of L. interrogans were determined using fecal source tracking (FST) assays for human (HF-183), canine (BacCan-UCD), equine (HoF597), and rat mitochondrial DNA NADH dehydrogenase subunit 2 gene (RatMt) markers.
