The objective of the present study was to mature and fertilize primary sheep oocytes in vitro and to develop them to the blastocyst stage. Ovaries from slaughtered Kıvırcık ewes were used. The ovaries were transported to the laboratory in a vacuum flask in which 30-35 °C PBS (phosphate buffered saline) was included. The cumulus-oocyte complexes were collected by rupturing and washing the follicle wall with oocyte washing medium. They were then selected under a stereo microscope and maturated for 23-24 hours in medium 199 supplemented with sodium pyruvate, follicle stimulating hormone (FSH), luteinizing hormone (LH) and 10% fetal calf serum …

Evaluation Of Echocardiographic Findings Via Doppler Technique In Rabbits, Melahat Toker, Vedat Sağmanligi̇l

Turkish Journal of Veterinary & Animal Sciences

In this study, echocardiograpic examination was performed on 20 healthy New Zealand rabbits. All rabbits were examined by the color Doppler technique using a 5-7.5 Mhz transducer. Images were obtained from parasternal long axis, parasternal short axis, apical four-chamber and modified four-chamber sections. The anatomy and function of hearts were examined in each rabbit individually. The values obtained were not significantly different from those obtained in dogs and cats. In addition, the changes seen in the right ventricle parameters were consistent with the changes in the left ventricle parameters.

Considering the relation between the endocrine and immune systems, the effects of zeranol, a synthetic oestrogen compound, on the growth rate and nonspecific immune system in rainbow trout (Oncorhynchus mykiss, Walbaum) were investigated. For this reason, fish were fed a diet containing 0, 2.5, 5, 10 and 20 ppm zeranol for 21 days. This study was conducted to determine the fattening performance of Sönmez type lambs weaned on average at 60±5 days old. Sixty lambs were put into four different pens, as single males (15), single females (15), twin males (15) and twin females (15), and the fattening period was 56 days in length. The daily weight gains were 282.14 g, 242.49 g, 207.31 g and 179.99 g for single males, single females, twin males and twin females, respectively. This study is based on the examination of 759 chubs from Karasu River (40° 45´ 33´´ E, 39° 56´ 16´´ N) between November 1995 and October 1997. This population comprised seven age groups of males and nine of females. The second age group was dominant in the population. Individuals were composed of 50.46% males and 49.54% females. Comparison Of In Vitro Dry Matter Degradation Of Four Maize Silages Using The Menke Gas Production Method, Adem Kamalak, Yavuz Gürbüz, Heather Finlayson

Four maize silages from the UK and France were evaluated using the gas production method to estimate the rate and extent of dry matter degradation in vitro. There were significant differences in gas production after 17 h incubation. The silages showed similar times to produce 50, 95 and 100% of total gas production, whereas the lag time for Ema was significantly (P < 0.001) lower than that for Volgate. The potential gas production value (a + b) for Nancis was significantly (P < 0.05) lower than that for Volgate. There was a significant correlation (r = 0.996, R^2 = 90.71, P < 0.001) between gas production and total VFA production. There were considerable differences in the fermentability of carbohydrates between different maize silages. Nancis and Ema had more readily fermentable carbohydrate, which yielded high propionate production and a shorter lag time. 6196 and Volgate had more slowly fermentable carbohydrate, which did become available to microorganisms but after a longer lag time.

In this study, the anaesthetic effects and reliability of the combination of etomidate, which is a non-barbiturate injectable anaesthetic, and alfentanil, which is a strong narcotic analgesic, in anaesthesia induction in dogs were investigated. Twenty dogs were used in this study. Following intramuscular administration of atropine at a dose of 0.05 mg/kg, diazepam was given intravenously at a dose of 0.5 mg/kg. Following tranquillisation, etomidate at a dose of 2mg/kg and alfentanil at a dose of 0.02mg/kg were injected intravenously. The Effect Of Concentrate Feeding Levels On The Postweanining Performance Of Holstein Friesian Calves, Mete Yanar, Olcay Güler, Bahri̇ Bayram

The influences of different levels of concentrate on the growth characteristics of Holstein Friesian calves reared under an original calf-rearing program were investigated in this study. Forty-one new-born calves (20 male and 21 female) were allocated to the four concentrate groups: 2, 2.5, 3 kg/calf and ad libitum. The calves were weaned at 5 weeks of age, and the trial lasted for 6 months. The results with regard to weights at 6 months of age revealed that an increasing amount of concentrate supplementation resulted in a significantly (P < 0.01) higher live weight. Furthermore, daily weight gains between birth and 6 months of age was also significantly higher (P < 0.01) in the ad libitum group than in the 2, 2.5 and 3 kg/day groups. The highest overall gains in body measurements, for example body length and heart girth, were obtained from calves fed concentrate ad libitum. However, feed efficiency ratios were not significantly influenced by the various amounts of concentrate offered to calves. The overall results suggest that the appropriate level of concentrate for the new calf-rearing program should be ad libitum.

In this paper, four diagnostic techniques were compared in terms of their ability to detect BVD virus in buffy coats frozen for more than one year. The samples were selected according to their initial test results. Forty-five BVDV positive and 15 BVDV negative buffy coats were used. The techniques used were the virus isolation-immunoperoxidase test (IPX), immunoperoxidase monolayer assay (IPMA), indirect immunoperoxidase monolayer assay (IIPMA) and reverse transcription-polymerase chain reaction (RT-PCR). We investigated the effects of chronic exposure to dietary cadmium (Cd) on the levels of lipid peroxidation (LPO) and reduced glutathione (GSH) associated with activities of glutathione peroxidase (GSH-Px) and the catalase (CAT) enzyme in the liver and kidneys of rabbits. Twenty 10-month-old clinically healthy New Zealand white rabbits (10/group) of 3545 ± 250 g average body weight were used. The control group was allowed to take food and tap water ad libitum, while the experimental group was given tap water containing 200 ppm CdCl_2/L for 28 weeks. Lipid peroxidation contents increased significantly in the liver (p < 0.001) and kidneys (p < 0.005) of Cd-treated rabbits. Glutathione levels decreased significantly in both the liver (p < 0.001) and the kidneys (p < 0.005) of the experimental group. Cadmium treatment caused a significant decrease in the activities of GSH-Px and CAT in the liver (p < 0.001, p < 0.01) and kidneys (p < 0.005, p < 0.1) of the rabbits. These results indicated that a Cd-induced increase in LPO was associated with reductions in GSH-Px and CAT enzyme activities, and in GSH levels in the liver and kidneys of rabbits.

Seasonal changes in serum concentrations of the testosterone in males, 17\beta-estradiol in females and triglyceride and cholesterol in both sexes of Capoeta capoeta umbla were determined. Peak concentrations of testosterone (2.828 ± 0.278 ng/mL) and 17\beta-estradiol (3.182 ± 0.116 ng/mL) occurred in May. The concentrations of testosterone and 17b-estradiol were significantly correlated with the gonadosomatic index in both sexes. After the spawning period, steroid hormone levels significantly diminished (P < 0.05). The triglyceride concentration was highest in April (607.611 ± 30.146 mg/dL). The serum cholesterol level showed a rising trend until April and reached peak values during this month (393.000 ± 8.991 mg/dL). In the spawning month (May), a significant decrease was observed in triglyceride and cholesterol concentrations (P < 0.05). These results suggest that seasonal changes in both serum lipids and steroid hormones were associated with reproductive activity in C. c. umbla.

During recent decades, the study of fatty acids (FAs) belonging to the n-3 family in seafood has intensified due to their beneficial effect on cardiovascular disease. This study was undertaken to determine the FA composition of muscle lipids of three different farm raised trouts (Salvelinus alpinus, Salmo trutta fario and Oncorhynchus mykiss) fed the same diet and raised under the same conditions. Palmitic acid (16:0) in total saturated fatty acid (SFA) and oleic acid (18:1 n-9) in monounsaturated fatty acid (MUFA) were the most abundant FAs and significant differences (P < 0.05) were observed between fish species. The differences in total SFA among the trouts were significant (P < 0.01), and O. mykiss had the highest (31.92%) SFA content while S. trutta fario showed the lowest in their edible muscle tissue. However, there was a significantly (P < 0.01) opposite observation for MUFA among the trouts, in which S. trutta fario had the highest content (41.90%) of MUFA in their muscle tissue while O. mykiss had the lowest (30.81%). Lens Abnormalities In Slaughtered Holstein And Brown Swiss Beef Cattle In Konya, Zeki̇ Oğurtan, Cengi̇z Ceylan, İlhami̇ Çeli̇k, Emrah Sur

A total of 1000 beef cattle were examined after being slaughtered, of which 373 were Holstein and 627 were Brown Swiss. The high incidence of lens abnormalities was observed to be as follows: coloboma (20.4%), microphakia (6%) and cataract (3.9%) in Holsteins for a total incidence rate of 30.3%, and coloboma (5.6%), microphakia (2.6%) and cataract (4.2%) in Brown Swiss for a total incidence rate of 12.4 %. Holsteins were more frequently affected than Brown Swiss with coloboma and microphakia, but not with unilateral cataract formation. Effects Of Progesterone And Testosterone On The Hyaluronidase Activities And Sperm Characteristics In Sheep, Sadetti̇n Tanyildizi, Tanzer Bozkurt

The aim of this study was to determine the effects of progesterone and testosterone hormones on the hyaluronidase activities of serum and semen and on sperm characteristics in sheep. Thirty healthy Akkaraman sheep were used. The progesterone and testosterone were injected intramuscularly at the rate of 0.5 mg/kg and 5 mg/kg, respectively. Then the blood and semen samples were obtained from the sheep at 1, 2, 4, 24, 48, 72 and 168 h. The serum hyaluronidase activities of ewes and rams in progesterone and testosterone groups increased significantly (p < 0.01, p < 0.001) when compared with the control groups at all times. Additionally, there were significant (p < 0.001) increases in the levels of semen hyaluronidase activity in the progesterone group except at 4 h. Furthermore, semen hyaluronidase activities in the testosterone group were detected to decrease significantly (p < 0.01) in comparison with the control rams except at 4 h. The use of progesterone and testosterone caused a significant (p < 0.05, p < 0.01) increase in spermatozoa motility, respectively. However, sperm concentrations were determined to decrease significantly (p < 0.01, p < 0.001) in both groups. Although the values for semen volume decreased significantly (p < 0.05, p < 0.01) in the progesterone group, the same values were observed to increase significantly (p < 0.01) in the testosterone group except at 48 h when compared with the control groups. The results of this study showed that there were no significant correlations between semen hyaluronidase activities and sperm characteristics. In conclusion, the use of both hormones did not have any deleterious effects on hyaluronidase activities but for the semen samples in the testosterone groups. However, progesterone and testosterone hormones cause harmful effects on the fertilization ability of rams due to decreased sperm concentration values.

Fish were exposed to three different sublethal doses in order to determine the effects of Cypermethrin on some biochemical parameters (Ca-Calcium, P-Phosphorus, Na-Sodium and TP-Total Protein) of rainbow trout (Oncorhynchus mykiss). After 15 days exposure, Ca and P decreased, while TP and Na took various values depending on the doses of synthetic pyrethroid.

Male Wistar albino rats, bred and fed under standard laboratory conditions, were administered for 20 days two dosages [Group I: 750 mg/kg body weight (n = 15) and Group II: 1500 mg/kg body weight (n = 15)] of oral hexachlorobenzene resolved in corn oil. Control group animals (n = 15) received only corn oil for 20 days. Twenty-four hours after the administration of the last dose, renal tissue samples, obtained by laparotomy, were fixed in 3% glutaraldehyde in phosphate buffer and 1% aqueous osmium tetroxide. A specimen of Squatina aculeata (sawback angelshark) was caught in a trawl on 15 May 1997 in Iskenderun Bay at about 120-200 m depth and recorded for the first time from the eastern Mediterranean coast of Turkey. Thus, a new species of fish was recently added to the chondrictyes fish fauna in the eastern Mediterranean Sea off Turkey.

In this study a series of weight-length relationships are presented for 13 fish species caught between September and December 2000 off the south coast of İskenderun Bay. The samples were collected with longlines. The values of the exponent b in the weight-length relationship, W = aL^b, ranged from 2.391 to 3.675 and the median value was 2.714.

A New Fish Record From Turkish Seas: The Blackfin Sorcerer Nettastoma Melanurum Rafinesque, 1810 (Nettastomidae), Nuri̇ Başusta, Cemal Turan, Mevlüt Gürlek

Two and five specimens of the blackfin sorcerer, Nettastoma melanurum, were trawled on 25 and 30 April 2001 respectively at Samandağ, Hatay (36º10'N, 35º20'E; 36º05'N, 35º40'E) at about 80-200 m depth. It is recorded for the first time from the eastern Mediterranean coast of Turkey. Thus, a new species and family have been added to the fish fauna in Turkish seas.

A female specimen of Dioctophyma renale was recovered from the abdominal cavity of a stray dog necropsied in Istanbul, Turkey on 8 June, 1999. The nematode was 55.5 cm in lenght, with a diameter of 0.7 (0.6-0.8) cm. This is the first case report of D. renale from Turkey.
