Swelling subsequent to hyposmotic conditions activates a process of volume regulation present in most cell types. This volume adjustment is accomplished by osmolyte translocation toward the extracellular space to reach a new osmotic equilibrium. Molecules involved in this homeostatic mechanism have been broadly classified into two categories: organic and inorganic osmolytes. Inorganic osmolytes comprise mainly the intracellular ions K+ and Cl–. Cell swelling–induced activation of separate K+ and Cl– channels has been described in most preparations. The cytochrome P450 (CYP) 2J2 arachidonic acid epoxygenase gene was down-regulated at a pre-translational level in human hepatoma-derived HepG2 cells incubated in a hypoxic environment; under these conditions, the expression of c-Jun and c-Fos mRNA and protein was increased. The 5′-upstream region of the CYP2J2 gene was isolated by amplification of a 2341 bp fragment and putative regulatory elements that resembled activator protein-1 (AP-1)-like sequences were identified. From transient transfection analysis, c-Jun was found to strongly activate a CYP2J2–luciferase reporter construct, but co-transfection with plasmids encoding c-Fos or c-Fos-related antigens, Fra-1 and -2, abrogated reporter activity. Nearly all human immunodeficiency virus (HIV) infections are acquired mucosally, and the gut-associated lymphoid tissues are important sites for early virus replication. Thus, vaccine strategies designed to prime virus-specific cytotoxic T lymphocyte (CTL) responses that home to mucosal compartments may be particularly effective at preventing or containing HIV infection. The Salmonella type III secretion system has been shown to be an effective approach for stimulating mucosal CTL responses in mice. The present invention provides a method to detect the presence of a microorganism or agent in an animal. The method encompasses placement of devices at various locations where the animal resides so as to induce the animal to initiate contact with the device. As a result of this contact, the animal deposits various microorganisms and agents on the device. The device is then tested for the presence of the particular microorganism or agent of interest.

D-Cycloserine (DCS) targets the peptidoglycan biosynthetic enzymes D-alanine racemase (Alr) and Dalanine: D-alanine ligase (Ddl). Previously, we demonstrated that the overproduction of Alr in Mycobacterium smegmatis determines a DCS resistance phenotype. In this study, we investigated the roles of both Alr and Ddl in the mechanisms of action of and resistance to DCS in M. smegmatis. We found that the overexpression of either the M. smegmatis or the Mycobacterium tuberculosis ddl gene in M. smegmatis confers resistance to DCS, but at lower levels than the overexpression of the alr gene. A high percentage of the world’s population are infected with herpes simplex virus type 1 (HSV-1), and infection can cause a variety of disorders (35, 187). Recurrent ocular HSV-1 is the leading cause of infectious corneal blindness in industrialized nations (190). In a murine model, ocular infection induces autoimmune disorders, leading to corneal antigen destruction and stromal keratitis (275). HSV-1 infections also cause gastrointestinal disorders, esophageal disorders, and approximately 25% of the genital herpes infections (67, 158). HSV-1 infections can cause sporadic encephalitis, but this is relatively rare compared to other diseases resulting from infection. Further evidence for its involvement …

Complete sequencing and annotation of the M. leprae genome has provided new information related to proteins constituting its hypothetical proteome. Since M. leprae can not be grown in vitro, novel approaches are needed to determine which proteins are expressed during infection and whether these proteins are related to pathogenesis. The aim of this study was to determine the diagnostic importance of erythrocyte glutathione peroxidase activity (GSH-Px) in lambs with white muscle disease (WMD) and the changes in some serum parameters (glucose, creatinine, total protein, albumin, cholesterol, triglyceride, Ca, inorganic P, Na, K, Cl, AST, ALT, ALP, LDH and CPK) in healthy and white muscle diseased lambs. In this study, which was conducted to detect the critical control points and to investigate the direct and cross-contamination sources in white pickled cheese production, 21 control points were examined for aerobe mesophilic counts, staphylococcus-micrococcus, coagulase-positive staphylococcus, enterobacteriaceae, coliform bacteria, E. coli, enterococcus, yeast and mould counts. It was determined that the high microbiological load of raw milk affected the microbiological quality of pasteurized milk negatively, and that the production pipe-line after pasteurization, and equipment such as vats, plastic wraps, pressing cloths, starter culture and the hands of the workers, were direct contamination sources. This study was conducted on sheep and goats to investigate theileriosis and tick infestations around Kayseri between August 1997 and October 1999. During this period a total of 300 small ruminants consisting of 250 sheep and 50 goats were microscopically examined for Theileria species. Blood smears were prepared from the punctured ear of each animal for microscopic examination. At the same time, ticks were collected from the animals. A total of 426 serum samples were examined from 9 broiler and 7 broiler breeder flocks showing clinical signs of swollen head syndrome (SHS). Turkey rhinotracheitis virus antibodies in 8 broiler flocks were between 31.8 and 75.9% and in broiler breeder flocks between 5.4 and 48.6%, with a commercial ELISA. Typical clinical symptoms were observed for SHS in broilers and broiler breeders. During the study, turkey flocks showing clinical signs were not detected. The antibodies to TRTV were between 5.5 and 80.0% in 7 out of 10 broiler flocks, and between 20.0 and 57.1% in 3 out of 4 fattening …

The aim of this study was to determine the effects of urea or urea plus molasses addition into corn harvested at the milk stage on the silage quality and in vitro digestibility of silage. To achieve this objective, four different corn varieties (Rx-947, 33-94, Frassino and Arifiye) harvested at the milk stage were utilized. This study was carried out to investigate the effects of age, sex and season change on electrocardiograms of Van cats. Thirty-eight (male and female) healthy Van cats, 3-15 months old were used. Electrocardiograms (ECGs) and heart radiographs were obtained from cats seperately in summer, autumn, winter and spring. ECGs were recorded as bipolar, augmented unipolar and precordial leads (CV_6LL, CV_6LU, V_10). Our purpose in this study was to investigate and compare the morphological relationship between the true chordae tendineae (CT) and the false chordae tendineae (FCT). One hundred and forty-seven hearts (8 humans, 23 dogs, 91 sheep, 25 goats) were investigated in our study. In terms of CT number, while the difference between humans and sheep and goats, and dogs and sheep and goats was significant statistically (p < 0.05), the difference between humans and dogs was less significant than the others. The difference between sheep and goats was not significant statistically (p > 0.05). The number of FCT were not significant (p > 0.05) in the other species while the difference between humans and dogs was significant statistically. Red blood cells, white blood cells, haematocrit and haemoglobin values (as haematological parameters) and glucose, total protein, albumin, globulin, urea, kreatinine, serum enzymes (ALP, AST, ALT, GGT, CK, amilase) (as biochemical parameters) and electrolytes (Ca, P, Mg, Zn, Cu, Na, K, Cl) levels were measured. It was concluded that the values obtained in this study could be used as reference values for ostriches.

In this study, 627 live freshwater mussels collected from the Koçkale and Pertek regions of Keban Dam Lake were examined for the presence of Salmonella spp. Homogenates prepared from the soft tissue of mussels, after pre-enrichment in Rappaport Vasilliadis, were inoculated onto Brilliant Green Phenol Red Lactose agar and Salmonella-Shigella selective media. Biochemical tests and DNA extraction were carried out on isolates suspected of containing Salmonella. Quality changes in common guitarfish fillets vacuum packed and non-vacuum packed in polyethylene material were investigated during 6 months' cold storage at -18 ^oC. Total yield of the common guitar fish was around 65%. pH, TVB-N, crude protein, moisture, crude ash and fat were 6.50-6.82, 19.87-48.62 mg/100 g, 16.63-22.63%, 75.83-79.88%, 1.00-1.65% and 0.2-0.7%, respectively. The pharmacokinetics of oxytetracycline (OTC) were investigated in rainbow trout in freshwater at 10.5 ^oC. A group of 70 fish received OTC, an intravascular injection (i.v.) of 5 mg/kg body weight. Another group of 105 fish were fed medicated feed containing a dose of 100 mg/kg body weight. OTC plasma concentrations were determined by high performance liquid chromatography. In this study, Rinderpest virus (RPV) recombinant hemagglutinine protein (rH) fused with protein A region of Staphylococcus aureus was expressed in Escherichia coli and purified by IgG affinity chromatography. rH protein was also used to establish enzyme immunoassay. This study was carried out to investigate the fertility traits, survival rate and growth characteristics of Karayaka and Bafra (Chios x Karayaka B_1) genotypes. Karayaka and Bafra genotypes are raised in the Black Sea region of Turkey. In the study, fertility results for Karayaka and Bafra were determined to be 92.3 and 93.7% for birth rates, and 1.08 and 1.78 for litter size, respectively. The survival rates of Karayaka and Bafra lambs at weaning (90 days) were 93.6 and 91.9%, respectively.
