The purpose of this study was to isolate Pasteurella spp and other aerobic bacterial agents from the lungs of sheep and goats with pneumonia and to identify Pasteurella multocida and Mannheimia haemolytica by both culture methods and polymerase chain reaction (PCR). In addition, mouse pathogenecity tests were carried out on suspected P. multocida isolates. In the examination of lung samples collected from sheep, 15 (4.3%) P. multocida and 8 (2.3%) M. haemolytica strains were isolated and identified. Lungs from 8222 cattle slaughtered at an abattoir in Elazığ were examined macroscopically, and pneumonia was detected in 500 (6.1%) lungs. These samples were inoculated onto blood agar supplemented with 7% sheep blood for isolation of bacterial agents. A polymerase chain reaction (PCR) based upon the use of species-specific primers was carried out on DNA samples extracted from suspected Pasteurella spp. isolates. In addition, a mouse inoculation test was carried out on suspected Pasteurella multocida isolates. This study was conducted to determine the ruminal degradation characteristics of nutrients with respect to pasture stage of maturity in Kars district. The nutritional parameters were dry matter (DM), organic matter (OM), crude protein (CP) and crude fiber (CF). Samples were collected from pastures of 9 vicinities of Kars district over a 28-day interval. Three adult Morkaraman rams were fitted with a rumen cannula and samples were incubated in the rumen for 4, 8, 16, 24 and 48 h. Effects Of Different Diets On The Growth Performance, Gonad Development And Body Composition At First Sexual Maturity Of Rainbow Trout (Oncorhynchus Mykiss), Devri̇m Memi̇ş, Hüseyi̇n Gün

Rainbow trout Oncorhynchus mykiss with an initial mean weight of 174 g (1+ year old) were fed on 3 different commercial diets (1 extruded pellet and 2 steam pellet) in the same environmental conditions until their first spawning. Changes in growth performance (weight gain, condition factor, specific growth rate and feed conversion rate), gonadosomatic index (GSI), hepatosomatic index (HSI) and body composition (protein, lipid, ash and dry matter) were investigated. The mean weight gain and condition factor for fish fed the extruded diet was significantly greater than for those fed the steam pellet diets (P < 0.05 ). There were no significant differences in specific growth rate, mean length increments and feed conversion rate among the groups, although the feed conversion rate for the extruded diet was slightly lower than the others. The HSI of the group fed an extruded diet was significantly greater than those of the others ( P < 0.05 ). The GSI values of female and male fish at the end of the study (October) did not differ significantly among the groups. Similarly, diet had no significant effects on the muscle composition of the trial fish, except that the muscle lipid of the group fed the extruded diet was significantly higher than those of the other groups (P < 0.05).

Thirty-three adult (2+ years old) male and 27 female Kars dogs, raised by different breeders in several villages of the Kars region, were used. This study was carried out to determine the effects of phenobarbital, administered orally and intraperitoneally over different periods, on paraoxonase, arylesterase, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities and total protein levels in the serum and liver. Thirty-six rats, 16-18 weeks old and weighing 240-260 g, were used. Phenobarbital was applied both orally and intraperitoneally at a dose of 100 mg/kg. Paraoxonase and arylesterase activities in serum and liver samples were measured spectrophotometrically using paraoxon and phenylasetate substrates, respectively. The aim was to estimate the genetic parameters for birth and weaning weights of Turkish Merino lambs by separating direct genetic, maternal genetic and maternal permanent environmental effects. Data and pedigree information of the Turkish Merino sheep used in this study were collected at the Marmara Animal Breeding Research Institute from 1995 to 2001. Variance components for birth and weaning weights were estimated by the REML technique. Six different animal models were fitted by including or excluding maternal effects. A 1-year-old cat with a suspected abortion was brought to the clinic of the Department of Obstetrics and Gynaecology, Faculty of Veterinary Medicine, Ankara University. The cat was examined because of a reoccurrence of a bloody vaginal discharge. Failure to detect a foetus by ultrasonography and the presence of an anechoic zone in the uterus revealed that there had not an abortion. Since the anechoic zone was large, it was thought that a pharmacological approach would not be successful. A 27.2 kb plasmid in Lactococcus lactis subsp. lactis ELL23, a 32.7 kb plasmid in L. lactis subsp. cremoris ELC5 and a 31.1 kb plasmid in L. lactis subsp. cremoris ELC19 were found to determine exopolysaccharide production (Eps^+) and lactose fermentation (Lac^+) abilities at the end of conjugal transfer studies. The objective of this study was to investigate the prevalence of Y. enterocolitica and other Yersinia species in raw ground beef samples and to determine their potential virulence. For this purpose, 61 ground beef samples purchased from 15 different butcher's shops and 3 supermarkets in Aydın province in Turkey were examined between June 2000 and January 2001. We investigated the effects of drop volume on the development of mouse presumptive zygotes to the completely hatched blastocyst stage. For this aim, we established 3 experimental groups. In the first group, we cultured hybrid presumptive zygotes to the hatched blastocyst stage in 10 presumptive zygotes per 5, 10, 20, 40 and 50 µl drops. The present study was conducted to monitor changes in milk progesterone concentrations at the time of oestrus and post-mating for diagnosing early pregnancy in Anatolian water buffaloes. Twenty-one buffaloes reared at the Kocatepe Agricultural Research Institute, Afyon, Turkey, were used. Regional impacts of agricultural policies have emerged as a key concern in Turkey for both domestic reasons and because of Turkey's desire to join the European Union. This study uses a spatial equilibrium model to analyze the regional impacts of 2 alternative dairy support policies that are currently being considered: a price premium policy and a target price/deficiency payment policy. Both policies are found to exacerbate the post-1980 trend toward a greater share of milk produced in the more prosperous western regions of Turkey. Cat semen was extended with 3% and 4% glycerol containing Tris extender and frozen in (0.25 ml) straws. Spermatological characteristics were examined in post-thaw semen samples. Five tom cats aged 2-3 were used. Semen was collected by electro-ejaculator under general anesthesia (xylazine-ketamine HCl combination). On a weekly basis a total of 8 ejaculates were collected from each tom cat. Spermatozoon concentration (x10^6/ml) was determined by the hemocytometric method. This research was carried out to determine the effects of vitamin E (DL-a tocopheryl acetate) supplementation on inter- and intramuscular fatty acid composition in the meat of Morkaraman lambs. The morphology of Heligmosomum costellatum, a nematode of field mice (Microtus epiraticus), was described by scanning electron microscope. The scanning electron microscopic view of this nematode revealed that the anterior end was surrounded by 2 cephalic vesicles. The 2 copulatory spicules of the male were enveloped in membrane and the male bursa was large. The female posterior end was characterized by a caudal spine. The body of this parasite had transverse ridges.

An Investigation Of Quality Changes In Anchovy (Engraulis Encrasicolus, L. 1758) Stored At Different Temperatures, Sevi̇m Köse, Mehmet Emi̇n Erdem

Quality changes in anchovy caught at different times during storage at ambient and refrigerated temperatures were investigated in 3 different batches. Sampling was carried out each day during storage. The results showed no significant differences among the anchovy batches except for bacteriological counts and trimethylamine (TMA) values at refrigerated temperatures. Storage time and temperature had a significant effect (P < 0.001, P < 0.05) on sensory, bacteriological and chemical values. Bacteriological counts and chemical values increased significantly (P < 0.05, P < 0.001) depending on time, while sensory results decreased (P < 0.05). According to sensory results, samples were acceptable for 1 day at ambient temperatures and for 2 days refrigerated. This study showed that refrigeration did not extend the shelf-life of fresh anchovies by more than a day compare to ambient temperature.

The reproductive activities of Silurus glanis were investigated over 12 months in Menzelet Reservoir, in the East Mediterranean region of Turkey. Total length of the examined 245 S. glanis varied from 33.4 cm (220 g) to 195.0 cm (48,000 g), and age distribution ranged between 1 and 14. Observations on the seasonal distribution of the maturity stages and variations in the gonadosomatic index (GSI) confirmed that Silurus glanis has a prolonged spawning period extending from the beginning of June to August. The aim of this study was to investigate the possibility of producing chimeric bovine embryos by reaggregating blastomeres of disaggregated day 4 bovine embryos. Oocytes were obtained from a slaughterhouse. Selected oocytes were matured, fertilized and cultured in vitro. Fertilization day was taken as day 0. Blastomeres of day 4 embryos were disaggregated and then pooled together. Chimeric embryos were randomly constructed by reaggregating blastomeres in empty zonae pellucidae. A common concern with official estimates of red meat production in Turkey is that they underestimate the level of production. One of the frequently cited reasons is that unregulated slaughter is undercounted. This study uses an estimation method based on the biology of red meat production to examine the potential impact of this undercounting. The production estimates obtained for 1991-1998 using this biological-based method exceed the numbers estimated by official organizations, but fall within the range of estimates obtained from other proposed approaches to estimating red meat production.

The aim of this study was to isolate Edwardsiella ictaluri from diseased rainbow trout obtained from a commercial farm in the Ankara region, Turkey. Growth of the microorganism after 36 h of incubation was higher and purer at 28 ºC than at 37 ºC on all the organs (liver, spleen, kidney) of the 10 fish examined. This study aimed to investigate whether the preovulatory LH surge mechanism is involved in sheep-to-sheep interaction for oestrous synchronisation. For that purpose, anoestrous ewes were inserted with intravaginal progestagen sponges for 13 days to induce ovulation. Inhibitory effects of some antibiotics on glucose 6-phosphate dehydrogenase from the erythrocytes of rainbow trout (Oncorhynchus mykiss Walbaum, 1792) were investigated. For this purpose, initially erythrocyte glucose 6-phosphate dehydrogenase was purified 1271.19-fold in a yield of 70.40% by using ammonium sulfate precipitation and 2',5'-ADP Sepharose 4B affinity gel. A constant temperature (+4 ºC) was maintained during the purification process. Enzyme activity was determined with the Beutler method by using a spectrophotometer at 340 nm. This method was utilized for all kinetic studies. Thiamphenicol, amikacin, gentamicin, and netilmicin were used as antibiotics. All the antibiotics exhibited inhibitory effects on the enzyme. …

This study was carried out to determine the seroprevalence of Babesia ovis around Afyon between June and September 2000. During this period, blood samples were collected to obtain serum samples from total of 204 Akkaraman sheep in different age groups in 9 different localities. This research was conducted to study the effects of artificial sowing with various plant species and different sowing dates on forage yield, grazing capacity and estimated carcass weight in rangelands under continental dry conditions. Liza abu stocks from the rivers Orontes, Euphrates and Tigris were investigated using genetic and morphometric data. Allozyme electrophoresis for genetic comparison and the truss network system for morphometric comparison were simultaneously applied to the same sample set. Highly significant morphological differences were observed between the 3 Liza abu stocks. The correct classification of individuals into their original samples for morphological characters was very high (100% of individuals). This study was designed to determine the health status of dairy cattle, and the rates of and reasons for culling on dairy farms in Kars. A 2-stage stratified sampling strategy was used to select localities and farms. The study involved an interview with the farmers and regular visits to the farms. The farm prevalence levels of abortion, foot- and -mouth disease (FMD), respiratory problems, leptospirosis, blackleg, hardware (TRP), bloat and anthrax were 46.7%, 57.8%, 4.4%, 13.3%, 4.4%, 11.1%, 8.8% and 0% in 2001 and 53.3%, 62.2%, 17.8%, 17.8%, 4.4%, 11.1%, 11.1% and 8.8% in 2002, respectively. We aimed to improve the selectivity of lauryl tryptose (LST) broth and HC (hemorrhagic colitis Escherichia coli strains) agar for the detection of E. coli O157:H7 in frozen ground beef samples. The selectivity of LST broth was superior to that of LST containing novobiocin (20 mg/l) (LST + n), but LST containing novobiocin, cefixime (0.05 mg/l) and tellurite (2.5 mg/l) (LST + nct) was found to be unsuitable as an enrichment medium. The sorbitol positive strains were significantly more resistant to novobiocin than were the sorbitol negative strains (P < 0.05). As a result, the enrichment of ground beef samples in a selective broth without antibiotics such as LST, and then spread plating a subsequent amount from this original enrichment culture and the first three 10-fold serial dilutions onto a rhamnose and sorbitol based selective medium such as cefixime-tellurite HC medium (CTR-HC) was found to be recommendable as a routine cultural method. Transferring the metallic sheen property of eosin methylene blue (EMB) agar to CTR-HC agar can represent an alternative to the available media, and this will shorten the analysis time by 1 day. Use of salicine or 4-methylumbelliferyl-b-D glucuronide appears unnecessary in routine cultural techniques.
