Fusion Of A Fluorescent Protein To The Pul25 Minor Capsid Protein Of Pseudorabies Virus Allows Live-Cell Capsid Imaging With Negligible Impact On Infection, Kevin P. Bohannon, Patricia J. Sollars, Gary E. Pickard, Gregory A. Smith

School of Veterinary and Biomedical Sciences: Faculty Publications

In order to resolve the location and activity of submicroscopic viruses in living cells, viral proteins are often fused to fluorescent proteins (FPs) and visualized by microscopy. In this study, we describe the fusion of FPs to three proteins of pseudorabies virus (PRV) that allowed imaging of capsids in living cells. Included in this study are the first recombinant PRV strains expressing FP–pUL25 fusions based on a design applied to herpes simplex virus type 1 by Homa and colleagues. The properties of each reporter virus were compared in both in vitro and in vivo infection models. PRV strains expressing FP–pUL25 …

Staphylococcal Response To Oxidative Stress, Rosmarie Gaupp, Nagender Ledala, Greg A. Somerville Jan 2012

Staphylococcal Response To Oxidative Stress, Rosmarie Gaupp, Nagender Ledala, Greg A. Somerville

School of Veterinary and Biomedical Sciences: Faculty Publications

Staphylococci are a versatile genus of bacteria that are capable of causing acute and chronic infections in diverse host species. The success of staphylococci as pathogens is due in part to their ability to mitigate endogenous and exogenous oxidative and nitrosative stress. Endogenous oxidative stress is a consequence of life in an aerobic environment; whereas, exogenous osidative and nitrosative stress are often due to the bacteria's interaction with host immune systems. To overcome the deleterious effects of oxidative and nitrosative stress, staphylococci have evolved protection, detoxification, and repair mechanisms that are controlled by a network of regulators. In this review, …

Koutango: Under Reported Arboviral Disease In West Africa, Jaime Matias De Araujo Lobo

LSU Doctoral Dissertations

Arthropod-borne viruses (arboviruses) are among the most common agents of human febrile illnesses worldwide. As crucially important emerging pathogens, they have caused multiple, notable epidemics of human disease and unnoticed epizootics over recent decades. Despite the public health relevance, very little is known about the geographic distribution of the agents and vectors, relative impact, and risk factors associated to the arboviral infection in many regions of the world and in the tropics in particular. Presented in this dissertation is an experimental study that explores the serology screening of serum samples from 151 patients whom were diagnosed with undifferentiated febrile illness …

Immune System Dysfunction In Broiler Chickens Experimentally Inoculated With Fowl Adenovirus Serotype-4 Associated With Inclusion Body Hepatitis Hydropericardium Syndrome, Iftikhar Hussain, Muhammad Shahid Mahmood, Muhammad Imran Arshad, Masood Akhtar, Fazal Mahmood, Azhar Rafique Jan 2012

Immune System Dysfunction In Broiler Chickens Experimentally Inoculated With Fowl Adenovirus Serotype-4 Associated With Inclusion Body Hepatitis Hydropericardium Syndrome, Iftikhar Hussain, Muhammad Shahid Mahmood, Muhammad Imran Arshad, Masood Akhtar, Fazal Mahmood, Azhar Rafique

Turkish Journal of Veterinary & Animal Sciences

Inclusion body hepatitis-hydropericardium syndrome (IBH-HPS) in broilers is caused by an adenovirus, here referred to as inclusion body hepatitis-hydropericardium syndrome virus (IBH-HPSV). This study describes the immune status of broiler chickens experimentally inoculated with an isolate of fowl adenovirus-4 serotype-1, involved in IBH-HPS in various trials. When compared with the unchallenged controls, IBH-HPSV-inoculated broilers had significantly higher atrophy of the bursa of Fabricius (BF) by up to 2-fold, as well as of the thymus (up to 9-fold) and spleen (up to 1.5-fold) (P < 0.05), with up to 60% mortality. Upon challenge with sheep red blood cells (SRBCs), IBH-HPSV-inoculated birds had from 2- to 3-log lower anti-SRBC antibody titers than the control. Response to phytohemagglutinin-P (PHA-P) was significantly reduced in IBH-HPSV-inoculated birds (P < 0.05) as compared to the control. Results of the studies revealed that the immune status of broilers is compromised significantly during IBH-HPSV infection in terms of lymphoid organ integrity, humoral, and cell-mediated immune responses, respectively. These findings suggest that immune dysfunction could be a contributing factor in the increased mortality of birds affected by IBH-HPS, which renders the birds immunosuppressed for the rest of their lives.

The Influence Of Egg Shell Crack Types On Hatchability And Chick Quality, Mozhdeh Moosanezhad Khabisi, Ahmad Salahi, Seyed Naser Mousavi Jan 2012

The Influence Of Egg Shell Crack Types On Hatchability And Chick Quality, Mozhdeh Moosanezhad Khabisi, Ahmad Salahi, Seyed Naser Mousavi

Turkish Journal of Veterinary & Animal Sciences

Fertile eggs were obtained from a commercial flock of Ross broiler breeders (51 weeks old) and candled to determine the presence of hairline or star cracks. Eggs (5400 total) were assigned in equal numbers to 3 treatments (hairline-cracked, star-cracked, and normal eggs) and incubated for 21 days. Upon hatching, 10 chicks per treatment were euthanized, and organ weights were measured. Compared with normal eggs, the presence of egg shell cracks (regardless of type) resulted in higher egg weight loss at the time of transfer; a decrease in hatchability, hatch of fertile eggs, chick length, yolk-free body mass (P < 0.01), and percentage of hatchling breast and liver weight (P < 0.05); and an increase in the contamination rate of eggs (P < 0.01). Weight loss in hairline-cracked eggs was significantly higher than in star-cracked eggs (P < 0.001). Percentage hatchability and body weight of chicks hatched from star-cracked eggs was higher than in chicks from hairline-cracked eggs (P < 0.001). Crack type did not have a significant effect on chick length and body weight uniformity; however, the Pasgar score as a quality indicator was higher in chicks hatched from star-cracked eggs than those from hairline-cracked eggs (P < 0.05). There were no significant differences among treatments in terms of heart and small intestine weight. The contamination rate of hairline-cracked eggs was higher than that of star-cracked (P < 0.01). A higher incidence of embryonic mortality was observed during days 1-8 of incubation for shell-cracked eggs (P < 0.001). Total embryonic mortality in hairline-cracked, star-cracked, and normal eggs was 51.1%, 36.6%, and 13.3%, respectively. This study showed that egg shell cracks reduced incubation parameters and chick quality. The negative effects of hairline cracks were more pronounced than those of star cracks.
