Development Of An Integrated Microfluidic System For Cell Sorting And Trapping—Considering Sorting Efficacy And Cell Viability, Thammawit Suwannaphan

This study is divided into two phases. The first phase is about the investigation of cell viability and damage of white blood cells after sorting in both a spiral microchannel and contraction and expansion array (CEA) at appropriate flow rates for cell separation. Three techniques such as Trypan Blue staining, Scanning Electron Microscopy (SEM) and Wright-Giemsa staining are employed in the experiments. After flowing the sample through a whole setup of spiral microchannel at 2 ml/min, 85% of cells are viable while 30% of cells are significantly deformed and 2% of cells are seriously damaged in their intracellular structures. การศึกษานี้แบ่งออกเป็นสองวัตถุประสงค์หลัก วัตถุประสงค์แรกเป็นการศึกษาผลกระทบของมุมของหลุมจุลภาครูปร่างทรงพีระมิดฐานสี่เหลี่ยมข้าวหลามตัดทั้ง 3 รูปร่างคือ มุมแหลม (66 องศา) มุมด้านเท่า (90 องศา) และมุมป้าน (106 องศา) ต่อการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อสำหรับการเลี้ยงที่ใช้การไหลแบบต่อเนื่อง โดยแต่ละหลุมจุลภาควางเรียงชิดกันเพื่อลดพื้นที่ว่างไม่ให้เซลล์ไปเกาะหรือเกิดการสูญเสียเซลล์ไป ในบริเวณก้นหลุมจุลภาคมีทรงกระบอกรัศมี 450 ไมโครเมตร ติดตั้งอยู่ ซึ่งเป็นตำแหน่งที่จะเกิดการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อขึ้น และทำให้ความลึกรวมทั้งหมดของหลุมจุลภาคเท่ากับ 550 ไมโครเมตร ในการทดลองได้ใช้อัตราการไหลเท่ากับ 10 ไมโครลิตรต่อชั่วโมง ทั้งจากผลการคำนวณ และผลการทดลองพบว่า ในกรณีหลุมจุลภาคที่มีมุม 66 องศา ของไหลที่ไหลผ่านหลุมมีการเบี่ยงเบนทิศทางเข้าสู่กึ่งกลางหลุมมากกว่าหลุมแบบอื่น และน่าจะทำให้เกิดการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อของเซลล์ลำบาก เพราะถูกรบกวนจากของไหลค่อนข้างสูง ในกรณีหลุมจุลภาคที่มีมุม 90 องศา ของไหลมีการเบี่ยงเบนทิศทางเข้าสู่กึ่งกลางลดลง ทำให้มีการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อได้ดีขึ้น อย่างไรก็ตาม เมื่อหลุมจุลภาคมีมุม 106 องศา ของไหลมีการเบี่ยงเบนทิศทางเข้าหากึ่งกลางหลุมน้อยลงมาก ทำให้เซลล์บางส่วนไม่ถูกพัดพามารวมกันที่บริเวณก้นหลุมทรงกระบอก โดยเซลล์บางส่วนติดอยู่บนบริเวณผนังด้านข้าง ทำให้การสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อลดต่ำลง ดังนั้นผลการศึกษาจึงบ่งชี้ว่าหลุมจุลภาคทรงพีระมิดฐานสี่เหลี่ยมข้าวหลามตัดมุม 90 องศา มีความเหมาะสมที่สุดในการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อ นอกจากนั้น ผลการทดลองยังแสดงให้เห็นด้วยว่าอัตราการรอดชีวิตของเซลล์มีแนวโน้มดีกว่าหลุมจุลภาคแบบอื่นอีกด้วย วัตถุประสงค์ที่สองคือการสร้างระบบของไหลจุลภาคที่ใช้ต้นทุนต่ำ และรวดเร็ว โดยการสร้างแม่พิมพ์ด้วยการพิมพ์สามมิติด้วยเรซิ่น สำหรับการพิมพ์สามมิติจะทำให้สร้างแม่พิมพ์ที่มีรูปร่างที่ซับซ้อนมากได้ แต่ขนาดที่เล็กสุดยังจำกัดอยู่ และความแข็งแรงทางกล และความทนทานต่อความร้อนไม่ดีนัก อย่างไรก็ตาม วิธีการที่พัฒนาที่ขึ้นสามารถใช้งานได้ดีในการหล่อสารพอลิไดเมทิลซิโลเซนสำหรับระบบของไหลจุลภาค ซึ่งใช้งานอย่างมากในงานวิจัยทางด้านนี้

ปัจจุบันประชากรในประเทศไทยนิยมเลือกใช้บริการร้านสะดวกซื้อมากขึ้น เนื่องจากร้านสะดวกซื้อมีทำเลที่ตั้งที่เข้าถึงได้อย่างสะดวกรวดเร็วเหมาะสมกับการใช้ชีวิตประจำวัน ดังนั้นการเลือกระบบปรับอากาศที่มีความเหมาะสมกับร้านสะดวกซื้อจึงเป็นสิ่งที่สำคัญในเรื่องการจัดการการใช้พลังงานของร้านสะดวกซื้อ งานวิจัยนี้มีจุดมุ่งหมายในการวิเคราะห์การใช้พลังงานของระบบปรับอากาศแบบแยกส่วนสำหรับร้านสะดวกซื้อ ซึ่งเป็นระบบปรับอากาศที่พบว่ามีการใช้งานภายในร้านสะดวกซื้อเป็นส่วนใหญ่ในสภาพอากาศแบบร้อนชื้น โดยการเปรียบเทียบการใช้พลังงานของค่าพารามิเตอร์ต่าง ๆ ในลักษณะของค่าเฉลี่ย ได้แก่ อุณหภูมิ ความชื้นสัมพัทธ์ กำลังไฟฟ้า และการใช้พลังงาน สามารถแบ่งการวิเคราะห์การใช้พลังงานได้ 2 รูปแบบ คือ การวิเคราะห์การใช้พลังงานในแต่ละโซนอุณหภูมิและการวิเคราะห์การใช้พลังงานในแต่ละเดือนของร้านสะดวกซื้อ ในการคำนวณการทำงานของระบบปรับอากาศตลอดทั้งปีนั้นจะใช้โปรแกรม EnergyPlus และโปรแกรม Openstudio โดยจะทำการจำลองพลังงานของระบบปรับอากาศสำหรับร้านสะดวกซื้อ โดยตัวอาคารแห่งนี้มีลักษณะเป็นสี่เหลี่ยมผืนผ้า อาคารเดี่ยว 1 ชั้น ติดตั้งระบปรับอากาศแบบแยกส่วนอยู่ภายในทั้งหมด 4 เครื่อง ในการสร้างโมเดลของร้านสะดวกซื้อจำลองแห่งนี้อ้างอิงจากแบบพิมพ์เขียวของร้านสะดวกซื้อและสร้างขึ้นโดยโปรแกรม SketchUp โดยนำข้อมูลสภาพอากาศที่ใช้ในการจำลองพลังงานจากกรมอุตุนิยมวิทยา สำหรับวัตถุประสงค์ในการเปรียบเทียบการใช้พลังงานของระบบปรับอากาศแบบแยกส่วน จะใช้ข้อมูลแผ่นข้อมูลจำเพาะของอุปกรณ์ที่อยู่ในระบบปรับอากาศที่มีการติดตั้งและใช้งานจริง ผลของการเปรียบเทียบค่าพารามิเตอร์ต่าง ๆ จากการแบ่งโซนอุณหภูมิ 8 ตำแหน่ง สามารถสรุปได้ 3 กลุ่ม คือ โซนที่อยู่บริเวณด้านหน้า ตรงกลาง และด้านหลังของร้านสะดวกซื้อ ซึ่งขึ้นอยู่กับตำแหน่งการติดตั้งเครื่องปรับอากาศ พบว่า บริเวณจุดอับลมมีสัดส่วนการใช้พลังงานสูงที่สุดคิดเป็นร้อยละ 28.1 และ 28.7 และสัดส่วนการใช้พลังงานตลอดทั้งปีของอุปกรณ์ต่าง ๆ ได้แก่ เครื่องปรับอากาศแบบแยกส่วน ตู้เย็น อุปกรณ์ต่าง ๆ และระบบแสงสว่าง คิดเป็นร้อยละ 15.8 39.5 22.3 และ 22.4 ตามลำดับ ถึงแม้จะพบว่าสัดส่วนการใช้พลังงานของระบบปรับอากาศแบบแยกส่วนมีค่าน้อยที่สุด แต่ก็ยังส่งผลกระทบต่อโซนอุณหภูมินั้น ๆ อีกด้วย โดยการใช้พลังงานโดยรวมของร้านสะดวกซื้อในเดือนมีนาคมมีค่ามากที่สุดเมื่อเปรียบเทียบตลอดทั้งปี นอกจากนี้การใช้พลังงานโดยรวมในแต่ละโซนอุณหภูมิต่อพื้นที่ใช้งานจริงสำหรับพื้นที่ปรับอากาศและสำหรับพื้นที่ทั้งหมดของร้านสะดวกซื้อตลอดทั้งปี มีค่าเท่ากับ 429.32 และ 403.22 kWh/m2 ตามลำดับ และยังพบว่า เดือนมีนาคมมีค่ามากที่สุด เท่ากับ 34.90 kWh/m2 ซึ่งมีความสอดคล้องกับการวิเคราะห์ผลการใช้พลังงานในแต่ละเดือนของร้านสะดวกซื้อแห่งนี้อีกด้วย

มันสำปะหลังเป็นพืชที่ปลูกง่ายและปลูกเป็นจำนวนมากในประเทศไทย เครื่องมือที่เกี่ยวข้องในการตรวจวัดคุณภาพมันสำปะหลังหลังการเก็บเกี่ยวจนไปถึงก่อนแปรรูปที่ใช้ในปัจจุบันเป็นเครื่องมือที่ใช้หลักการวัดทางกลพื้นฐานและใช้ความรู้สึกของผู้ประเมินร่วมด้วย ทำให้มีโอกาสที่จะได้ผลประเมินที่ไม่เป็นความจริง วิทยานิพนธ์นี้เสนอแนวทางการตรวจวัดคุณภาพมันสำปะหลังในห่วงโซ่อุปทานโดยใช้กล้อง Kinect ซึ่งเป็นกล้อง RGB-D ถ่ายรูปภาพของหัวมันสำปะหลังเพื่อหาปริมาตรของมันสำปะหลัง การศึกษาทำโดยถ่ายภาพมันสำปะหลังพันธุ์เกษตรศาสตร์ 50 จำนวน 90 ตัวอย่างแล้วนำมาวิเคราะห์หาปริมาตรด้วยโปรแกรม MATLAB และใช้วิธีการประมาณปริมาตรมันสำปะหลัง 2 วิธีได้แก่ การประมาณปริมาตรโดยใช้ Depth image และการประมาณปริมาตรโดยใช้ Point Cloud image ทั้งสองวิธีการนี้มีพื้นฐานมาจากการหาปริมาตรของรูปทรงเรขาคณิต ผลการหาปริมาตรของมันสำปะหลังจากการประมาณด้วยวิธีการ depth image และวิธีการ point cloud image เมื่อนำมาพล็อตกับปริมาตรของมันสำปะหลังที่ได้จากวิธีการแทนที่น้ำให้ค่าความคลาดเคลื่อน RMSE เท่ากับ 66.94 มิลลิลิตรและ 41.4 มิลลิลิตร หรือคิดเป็นร้อยละ 21.12 และ 13.06 ตามลำดับเมื่อเทียบกับค่าเฉลี่ยของปริมาตรของหัวมันสำปะหลังจากวิธีการแทนที่น้ำ และค่า R-squared ที่ได้จากการประมาณทั้งสองวิธีมีค่า 0.9561 และ 0.9626 ดังนั้นจึงสรุปได้ว่าการประมาณปริมาตรหัวมันสำปะหลังด้วยวิธีการใช้รูป point cloud จะดีกว่าจากค่า R-squared ที่มากกว่าและค่า RMSE ที่น้อยกว่า ปริมาตรของมันสำปะหลังที่ประมาณด้วยรูปถ่ายนี้สามารถนำไปประยุกต์หาเปอร์เซ็นต์แป้งมันสำปะหลังได้

ระบบหุ่นยนต์หลายตัวส่วนใหญ่ถูกนำมาใช้เพื่อเพิ่มประสิทธิภาพในการทำงานหรือถูกนำมาใช้เมื่อไม่สามารถทำงานด้วยหุ่นยนต์ตัวเดียวได้ วิทยานิพนธ์ฉบับนี้กล่าวถึง ระบบหุ่นยนต์หลายตัวสำหรับขนส่งแผงโซล่าเซลล์ที่มีขนาดใหญ่ซึ่งไม่สามารถขนส่งด้วยหุ่นยนต์ตัวเดียวได้ ซึ่งจะเป็นการขนส่งโดยใช้หุ่นยนต์ 2 ตัวในการขนส่ง หุ่นยนต์แต่ละตัวนั้นจะเคลื่อนที่อยู่ภายในรางและเคลื่อนที่ในแนวตรง การควบคุมการเคลื่อนที่ระหว่างหุ่นยนต์เป็นการควบคุมความเร็วในการเคลื่อนที่ของหุ่นยนต์ให้อยู่ในระดับเดียวกันผ่านการวัดมุมที่เกิดขึ้นระหว่างการเคลื่อนที่ของหุ่นยนต์ 2 ตัวด้วยการติด Encoder วัดมุมด้านบนตัวหุ่นยนต์ที่อุปกรณ์จับแผงโซล่าเซลล์ การควบคุมดังกล่าวจะช่วยลดระยะความต่างของระยะห่างที่หุ่นยนต์เคลื่อนที่ตามกัน นอกจากนี้ระบบหุ่นยนต์ได้รับการพัฒนาระบบการสื่อสารไร้สายด้วยเครือข่ายไร้สาย Mesh network เพื่อช่วยพัฒนาการเคลื่อนที่ให้ดียิ่งขึ้นด้วยการส่งข้อมูลสถานะการเคลื่อนที่เพื่อให้หุ่นยนต์ทั้ง 2 ตัวรับทราบการเคลื่อนที่ ณ เวลานั้น ๆ และจากการทดลองทั้งหมดพบว่า ระยะที่หุ่นยนต์เคลื่อนที่ตามกันมีระยะไม่เกิน 3 เซนติเมตร

Cystic Fibrosis (CF) is a degenerative disease, which causes thickening of the airway surface liquid and reduced mucociliary clearance, which provides an ideal habitat for bacterial infections. Early treatment of CF in children can prevent chronic infection, improve quality of life, and increase life expectancy. The most predominant bacteria found in CF-diseased lungs is Pseudomonas aeruginosa (Pa), which can be treated with inhaled tobramycin. The main operating principle consists of an actuator which provides a stimulus, and a sensor to receive a signal traveling to a fluid domain. Water heating in residential buildings, also known as domestic hot water (DHW), is the third largest use of energy after appliances and space conditioning. About 90% of the residential buildings in the state use natural gas fueled water heaters, 6% use electricity, and a small percent use liquefied petroleum gas (LPG) or solar water heaters. Perovskites are solution-processed, high-performance semiconductors of interest in low-cost photovoltaics. The interfaces between the perovskite photoactive layers and the top and bottom contacts are crucial for efficient charge transport and minimizing trapping. Control of the collection of charge carriers at these interfaces is decisive to device performance. Here, we review recent progress in the realization of efficient perovskite solar cells using cheap, easily processed, stable, carbon-based interlayers. Interface materials including graphene, carbon nanotubes, fullerenes, graphene quantum dots and carbon dots are introduced and their influence on device performance is discussed.

As technology advances, higher performance cooling systems are desired to meet numerous thermal management challenges. Extended surfaces, also known as heat sinks or fins, are commonly used to passively dissipate unwanted heat from mechanical and electrical systems. Originally discovered in condensed matter systems, topological insulators (TIs) have been ubiquitously extended to various fields of classical wave physics including photonics, phononics, acoustics, mechanics, and microwaves. In the bulk, like any other insulator, electronic TIs exhibit an excessively high resistance to the flow of mobile charges, prohibiting metallic conduction. The introduction of in-use emissions regulations by the United States Environmental Protection Agency (U.S.EPA) requires medium-duty (MD) and heavy-duty (HD) engine manufacturers to demonstrate emissions compliance during in-fleet operation. In the United States (U.S.), the Not-to-Exceed (NTE) method is used to evaluate real-world emissions compliance from on-highway MD and HD trucks. High-temperature reacting systems are central to many fields including propulsion, power generation, and transportation. Studying such systems requires the use of experimental facilities such as shock tubes to obtain the relevant high temperature conditions, and non-intrusive diagnostic tools for monitoring parameters of interest in the reaction zone, including temperature and species concentration time-histories. Laser absorption spectroscopy offers high-speed, in-situ measurements of the reacting flow field and provides direct measurements of species concentrations and temperature. Droplet Microfluidics can be utilized to control the production of single droplets and double emulsions in biomedical application. This dissertation analyzes the droplet formation in hydrophobic and hydrophilic microchannels, encapsulation of cells in flow-focusing channels, and deformation of cell-laden droplets in constricted microcapillaries. The formation of droplets is studied in hydrophobic and hydrophilic channels in order to investigate the role of contact angle on flow structures and droplet generation patterns. One method of increasing local heat fluxes is the fabrication of superhydrophilic materials via micro/nanostructuring of the surface. Superhydrophilic surfaces act in a self-pumping capacity, spreading fluid beyond its intrinsic meniscus length to form thin-films, a process known as "hemiwicking", with practical applications in evaporative cooling, as well as flow, pool, and thin-film boiling.
