As the obesity epidemic increases, the prevalence of obesity-related diseases grows simultaneously. The internal environment of chronic low-grade inflammation, which characterizes obesity, leaves individuals vulnerable to disease. Cardiovascular dysfunction represents a consequential effect of obesity. An early indicator of cardiovascular disease is an impaired stress response. Further, the type of stress (active versus passive) may be differentially affected representing additional distinctions in impairment. In the present study, we have used two types of stress (active and passive) to investigate obesity-instigated alterations in cardiovascular, immune and endocrine response to stress. Major depressive disorder (MDD) is the most common mood disorder in the United States and European Union; however, the limitations of clinically available antidepressant drugs have led researchers to pursue novel pharmacological treatments. Clinical studies have reported that monotherapy with the atypical antipsychotic drug quetiapine produces a rapid reduction in depressive symptoms that are apparent following one week of quetiapine treatment, and it is possible that the active metabolite N-Desalkylquetiapine, which structurally resembles an antidepressant drug, produces antidepressant effects. Impulsive choice refers to the inability to delay gratification and is associated with increased drug abuse vulnerability. Understanding the underlying neural mechanisms linking impulsive choice and drug abuse can contribute to improved treatment options for individuals with substance use disorders. Evidence suggests a major role for nucleus accumbens core (NAcc) in impulsive choice and the reinforcing effects of drugs of abuse. Huntington’s disease (HD) is a heritable, neurodegenerative disorder characterized by motor, cognitive, and psychiatric disturbances. An unstable CAG expansion within the gene normally encoding for the Huntingtin protein is responsible. The expanded mutant form of Huntingtin and the putative protein co-factor Rhes interact and cause cell death within the striatum. We hypothesized chronic treatment with simvastatin, a cholesterol lowering drug, would disrupt the biosynthetical pathway which gives both Rhes and its target cells binding sites and render Rhes inactive. Attention deficit hyperactivity disorder (ADHD) is a heritable disorder, which has detrimental effects on childhood development and is associated with maladaptive functioning in adulthood. Despite this, we are far from an understanding of the etiology and possible trajectories of ADHD, possibly due to investigations focusing on the contribution of single genes. In fact, single genes are likely not influential enough to alter behavior, but the additive effect of many genes may predispose an individual toward certain behaviors. The Hypothalamic-Pituitary-Adrenal (HPA) and –Gonadal (HPG) axes have been considered mutually inhibitory; however, emerging evidence supports the proposition that this might not necessarily be the case. This idea is termed “coupling,” in which the HPA-HPG axis are mutually activated or deactivated. Coupling is examined across three data sets with different time-courses of stress exposure, and results demonstrate HPA-HPG co-activation occurs. Furthermore, stress exposure influences this relationship. The discussion shows how it is physiologically possible to have positive coupling or co-activation between these axes according to complex regulatory feedback systems and overlapping neural structures. Among several commonly used atypical antipsychotic drugs, olanzapine and risperidone cause a sensitization effect in the conditioned avoidance response (CAR) and phencyclidine (PCP)- induced hyperlocomotion paradigms – two well established animal tests of antipsychotic drugs, whereas clozapine causes a tolerance effect. Asenapine is a novel antipsychotic drug recently approved for the treatment of schizophrenia and manic disorders. It shares several receptor binding sites and behavioral features with other atypical antipsychotic drugs. Iptakalim is an ATP-sensitive potassium channel opener, as well as an a4b2-containing nicotinic acetylcholine receptor (nAChR) antagonist. Pretreatment with iptakalim diminishes nicotine-induced dopamine (DA) and glutamate release in the nucleus accumbens. This neuropharmacological profile suggests that iptakalim may be useful for treatment of nicotine dependence. Thus, we examined the effects of iptakalim in two preclinical models. First, the impact of iptakalim on the interoceptive stimulus effect of nicotine was evaluated by training rats in a discriminated goal-tracking task that included intermixed nicotine (0.4 mg/kg, SC) and saline sessions. This investigation was an analysis of the effects of methylphenidate (MPH; trade name: Ritalin) on drug reward using the conditioned place preference (CPP) behavioral paradigm in a rodent model and underlying mechanisms of this effect. Animals were conditioned in adolescence from postnatal day (P)33-39) or P44-49 with saline, 1 or 5 mg/kg MPH. Rats administered 5 mg/kg but not 1 mg/kg MPH, resulted in a significant preference that was more robust in younger male adolescent rats. The 5 mg/kg dose of MPH also resulted in a significant decrease of the dopamine transporter in both the nucleus accumbens and striatum, revealing …

In nocturnal rodents, brain areas that promote wakefulness have a circadian pattern of neural activation that mirrors the sleep/wake cycle, with more neural activation during the active phase than during the rest phase. To investigate whether differences in temporal patterns of neural activity in wake-promoting regions contribute to differences in daily patterns of wakefulness between nocturnal and diurnal species, we assessed Fos expression patterns in the tuberomammillary (TMM), supramammillary (SUM), and raphe nuclei of male grass rats maintained in a 12:12 h light-dark cycle. It is generally thought that macronutrients stimulate intake when sensed in the mouth (e.g., sweet taste) but as food enters the GI tract its effects become inhibitory, triggering satiation processes leading to meal termination. Here we report experiments extending recent work (see [1]) showing that under some circumstances nutrients sensed in the gut produce a positive feedback effect, immediately promoting continued intake. In one experiment, rats with intragastric (IG) catheters were accustomed to consuming novel flavors in saccharin daily while receiving water infused IG (5 ml/15 min). The perception of and actual ability to control emotional responses during stressful, taxing situations are important to an individual’s well-being. Studies have shown that both low perceived control and a low actual ability for emotional control are related to internalizing and externalizing problems in youth. Light influences the daily patterning of behavior by entraining circadian rhythms and through its acute effects on activity levels (masking). Mechanisms of entrainment are quite similar across species, but masking can be very different. Specifically, in diurnal species, light generally increases locomotor activity (positive masking), and in nocturnal ones, it generally suppresses it (negative masking). The intergeniculate leaflet (IGL), a subdivision of the lateral geniculate complex, receives direct retinal input and is reciprocally connected with the primary circadian clock, the suprachiasmatic nucleus (SCN). The zebrafish (Danio rerio) is accepted in the developmental and genomic communities as a model organism. However, the capacity for the zebrafish as a behavioral model has yet to be fully acknowledged. The research presented provides evidence validating the novel task, aids in gaining a better understanding of the learning processes, and identifies individual differences. The novel associative learning task differs from any present well established behavioral model and lends itself to future development. Socio-ecological theorists tie primate social structure diversity to variations in habitats within which primate species reside. This premise permits laboratory researchers to investigate specific factors that influence or relate to social structure formation and maintenance. The focus of the current investigation was three fold. We first aimed to determine the relationships between traditional and non-traditional behavioral measures of dominance, then evidence for dominance hierarchy formation was examined, and various hypotheses were tested to discern if adjustments in the foraging context altered social behaviors in Garnett’s bushbaby (Otolemur garnettii). Reconciliation is the occurrence of friendly behaviour between opponents shortly after an aggressive conflict. In primate groups, reconciliation reduces aggression and post-conflict arousal. Aggression within a group can also increase arousal of bystanders (e.g. increase bystanders’ rates of self-directed behaviour). Since reconciliation reduces aggression between opponents, we tested whether it also reduces self-directed behaviour in bystanders. Following aggression in a captive group of hamadryas baboons, one observer conducted a focal sample on one of the combatants to document reconciliation and a second observer simultaneously conducted a focal sample on a randomly selected bystander. In 2011, the U.S. reported 3 million child maltreatment cases, an uncomfortably high but recurring figure each year. Research shows exposure to early life stress (ELS) increases an individual’s susceptibility to substance abuse, specifically of nicotine, alcohol, and cocaine. Increased susceptibility may result from dysregulation of the HPA axis sustaining activation into adulthood after ELS. Hyperactivation of the HPA axis significantly reduces hippocampal BDNF, a neurotrophin involved in neuronal growth and plasticity. Reduced hippocampal BDNF may be a factor in substance abuse vulnerability. Additionally, research shows exercise protects hippocampal BDNF from stress induced down-regulation. Humans often make use of the neurotransmission altering effects of drugs such as caffeine, ephedrine, and ethanol. These drugs induce changes in memory and learning ability, specifically when used for sleep deprivation. English white mice (Mus musculus) have been shown to exhibit similar cognitive changes when administered drugs while sleep deprived. A sample of 20 mice was divided into three experimental groups (each group receiving a different drug) and one control group. Maze-testing demonstrated that mice performance decreased with sleep deprivation (Treatment-1) but all groups returned to baseline performance when injected (IP) with selected drugs (Treatment-2), regardless of …

Mindfulness meditation is a practice of focus, awareness, and non-judgmental acceptance of one's thoughts (Deyo et al., 2009; Kenny et al., 2007). Rumination is a maladaptive pattern of thought that is common in people with depression and other mood disorders. It can lead to further episodes of depression, and can be very destructive in that way (Nolen-Hoeksema, 2008). Anxiety disorders are the most prevalent class of mental disorders, often characterized by a chronic course and comorbid psychopathology. The anxiety-stress literature utilizing cortisol as a biomarker of the hypothalamus pituitary adrenal (HPA) axis has been inconsistent. The establishment of salivary alpha-amylase (sAA) as a biomarker of sympathetic nervous system (SNS) activation allows researchers an opportunity to examine the stress response more fully.
