In this study,antibodies against Brucella melitensis in aborted sheep were investigated with enzyme-linked immunosorbent assay (ELISA), standard tube agglutination test (SAT), rose bengal plate test (RBPT), complement fixation test (CFT), 2-mercaptoethanol microagglutination test (2-ME-MAT)and dithiothreitol microagglutination test (DTT-MAT). The sensitivity and specificity of the tests were calculated by using CFT results as the gold standard. A Kappa value was used to measure the agreements between ELISA and the other tests. Sonicated antigen prepared from Brucella melitensis 16M strain,the peroxidase labelled anti-sheep IgG conjugate and ortho-phenylenediamine substrate were used in ELISA. This study was carried out to determine the pathomorphologic changes and the incidence of these changes in the uterus, cervix, vagina and related ligaments of ewes slaughtered at Elazığ abattoirs. For this purpose,the genital organs of 6100 ewes from different breeds and ages were examined.There were pathomorphologic changes in 274 cases (4.49%)in the uterus,70 cases (1.14%)in the cervix,and 61 cases (1.00%)in the vagina.Inflammatory changes were primarily lesions (2.68%)observed in the uterus.These changes were catarrhal endometritis (0.52%), purulent metritis (0.32%),necrotic endometritis (0.11%), chronic nonpurulent endometritis (0.95%),granulomatous endometritis (0.13%),pyometra (0.11 %)and parametritis (0.52%). In this study, 14 adult albino guinea pigs, weighing 300-350 g, were divided into two equal groups. The small intestinal segments (distal ileum)taken from the animals were kept in vitro in the isolated organ bath, and the isotonic contractions of the tissues produced by amitraz and acetylcholine were recorded on a polygraph. The chicks were divided into four groups,one control and three experimental groups (II,III,IV). The Development Of European Sea Bass (Dicentrarchus Labrax L., 1758) Eggs In Relation To Temperature, Şahi̇n Saka, Kürşat Firat, H. Okan Kamaci

Turkish Journal of Veterinary & Animal Sciences

In this study, the development of European Sea Bass (Dicentrarchus labrax L., 1758) eggs was investigated at different temperatures. The experiments were carried out at 15ºC and 17ºC. The average diameters of the eggs were determined tobe approximately 1.162±0.004 mm. The embryonic development of the eggs was observed every hour and then photographed. The hatching rates were found to be 84.8-89.4% at 15ºC and 82.3-86.3% at 17ºC, respectively. Hatching of the larvae took 87 hours at 15ºC and 69 hours at 17ºC.

This study was carried out to determine the prevalence of Cryptosporidium spp. oocysts in diarrhoeic calves in Kars province. The study was conducted between February 1999 and June 1999, involving 140 diarrhoeic calves from 8 different localities (Kars-Central district, Karakaş, Bulanık, Karakale, Hacıhalil, Boğazköy, Susuz-Central district, and Arpaçay). These localities were visited once during the study period, and fresh faecal samples were taken from the rectums of the calves up to 3 months old. A total of 200 wild pigeons, 82 young and 118 adults, captured from 12 localities in Ankara District, were examined for blood parasites. Haemoproteus columbae was found in 57% (114/200) of the pigeons examined, and no other blood parasites were detected.

In this study, the induced antibody level of rainbow trout immunized by Yersinia ruckeri bacterin was determined using two serological techniques, namely, indirect fluorescent antibody (IFAT) and enzyme-linked immunosorbent assay (ELISA). The induced humoral antibody levels were studied after intraperitonal injection of Yersinia ruckeri bacterin inoculates containing Freund's incomplete adjuvant or PBS in rainbow trout during the experimental period of 20 weeks. In this research, the pathogenesis and presence of transplacental pestivirus infection were investigated in different flocks. For this purpose, a total of 661 pregnant ewes and 29 rams were used. 174 of 661 pregnant ewes were sampled twice in the second or the third month of gestation and as well as on the day of gestation, whereas 487 of them were sampled only on the day of delivery. Some 14 ethanol extracts from 13 plant species collected from various regions of Turkey were assayed for in vitro antibacterial activity against two Gram positive and 5 Gram negative bacteria, using the disc-diffusion and dilution methods. The diameter of the inhibition zone in the disc-diffusion method shows that 12 extracts from the 14 plants studied have antibacterial activity in different levels. The broadest spectrum of activity was exhibited by Malva sylvestris against five bacteria, whereas the Verbascum spp. extract only showed activity against one. In Vitro Antibacterial Activities Of Tilmicosin And Cefquinome Alone Or In Combination Against Certain Poultry Pathogens, Seyyal Ak, Oya Keleş, Tülay Bakirel

The antimicrobial activities of cefquinome and tilmicosin were determined in vitro against P. multocida and O. rhinotracheale of poultry origin by serial broth macrodilution. Both antibiotics exhibited good in vitro antibacterial activity against the strains tested. The minimum inhibitory concentration (MIC) for cefquinome was 1.562 µg/ml and for tilmicosin was 0.78 µg/ml for both microorganisms. To investigate the effects of a combination of cefquinome and tilmicosin, the fractional inhibitory concentration (FIC) index was determined by checkerboard assay. The antimicrobial combination was additive against P. multocida (FIC=1) and indifferent against O. rhinotracheale (FIC=2).

In this research, some physical characteristics and uses of the coarse fibers obtained from hair goats raised in some provinces of Turkey were examined. For the general provinces, the diameter of fiber and natural length of fiber were 92.1±0.56 µ and 11.6±1.37 cm resepctively. According to the findings, the coarse fibres of hair goats can be used in products such as tents, stiffening cloth, card weaving, floor weaving (çul ve cicim), saddle-bags, bags, sacks, animal feeding bags and socks.

Genetic determinants of lactose fermentation, proteolytic activity and bacteriocin production in the strains L. lactis subsp. lactis MPL114, L. lactis subsp. cremoris MPC112 and L. lactis subsp. lactis biovar. diacetylactis MPD166 were studied. As a result of mutation and genetic transfer studies, it was determined that all of these characters were encoded by the 65.4 kb plasmid in L. lactis subsp. cremoris MPC112. This study was conducted to evaluate the pharmacologic effects of Polygonum lapathifolium L. (Polygonaceae), growing naturally in Turkey. The antiinflammatory activity of the ethanol extract of the root and rhizome of P. lapathifolium was evaluated using carrageenan-induced paw edema in rats. Intraperitoneal pretreatment with extract at doses ranging from 100 to 500 mg/kg significantly inhibited the acute phase of this experimental model of the inflammatory process in a dose-dependent manner. This study was conducted to investigate Serratia liquefaciens infection in juvenile cultured rainbow trout (Oncorhynchus mykiss Walbaum) during June and July in 1996 about 20 days after flooding in three farms in Erzurum. Bacterial isolates were identified from each farm and tested to determine sensitivity against 18 chemotherapeutants. A twelve-week nutrition trial was carried out to evaluate the efficacy of different carbohydrate levels on digestibility, feed intake, growth performance, carcass and muscle composition and plasma glucose level in the rainbow trout, Oncorhynchus mykiss. The objectives of this study were to (i) compare the effects of two staining techniques and one fixative solution on the morphological features of ram spermatozoa and (ii) evaluate the morphological features of dead/live spermatozoa by using Eosin-Nigrosin staining. The morphological features of raw spermatozoa were evaluated (by Phase Contrast microscopy) after staining with Eosin-Nigrosin mixture or Nigrosin (alone), or after fixation with Hancock's buffered formol saline solution. 42 raw pooled semen samples from 6 Kıvırcık rams were used. The possible use of a genus-specific polymerase chain reaction (PCR) for the identification of Salmonella colonies was tested on 14 reference strains and 38 clinical Salmonella strains isolated from chickens by the standard method of the National Poultry Improvement Plan and Auxiliary Provisions (NPIP), USDA, U.S.A. All the PCRs using primer pairs InvA1 and InvA2 (5) gave 281 bp amplification product specific for the Salmonella genus and produced results identical (100%) to those obtained with biochemical and serological methods. In this report, the isolation of a Salmonella typhimurium strain as pure culture from abcess foci on the liver, spleen, kidney, stomach and intestines, and lungs of an 8-month-old, male, Thoroughbred foal, which had died in a racehorse breeding farm, is presented. Heavy respiratory infection and prolonged antibiotic therapy were considered to be stress factors that played a significant role in this case.

Seroprevalence of Babesia ovis in Awassi sheep was studied in Urfa and environs between November 1997 and October 1998. A total of 607 serum samples were collected from sheep in 12 different localities, and tested for the presence of antibodies to B. ovis by using an indirect ELISA. Blood smears were also prepared from 110 sheep. The overall prevalence of B. ovis infection was 41.02%. B. ovis was only detected in two blood smears examined (1.82%). The purpose of the present study was to examine the effects of vitamin A and zinc interaction in laying hens fed on diets supplemented with two different levels of vitamin A (0, 10.000 IU/kg) in combination with five different levels of zinc (0, 25, 50, 100, 200 mg/kg) on egg yield, plasma and yolk concentration of zinc, and the levels of plasma triglyceride and phosphorus for 12 weeks.
