Turkish Journal of Veterinary & Animal Sciences
This investigation was carried out to evaluate the use of allergic skin tests in the diagnosis of pseudotuberculosis in sheep. For this purpose, two different allergens were prepared by two different methods. Allergens were prepared from C. pseudotuberculosis NVH 3368 strain. The study was carried out on farms with a total of 300 sheep in the province of Konya. The effects of two different allergens injected into four different sites in all three groups (infected, suspected and healthy) were examined (P < 0.05). At the end of the allergic skin test it was found that 73 (73%) of infected sheep tested positive. Sixty-one (83.5%) of these 73 were tested by SAT, 60 (82.1%) by postmortem examination and 38 (52.0%) by bacteriological examination (P < 0.05). All tests gave negative results for the healthy group. Moreover, in the infected animals, the sensitivity, specificity and predictive values from SAT, postmortem examination and bacteriological examination were investigated. The values were 100.0%, 29.0% and 75.5% for the axillary site. It was confirmed that the axilla is the best injection site in allergic tests. In conclusion, it was found the allergic skin test was a reliable method for the diagnosis of pseudotuberculosis in sheep in the field.

Transgenic Bovine Nuclear Transfer Embryos From Adult Somatic Cell Lines*, Sezen Arat, S. Jacek Rzucidlo, Steven L. Stice
Turkish Journal of Veterinary & Animal Sciences
This study was performed to examine whether adult fibroblast cells can be manipulated genetically and whether clonal lines derived from those cells can support embryonic development. A primary adult cell line was established from the ear of an aged cow. Adult cells with a plasmid containing the enhancer green fluorescence protein (EGFP) gene, and non-transfected cells were used for cloning. Green fluorescence expression was observed in 35/49 (71.4%) adult clones. The developmental rates of embryos were significantly lower for cell lines expressing EGFP (11.4%) than for non-transfected cells (20.1%, P < 0.05). However, there was no decrease in nuclear transfer (NT) developmental rates (21.5%) when donor nuclei from EGFP transfected cell lines not expressing EGFP but retaining neomycin-resistance gene expression were used as donor nuclei. The NT embryos from transfected and non-transfected cell lines had similar morphology and cell numbers. The results indicated that adult cells can complete clonal propagation including transfection and selection and can be used to produce transgenic NT embryos.

The Application Of A Phosphorus Budget Model Estimating The Carrying Capacity Of Kesikköprü Dam Lake, Serap Pulatsü
Turkish Journal of Veterinary & Animal Sciences
The aim of this study was to estimate the carrying capacity of Kesikköprü Dam Lake, Ankara, where cage farms for the intensive culture of rainbow trout are located. For this purpose Dillon and Rigler's phosphorus budget model was applied in a series of steps and the carrying capacity of the lake was found to be 3335 tonnes per year. This estimated value was about 10 times higher than the present production level of the lake. It seems possible to orientate the fish culture in cages in inland waters by using the phosphorus budget model recommended in this study.

This study was intended to establish whether there is any relation between immune deficiency and cryptosporidiosis by using an animal model. Forty Rattus norvegicus rats obtained from the Experimental Animal Laboratories of the Ege University Faculty of Medicine were divided into two groups. The first group (n = 20) received corticosteroids (dexamethasone, 1.5 mg, subcutaneously, twice weekly) for eight weeks for immunosuppression, while the control group (n = 20) received no treatment. Drinking water was supplemented with tetracycline (500 mg/L) to avoid secondary infections. The animals were kept in groups of five in separate cages under the same environment and …

Longissimus dorsi muscle obtained from beef carcasses was used in this study. Meat cores 2.54 cm in diameter were marinated in 0.34, 0.68 and 1.02 M sodium chloride (NaCl) and 0.05, 0.1 and 0.15 M calcium chloride (CaCl_2) solutions at 5 °C for 72 h. Warner Bratzler Shear (WBS) values were significantly different between samples. The samples marinated with CaCl_2 held less water than those marinated in NaCl. Cooking losses were lower in the control samples than in the marinated samples. Differential scanning calorimetry was employed to determine the amount of bound water in the meat samples. The latent heat …

Alterations In Hematological Parameters Of Rainbow Trout (Oncorhynchus Mykiss) Exposed To Mancozeb, Muhammed Atamanalp, Telat Yanik
Turkish Journal of Veterinary & Animal Sciences
Rainbow trout reared in fresh water were exposed to a sublethal concentration of mancozeb (1/2 of LC_{50} = 1.1 mg/l) at 24 h intervals, for 3 weeks in tanks containing 780 l water under natural light conditions with a constant flow (1.5 l min^{-1}) of aerated dechlorinated tap water at 9-11 °C and with no recirculation. Approximately 2 cc venous blood was drawn from each subject (after weekly exposure) for the determination of hematological parameters, using heparin as an anticoagulant. Hematological parameters were investigated weekly. The findings of this study showed that mancozeb had some effect on the hematological parameters …

Turkish Journal of Veterinary & Animal Sciences
The objective of this study was to demonstrate the specific blood velocity parameters called peak systolic velocity (PSV), end diastolic velocity (EDV) and the resistive index (RI) in the internal ophthalmic artery (IOA) and the long posterior ciliary artery (LPCA) with color Doppler imaging (CDI) in clinically normal dogs. Eight healthy dogs were used in the study. Color Doppler ultrasonography was performed on both eyes. PSV, EDV and RI measurements were obtained for both eyes. The mean RI of the IOA was 0.57 ± 0.095 cm/s and that of the LPCA was 0.58 ± 0.083 cm/s. The RI values did …
