In this research, the primary objective was to develop a microfluidic platform utilized for the size-based single cell sorting of canine cutaneous mast cell tumor (MCT) cells which were categorized into 4 groups; I, II, III and IV, upon their individual sizes ranged from 5-9, 10-14, 15-19, and equal to or over than 20 mm respectively. And the secondary aim was to establish the microfluidic-based entrapping microdevice which had a capacity to ensnare single MCT cells into its triangular microwell array at the ratio of one cell per one microwell. This study was planned to determine the prevalences of the Nosema spp. and the black queen-cell virus (BQCV) among honeybees (Apis mellifera) raised in the province of Van by PCR and to determine the molecular characteristics of the determined isolates. A total of 260 adult worker bees from 26 colonies at 5 apiary locations belonging to the province of Van in April and May 2015 were collected for this reason. Samples were examined microscopically. Various treatments are currently available for the control of Varroa mites. Some of these treatments depend on using natural and nonchemical compounds. In this study, the impacts of some materials (oxalic acid, thymol, and potassium citrate) on the survival of honey bees and some quality parameters were investigated under laboratory conditions. The potential impacts of different feeding types (sugar syrup, sugar candy, honey candy, honey jelly, and creamed honey) mixed with the tested Varroa control materials on honey bees were also studied. We investigated the effects of sex and breeding season on the morphological and histological structures of the interdigital gland in Awassi sheep (Ovis aries). Forelimbs and hind limbs were collected from 10 males and 10 females in both the breeding (June-July) and nonbreeding (November-December) seasons. The interdigital glands were located between the first and second phalanxes and were present on all of the limbs. The mean weight of the gland was 1.03 ± 0.03 g. This study was carried out to determine flea species infesting small, wild rodents and insectivorous wild mammals of the Aegean Region in Turkey. A total of 52 small, wild mammals, comprising one insectivorous and four rodent species, namely Crocidura suaveolens, Mus macedonicus, Apodemus flavicollis, Microtus levis, and Apodemus witherbyi, were captured. Fleas were collected, preserved in 70% alcohol, and labelled for identification in the field. In the laboratory, they were cleared in 10% KOH and mounted with Hoyer's medium. Fowl adenovirus serotype 4 (FAdV-4) is a new emerging viral disease of chickens worldwide. It causes inclusion body hepatitis and hepatitis-hydropericardium syndrome. Little is known about its prevalence in the Middle East. Here we report the prevalence of FAdV-4 in five chicken farms in eastern Saudi Arabia. High mortality rates were reported from birds from those five farms at 15 weeks of age. Gross examination revealed typical hydropericardium syndrome and accumulation of jelly-like materials in the pericardial cavities. We isolated FAdV-4 by using embryonated chicken egg inoculation. Contrary to popular belief, pet overpopulation is caused more by owners failing to keep their dog than with accidental or intentional breeding (Moulton et al., 1991). Factors related to relinquishment have been researched, and concluded that behavioral problems are the most reoccurring reasons observed (Bailey, 1992; Miller at al., 1996; Patronek et al., 1996; Serpell, 1966; Salman et al., 1998, 2000; Scarlett et al, 1999; New et al., 2000; Shore et al., 2003, 2005; Mondelli 2004, Blackwell et al., 2008). The main problem of BVDV is the immune suppression that is associated with lymphoid depletion shortly post infection. The mechanism of this immune suppression is not well understood. Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically devastating virus that causes a persistent infection and spreads quickly. The virus causes abortions in sows and respiratory diseases in young piglets. Current PRRSV vaccines appear to provide limited cross-protection. A virus-like particles (VLPs) vaccine is made up of key PRRSV structural proteins, which could be cross protective among the different strains. The purpose of this project was to define monoclonal antibodies against viral proteins from Epizootic Hemorrhagic Disease Virus (EHDV) to be used in quality control of vaccines produced by an industrial partner, and development of tools to identify EHDV. EHDV is an Orbivirus, which also includes Bluetongue Virus and African Horse Sickness, which are all transmitted through arthropod vectors. EHDV causes significant morbidity and mortality in white-tailed deer, but has recently been found to infect cattle. Senecavirus A (SVA) is a positive sense single-stranded virus that belongs to the genus Senecavirus within the family Picornaviridae. Senecavirus has been present in US swine population since 1988. However, there have been only a few reports of SVA associated with disease in last two decades. In late 2014 early 2015, a sudden increase in the number of cases of Senecavirus A associated with vesicular disease was observed in Brazil, the US and China. Accurate and rapid diagnostic tests are essential for the management and control of emerging infectious diseases. Therefore, the overall goals of studies presented in this dissertation were to provide improved diagnostic and surveillance capabilities for several emerging viral diseases impacting the livestock industry and to provide insight into appropriate control strategies. Recent, severe outbreaks of porcine epidemic diarrhea virus (PEDV) in Asia and North America highlighted the need for well-validated diagnostic tests for identification of PEDV infected animals and evaluation of their immune status. The role of the renin-angiotensin aldosterone system in the genesis of hypertension between nonhunting dogs and hunting dogs of various breeds was comparatively studied in the city of Tuzla (Bosnia and Herzegovina). Sixty dogs were divided into four subgroups based on different age categories. Hypertension was recorded in 21 dogs or in 35% of examined dogs. Fourteen dogs had mild (23.3%), 5 dogs had moderate (8.3%), and 2 dogs had severe hypertension (3.3%). The most cases of hypertension (n = 9) were recorded in hunting dogs aged from 9 to 12 years. This study aimed to determine Salmonella enterica subspecies enterica serovar Enteritidis (SE) presence by conventional serotyping and SE-specific real-time PCR (SE-rPCR) in poultry-derived food and avian isolates in our laboratory Salmonella spp. collection. In this study, Salmonella strains from dairy cows, calves with diarrhea, and buffalo and camel feces were isolated and serotyped. There were 869 feces samples (437 calves, 287 dairy cows, 100 buffalo, and 45 camels) collected from 13 provinces and 21 farms. Preenrichment feces samples were added to Rappaport-Vassiliadis medium. Samples were then subcultured on xylose lysine tergitol-4 agar and plates. Suspected colonies of Salmonella were confirmed by latex agglutination test. In order to assess the prevalence of Trypanosoma evansi, a parasitological, molecular, and serological-based investigation was carried out in randomly sampled horses (n = 375) belonging to different age groups, sexes, and localities from the district of Bahawalpur, Pakistan. The diagnostic performance of applied tests was also compared. The prevalence was recorded as 0.5% with Woo's test, 1.3% with both ITS CF/BR PCR and RoTat 1.2 PCR, and 14.4% with CATT/T.evansi. Based on CATT/T.evansi, significant differences (P ≤ 0.05) were observed for prevalence estimates according to different localities, sexes, body condition scores, and origins. The aim of this study was to look at four ultrasonographic approaches that diagnose acquired reticular diaphragmatic hernia (RDH) using either left or right caudal thoracic approaches (4th/5th intercostal space) and compare them to those made more cranially (1st/2nd intercostal space). Each sonographic observation was made in 27 unsedated, standing animals (22 buffaloes and 5 cattle) that were surgically confirmed for RDH. To identify metabolic indicators in blood and urine for early pregnancy in dairy cows (32 crossbred and 12 purebred zebu cows), blood and urine samples were collected on days 0, 15, 20, 24, and 30 postbreeding. The plasma progesterone (P4) concentration was significantly (P < 0.05) higher in pregnant cows than in nonpregnant cows after day 15. The plasma blood urea nitrogen (BUN) concentration remained significantly (P < 0.05) higher in pregnant cows on day 15. The plasma concentrations of cholesterol, glucose, protein, and nonesterified fatty acid (NEFA) did not differ between pregnant and nonpregnant cows. The urine protein concentration was significantly higher (P < 0.05) in pregnant zebu cows compared to nonpregnant zebu cows. Plasma P4 was significantly (P < 0.001) and positively associated with urine glucose levels in nonpregnant crossbred cows. In nonpregnant zebu cows, the correlation between P4 was positive (P < 0.05) with plasma cholesterol, glucose, urine glucose, and protein. In pregnant zebu cows, P4 was negatively associated with plasma BUN and positively (P < 0.05) associated with both plasma and urine glucose levels. Thus, urinary proteins can be used as a noninvasive tool for early nonpregnancy confirmation in cows with very high accuracy at around 24 days after artificial insemination.

The aim of the study was to determine the seroprevalence of foot-and-mouth disease (FMD) and isolation of the FMD virus (FMDV) in the milk of apparently healthy Asian buffaloes. A cross-sectional study was conducted in periurban dairy farms (n = 20) on ≥20 animals per farm in the suburbs of Islamabad, Pakistan. Sera samples (n = 200) were collected to monitor FMDV seroconversions. The analysis of serum samples using ELISA indicated a high seroprevalence of FMD (78%; 156/200) in Asian buffaloes. The purpose of the study was to use decision trees to predict increased levels of somatic cells in cow's milk. The material for the study comprised data collected in 2012-2014 from five farms in Poland equipped with an automatic milking system. Data on 803 Polish Holstein-Friesian cows were collected. In order to predict somatic cell count data mining techniques were used to build a graphical model of a decision tree. The aim of this study was to determine the slaughter and carcass characteristics of Herik lambs finished under intensive husbandry conditions. Twenty single male lambs, ten of which had long, semifat tails and ten of which had short, round, and fat tails, all with an average live weight of 20 kg, were used in the study. Thirty Holstein calves presented with infectious diarrhea during the second week following birth were utilized to evaluate the effects of a standardized product containing a combination of probiotics, ß-glucans, and glyconutrients (GLY) in support of antibiotic therapy during the early rearing phase (day 7 to day 45 following birth). Treatments were 1) antibiotic therapy (trimethoprim and sulfas; ANTIB), and 2) ANTIB plus 5 g/calf per day GLY. Treatments did not affect diarrhea duration, averaging 16.7 days. However, GLY support decreased (P < 0.05) the mortality rate and increased (P < 0.05) live weight gain. Increased weight gain was attributable to enhanced (P < 0.05) consumption of calf starter. Supplemental GLY increased (P < 0.05) serum glucose and lowered (P < 0.05) serum urea nitrogen, nonesterified fatty acids, and cortisol concentration. We conclude that daily provision of 5 g of a standardized product containing probiotics, ß-glucans, and glyconutrients to diarrheic calves under antibiotic therapy will promote health and enhance early growth performance.

Mitochondria play a fundamental role in the regulation of cell death during accumulation of oxidants. High concentrations of atmospheric oxygen (hyperoxia), used clinically to treat tissue hypoxia in premature newborns, is known to elicit oxidative stress and mitochondrial injury to pulmonary epithelial cells. A consequence of oxidative stress in mitochondria is the accumulation of peroxides which are detoxified by the dedicated mitochondrial thioredoxin system. This system is comprised of the oxidoreductase activities of peroxiredoxin-3 (Prx3), thioredoxin-2 (Trx2), and thioredoxin reductase-2 (TrxR2). Intrauterine growth restriction (IUGR) is a leading cause of perinatal morbidity and mortality. Low birth weight resulting from preterm birth and/or IUGR is an underlying factor in 60–80% of perinatal death worldwide, and is particularly common in developing countries (UNICEF, 2008). Furthermore, studies have linked IUGR and the associated fetal malnutrition to increased incidence of metabolic syndrome in adult life (Barker et al., 1993; Godfrey and Barker, 2000). Poxviruses have evolved multiple strategies to subvert signaling by Nuclear Factor κB (NF-κB), a crucial regulator of host innate immune responses. Here, we describe an orf virus (ORFV) virion-associated protein, ORFV119, which inhibits NF-κB signaling very early in infection (≤30 min post infection). ORFV119 NF-κB inhibitory activity was found unimpaired upon translation inhibition, suggesting that virion ORFV119 alone is responsible for early interference in signaling. A C-terminal LxCxE motif in ORFV119 enabled the protein to interact with the retinoblastoma protein (pRb) a multifunctional protein best known for its tumor suppressor activity. Bovine skin and teat papillomas were examined for immunohistochemical expression of bovine papillomavirus (BPV), proliferating antigens (PCNA, Ki-67), CD3+ T lymphocytes, and some of the type III intermediate filaments. Papillomas showed epidermal hyperplasia, koilocytes, and hyperkeratosis with prominent dermal connective tissue. Immunohistochemistry found the presence of BPV antigens in the nuclei of granulosa cells. PCNA and Ki-67 revealed positive nuclear staining in the dermis and epidermis. Perivascular cuffing in the dermis predominantly consisted of CD3+ T cells. In this experimental study, effects of bovine amniotic fluid and hyaluronic acid were compared on an osteoarthritic model induced in rabbit stifle joints. The study was performed on 21 rabbits undergoing anterior cruciate ligament transaction for establishing osteoarthritis. Four weeks later, the rabbits were divided into 3 equal groups of 7 animals each.
