The Effect Of Maternally-Derived Antibodies Against Antibody Production In Vaccinated Piglets With Modified-Live Porcine Reproductive And Respiratory Syndrome Virus Vaccines, Sunit Mebumroong
Chulalongkorn University Theses and Dissertations (Chula ETD)
The humoral antibody responses following vaccination with different lineage of modified live genotype 2 porcine reproductive and respiratory syndrome virus (PRRSV) vaccines (MLVs) were investigated. Moreover, the mortality and growth performance in pigs were observed. Twenty-four pigs with high maternally derived antibody (MDA) levels (SN titer ≥ 8) from each PRRSV positive Herd 1 and Herd 2 were allocated to the H1, H1-1, and H1-2 groups (Herd 1) and H2, H2-1, and H2-2 groups (Herd 2). Twenty-four pigs with low MDA levels (SN titer <8) were allocated to the L1, L1-1, and L1-2 (Herd 1) and L2, L2-1, and L2-2 groups (Herd 2). Twenty-four pigs from the negative herd (Herd 3) were allocated to 3 groups: N, N-1, and N-2 groups. The H1, H2, L1, L2, and N groups were unvaccinated. The H1-1, H2-1, L1-1, L2-1, and N-1 groups were vaccinated with MLV (Ingelvac® PRRS MLV) at 2 weeks of age. The 5 other groups: H1-2, H2-2, L1-2, L2-2, and N-2 groups were vaccinated with MLV2 (Prime Pac® PRRS MLV) at 2 week of age. The antibody responses were measured by ELISA and serum neutralization (SN). The mortality was not observed in all vaccinated groups. Additionally, pigs with high MDA levels from the H1 and H2 groups did not exhibit the mortality throughout the study. ADG in pigs vaccinated with each MLV1 and MLV2 was significant higher than that in unvaccinated pigs. However, pigs vaccinated with MLV2 showed the highest values of ADG. Seroconversion was observed in all vaccinated groups with no significance difference in antibody titers between pigs with low and high MDA levels. However, pigs with low MDA levels showed the earliest and highest antibody level post vaccination. The highest SN-titer against MLV1 isolate and MLV2 isolate was observed in pig from the L1-1 and L2-2 groups, respectively. The field PRRSV-2 isolates: THA_SP/RB_S1/P1/0120-18 and THA_WC/RB_F165/20-22, the SN titers against these isolates were not observed in vaccinated groups. These results indicated that MDAs might not affect the immune response post vaccination. PRRSV-2 MLVs reduced the mortality and improved growth performance of pigs in the endemically PRRSV infected herds. Although, the vaccine was changed in weaning pigs, the antibody responses were detected post vaccination in both pigs with low and high level of MDAs. Moreover, the genetic similarity between vaccine and field virus was not related to the protection. Vaccine selection should depend on the induction of immune response and protection against heterologous PRRSV infection.

Relationship Between Insulin-Like Growth Factor-1 And Semen Quality In Asian Elephants (Elephas Maximus), Yuqing Yang
Chulalongkorn University Theses and Dissertations (Chula ETD)
Evidence has been increasingly suggested that insulin-like growth factor-1 (IGF-1) is related to steroidogenesis and spermatogenesis. This may be conducive to find the causes of poor sperm quality in Asian elephants (Elephas maximus). The present study aimed to investigate whether correlations among serum IGF-1 concentration, serum testosterone level and semen variables exist in elephants. A total of 17 ejaculates (1–3 ejaculates/bull) were collected by performing transrectal massage. Before each ejaculate, blood samples were obtained (n = 17). Subsequently, semen characteristics of each ejaculate were evaluated. Assessments of precision (calculations of the intra- and inter-assay coefficient of variation, CV) and accuracy …

Background: Aminorex, (RS)-5- Phenyl-4,5-dihydro-1,3-oxazol-2-amine, is an amphetamine-like anorectic and in the United States a Drug Enforcement Administration [DEA] Schedule 1 controlled substance. Aminorex in horse urine is usually present as a metabolite of Levamisole, an equine anthelmintic and immune stimulant. Recently, Aminorex identifications have been reported in horse urine with no history or evidence of Levamisole administration. Analysis of the urine samples suggested a botanical source, directing attention to the Brassicaceae plant family, with their contained GlucoBarbarin and Barbarin as possible sources of Aminorex. Since horsepersons face up to a 1 year suspension and a $10,000.00 fine for an Aminorex …

Antimicrobial resistance is a threat to agricultural production and public health. In this proof-of-concept study, we investigated predicting antimicrobial sensitive/resistant (S/R) phenotypes and host sources of Escherichia coli (n = 128) based on differential fatty acid abundance. Myristic (14:0), pentadecanoic acid (15:0), palmitic (16:0), elaidic (18:19) and steric acid (18:0) were significantly different (α = 0.05) using a two-way ANOVA for predicting nalidixic acid, ciprofloxacin, aztreonam, cefatoxime, and ceftazidime S/R phenotypes. Additionally, analyses of palmitoleic (16:1), palmitic acid (16:0), methyl palmitate (i-17:0), and cis-9,10-methyleneoctadecanoic acid (19:0Δ) showed these markers were significantly different (α = 0.05) between isolates obtained from cattle …

Spaying of female dogs is a widespread practice, performed primarily for population control. While the consequences of early spaying for health are still being debated, the consequences for behaviour are believed to be negligible. The current study focused on the reported behaviour of 8981 female dogs spayed before 520 weeks (ten years) of life for reasons other than behavioural management, and calculated their percentage lifetime exposure to gonadal hormones (PLGH) as a proportion of their age at the time of being reported to the online Canine Behavioral Assessment and Research Questionnaire (C-BARQ). We found that 23 behaviours differed between entire …

Hyarulonicacid (HA) is an abundant molecule found in synovial fluid and plays an important role as a lubricant and shock absorber for the joints. Nonetheless, the effects of synovial fluid replacement with HA after canine stifle surgery remains elusive. A retrospective study was conducted on 54 small breed dogs with either medial patellar luxation (MPL) or cranial cruciate ligament ruptures (CCLR). Dogs were classified as HA (21 with MPL-HA and 16 with CCLR-HA) or control (10 with MPL-C and 7 with CCLR-C) groups. The weight bearing score, lameness score, and pain scores were compared between HA and control groups on …

Tall fescue is the primary cool season perennial forage grass in southeastern US for grazing livestock. Tall fescue the is known to be drought tolerant , disease resistant, grazing tolerant and overall very hardy. Many of these attributes common to fescue are due to a symbiotic relationship between the fescue plant and an endophyte, Epichloë coenophiala , which produces a class of mycotoxins known as ergot alkaloids. These ergot alkaloids are similar in structure to several biogenic amines, such as epinephrine, norepinephrine, and dopamine. When consumed, ergot alkaloids cause a syndrome known as fescue toxicosis which causes problems with weight …

Oropharyngeal swabs of 262 free-ranging macaques (Macaca fasicularisand M. mulatta) were tested for Herpesviridae. The occurrence was 24.4% (64/262), and no macaques showed clinical signs of the disease. The virus was restricted to the northern and north-eastern regions of Thailand. DNA polymerase genes of the 64 positive samples were then characterized and analyzed. Eight samples (3.1%) were found to be closely related to retroperitoneal fibromatosis-associated herpesvirus (RFHV) and were classified in the genus Rhadinovirus, subfamily Gammaherpesvirinae. The phylogram of the virus revealed it to be species-specific within the macaque population. A total of 56 samples (21.4%) exhibited a nucleotide relationship …

Rubber Band Syndrome In A Dyspneic Dog, Nithida Boonwittaya, Saroch Kaewmanee
The Thai Journal of Veterinary Medicine
The rubber band syndrome has been reported inboth human and animal patients but the number of cases shows it is an uncommon condition. Most cases live in communities where a rubber band is commonly used for a wide variety of purposes. Here we report a case of a dyspneic dog with a linear constricting scar and a discharging sinus on the ventral neck. The radiography showed constructive signs. Surgical exploration found a colored rubber band burrowing into the tracheal lumen. Following surgery, the dog recovered without any complications in the postoperative follow-up period. This case emphasized that rubber band syndrome …

The Thai Journal of Veterinary Medicine
Although the major dog population in Thailand is the Thai native dog, its anatomy has rarely been studied. This study aimed to investigate anatomical variations of the canine coeliac trunk (CT) in Thailand by observing 62 cadavers including 46 Thai native dogs and 16 non-Thai native dogs. All cadavers were preserved in formalin solution, and the arteries were illustrated by injection of red coloured latex. Variations of CT were initially classified into 2 main types and 7 subtypes (type 1a-e and 2a-b) basedon the previous reports. Type 1 was described as CT separated from the cranial mesenteric artery (CMA) while …

Staphylococcus aureusis the leading cause of mastitis in dairy goats and is globally recognized as a significant mastitis-causing pathogen in dairy animals. Previous studies have established it as the causative agent of various human disorders, including food poisoning and Staphylococcal enterotoxins. Furthermore, poor sanitary conditions suit infection by this bacterium. This study determined the occurrence of S. aureusin infected dairy goats by molecular analysis. A hundred raw milk samples were collected from two infected dairy goat breeds (Beetal = 71; Teddy = 29) and cultured on blood agar media. The strain identified as S. aureusby morphological method (Gram staining), biochemical …

Competitive exclusion (CE) relies on the activity of normal flora to limit intestinal colonization by enteric pathogens. This study aims to investigate the effects of CEon Salmonellainfectionin broilers. The protective effect of CE on Salmonella cecal colonization was evaluated in 1 day-old chicks. In the laboratory trial, CE products were administered by oral inoculation, drinking water and whole body spray. Three days after treatment, the chickens were challenged with 107cfu/mL of SalmonellaEnteritidisorally, and were evaluated for 10 days. The CE-treated chickens showed comparable protection andthe number of Salmonella in the cecal contents significantly decreased(P<0.05)compared with those of the positive control. In the farm trial,CE was administeredby whole body spray at the hatchery and the second was administered by drinking water and evaluated at 32 and 42 days, respectively. The CE significantly reduced Salmonellacontamination in the farm trial andno Salmonellawas detected in the cecal contents compared with the control group. Average body weight gains, feed conversion ratioand theperformance indexof the chickens were greater than thecontrol group. Thus, the use of CEhad a positive effect on broiler performance. Furthermore, the CE-treated chickens displayed greater intestinal histology including villous height, submuscular layerand cell mitosis. These studies demonstrate that CEwas able to protect chickens from Salmonella cecalcolonization and it is apparent that new methodologies associated with the development of a workable CE program are needed in thepoultry industry.

The underlying mechanism of porcine reproductive and respiratory syndrome virus (PRRSV) causing reproductive failure and re-circulation in herds has remained unclear. Endometrial cells primarily infected with PRRSV may serve as a significant target for PRRSV eradication. Primary endometrial (PE) cells from the porcine uterus were isolated and cultivated to pursue this possibility. Immunocytochemistry analysis revealed the protein expression of classical estrogen receptors (ER-and ER-), but not PRRSV receptors, CD163 and sialoadhesin in PE cells. PE cells were apically/basolaterally inoculated with PRRSV type I/type II isolated from PRRSV infected lungs or mock infection. Cytopathic effects (CPE) and PRRSV-GP5 positive cells were …

Graduate Theses and Dissertations
Advancements in poultry research continue to add knowledge and understanding to an already technical industry. Such information has led to new developments in early feeding strategies which provide nutrients to broiler chicks while in the egg or during transport. This wealth of knowledge in poultry research extends beyond chickens and is utilized for other species, such as quail and pheasants, including how fertile eggs are stored and incubated. However, methods of early feeding broilers have not been investigated in industry relevant approaches to understand its effects, including how timing of feed access interacts with the hatch window. As well, quail …
