Introduction:
A pressure gradient is where high and low pressure meet. It is responsible for displacement and irritation of the skin commonly caused by tight bandages, bracelets, or socks. The irritation is at the edge where the skin is displaced by the high and low pressure. An analysis of the effect pressure gradients have on discomfort would have significant medical relevance for any device, fixture, or clothing pressed against the skin at high pressure. This primarily includes prosthetic and orthotic devices.

The combination of outsole, midsole and insole layers of new shoes provide proper cushioning to the foot during the daily activities of life. Subsequently, the wear-based degeneration or damage to one or more of these sole layers can reduce the cushioning effect to the plantar surface of the foot. This reduction of cushioning effect can impact the distribution of stress concentration regions along the foot plantar surfaces and lead to foot pathologies. Osteoarthritis (OA) is a musculoskeletal disorder specified as a joint disease that affects mostly human joints worldwide. Total knee arthroplasty (TKA) is performed to restore the affected joint and relieve the symptoms. However, tibial malrotation, which is one of the most common errors in TKA, results in poor function of the implant and pain after the procedure. Macrophages are an important cell type in the body responsible for orchestrating the inflammatory response and return to homeostasis. Macrophages are often implicated in a multitude of disease pathologies including delayed wound healing, excessive fibrosis, autoimmune disease, and immunosuppression within the tumor microenvironment. Although macrophages have been targeted for therapeutic applications, success in utilizing them has remained elusive due to their relative ease of phenotype switching between inflammatory and anti-inflammatory. Total shoulder arthroplasty (TSA) is often considered a surgery of last resort for patients with debilitating pathologies of the glenohumeral joint such as osteoarthritis, humeral head fracture, and advancing rotator cuff tears. TSA replaces the articulation between the humeral head and the glenoid fossa, with the goals of relieving pain and restoring function. Two types of total shoulder implants are available: Anatomic Total Shoulder Arthroplasty (aTSA) and Reverse Total Shoulder Arthroplasty (rTSA). Tennis, a widely played sport across various age and skill groups, prompts continual skill improvement among competitive players seeking a competitive edge. This study explores two approaches for enhancing playing style: motion capture (MoCap) and surface electromyographic (sEMG) signals. The study addresses a gap in simultaneous examination of MoCap and entire dominant leg muscle activation, particularly concerning the influence of skill level and gender on various tennis strokes. This research developed methods for culturing self-assembling capillaries in an injectable gel as a potential method for vascularizing tissue-on-a-chip models to mimic physiological drug delivery. Additionally, a mathematical model was developed as a tool for understanding nutrient delivery and comparison of potential delivery systems. Organs-on-a-chip provide novel platforms for studying biology and physiology in 3D, allow exploration of tissue engineering on a manageable scale, and serve as models for drug screening and drug-delivery testing.

This paper presents a non-motorized foot and ankle exoskeleton to assist persons who have no lower limb or abdominal function. Exoskeletons are defined as “an artificial external supporting structure” [1], they are wearable devices that can be used to assist the capabilities of the human body. Reverse total shoulder arthroplasty (RTSA) is a procedure that aims to fix irreversible rotator cuff damage and degenerative glenohumeral joint diseases that hinder arm abduction. One major complication of RTSA is glenoid baseplate failure resulting from increased movement at the bone-baseplate interface. This study examined 36, 12 PCF composite Sawbones® scapulae. The specimens were split equally into 6 study groups, each having different implant configurations with varying baseplate lateralization and central screws or posts. Each specimen was subjected to cyclic shear loading and failure testing on an Instron to determine implant micromotion and load values.

Bladder outlet obstruction (BOO) is a prevalent urological condition and can be characterized by the presence of lower urinary tract symptoms (LUTS), such as hesitancy, weak stream, and nocturia. The main cause of BOO is a partial blockage of the urethra, which results in elevated voiding pressure, high storage pressure, and tissue ischemia, which are thought to be the triggers of bladder inflammation and subsequent fibrosis. Clarifying multifactorial musculoskeletal disorder etiologies supports risk analysis and development of targeted prevention and treatment modalities. Deep learning enables comprehensive risk factor identification through systematic analysis of disease datasets but does not provide sufficient context for mechanistic understanding, limiting clinical applicability for etiological investigations. Conversely, multiscale biomechanical modeling can evaluate mechanistic etiology within the relevant biomechanical and physiological context. The US has seen a greater number of mass casualty events, and conflicts across the globe have led to an increased use of tourniquets. Large vessel occlusion during acute ischemic strokes requires mechanical thrombectomy (MT) procedures using stent retrievers (SR). However, complete clot removal typically takes several attempts, which can amplify endothelial cell injury (EI). This project aims to characterize the extent of EI during MT using an in vitro vascular platform and a finite-element model (FEM). Polymer nanofibers have attracted high interest in a wide array of applications, including biomaterial scaffolding, textiles, sensors, reinforcement in composites, and filtration systems. Many of these applications combine nanofibers into non-woven fabrics for better properties and easier manipulation. Physiological abnormalities in pulmonary granulomas–pathological hallmarks of tuberculosis (TB)–compromise the transport of oxygen, nutrients, and drugs. In prior studies, we demonstrated mathematically and experimentally that hypoxia and necrosis emerge in the granuloma microenvironment (GME) as a direct result of limited oxygen availability. Building on our initial model of avascular oxygen diffusion, here we explore additional aspects of oxy- gen transport, including the roles of granuloma vasculature, transcapillary transport, plasma dilution, and interstitial convection, followed by cellular metabolism. Approximate analytical solutions are provided for oxygen and glucose concentration, interstitial fluid velocity, interstitial fluid pressure, and the thickness of the convective zone. …

Rock climbing is a total body sport testing the mental and physical capabilities of athletes. Efficiency of movement can improve the ability of climbers to perform at a higher capacity and reduce the risk of injury. Data on the relationship between the continuous relative phase (CRP) of contralateral joint rotations and time to completion of a bouldering move are ways to assess rock climbing. While 3D direct linear transformation is a versatile motion capture method, care must be taken so that the cameras are not bumped or adjusted between the object space calibration phase and the end of video recording. Disturbing the cameras during this time will misalign the view, making the movement data invalid for the associated calibration. This study aimed to assess the effects of whole-body vibration (WBV) on the brain through changes in blood protein concentrations, seat and head accelerations, and symptomatology over time. Subjects were assigned to control, 1-hour, or 8-hour vibration exposure groups. Glial fibrillary acidic protein (GFAP) and S100B concentrations were measured. Root mean square average weighted vibration (Aws), vibration dose value (VDV), and seat-to-head transmissibility (STHT) were calculated. Under normal physiological conditions, bone remodeling is balanced by bone-forming osteoblasts and bone-resorbing osteoclasts, and the activity of osteoblasts and osteoclasts is orchestrated by osteocytes. Osteocytes are the most abundant cell type found in bone and ample evidence suggests they are bone’s main mechanosensory cell type. Therefore, understanding osteocyte biology and responses to mechanical signals, or lack thereof during unloading, may lead to new therapeutic targets to combat bone loss during disuse. Mechanical stiffness of the extracellular matrix (ECM) impacts many cellular functions such as proliferation, migration, and differentiation. ECM stiffness is sensed by a cell via integrin-based focal adhesions (FAs) by changing conformation and biochemical activities of molecules within FAs by the exchange of the force between the ECM and filamentous actin (F-actin). Cells in turn respond to this stiffness by generating traction force that plays an important role in many biological events such as tissue morphogenesis, stem cell differentiation, wound healing, and cancer cell metastasis. The lymphatic system plays a vital role in maintaining fluid balance in living tissue and serves as a pathway for the transport of antigen, immune cells, and metastatic cancer cells. In this study, we investigate how the movement of cells through a contracting lymphatic vessel differs from steady flow, using a lattice Boltzmann-based computational model. Our model consists of cells carried by flow in a 2D vessel with regularly spaced, bi-leaflet valves that ensure net downstream flow as the vessel walls contract autonomously in response to calcium and nitric oxide levels regulated by stretch and shear stress levels. Contemporary total knee arthroplasty (TKA) has not fully restored natural patellofemoral (P-F) mechanics across the patient population. Previous experimental simulations have been limited in their ability to create dynamic, unconstrained, muscle-driven P-F articulation while simultaneously controlling tibiofemoral (T-F) contact mechanics. The purpose of this study was to develop a novel experimental simulation and validate a corresponding finite element model to evaluate T-F and P-F mechanics. Urban farming can enhance the lives of communities and help reduce food scarcity. This paper presents a conceptual prototype of an efficient urban farming community that can be scaled for a single apartment building or an entire community across all global geoeconomics regions, including densely populated cities and rural, developing towns and communities. When deployed in coordination with smart crop choices, local farm support, and efficient transportation then the result isn’t just sustainability, but also increasing fresh produce accessibility, optimizing nutritional value, eliminating the use of ‘forever chemicals’, reducing transportation costs, and fostering global environmental benefits.
