Hybrid structures formed between single-stranded DNA (ssDNA) and single-walled carbon nanotubes (SWCNT) have emerged as promising components in future direct DNA sequencing technologies due to their unique morphological and electronic properties. The nanoscale configuration of these hybrids, including the DNA wrapping angle, pitch, and contour behavior, directly influences their potential for stable electronic readout. This thesis presents an experimental study of DNA-SWCNT hybrid morphology using a fixed, reproducible wrapping protocol designed to minimize environmental variability and isolate the influence of DNA nucleotide sequence on hybrid structure. As described in prior work, this protocol produces well-dispersed hybrids suitable for high-resolution imaging. …

Physical forces drive many cellular processes such as migration and proliferation. A metastatic tumor will have invasive strands/chains that extend from the main tumor. These chains of cells collectively migrate away from the main tumor to establish new colonies elsewhere in the body. However, how physical forces drive this collective invasion and the required energy for this process is not well understood. Invasion of a metastatic tumor to surrounding tissues leads to a majority of cancer-associated death and is often a collective effort among cells, it is therefore important to fully understand the process behind collective cancer invasion.

Macrophages are an important cell type in the body responsible for orchestrating the inflammatory response and return to homeostasis. Macrophages are often implicated in a multitude of disease pathologies including delayed wound healing, excessive fibrosis, autoimmune disease, and immunosuppression within the tumor microenvironment. Although macrophages have been targeted for therapeutic applications, success in utilizing them has remained elusive due to their relative ease of phenotype switching between inflammatory and anti-inflammatory. Injectable hydrogels offer numerous advantages in various areas, which include tissue engineering and drug delivery because of their unique properties such as tunability, excellent carrier properties, and biocompatibility. These hydrogels can be administered with minimal invasiveness. In this study, we synthesized an injectable hydrogel by rehydrating lyophilized mixtures of guar adamantane (Guar-ADI) and poly-β-cyclodextrin (p-βCD) in a solution of phosphate-buffered saline (PBS) maintained at pH 7.4. The hydrogel was formed via host-guest interaction between modified guar (Guar-ADI), obtained by reacting guar gum with 1-adamantyl isocyanate (ADI) and p-βCD. The mitral valve serves a critical role in healthy cardiac function by ensuring the unidirectional flow of oxygenated blood from the left atrium into the left ventricle. It experiences the highest pressures found within the heart and its closure is the result of a complex interaction of several different structures that, furthermore, are unique to each individual. Despite the valve’s vital role however, the specific function of these constituent structures is not fully understood. Dissipative particle dynamics (DPD), a coarse grain simulation method, was applied to the membrane formation process of non-solvent induced phase separation (NIPS) to gain further insight on the mechanism of certain variables and how they affect the final morphology. NIPS involves two solutions, an organic polymer dissolved in an organic solvent colloquially called the dope and an aqueous coagulation bath, brought into contact with one another. The solvents then mix, causing the polymer to fall out of solution as an asymmetric membrane with a dense surface layer and a more open subsurface layer in response to the decreasing solubility. Volumetric muscle loss (VML) overwhelms muscle’s robust capacity for regeneration. A key event in the etiology of VML injury is the bulk loss of structural cues provided by the underlying extracellular matrix (ECM). While muscle is a highly structured tissue, with cell and ECM alignment in the direction of contractile force production, the impact of scaffold alignment on recovery remains unclear. Bulk human decellularized skeletal muscle (DSM) tissues were sectioned into 10 x 1-2 mm fibers. My primary project was to modernize an abandoned pulse duplicator device and protocol to allow the company to re-enter a market area which they left nearly 15 years prior: heart valve testing. The company halted this initiative due to changes in international testing standards in previous years. Microneedle patches are a promising source for transdermal diffusion of macromolecules and are designed to painlessly penetrate the skin. In this study, a biodegradable chitosan microneedle patch to deliver meloxicam for managing pain in cattle was tested. The potential of reuse of the polymeric solution to fabricate the patches, optimization of fabrication, morphological analysis of the microneedle patch and analysis of preservation of the chemical composition after sterilization were evaluated. This increase in friction causes a fraction of them to give out within ten years. Therefore, to minimize the probability of this happening, recent research has been conducted regarding laser surface texturing of the cobalt-chromium-molybdenum alloy components of artificial joints. Venous valve failure allows for the retrograde, or backward, flow of blood into the lower extremities, which leads to Chronic Venous Insufficiency (CVI). CVI infringes upon quality of life through ulceration and can result in death due to Deep Vein Thrombosis (DVT), or blood clots, causing pulmonary embolism. A successful treatment of CVI restores valve function and prevents retrograde blood flow; however, current bioprosthetic venous valves exhibit low patency and high calcification. The focus of regeneration therapy for traumatic brain injuries and Alzheimer's disease is on the promotion and growth of neuronal cells. In vitro research attempts to improve this by modifying the stiffness and topography of the extracellular matrix (ECM). However, the limitations of in vitro experiments make it difficult to control the individual factors influencing neuronal cell growth. Virus filtration is an integral part of the downstream purification of mammalian cell culture-derived biotherapeutics to assure the viral safety of the products. Virus filtration membranes remove viruses based on a size-exclusion mechanism. Commercial parvovirus filers possess unique membrane structure and are designed to remove smaller non-enveloped parvoviruses with size 18-26 nm. However, some filters face issues, such as pre-mature fouling, the decline of filtrate flux, and reduction in virus retention. For this Honors Thesis project, I designed and fabricated a ventilation and recirculation airway sub-system prototype for a hydroponic demonstration unit to be used as an educational demonstration for prospective students, illustrating the capabilities of biological engineers. This thesis focuses on the design of an autonomous air system that toggles between ventilation and recirculation modes based upon sensor output. In the literature review I discuss how ventilation can be used to achieve a targeted climate, the benefits of hands-on projects for education, and the optimum parameters for growing herbs in a hydroponic system.

Studies show that inductive teaching methods for Biomedical Transport Phenomena greatly benefit from an accompanying visual aid. The following project aimed to develop a MATLAB GUI application that illustrates steady-state heat transfer with a graph and heat map using user-defined boundary conditions and numerical parameters. The application was evaluated using a survey that first familiarized the user with the GUI by running through heat transfer exercises, then allowed the user to experiment with the application, and finally asked users about their experiences using a questionnaire. The renin-angiotensin system (RAS) plays a crucial role in the regulation of renal, cardiac, and vascular physiology. This system regulates in vivo blood pressure and fluid balance. As renal blood flow decreases, the kidneys convert prorenin into renin and secrete it into the circulatory system. Renin then converts angiotensinogen into angiotensin I (ang-I). The ang-I is then converted into angiotensin II (ang-II) by the angiotensin-converting enzyme (ACE). Ang-II, a vasoconstrictor, increases blood pressure by causing the blood vessels to narrow. Recent evidence suggests that RAS may be involved in the progression of valve disease, most notably, aortic stenosis.

Volumetric muscle loss affects both military and civilian persons. The hallmark of this injury is incomplete muscle regeneration, excessive fibrosis, and chronic inflammatory signaling resulting in permanent functional loss. Since permanent functional loss drastically reduces quality of life, many studies have been conducted to improve force recovery. Current scientific literature considers a repair strategy of either devitalized scaffolds infused with growth factors or viable tissue plus activating factors to be the more promising interventions for optimal force recovery. Traumatic brain injuries (TBIs) are a major global health concern that have an economic impact of $60 billion in the United States in related costs annually. Developing drugs for TBI treatment is an approach that currently faces limitations involving the permeability of the blood-brain barrier (BBB). The BBB naturally limits molecules from reaching the brain as a protective mechanism against disease, acting as a barrier during drug delivery. This paper details an investigation into methods and designs of 3D printing a microfluidic system capable of droplet emulsion using NinjaFlex filament. The specific field in which this paper’s experiment is rooted is dubbed “BioMEMS,” short for bio microelectromechanical systems. Angiotensin-II (Ang-II), a peptide hormone, is a potent vasoconstrictor and cell mitogen. It has been implicated in the development of hypertension as well as atherosclerosis. Recent work has shown that sclerotic aortic valves possess expression of angiotensin type I receptor (AT-1R) and angiotensin converting enzyme (ACE), suggesting altered angiotensin signaling during disease. The role of altered angiotensin signaling on aortic valve mechanics, however, is not clearly understood.
