Scoliosis is an abnormal spinal curvature of greater than 10 degrees. Severe scoliotic deformities are addressed with highly invasive procedures: anterior or posterior spinal fusion approaches. This invasiveness is due, in part, to the constraints of current surgical planning, which utilizes computed tomography (CT) scans unable to discern spinal ligaments that are dissected to make the spine sufficiently compliant for correction. If localization of ligaments and soft tissues were achieved pre-operatively, corrective procedures could become safer and more efficient by using finite element (FE) biomechanical simulations to determine decreased incidences of ligament releases. Musculoskeletal modeling provides an alternative to in-vivo characteristics that are difficult to directly measure for movements such as running, especially for trunk muscles and joints. The full-body-lumbar-spine (FBLS) model by Raabe and Chaudhari, 2016 is an OpenSim model created for simulations of jogging. The lifting full-body (LFB) model by Beaucage-Gauvreau et al., 2018 is an adaptation of the FBLS created for estimating spinal loads during lifting. PURPOSE: Determine validity of the FBLS and LFB models in simulating pelvis and lumbar spine kinematics during running. This study examines several different kinetic variables in relation to pitch velocity and elbow varus torque in collegiate baseball pitchers using force plates, an inertial measurement unit, and a radar unit. The purpose of this study is to investigate the kinetic variables being measured and their relationship to pitch velocity and loads being placed on the medial elbow. Twelve collegiate baseball pitchers participated in this study, which was approved by the IRB. Peripheral Artery Disease (PAD) is a pathology that can decrease blood flow to the lower limbs, resulting in the inability to regulate body temperature [1]. The effects of impaired blood flow caused by PAD on body temperature are unclear. Coordination is foundational to human movement. One prominent model of coordination is the Haken-Kelso-Bunz which predicts change in relative phase between two oscillators during movement. Use of this model has shown that the body has a natural tendency to prefer certain coordination patterns over others. The generalizability of the model has sparked its use throughout the scientific community to observe movement through the lens of dynamical systems theory. We contend that this model can be advanced further through incorporation of visually perceived spatial asymmetries, a component not currently accounted for by the model. Robotic exoskeletons can reduce metabolic cost in healthy individuals and restore mobility in patients with peripheral artery disease (PAD). PAD is a cardiovascular disease produced by atherosclerosis of the leg arteries. The primary symptom of PAD is claudication or pain in the legs during walking, which severely shortens the distance a patient can walk. Knowing that up to 40% of the metabolic cost of walking comes from the hip muscles, different groups have been developing rigid exoskeletons and soft exosuits that assist the hip. Imposing a perturbation can induce an unstable condition during gait. Measuring the compensatory responses may be an accurate indicator of the ability to control stability. Responses to a perturbation can be quantified through the perturbed walking pattern deviations from an unperturbed condition. Aerodynamic efficiency behind the annual migration of monarch butterflies, the longest among insects, is an unsolved mystery. Monarchs migrate 4000 km at high-altitudes to their overwintering mountains in Central Mexico. The air is thinner at higher altitudes, yielding reduced aerodynamic drag and enhanced range. However, the lift is also expected to reduce in lower density conditions. To investigate the ability of monarchs to produce sufficient lift to fly in thinner air, we measured the climbing motion of freely flying monarchs in high-altitude conditions. Bone adapts its architecture to the applied load; however, it is still unclear how bone mechano-adaptation is coordinated and why potential for adaptation adjusts during the life course. Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. Low back pain (LBP) is a significant issue related to spinal stability and, therefore, to trunk stiffness. Due to the nature of their work, nursing personnel are exposed to potential risk factors for LBP, such as lifting and trunk flexion, which have been reported in the literature to lead to decreases in trunk stiffness. Hydrogels are widely used tools for tissue engineering and regenerative medicine. Characterized as biofunctional, water-based polymer matrices with tunable mechanical properties, hydrogels have promising but limited applications in biomedical engineering, due to poor and static matrix strength. Here we plan to rectify this issue by introducing a new hydrogel made from a composite of gelatin and silk fibroin crosslinked by microbial transglutaminase (mTG) instantly and beta sheet formation gradually, respectively. Mechanical loading is essential for maintaining bone tissue. Reduced mechanical loading has been shown to have a negative effect on bone, and can result in the development of disuse osteoporosis. Disuse models of muscle inactivity and immobilization, like the Botox model used in this study, result in changes in the bone microarchitecture, the mechanisms behind which are not fully understood. In a previous four-week Botox disuse study, skeletally mature 20- week-old rats experienced degradation of intracortical bone, increased vascular porosity, and decreased osteocyte lacunar density in the tibiae. Subarachnoid hemorrhage is a potentially devastating pathological condition in which bleeding occurs into the space surrounding the brain. One of the prominent sources of subarachnoid hemorrhage are intracranial aneurysms (IA): degenerative, irregular expansions of area(s) of the cerebral vasculature. In the event of IA rupture, the resultant subarachnoid hemorrhage ends in patient mortality occurring in ~50% of cases, with survivors enduring significant neurological damage with physical or cognitive impairment. Anti-resorptive drugs, principally bisphosphonates (BPs), are the mainstay of osteoporosis treatment. They work by inhibiting bone resorption/remodeling, thus preventing bone loss. However, long-term suppression of bone resorption adversely affects bone tissue mechanical properties, even while conserving bone mass. Lack of remodeling leads to accumulation of fatigue-induced microdamage, altered matrix mineralization and reduction in normal bone tissue heterogeneity, causing impaired strength and fracture toughness. The most severe consequence to patients, while rare, is Atypical Femur Fractures (i.e., complete fatigue fractures of the femoral shaft). Current face shields used in home and institutional healthcare settings create hardships for their wearers, which makes normal work routines more difficult. Recent mandates require healthcare workers to wear both surgical masks as well as plastic face shields when tending to patients. Unfortunately, the majority of face shields have been designed for hospital settings, which does not address the specific requirements for in-home therapist use. While total hip arthroplasty is considered a successful procedure, dislocation remains a serious complication as recurrent dislocations may require additional surgeries. Knowledge on dislocation events as they occur in vivo are limited, therefore researchers rely on experimental and computational methods. A custom MATLAB script and an experimental procedure utilizing a six-degree of freedom actuator were developed to further understand how various surgical considerations affect dislocation mechanics in total hip arthroplasty. The objective of this study is to determine the feasibility of improving the gait of above-knee (AK) amputees by performing daily core exercises aimed to provide an efficient and stable walking pattern. The goal of the exercise is to strengthen core muscles and form temporary neural connections in the brain aimed at improving metabolic efficiency and stability. Unstable aortic aneurysms and dissections are serious cardiovascular conditions associated with high mortality. The current gold standards for assessment of stability, however, rely on simple geometric measurements, like cross-sectional area or increased diameter between follow-up scans, and fail to incorporate information about underlying aortic mechanics. Displacement encoding with stimulated echoes (DENSE) magnetic resonance imaging (MRI) has been used previously to determine heterogeneous circumferential strain patterns in the aortas of healthy volunteers. Tissue engineering is a multidisciplinary field that investigates and develops new methods to repair, regenerate and replace damaged tissues and organs, or to develop biomaterial platforms as in vitro models. Tissue engineering approaches require the fabrication of scaffolds using biomaterials or fabrication of living tissues using cells. As the demands of customized, implantable tissue/organs are increasing and becoming more urgent, conventional scaffold fabrication approaches are difficult to meet the requirements, especially for complex large-scale tissue fabrication. For this Honors Thesis project, I designed and fabricated a ventilation and recirculation airway sub-system prototype for a hydroponic demonstration unit to be used as an educational demonstration for prospective students, illustrating the capabilities of biological engineers. This thesis focuses on the design of an autonomous air system that toggles between ventilation and recirculation modes based upon sensor output. In the literature review I discuss how ventilation can be used to achieve a targeted climate, the benefits of hands-on projects for education, and the optimum parameters for growing herbs in a hydroponic system.

Virus filtration is an integral part of the downstream purification of mammalian cell culture-derived biotherapeutics to assure the viral safety of the products. Virus filtration membranes remove viruses based on a size-exclusion mechanism. Commercial parvovirus filers possess unique membrane structure and are designed to remove smaller non-enveloped parvoviruses with size 18-26 nm. However, some filters face issues, such as pre-mature fouling, the decline of filtrate flux, and reduction in virus retention. Diabetes accelerates cancer cell proliferation and metastasis, particularly for cancers of the pancreas, liver, breast, colon, and skin. While pathways linking the 2 disease conditions have been explored extensively, there is a lack of information on whether there could be cytoarchitectural changes induced by glucose which predispose cancer cells to aggressive phenotypes. This study identifies and addresses three voids and potential shortcomings associated with the classification, assessment and management of Cerebral Palsy (CP). These concerns should not undermine the significant advancement that has been made in this specific branch of rehabilitation, but addressing them is necessary. Cerebral Palsy is a group of motor impairments due to hypoxic-ischemic brain damage around the time of birth and very commonly manifests as excessive muscle tone and poor motor control. There are classifications within CP including spasticity and dystonia.

Candida spp. pathogens continue to be a major health care burden with high mortality and exceeding a billion dollars in terms of healthcare cost. Candidainfection ranges from superficial dermatological infection to more serious blood stream infection in debilitated patients. One of the major lines of innate immune defense against Candidais phagocytosis. Dectin-1 is the antifungal receptor in myeloid cells responsible for most immune responses against fungi including phagocytosis. We looked into the signaling pathway coordinating this mechanical process downstream of Dectin-1. We found that Dectin-1 activation gives rise to a significant mechanical force generation mediated through RHOA-ROCK-MyosinII pathway. …
