The Asiatic clam Corbicula fluminea is a filter-feeding habitat generalist. Carbon isotopic composition (δ13C) of C. fluminea body tissue was compared between reaches of Kentucky’s upper Green River that differed in Cladophora growth. Corbicula fluminea from an upstream reach with little Cladophora was translocated to a downstream reach with high Cladophora levels. Individuals from both reaches were placed in mussel silos in the same downstream reach in autumn 2012 and again in 2013 for 77 and 119 days, respectively. Flow during 2012 consisted of no high flow events until late autumn. Flow patterns in 2013 were consistently higher and more …

Cabbage is considered an excellent source ofpolyphenolswith substantial antioxidant properties associated with the alleviation of oxidative stress and the prevention of free-radical mediated diseases. Many cabbage varieties are typically blanched prior to consumption mainly to enhance associated sensory attributes. Conventional hot water (80-100°C) or steam blanching are the most industrially applied methods. Blanching causes adverse losses in the antioxidant capacity of cabbage with over 70% resultingwithin the first few minutes. Blanching time, water to cabbage ratio and cabbage variety are the main determinants of the extent of antioxidant losses. Consumers’ demands for ω-3 polyunsaturated fatty acids (PUFAs) are at all-time high. Algae, a common source of PUFAs, and antioxidants are both used as supplements in livestock feeds, are known to affect the overall quality of meat. To implement PUFA deposits into broiler meat, this study evaluated combining antioxidants and algae in broiler feed to enhance the breast meat quality. Broilers were fed diets supplemented with 50 IU Vitamin E or 200 g/ton EconomasE (EcoE, an antioxidant pack) plus 10 IU Vitamin E, with or without 0.5% algae extract (SP-1). This study investigated the antimicrobial efficacy of bioactive plant compounds along with high pressure processing (HPP) against pathogens Bacillus cereus and Cronobacter sakazakii in infant formula and infant rice cereal. The influence of these applications on antimicrobial activity, shelf-life and sensory attributes of infant foods were examined.
Trans-cinnamaldehyde (TC), (-)-Epigallocatechin gallate (EGCG) and [10]-Gingerol (GI) were incorporated (0.05%) in infant rice cereal reconstituted with infant formula. A novel combination of extraction and detection methods is demonstrated for pesticide residue analysis in vegetable samples. Acetylcholinesterase (AChE) inhibition was used as a simple colorimetric test for organophosphates/carbamates (OP/C), and was tested with extracts from the widely-used QuEChERS extraction method. In the absence of pesticide, diluted (50% with water) acetonitrile did not inhibit enzyme activity, demonstrating the compatibility of this extraction solvent with the AChE inhibition test. QuEChERS extraction of chlorpyrifos-spiked tomato, spinach and lettuce samples indicated a high sensitivity to OP/C, with AChE inhibition occurring in the ppb range. Species substitution is a form of seafood fraud for the purpose of economic gain. DNA barcoding utilizes species-specific DNA sequence information for specimen identification. Previous work has established the usability of short DNA sequences—mini-barcodes—for identification of specimens harboring degraded DNA. This study aims at establishing a DNA mini-barcoding system for all fish species commonly used in processed fish products in North America. The objective of this study was to investigate the genetic characteristics of Listeria monocytogenes that contribute to the threat it poses to the food processing industry. Transcriptomic analysis of a persistent L. monocytogenes isolate following exposure to an industrial detergent identified key upregulations in biological processes involved m peptidoglycan biosynthesis, chemotaxis and motility. Two bacteriophages against serotype 4b and 4e isolates of L. monocytogenes were isolated and sequenced. Comparative genomic and functional analysis identified the receptor binding proteins in each phage, whi]e transmission electron microscopy demonstrated a conserved baseplate architecture which also exists in other Grampositive phages. The work presented in this thesis describes the isolation and characterisation of novel bacteriophages, and the subsequent exploitation of their bactericidal properties against two opportunistic food related pathogens, Mycobacterium avium subsp. paratuberculosis (MAP) and Cronobacter sakazakii.
MAP is a proven animal pathogen known to cause Johne’s disease in cattle but has been implicated as a causative agent of Crohn’s disease in humans. Despite the fact that this association has yet to be proven, significant focus has been directed towards evaluating the consequences of consuming milk contaminated with MAP. The main limitation of meals from canola and other Brassica spp. is the presence of glucosinolates (GLS), which are anti-nutritional and can even be toxic at high ingestion levels. Furthermore, large amounts of GLS can reduce palatability for livestock and thus reduce intake and growth rates. For this reason canola was bred to contain lower levels of GLS (<30 >μmol/g) and erucic acid (< 2%). However, feed inclusion rates are still limited to ~30%, and this reduces the value of canola meal. The goal of this thesis was to optimize a pretreatment and fungal conversion process to enhance the nutritional value of canola meal. Various combinations of physical/chemical pretreatments, fungal cultures, and incubation methods were investigated to metabolize GLS into cell mass, CO2, or other non-toxic components. These treatments also served to hydrolyze canola meal fiber into carbohydrates which were then metabolized by the fungi into single cell protein. Solid-state incubation conditions were initially tested, since filamentous fungi are well adapted to grow at lower moisture levels, and this would potentially reduce contamination issues with bacteria. Flask trials were performed using 50% moisture, hexane extracted (HE) or cold pressed (CP) canola meal, with incubation for 168 h at 30ºC. On HE canola meal Trichoderma reesei (NRRL-3653) achieved the greatest increase in protein content (23%), while having the lowest residual levels of sugar (8% w/w) and GLS (0.4 μM/g). Pitting corrosion of stainless steel (SS) is observed in many different industries including the dairy industry. It is of concern, because it weakens the steel, and can cause cracking. Any replacement or repair of SS equipment is very costly and also causes delays in product manufacture. Microbial Induced Corrosion (MIC) is a possible accelerator of natural corrosion seen in galvanized steel pipes. The term probiotic was technically defined as “live microorganisms which upon ingestion in certain numbers exert health benefits beyond inherent nutrition” (FAO/ WHO 2001). This definition requires that the microorganisms must be alive and present in high numbers, generally more than 109 cells per daily ingested dose. Probiotic food products are considered as functional foods which are defined to contain health-promoting components beyond traditional nutrients and the addition of probiotic cultures is one approach in which foods could be modified to become functional.

Novel molecular method use is leading to better identification and understanding of microbial presence in food and food processing environments. The application of molecular methods targeting foodborne pathogens provides genetic profiling and targeted intervention methods. Whole-genome sequencing (WGS) provides information on the genomic uniqueness of specific pathogens and is being implemented in epidemiological investigations. Transcriptomic profiling allows for tracking the differential expression of genes and elucidating stress responses. Effective methods and treatments require an understanding of intervention (e.g. sanitizers) and treatment (e.g. antibiotics) mechanisms and efficacies. Explore traditionally used medicinal plants for diabetes mellitus would be beneficial to find an alternative option for controlling global diabetic prevalence. Present study aims to compare and identify potential antidiabetic activity in Tinospora cordifolia, Gymnema sylvestre and Aegle marmelos. Fifty percent acetone and ethyl acetate extracts of each plant were studied for anti-diabetic activity using different assays (ORAC, DPPH, ABTS, TPC, α-glucosidase inhibition). Total phenolic content and anti oxidant properties were significantly different among three plants. Tinospora cordifolia ethyl acetate extract possessed significantly higher anti oxidant activity and total phenolic content. DPPH and ABTS results were significantly correlated with each …

A total of 138 Listeria isolates, including 58 Listeria welshimeri, 44 Listeria monocytogenes, and 36 Listeria innocua, from retail meat were characterized by antimicrobial susceptibility tests against nine antimicrobials. In addition, the 44 L. monocytogenes were analyzed by serotype identification using PCR and genotyping using pulsed-filed gel electrophoresis (PFGE). Resistance to one or two antimicrobials was observed in 32 Listeria (23.2%). No multidrug resistance was identified. Tetracycline resistance was the most common resistance phenotype and identified in 22 Listeria isolates. Low prevalence of resistance to ciprofloxacin, erythromycin, gentamicin, and vancomycin was also detected. L. innocua demonstrated the highest overall prevalence …

The organic industry is full of opinions and perceptions, which influence consumer purchasing decisions. A convenience sample of 69 college students between the ages of 19-23 registered in a basic food preparation class participated in this study. A sensory analysis of organic and conventional fruits, vegetables, meat and poultry, eggs and dairy measuring categorical taste differences between the two types of food. Student perceptions of organic foods in relation to health, environment and pricing and the likelihood of purchase were also studied. The influence of Diet and Well-being on organic purchases predominantly mattered somewhat to students, while price mattered completely. …

Conjugated linoleic acid (CLA) is an 18-carbon fatty acid with multiple health benefits, including anti-obesity and anti-carcinogenic properties. CLA-rich soy oil (CLARSO) can be produced through a heterogeneous catalysis process, and this oil was previously used to produce CLA-rich margarines and shortenings. The objective of this study was to produce CLA-rich chocolate pastes by replacing a portion of the fat with CLARSO and compare the rheological (flow), textural, and thermal properties of these pastes to controls made with either soy oil or traditional fats. Mathematical modelling of transport phenomena in food processes is vital to understand the process dynamics. In this work, we study the process of double sided cooking of meat by developing a mathematical model for the simultaneous heat and mass transfer. The constitutive equations for the heat and mass transport are based on Fourier conduction, and the Flory–Huggins theory respectively, formulated for a two-phase transport inside a porous medium. We investigate a reduced one-dimensional case to verify the model, by applying appropriate boundary conditions. The overall purpose of this preparatory course textbook is to help students familiarize with some terms and some basic concepts they will find later in the Human Anatomy and Physiology I course.
The organization and functioning of the human organism generally is discussed in terms of different levels of increasing complexity, from the smallest building blocks to the entire body. This Anatomy and Physiology preparatory course covers the foundations on the chemical level, and a basic introduction to cellular level, organ level, and organ system levels. There is also an introduction to homeostasis at the beginning.

This experiment exposed third and fourth year undergraduates and graduate students taking a course in advanced food analysis to DNA extraction, polymerase chain reaction (PCR), and DNA sequence analysis. Students provided their own fish sample, purchased from local grocery stores, and the class as a whole extracted DNA, which was then subjected to PCR, gel electrophoresis and Sanger sequencing. Students retrieved their sequences and identified their fish using the NCBI BLAST nucleotide database.
