Regulatory agencies worldwide have implemented stringent measures to monitor and reduce Salmonella contamination in poultry products. Rapid quantitative detection methods enable producers to identify contamination early, implement corrective actions, and enhance food safety. This study aimed to develop and optimize a surface plasmon resonance (SPR) biosensor for the quantitative detection of Salmonella Typhimurium in ground chicken. The sensor surface was functionalized with a well-characterized monoclonal antibody specific to Salmonella flagellin, and an SPR workflow was established for quantitative analysis. Food safety strategies are being developed in the hopes of combating foodborne pathogens and their risks. Previous techniques and technologies are struggling to meet the needs of the public, leading to the demand for new, innovative methods. Mild high-pressure processing (HPP) of 350 MPa was used in a synergetic combination with mild heat of 60°C and the antimicrobial carvacrol for the decontamination of non-typhoidal Salmonella serovars that resulted in a >5 log CFU/mL reduction. Overexploitation of antibiotics in the animal industry has driven the need for reduced usage, but this reduction has intensified the challenge posed by Salmonella enterica and Campylobacter jejuni to livestock systems and public health. This situation underlines the need for sustainable antimicrobial alternatives. Although cyanobacteria are promising in this respect, since they may produce a variety of bioactive metabolites, the amounts of these metabolites in their natural state are insufficient to elicit potent antibacterial action. This study evaluated the antibacterial efficacy and growth-promoting potential of Moringa oleifera and Thymus vulgaris methanolic extracts in broiler chickens challenged with Escherichia coli O78. In vitro antibacterial screening using agar well diffusion and disc diffusion assays revealed that ciprofloxacin exhibited the strongest inhibitory effect, followed by tetracycline and enrofloxacin, whereas among plant extracts, T. vulgaris was more effective than M. oleifera. The optimal combination (M100T150; 100 mg M. oleifera + 150 mg T. vulgaris) produced the largest inhibition zones against E. coli strains. This study evaluated the effects of replacing inorganic trace minerals with organic glycinate forms at two inclusion levels in layer breeders during the late laying cycle over 6 weeks (66 to 72 weeks of age). For this, a total of 180 layer breeders (162 hens and 18 males; Lohmann LSL Ultralite) were randomly divided into three treatment groups having six replicates of nine hens each and one male for each replicate following a completely randomized design (CRD). Obesity remains a major global health concern, contributing significantly to chronic diseases. While diet is a well-known driver of obesity, emerging evidence suggests the key role of gut microbiota in mediating diet-related metabolic outcomes. Numerous epidemiological and microbiome studies suggested diet-microbiome-obesity associations. However, consistent microbial markers and mechanistic insights remain limited. To address this gap, this study aimed to identify consistent microbial species associated with obesity and investigate how diet influences their growth and function. A review of over 200 publicly available 16S rRNA and shotgun metagenomic studies revealed consistent microbial patterns in obesity, including members of Firmicutes and Bacteroidetes. …

To investigate the impacts of dietary Bacillus-based probiotics and yeast-derived prebiotics on the hepatic transcriptome profile, 500 Hisex White laying hens were randomly allotted into five dietary treatments from 37 to 52 weeks of age: control; control + Bacillus subtilis; control + Bacillus subtilis and Bacillus licheniformis; control + Bacillus coagulans; and control + Saccharomyces cerevisiae yeast cell wall. Transcriptome analysis revealed a substantial number of differentially expressed genes exclusively between the control and prebiotic groups, identifying 2221 genes (FDR ≤ 0.05), with 980 genes upregulated (log2 fold change 0.69 to 24.62) and 1241 downregulated (log2 fold change −0.74 to …

Highly pathogenic avian influenza (HPAI) remains a persistent threat to global poultry production and public health. Current vaccine platforms show limited cross-clade efficacy and often fail to induce mucosal immunity. Recent advances in microbiome research reveal critical roles for gut commensals in modulating vaccine-induced immunity, including enhancement of mucosal IgA production, CD8+ T-cell activation, and modulation of systemic immune responses. Engineered commensal bacteria such as Lactococcus lactis, Bacteroides ovatus, Bacillus subtilis, and Staphylococcus epidermidis have emerged as promising live vectors for antigen delivery. In the current study, the combined effects of essential oils (EOs) and polyphenols (PPs) were investigated as potential antibiotic alternatives. Thymol and carvacrol at a ratio of 1:1 was used as the EO due to their well-proven synergistic antimicrobial effect. The PPs were an extracted and freeze-dried product from grape pomace. The treatment solutions were prepared at the EO to PP ratios of 0:10, 3:7, 5:5, 7:3, and 10:0, at total concentrations ranging from 0 to 1000 ppm. Foodborne pathogens pose significant risks to public health and food safety, necessitating innovative control strategies. High-pressure processing (HPP) was evaluated as an intervention against Listeria monocytogenes in cold-smoked rainbow trout, with treatments at 500 MPa and 4.4 °C and 60.0 °C achieving up to a 5.03 log CFU/g reduction. The efficacy of nisin in suppressing Listeria monocytogenes and Listeria innocua during refrigerated storage was also examined, demonstrating significant microbial inhibition, particularly in the early storage phase, supporting its application as a hurdle technology. Hepatitis A viral outbreaks continue to occur. It can be transmitted through aerosolized droplets and thus can contaminate surfaces and the environment. Ultraviolet light emitting diode (UV-C LED) systems are used for inactivation of microbes, though research is needed to determine optimal doses for aerosolized HAV inactivation. This study evaluates the UV-C LED doses for the inactivation of aerosolized hepatitis A virus (HAV) deposited on stainless-steel and glass discs. Background/Objectives: Porcine reproductive and respiratory syndrome virus (PRRSV) significantly impedes swine production due to rapid genetic variation and suppression of antiviral interferon (IFN) responses, leading to ineffective immunity. To address this, we developed IFNmix, a replication-competent PRRSV modified live vaccine (MLV) candidate co-expressing three Type I IFN subclasses (IFNα, IFNβ, IFNδ) to enhance antiviral immunity. Methods: In two independent in vivo experiments, we compared the protection of IFNmix and a commercial PRRSV MLV vaccine during challenge with a virulent PRRSV strain. Many countries have established regulatory frameworks to monitor and mitigate Salmonella contamination in poultry products. The ability to rapidly quantify Salmonella is critical for poultry processors to facilitate early detection, implement corrective measures, and enhance product safety. This study aimed to develop an Immunomagnetic Chemiluminescent Assay (IMCA) for the quantification of Salmonella Typhimurium in ground chicken. Immunomagnetic microbeads functionalized with monoclonal antibodies were employed to selectively capture and concentrate Salmonella from ground chicken samples. In this study, four underutilized spices (fruit and ulb of Aframomum sulcatum, fruit of Xylopia africana, and bark of Hypodaphnis zenkeri) were analyzed for their physical properties, nutrients composition, and profile of volatile compounds using standard analytical methods. The protein, fat, sugar, and dietary fiber contents were analyzed. The spices were revealed to be a good source of essential amino acids and fatty acids including pentadecanoic acid. Several minerals (Mn, Mg, Ca, Fe, and K) of nutritional importance were found at various proportions. The main objective of this study was to investigate the effectiveness of ultraviolet light (UV-C) emitting diodes for the decontamination of stainless steel food contact surfaces. Listeria monocytogenes (ATCC 19115), Escherichia coli (ATCC 25922), and Salmonella enterica serovar Typhimurium (ATCC 700720) were chosen as challenge microorganisms. Target microorganisms were subjected to UV-C dosages of 0, 2, 4, 6, and 8 mJ cm−2 at an average fluence of 0.163 mW/cm2 using a near-collimated beam operating at 279 nm wavelength. Fermentation of food waste into 2,3-butanediol (2,3-BDO), a high-value chemical, is environmentally sustainable and an inexpensive method to recycle waste. Compared to traditional mesophilic fermentation, thermophilic fermentation can inhibit the growth of contaminant bacteria, thereby improving the success of food waste fermentation. However, the effects of sugar and nutrient concentrations in thermophilic food waste fermentations are currently unclear. The consumption of non-dairy milk is on the rise due to health benefits. Although there is increasing inclination towards milk alternatives (MA), there is limited data on antibiotic resistant bacteria in these substitutes. The aim of this study was to investigate antimicrobial resistance of bacteria isolated from MA. A total of 138 extracts from almonds (n = 63), cashew nuts (n = 36), and soybeans (n = 39) were analyzed for Enterobacteriaceae. Conversion of food waste into 2,3-butanediol (2,3-BDO) via microbial fermentation provides a promising way to reduce waste disposal to landfills and produce sustainable chemicals. However, sterilization of food waste, an energy- and capital-costly process, is generally required before fermentation to avoid any contamination, which reduces the energy net output and economic feasibility of food waste fermentation. Current study investigated effects of elevated hydrostatic pressure exposure in the presence of mild heat and natural antimicrobials against Staphylococcus aureus. Hydrostatic pressure of 350 to 550 MPa with nisin (5000 IU/mL), carvacrol, or caprylic acid (0.5% v/v) were applied for the reduction in four-strain mixture of S. aureus in HEPES buffer at 4 and 40 °C for up to 7 min. Results were statistically analyzed by ANOVA and D-values were additionally calculated using best-fitted linear model. Prior to exposure to treatments at 4 °C, counts of the pathogen were 7.95 ± 0.4 log CFU/mL and were reduced (p < 0.05) to 6.44 ± 0.3 log CFU/mL after 7 min of treatment at 450 MPa. This study developed and evaluated risk communication messages for ready to eat (RTE) foods targeted towards consumer storage practices in a food safety health campaign. Concepts were determined from a fractional factorial design of five categories of attributes potentially present in health promotion: title, message, graphic, slogan, and icon. Consumers viewed a subset of concepts and scored how useful the concept was in remembering to throw away RTE foods that were stored too long. This study investigated the prevalence of antimicrobial-resistant bacteria in retail edible offal and muscle meats in Nashville, Tennessee. A total of 348 retail meats (160 edible offal and 188 muscle) were analyzed for Salmonella enterica serovar, Campylobacter, Escherichia coli, E. coli O157:H7, and enterococci. Bacteria was identified using biochemical and PCR methods. Salmonella enterica serovar (4.4% and 4.3%), Campylobacter (1.9% and 1.1%), E. coli (79.4% and 89.4%), and enterococci (88.1% and 95.7%) was detected in offal and muscle meats, respectively. Chicken liver (9.7%) was most frequently contaminated with Salmonella enterica serovar, followed by ground chicken (6.9%) and chicken wings (4.2%). …

The efficacy of a UV-A light emitting diode system (LED) to reduce the concentrations of aflatoxin B1, aflatoxin M1 (AFB1, AFM1) in pure water was studied. This work investigates and reveals the kinetics and main mechanism(s) responsible for the destruction of aflatoxins in pure water and assesses the cytotoxicity in liver hepatocellular cells. Irradiation experiments were conducted using an LED system operating at 365 nm (monochromatic wave-length). Known concentrations of aflatoxins were spiked in water and irradiated at UV-A doses ranging from 0 to 1,200 mJ/cm2. The anti-inflammatory effects of phytochemicals, bioactive components from plants having health benefits, have been heavily investigated in the last several decades. However, the gap between the high dosage demands (μM) of phytochemicals in vitro studies and the low bioavailability (nM) of most phytochemicals after consuming relevant foods/supplements in humans undermines the application of these phytochemicals in the prevention of chronic inflammation and its related chronic diseases in humans. Bitter melon (Momordica charantia L.) has been used for anti-diabetes treatment for decades. Indian and Chinese bitter melons (BM) are two commonly produced cultivars in the US market. This study has comparatively evaluated the effects of two processing methods (fresh and freeze-drying) on Chinese and Indian BM by measuring their bioactivity in terms of total phenolic content (TPC), total triterpene content (TTC), antioxidant activity, and antidiabetic properties using the DPPH free radical scavenging and reducing power assays, and the α-amylase and α-glucosidase inhibition assays. The efficacy of UV-C irradiation as a nonthermal processing method for skim milk (SM) was investigated. SM inoculated with two surrogate viruses (MS2 and T1UV), and three bacteria (Escherichia coli ATCC 25922, Salmonella enterica serovar Typhimurium ATCC 13311, and Listeria monocytogenes ATCC 19115) was treated with Deanflow UV-C irradiation, a spiral reactor with the fluid pumped around a central low-pressure mercury UV lamp (40 W) emitting at 254 nm wave-length.
