Fucoxanthin is a bioactive compound found in one of the most prolific and sustainable organisms on the planet, alga. Its efficacy and potential in terms of health applications have been widely reported in clinical studies. Technical modifications, such as encapsulation, and sensory trials must be undertaken before fucoxanthin can be successfully utilised as a functional food ingredient. Factors to consider include solubility in the food matrix, organoleptic effects, stability, preservation against oxidation, consumer acceptability, bioavailability, and toxicity risk. Storage of grain in bags is common in Africa, Asia, and many other less developed countries making a bag probing method well-suited for moisture content (MC) measurement. A low-cost meter was developed under a USAID project to reduce post-harvest loss (PHL). The meter, referred to as the PHL meter, measures the MC of maize and other grains based on relative humidity (RH) and temperature (T) measurements obtained by a small digital sensor located in the tip of a tubular probe that can be inserted into bags of grain or other grain bulks. งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาผลของควิโนนต่อสมบัติของฟิล์มเจลาติน และศึกษาผลของการบ่มสารละลายฟิล์มและการบ่มแผ่นฟิล์มด้วยความร้อนต่อสมบัติของฟิล์มเจลาตินที่เติมควิโนน ในขั้นตอนแรกของงานวิจัยเป็นการศึกษาผลของชนิดและความเข้มข้นของควิโนนต่อสมบัติของฟิล์มเจลาติน แปรชนิดของควิโนนเป็น 2 ชนิด คือ ไฮโดรควิโนน และพารา-เบนโซควิโนน และแปรความเข้มข้นของควิโนนเป็น 4 ระดับ (0.5, 1.0, 1.5 และ 3.0% โดยน้ำหนักของเจลาติน) พบว่าเมื่อความเข้มข้นของควิโนนเพิ่มขึ้นในช่วง 0.5-1.5% ฟิล์มเจลาตินมีความต้านทานแรงดึงขาดและการยืดตัวถึงจุดขาดเพิ่มสูงขึ้น แต่เมื่อความเข้มข้นของควิโนนเพิ่มเป็น 3.0% ฟิล์มเจลาตินกลับมีความต้านทานแรงดึงขาดและการยืดตัวถึงจุดขาดลดต่ำลง เมื่อเปรียบเทียบระหว่างชนิดของควิโนนพบว่าพารา-เบนโซควิโนนมีความสามารถในการปรับปรุงสมบัติเชิงกลของฟิล์มเจลาตินสูงกว่าไฮโดรควิโนน นอกจากนี้ยังพบว่าการเติมควิโนนมีผลสำคัญต่อสมบัติเชิงแสงของฟิล์มเจลาติน โดยฟิล์มที่เติมควิโนนมีค่า L* ลดลงในขณะที่ a* และ b* มีค่าเพิ่มขึ้น การเติมควิโนนส่งผลให้มุมสีมีค่าลดต่ำลงจนเข้าใกล้มุมสีของสีแดง นอกจากนี้ยังมีผลให้ความเข้มสีสูงขึ้นและความโปร่งใสลดลงเมื่อเทียบกับตัวอย่างควบคุมที่ไม่เติมควิโนน นอกจากนี้การเติมควิโนนยังทำให้สภาพให้ซึมผ่านของไอน้ำและความสามารถในการละลายน้ำของฟิล์มเจลาตินมีค่าลดลง ในขณะที่มุมสัมผัสระหว่างหยดน้ำกับผิวฟิล์มมีแนวโน้มเพิ่มขึ้นซึ่งแสดงให้เห็นว่าผิวฟิล์มมีสมบัติไม่ชอบน้ำมากขึ้น การติดตามพฤติกรรมการดูดความชื้นที่อุณหภูมิคงที่ 25ºC ของฟิล์มที่เติมไฮโดรควิโนนและพารา-เบนโซควิโนนเข้มข้น 1.5% พบว่าตัวอย่างฟิล์มมีเส้นพฤติกรรมการดูดความชื้นชนิด type II และวอเตอร์แอกทิวิตีที่เปลี่ยนแปลงไปส่งผลต่อสมบัติเชิงกลของฟิล์มอย่างมีนัยสำคัญ (p≤0.05) การวิเคราะห์รูปแบบของแถบโปรตีนด้วยวิธี SDS-PAGE ยืนยันการเกิดการเชื่อมข้ามของโปรตีนในตัวอย่างที่เติมควิโนน นอกจากนี้จากการติดตามโดยเทคนิคทางฟูเรียร์แทรนสฟอร์มอินฟราเรดสเปกโทรสโกปียังยืนยันการเกิดพันธะโควาเลนต์ C-N ในขั้นตอนที่สองของงานวิจัยเป็นการศึกษาผลของการบ่มสารละลายฟิล์มด้วยความร้อนต่อสมบัติของฟิล์มเจลาตินที่เติมไฮโดรควิโนนและพารา-เบนโซควิโนนเข้มข้น 1.5% แปรอุณหภูมิการบ่มเป็น 3 ระดับ (45, 55 และ 65ºC) และระยะเวลาการบ่มเป็น 3 ระดับ (10, 20 และ 30 นาที) โดยทั่วไปพบว่าการบ่มสารละลายฟิล์มทำให้ฟิล์มที่ได้มีความต้านทานแรงดึงขาดเพิ่มสูงขึ้นและมีการยืดตัวถึงจุดขาดลดต่ำลงเมื่อเปรียบเทียบกับตัวอย่างควบคุมซึ่งเป็นฟิล์มที่เติมควิโนนชนิดเดียวกันแต่ไม่มีขั้นตอนการบ่มสารละลายฟิล์ม นอกจากนี้ยังพบว่าการบ่มสารละลายฟิล์มทำให้ฟิล์มที่ได้มีความสว่างและความโปร่งใสลดลงและมีความเข้มสีเพิ่มขึ้น การบ่มสารละลายฟิล์มด้วยภาวะที่ไม่รุนแรงไม่มีผลสำคัญต่อสภาพให้ซึมผ่านได้ของไอน้ำ ในขณะที่การบ่มที่ภาวะรุนแรง (ได้แก่ อุณหภูมิสูง และ/หรือ ระยะเวลายาวนาน) ทำให้สภาพให้ซึมผ่านได้ของไอน้ำและมุมสัมผัสระหว่างหยดน้ำกับผิวฟิล์มมีค่าลดต่ำลง นอกจากนี้ยังพบว่าฟิล์มที่ผลิตโดยมีขั้นตอนการบ่มสารละลายฟิล์มมีความสามารถในการละลายน้ำลดต่ำลงเมื่อเปรียบเทียบกับตัวอย่างควบคุม การเกิดพันธะไดซัลไฟด์ยืนยันได้จากการวิเคราะห์ด้วยเทคนิครามานสเปกโทรสโกปี โดยพบพีคในช่วงรามานชิฟท์ที่เกี่ยวข้องกับการยืดของพันธะ S-S ในขั้นตอนที่สามของงานวิจัยเป็นการศึกษาผลของการบ่มแผ่นฟิล์มด้วยความร้อนต่อสมบัติของฟิล์มเจลาตินที่เติมไฮโดรควิโนนและพารา-เบนโซ ควิโนนเข้มข้น 1.5% แปรอุณหภูมิการบ่มเป็น 3 ระดับ (45, 55 และ 65ºC) และระยะเวลาการบ่มเป็น 3 ระดับ (60, 180 และ 300 นาที) พบการเปลี่ยนแปลงในทำนองเดียวกันกับการบ่มสารละลายฟิล์มด้วยความร้อน …

Ochratoxin A (OTA) is a toxic metabolite produced by several species of fungal genera Penicillium and Aspergillus. The major OTA producer in temperate regions (i.e. U.S., Canada, Europe) is Penicillium verrucosum. Oat (Avena sativa), like any other cereal grain, can be contaminated with OTA when storage conditions are favorable for fungal growth. All cells contain proteases which hydrolyze the peptide bonds between amino acids in a protein backbone. Typically, proteases are prevented from nonspecific proteolysis by regulation and by their physical separation into different subcellular compartments; however, this segregation is not retained during cell lysis, which is the initial step in any protein isolation procedure. Prevention of proteolysis during protein purification often takes the form of a two-pronged approach; firstly inhibition of proteolysis in situ, followed by the early separation of the protease from the protein of interest via chromatographical purification. Near infrared hyperspectral imaging has been applied to grape seeds in order to select a representative subset of samples according to their spectral features in the 900-1700nm range. Afterwards, selected grape seeds have been classified according to their total phenol and flavanol extractabilities. In this way, samples were sorted in three different groups identified as low, medium and high extractability levels. In order to establish the chemical structures which can be responsible for the different extractabilities, vibrational spectroscopy has been applied to the non-extracted material after seed extractions. Fresh beef color is critical to consumers’ purchase decisions, and beef color stability is muscle-specific. Sarcoplasmic proteome plays a critical role in beef color stability. This dissertation focuses on the proteome basis of inter- and intra-muscular variations in beef color.

The first experiment examined the sarcoplasmic proteome of three beef muscles with differential color stability, i.e., longissimus lumborum (LL), psoas major (PM), and semitendinosus (ST), during wet-aging. The research within this thesis sought to determine the ability of various animal derived fats and plant derived oils to support the survival of several pathogenic cocktails over a multitude of storage times. The Salmonella study explored the survival rate of a four strain Salmonella cocktail in beef tallow, pig lard, duck fat, coconut oil, and extra virgin olive oil over seven days at 26˚C and 37˚C storage. However, there are limited data available on the microbial safety related to this sector of agriculture. The objective of the present study was to examine the prevalence and antimicrobial susceptibilities of E. coli and Enterococcus in soil and vegetables associated with urban agriculture.

Differential protein precipitation is a rapid and economical step in protein purification and is based on exploiting the inherent physicochemical properties of the polypeptide. Precipitation of recombinant proteins, lysed from the host cell, is commonly used to concentrate the protein of choice before further polishing steps with more selective purification columns (e.g., His-Tag, Size Exclusion, etc.). Recombinant proteins can also precipitate naturally as inclusion bodies due to various influences during overexpression in the host cell. This study focused on investigating the optimal stability conditions for R.intestinalis that can associate with an increase in growth. This study investigated the feasibility of oxidative modification with glucose oxidase (GluOx) to enhance the rheological and gelling properties of myofibrillar protein. Differential oxidative modifications of myofibrillar protein (MP) by hydroxyl radicals generated in an enzymatic system with glucose oxidase (GluOx) in the presence of glucose/FeSO4 compared to a Fenton system (H2O2/FeSO4) were investigated. Firmer and more elastic MP gels were produced by the GluOx-oxidizing system than by the Fenton system at comparable H2O2 levels due to an altered radical reaction pathway.

A challenge study to validate a 5 log10 CFU/g reduction of non-O157 Shiga-toxin producing Escherichia coli (STEC) in dry fermented sausage (DFS) was performed. A 4.49 ± 0.474 log10 CFU/g was achieved over two trials. The results indicated that the process was not effective in reducing the pathogen to the level required of most pathogens by the USDA.

Lactobacillus salivarius L28 (L28) was screened in vitro for the ability to inhibit STEC utilizing the paper disk diffusion method. This strain is a known bacteriocin producer. The results revealed that L28 would be a good candidate for use as …

A rapid quantitative PCR (qPCR) method was developed for the detection and quantification of fungi that are potentially present in dairy commodities. Genes of interest that were considered and used in the method development were the following: 18S rRNA, actin, beta-tubulin, and elongation factor 1-alpha. The following organisms were screened in this method development: Galactomyces candidus, Debaryomyces hansenii, Yarrowia lipolytica, Penicillium roqueforti, Penicillium verrocosum and Cladosporium cladosporioides. Nutritional concerns have impacted the protein market, decreasing red meat consumption as well as prompting the advent of lean and extra lean ground beef. However, such lean blends of ground beef may suffer in palatability. This study seeks to bridge the gap between perceived health and palatability. Participants were asked to identify the relative importance of characteristics commonly used in purchasing ground beef and select a preferred package of ground beef from labeled and unlabeled sections consisting of 4%, 10%, 20%, and 27% fat content. This study addresses the overarching topic of food insecurity by giving voice to individuals who are dedicating their careers toward combating child food insecurity throughout their communities. Voices are uplifted through the representation of narratives by volunteer coordinators overseeing child feeding programs, which have been established throughout Kentucky as an effort to help alleviate child hunger. This study is guided by London’s theory on career motivation, with the outcomes of this study serving as a pilot for future research centered around individuals working to combat child food insecurity.
