งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาเครื่องดื่มเยลลี่โปรตีนสูงจากโปรตีนเวย์ โดยใช้โปรตีนเวย์ไอโซเลท (whey protein isolate; WPI) เป็นแหล่งของโปรตีน ใช้เจลาติน (gelatin; GT) และบุก (konjac glucomannan; KGM) เป็นสารที่ทำให้เกิดเจล ในขั้นต้น พัฒนาสูตรเครื่องดื่มเยลลี่ที่มีโปรตีน 2 ระดับ คือร้อยละ 6 และร้อยละ 10 (โดยน้ำหนัก) โดยมีส่วนประกอบของสารปรับกรด (กรดซิตริก:กรดมาลิก; 1:1) สารให้ความหวาน (ซูคราโลส:อะซิซัลเฟม โพแทสเซียม; 1:1) และสารให้กลิ่นรส ร้อยละ 0.025 (โดยน้ำหนัก) ในสูตรเครื่องดื่มเยลลี่ทั้งสอง 2 ความเข้มข้น จากนั้นแปรอัตราส่วนของสารที่ทำให้เกิดเจล (GT:KGM) ที่ความเข้มข้นร้อยละ 1.5 (โดยน้ำหนัก) เป็น 5 อัตรส่วน ได้แก่ 100:0 75:25 50:50 25:75 และ 0:100 เพื่อวิเคราะห์สมบัติทางเคมีกายภาพและลักษณะทางประสาทสัมผัส ในการคัดเลือกตัวอย่างเครื่องดื่มเยลลี่โปรตีนสูง 2 ระดับ พบว่าเครื่องดื่มเยลลี่ที่มีโปรตีนร้อยละ 6 และ 10 (โดยน้ำหนัก) ถูกเลือกจากคะแนนความชอบโดยรวมของผลิตภัณฑ์สูงสุดที่มีนัยสำคัญ (p £ 0.05) จากผู้ทดสอบจำนวน 30 คน ซึ่งได้อัตราส่วนที่เหมาะสมของ GT:KGM ที่ 75:25 และ 25:75 ตามลำดับ โดยมีปริมาณของแข็งทั้งหมดเท่ากับ 9.10 และ 12.50 °Brix ค่า pH เท่ากับ 3.41 และ 3.43 ค่าความแข็งแรงของเจลเท่ากับ 58.02 และ 35.00 g (force) และความสามารถในการอุ้มน้ำเท่ากับร้อยละ 93.00 และ 83.90 ตามลำดับ ทั้งนี้สมบัติทางการไหลของเครื่องดื่มเยลลี่ที่มีโปรตีนร้อยละ 6 …

งานวิจัยนี้มีวัตถุประสงค์ เพื่อพัฒนาแนวทางการใช้สับปะรดตกเกรดผลิตเซลลูโลสจากแบคทีเรียที่อุดมด้วยจุลินทรีย์โพรไบโอติก เพื่อเป็นเครื่องดื่มฟังก์ชั่น งานวิจัยประกอบด้วย (i) ประเมินความเป็นไปได้ในการใช้น้ำสับปะรดเต็มส่วน (pH 4.0) เพื่อผลิต Bacterial cellulose (BC) โดยเติมเพียงแอมโมเนียมไดไฮโดรเจนฟอสเฟต (0.6 % (w/v)) สำหรับชักนำการทำงานของเอนไซม์ cellulose synthase โดยทดลองหมักด้วยแบคทีเรียมาตรฐาน 4 สายพันธุ์ ได้แก่ Gluconacetobacter xylinus TISTR 1064 (ATCC 23767), Komagataeibacter xylinus TISTR 086, K. xylinus TISTR 428 และ K. xylinus TISTR 1061 พบว่า แต่ละสายพันธุ์ผลิต BC ในน้ำสับปะรดได้แตกต่างกัน โดย TISTR 428 ผลิต BC ได้มากที่สุด (105.10±0.08 g/L) (p<0.05) จึงเลือกมาประเมินสภาวะการหมัก โดยแปรพื้นที่ผิวสัมผัสอากาศ และปริมาตรของน้ำสับปะรด พบว่า ค่า BC yield (y) มีความสัมพันธ์เชิงเส้นตรงกับอัตราส่วนระหว่างพื้นที่ผิวสัมผัสอากาศต่อปริมาตรน้ำหมัก (x) ดังสมการ y =19.422x + 25.936 (R2 = 0.9346) และพบว่า เมื่ออัตราส่วนระหว่างพื้นที่ผิวสัมผัสอากาศต่อปริมาตรของน้ำหมักเพิ่มขึ้น Hardness และ Cohesiveness มีค่าลดลง (p<0.05) ในขณะที่ Springiness, Gumminess และ Chewiness มีค่าเพิ่มขึ้น (p<0.05) (ii) คัดแยกแบคทีเรียผลิต BC เพื่อพัฒนาเป็นสายพันธุ์จำเพาะในการหมักน้ำสับปะรด โดยคัดแยกจากน้ำหมักสมุนไพรที่มีส่วนผสมของน้ำและ/หรือเนื้อสับปะรดที่มีอายุการหมัก 8 ปี 5 ปี และ 10 เดือน พบว่า สามารถคัดแยกแบคทีเรียที่มีสมบัติสร้างสารคล้าย BC ในน้ำสับปะรดทั้งหมด 4 ไอโซเลท จากน้ำหมักอายุ 10 เดือน เมื่อระบุชนิดด้วยการวิเคราะห์ลำดับนิวคลีโอไทด์ที่ตำแหน่ง 16s rDNA/RNA พบว่า ไอโซเลท K1, K4, K8 และ K3 ที่สร้างสารคล้าย BC คือ Kodamaea ohmeri (98.33%) Acinetobacter sp. (73.97%) Enterobacter cloacae (82.31%) และ G. xylinus (80.16%) ตามลำดับ เลือก K3 ที่มีสมบัติทางจีโนไทป์ที่ตรงกับสายพันธุ์ทางการค้า และเมื่อนำมาศึกษาสมบัติการสร้าง BC ในน้ำสับปะรด พบว่า สามารถสร้าง BC ได้มากกว่าและเร็วกว่าสายพันธุ์ TISTR ถึง 2 เท่า จึงนำไอโซเลท K3 มาพัฒนาเป็นสายพันธุ์จำเพาะในการผลิต BC ในน้ำสับปะรด โดยเพาะเลี้ยงภายใต้สภาวะที่ประเมินได้จาก (i) และแปรปริมาณแหล่งคาร์บอน (ซูโครส) ปริมาณแหล่งไนโตรเจน (เพพโทน) และปริมาณสารชักนำ (โซเดียมไดไฮโดรเจนฟอสเฟต) โดยใช้แผนการทดลองแบบ 3x2x2 แฟคทอเรียล พบว่า สภาวะที่มีปริมาณซูโครส เพพโทน และโซเดียมไดไฮโดรเจนฟอสเฟตที่ความเข้มข้น 5, 0.5 และ 0 % (w/v) ตามลำดับ มีปริมาณ BC ภายหลังการหมักสูงที่สุด (188.27±0.50 g/L) BC ที่ได้มีปริมาณความชื้นและค่า Aw สูงกว่าสภาวะอื่น (p<0.05) และเมื่อวิเคราะห์เนื้อสัมผัส (Texture profile analysis) พบว่า มีค่า Hardness ต่ำที่สุด และค่า Springiness สูงที่สุด น้ำสับปะรดหลังหมักประกอบด้วย กรดอินทรีย์หลักมากที่สุด คือ กรดซิตริก กรดมาลิก และกรดแอซิติก ตามลำดับ น้ำตาลประกอบด้วยฟรักโทสมากที่สุด รองลงมา คือ กลูโคส และซูโครส ตามลำดับ อีกทั้งมีปริมาณสารประกอบฟีนอลิกสูงที่สุด (p<0.05) และมีฤทธิ์ต้านออกซิเดชั่นสูงที่สุด (p<0.05) (iii) นำสภาวะที่ประเมินได้จาก (ii) มาใช้ในการผลิต BC ที่อุดมด้วยโพรไบโอติก โดยการเพาะเลี้ยง K3 ร่วมกับจุลินทรีย์ Lactobacillus plantarum FTCU10621 พบว่า BC ที่ได้มีปริมาณน้อยลง (26.40±0.80 g/L) และเป็น young BC และตรวจวัด L. plantarum FTCU10621 ในส่วน BC พบจำนวน 8.42±0.52 log CFU/g BC และในน้ำสับปะรดจำนวน 8.33±0.03 log CFU/ml ภาพถ่ายทางสัณฐานวิทยาของผิวหน้า BC พบเซลล์โพรไบโอติกถูกตรึงอยู่ใน BC คิดเป็น 7.08 log cell/g BC เมื่อวิเคราะห์โครงสร้างทางเคมีของ BC ด้วยเทคนิค FTIR พบว่า สเปกตรัม IR ของหมู่ไฮดรอกซิลมี % transmittance ต่ำ ส่งผลให้ BC มีลักษณะอ่อนนุ่ม และน้ำสับปะรดหลังหมักประกอบด้วยกรดอินทรีย์หลักมากที่สุด คือ กรดแลคติก กรดซิตริก กรดซัคซินิค และกรดมาลิก ตามลำดับ น้ำตาลประกอบด้วยฟรักโทสมากที่สุด รองลงมา คือ กลูโคส และซูโครส ตามลำดับ

Moringa oleifera leaf is considered an inexpensive source of protein. As leaf protein concentrate (LPC), protein content offered is higher and easier for use than in leaf. Therefore, this study aimed to prepare moringa leaf protein concentrate (MoLPC) from dried moringa leaves, compare their physico-chemical and functional properties and determine enhanced nutritional value of pancakes fortified with MoLPC compared to unfortified pancake. Using two varieties (Thai (TMo) and Indian (IMo) moringa from Nan area), LPC from each variety was prepared by alkali solubilization (pH 9) followed by acid-precipitation at pH 4.5. This research focuses on the optimization of ultrasound-assisted extraction (UAE) conditions of marigold flower extract (MFE) using response surface methodology (RSM) and to produce microcapsule using spray drying with different wall materials and ratios. The extraction factors including temperature (30-50 ℃), time (5 -15 min), and ethanol concentration (60 - 100% v/v) were investigated. The optimum condition was investigated according to the maximum concentration of total phenolic, total flavonoid, total carotenoid, and antioxidant activity determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical and ferric reducing antioxidant power (FRAP) assay. The rumen is the first digestion site of masticated food material in cattle and provides 70% of energy to host through fermentation of forage. The prevalence of obesity is rising steadily across the world, which increases the risk of many metabolic diseases and life-threatening illnesses. A specific strain, Clostridium cochlearium (C. cochlearium), reported as a butyrate producer, could have potential probiotic effects against obesity. The objective of this study was to evaluate the effects of dietary supplementation of C. cochlearium on a high-fat diet-induced obese (DIO) mouse model. Whey protein isolate supplementation has been recognized as having potential for regulating appetite, thereby potentially improving mood and food intake. The objectives of this project were to 1) analyze the effects of high-quality whey protein intake on overall diet and 2) identify and examine a correlation between tryptophan levels and mood regulation. Rates of obesity and other health disparities are exceptionally high in rural Appalachian counties compared to the nation as a whole. One causal factor of these health disparities in Appalachian counties is the inequitable allocation of healthy food. Food insecurity and the local food environment are large drivers for obesity experienced by Martin County, Kentucky residents. Successful socioecological model (SEM) and policy, systems, and environmental (PSE) interventions have shown promising results in reducing obesity and enhancing food security in this population. Through the transcription of semi-structured focus group interviews, thematic analysis aimed to obtain perspectives on the local food system. …

In this study, a novel value-added dairy-based health formulation was developed using whey protein hydrolysate and probiotic organisms. In the first part of the study, different forms of whey proteins, concentrate WPC80, isolate WPI90, and hydrolysates WPH10, WPH15 and WPH20 were screened for bioactivities (antimicrobial, antioxidant, and antihypertensive activity). Hydrolysate WPH10, exhibiting the highest bioactivities was conjugated with maltodextrin. A batch of 2L conjugated solution was spray dried in a Niro drier with an inlet and outlet temperature of 200⁰C and 90⁰C, and alternatively, freeze dried at −80 °C under 50 mTorr vacuum. In an effort to understand the apparent trade-off between the continual push for growth performance and the recent emergence of muscle pathologies, shotgun proteomics was conducted on breast muscle obtained at similar to 8 weeks from commercial broilers with wooden breast (WB) myopathy and compared with that in pedigree male (PedM) broilers exhibiting high feed efficiency (FE). Many microbreweries practice serial fermentation, or serial repitching. Serial repitching is the process by which one yeast culture is reused for multiple batches of beer. Each batch of subsequent beer is called a “pitch.” This technique helps breweries limit production costs. However, fermentation is difficult to predict throughout serial fermentation. This leads to beer quality issues such as fluctuation in alcohol production, buildup of unwanted flavor compounds, and decreased carbon dioxide production, which results in reduced profit. High concentrations of carotenoids are protective against cardiometabolic risk traits (CMTs) in adults and children. We recently showed in non-diabetic Mexican American (MA) children that serum α-carotene and β-carotene are inversely correlated with obesity measures and triglycerides and positively with HDL cholesterol and that they were under strong genetic influences. Additionally, we previously described a Pediatric Metabolic Index (PMI) that helps in the identification of children who are at risk for cardiometabolic diseases. Zygnematophyceae green algae (ZGA) have been shown to be the closest relatives of land plants. Three nuclear genomes (Spirogloea muscicola, Mesotaenium endlicherianum, and Penium margaritaceum) of ZGA have been recently published, and more genomes are underway. Here we analyzed two Zygnema circumcarinatum strains SAG 698-1a (mating +) and SAG 698-1b (mating −) and found distinct cell sizes and other morphological differences. The molecular identities of the two strains were further investigated by sequencing their 18S rRNA, psaA and rbcL genes. These marker genes of SAG 698-1a were surprisingly much more similar to Z. cylindricum (SAG 698-2) than to SAG 698-1b. …

To determine changes in the grain components between historical and modern wheat (Triticum aestivum L.) cultivars, wholemeal flours from 19 wheat cultivars and 2 landraces released or introduced between 1870 and 2013 and grown over two crop years were extracted using hydroalcoholic solution and analyzed using one dimensional 1H NMR spectral profiling. Grain yield, grain volume weight (GVW), and grain protein concentration were also measured. Grain yield increased while protein concentration decreased by release year (p < 0.001). Increasing trends (p < 0.01) were observed for tryptophan, sum of the measured amino acids, chlorogenic acid, ferulic acid, vanillic acid, and sum of the measured phenolic acids. Grain yield, phenolic acids, and tryptophan were mainly associated with modern cultivars, whereas grain protein concentration and GVW were associated with historical cultivars. The findings from this study showed changes in concentration of grain components over a century of breeding that may have implications for grain quality and human health.

The family Apiaceae includes many important vegetables and medicinal plants. Auxin response factors (ARFs) play critical roles in regulating plant growth and development. Here, we performed a comprehensive analysis of the ARF gene family in three Apiaceae species, celery, coriander, and carrot, and compared the results with the ARF gene family of lettuce, Arabidopsis, and grape. We identified 156 ARF genes in all six species and 89 genes in the three Apiaceae species, including 28, 34, and 27 in celery, coriander, and carrot, respectively. The intestinal microbiota is a critical component of mucosal health as evidenced by the fact that alterations in the taxonomic composition of the gastrointestinal microbiota are associated with inflammatory bowel diseases. To better understand how the progression of inflammation impacts the composition of the gastrointestinal microbiota, we used culture independent taxonomic profiling to identify temporal changes in the cecal microbiota of C3Bir IL-10-/- mice concomitantly with the onset and progression of colitis. This analysis revealed that IL-10-/- mice displayed a biphasic progression in disease severity, as evidenced by histopathological scores and cytokine production. Background: Preterm birth is a major determinant of neonatal survival and morbidity, but the gut microbiome and associated enteric inflammation are also key factors in neonatal development and the risk of associated morbidities. We prospectively and longitudinally followed two cohorts of preterm infants, one of which was fed activated Bifidobacterium longum subsp. infantis (B. infantis) EVC001 8 × 109 CFU daily, and the other was not fed a probiotic. Hospital feeding protocol assigned all infants born at < 1500 g and/or < 32 weeks corrected gestational age to the probiotic feeding protocol, whereas infants born at > 1500 g and/or > 32 weeks corrected gestational age were not fed a probiotic. Background and objectives: Refrigerated dough products have the potential to be a safety hazard to consumers because they could be consumed raw or undercooked. The objectives of this study were designed to evaluate the microbial and functionality changes in high pressured sugar-cookie dough as a function of aw (0.80–0.87), pressure level (100–600 MPa), and holding time (1–6 min).
Findings: Endogenous microbial populations were marginally reduced (0.2–0.5 log CFU/g) by pressure treatments. However, treating the dough at 600 MPa for 6 min significantly reduced counts of inoculated Escherichia coli by as much as 2.0 log CFU/g. Increasing the aw …

The last few decades have seen an outpouring of gastrointestinal (GI) microbiome studies across diverse host species. Studies have ranged from assessments of GI microbial richness and diversity to classification of novel microbial lineages.
