Despite the successes provided by vaccination, many challenges still exist with respect to controlling new and re-emerging infectious diseases. Innovative vaccine platforms composed of adaptable adjuvants able to appropriately modulate immune responses, induce long-lived immunity in a single dose, and deliver immunogens in a safe and stable manner via multiple routes of administration are needed. This work describes the development of a novel biodegradable polyanhydride nanoparticle-based vaccine platform administered as a single intranasal dose that induced long-lived protective immunity against respiratory disease caused by Yesinia pestis, the causative agent of pneumonic plague. Relative to the responses induced by the …
Angiostatin Like Peptides In Milk: Potential Development For Dairy Products Capable Of Cancer Prevention, Erin Stefanutti
Angiostatin Like Peptides In Milk: Potential Development For Dairy Products Capable Of Cancer Prevention, Erin Stefanutti
Master's Theses
For the past 40 years, antiangiogenic approaches have been of major interest in the development of methods to cure and prevent cancer. Angiogenesis, the development of blood vessels from pre-existing vascularization, is essential for cancer growth and spread of metastasis through the delivery of nutrients and oxygen essential to sustain the metabolic activity of these malignant cells. Blocking access to blood will cause cancerous cells to assume a dormant state creating inactive micro-tumors innocuous to the host. Angiostatin, the internal fragment of the fibrinolytic zymogen plasminogen, has shown great potential in reducing cancer size and number of metastatic colonies in …
Host-Microbial Symbiosis In The Vertebrate Gastrointestinal Tract And The Lactobacillus Reuteri Paradigm, Jens Walter, Robert A. Britton, Stefan Roos
Host-Microbial Symbiosis In The Vertebrate Gastrointestinal Tract And The Lactobacillus Reuteri Paradigm, Jens Walter, Robert A. Britton, Stefan Roos
Department of Food Science and Technology: Faculty Publications
Vertebrates engage in symbiotic associations with vast and complex microbial communities that colonize their gastrointestinal tracts. Recent advances have provided mechanistic insight into the important contributions of the gut microbiome to vertebrate biology, but questions remain about the evolutionary processes that have shaped symbiotic interactions in the gut and the consequences that arise for both the microbes and the host. Here we discuss the biological principles that underlie microbial symbiosis in the vertebrate gut and the potential of the development of mutualism. We then review phylogenetic and experimental studies on the vertebrate symbiont Lactobacillus reuteri that have provided novel insight …

Sterile apple juice inoculated with S. cerevisiae ATCC 9763 (103 CFU/mL) was processed in a bubble column with gaseous ozone of flow rate of 0.12 L/min and concentration of 33-40 µg/mL for 8 min. The growth kinetics of S. cerevisiae as an indicator of juice spoilage was monitored at 4, 8, 12 and 16 ºC for up to 30 days. The kinetics were quantitatively described by the primary model of Baranyi and Robert’s and the maximum specific growth rate was further modeled as a function of temperature by the Ratkowsky type model. The developed model was successfully validated for …

Sorghum Clear Beer Value Chain, Bernadette C. Chimai
INTSORMIL Presentations
The markets and industrial utilization of agricultural commodities have undergone considerable change in the last few decades.
In Zambia, sorghum was considered a traditional crop with limited industrial uses.
Research and transformations in consumer tastes and preferences have led to the development of new uses for sorghum.
Potential markets for industrial utilization of sorghum have been identified as food processing, beer brewing, feed concentrates and energy production.
One of the success stories in industrial utilization of sorghum has been Eagle, a sorghum based clear beer produced by Zambian breweries.
The study reported in this paper aims to examine the main …

Characterization Of Igg And Ige Binding To Parvalbumin Derived From Commercially Important Fish Species, P. Lee, Julie A. Nordlee, S. J. Koppelman, J. L. Baumert, Steve L. Taylor
Department of Food Science and Technology: Faculty Publications
Rationale: Parvalbumin is recognized as pan-allergen in fish and frog. However, previous studies demonstrated that the IgE- and IgG-binding patterns to parvalbumins vary depending on the fish species. We aimed to use 3 anti-parvalbumin IgG and human IgE to investigate the contributing factors for the binding differences.
Methods: Indirect enzyme-linked immunosorbent assay (ELISA) and IgG immunoblotting were used to determine the reactivity of the polyclonal anti-cod parvalbumin antibody and the commercially available, monoclonal anti-frog and anti-carp parvalbumin antibodies against raw muscle extracts of 25 fish species. Additionally, sera from 46 individuals with clinical history of fish allergy were analyzed for …

Soil Composition Of Community Gardens: Are There Quality Concerns?, Shahla Wunderlich, Charles Feldman, K. Latif, P. Punamiya
Department of Nutrition and Food Studies Scholarship and Creative Works
It is suggested, but not confirmed, that soil composition and minerals in soil can affect the nutritional quality of the produce. Determining soil composition of community gardens has therefore become significant for foods that are grown in these gardens as the popularity of community gardens are on the rise. A comparative study of five (5) community garden soils from different regions of New Jersey and New York over a period of 4 months was conducted. A total of nine (9) elements including Al, Cr, Fe, Ni, Cu, Zn, As, Cd, and Pb were analysed using the EPA Method 3051A. Among …
Biodiesel Production From Crude Cottonseed Oil: An Optimization Process Using Response Surface Methodology, Xiaohu Fan, Xi Wang, Feng Chen
Biodiesel Production From Crude Cottonseed Oil: An Optimization Process Using Response Surface Methodology, Xiaohu Fan, Xi Wang, Feng Chen
Publications
Biodiesel, known as fatty acid methyl ester (FAME), was produced from crude cottonseed oil (triglycerides) by transesterification with methanol in the presence of sodium hydroxide. This process was optimized by applying factorial design and response surface methodology (RSM) with SAS and PSIPLOT programs. A second-order mathematical model was obtained to predict the yield as a function of methanol/oil molar ratio, catalyst concentration, reaction temperature, and rate of mixing. Based on ridge max analysis and RSM, as well as economic cost consideration, the practical optimal condition for the production of biodiesel was found to be: methanol/oil molar ratio, 7.9; temperature, 53 …

Differential Precipitation And Solubilization Of Proteins., Barry Ryan
Differential Precipitation And Solubilization Of Proteins., Barry Ryan
Books/Book Chapters/ Proceedings
Differential protein precipitation is a rapid and economical step in protein purification and is based on exploiting the inherent physico-chemical properties of the polypeptide. Precipitation of recombinant proteins, lysed from the host cell, is commonly used to concentrate the protein of choice before further polishing steps with more selective purification columns (e.g. His-Tag, Size Exclusion etc.). Recombinant proteins can also precipitate naturally as inclusion bodies due to various influences during over-expression in the host cell. Although this phenomenon permits easier initial separation from native proteins, these inclusion bodies must carefully be differentially solubilised so as to reform functional, correctly folded …
Do Organic Cherry Vine Tomatoes Taste Better Than Conventional Cherry Vine Tomatoes? A Sensory And Instrumental Comparative Study From Ireland, Clare Gilsenan, Roisin Burke, Catherine Barry-Ryan
Do Organic Cherry Vine Tomatoes Taste Better Than Conventional Cherry Vine Tomatoes? A Sensory And Instrumental Comparative Study From Ireland, Clare Gilsenan, Roisin Burke, Catherine Barry-Ryan
Articles
A consumer panel was able to distinguish a perceptible difference between organically farmed and conventionally produced tomatoes, and preferred the taste of the conventional tomatoes. The sensory evaluation results of the trained panel revealed that the conventional tomatoes were sweeter and less sour than the organic tomatoes. In addition to this, the conventional tomatoes showed significant differences for oBrix, reducing sugars and electrical conductivity. No significant differences were observed between the organic and conventional tomato samples for color, size, firmness, pH and dry matter values.
Avoiding Proteolysis During Protein Chromatography., Barry Ryan
Avoiding Proteolysis During Protein Chromatography., Barry Ryan
Books/Book Chapters/ Proceedings
All cells contain proteases which effect catalytic hydrolysis of the peptide bond between amino acids in the protein backbone. Typically, proteinases are prevented from non-specific proteolysis by regulation and physical separation into different sub-cellular compartments; however, this segregation is not retained during cell lysis to release a protein of interest. Prevention of proteolysis during protein purification often takes the form of a two-pronged approach; firstly inhibition of proteolysis in situ, followed by the separation of the protease from the protein of interest via chromatographical purification. Proteinase inhibitors are routinely used to limit the effect of the proteinases before they are …
