The Pseudomonas syringae type III secretion system (TTSS) translocates effector proteins into plant cells. Several P. syringae effectors require accessory proteins called type III chaperones (TTCs) to be secreted via the TTSS. We characterized the hopO1-1, hopS1, and hopS2 operons in P. syringae pv. tomato DC3000; these operons encode three homologous TTCs, ShcO1, ShcS1, and ShcS2. ShcO1, ShcS1, and ShcS2 facilitated the type III secretion and/or translocation of their cognate effectors HopO1-1, HopS1, and HopS2, respectively. ShcO1 and HopO1-1 interacted with each other in yeast two-hybrid and coimmunoprecipitation assays. Interestingly, ShcS1 and ShcS2 were capable of substituting for …

Pseudomonas syringae is a gram-negative bacterial plant pathogen that is dependent on a type III protein secretion system (TTSS) and the effector proteins it translocates into plant cells for pathogenicity. The P. syringae TTSS is encoded by hrp-hrc genes that reside in a central region of a pathogenicity island (Pai). Flanking one side of this Pai is the exchangeable effector locus (EEL). We characterized the transcriptional expression of the open reading frames (ORFs) within the EEL of P. syringae pv. tomato DC3000. One of these ORFs, PSPTO1406 (hopB1) is expressed in the same transcriptional unit as hrpK. …

The present study describes the reproductive ecology of the prostrate shrub Juniperus sabina and the dwarf shrub Artemisia santolinifolia in dry mountain steppes of southern Mongolia. Whereas stands of the juniper are located at the drought limit of the genus’ distributional range, the genus Artemisia is typical of central Asian drylands. Both species produce large numbers of reproductive organs. For Juniperus sabina, however, only 2.5% of the morphologically intact seeds were found to be viable. Correspondingly, neither seedlings nor saplings were found in the field, and recruitment by reseeding happens at most rarely under the present climatic conditions.

Plant populations are often spatially structured owing to limited dispersal of pollen and seed. Mating between neighboring individuals in such populations often leads to reduced reproductive performance relative to matings between distant individuals. This response, which may be a result of inbreeding depression or prezygotic mating barriers, was investigated for slickspot peppergrass, Lepidium papilliferum L. (Brassicaceae), a rare insect-pollinated mustard endemic to sagebrush–steppe habitat in southwestern Idaho. Through hand pollination experiments we found that individual plants receiving pollen from distant sources (75–100 m and 6.5–20 km away) had significantly higher percent fruit sets than those relying on pollen from neighboring …

We estimated levels of developmental instability in leaves, as indicated by fluctuating asymmetry (i.e., random deviations from perfect bilateral symmetry), across a broad hybrid zone between two taxa in the Piriqueta caroliniana complex. Previous studies have indicated that this hybrid zone was initiated in southern Florida and has expanded northward. We found that plants from the hybrid zone generally had higher levels of leaf developmental instability than plants from allopatric regions, but the increase in asymmetry was not substantial. Although it is usually assumed that developmental instability is associated with lower fitness, these Piriqueta hybrids from central Florida are similar …

Many Gram-negative bacterial pathogens of plants and animals are dependent on a type III protein secretion system (TTSS). TTSSs translocate effector proteins into host cells and are capable of modifying signal transduction pathways. The innate immune system of eukaryotes detects the presence of pathogens using specific pathogen recognition receptors (PRRs). Plant PRRs include the FLS2 receptor kinase and resistance proteins. Animal PRRs include Tolllike receptors and nucleotide-binding oligomerization domain proteins. PRRs initiate signal transduction pathways that include mitogen-activated protein kinase (MAPK) cascades that activate defencerelated transcription factors. Use of small grain forage crops for stocker cattle production was extensively evaluated in two separate three-year research projects at the Livestock and Forestry Branch Research Station near Batesville, Ark. The first section of this Research Report presents results of a study in which 216 commercial crossbred steers (Avg. body weights 463 lb) grazed forage of wheat, oats, rye, ryegrass, wheat + rye, wheat + ryegrass, rye + ryegrass, and wheat + rye + ryegrass during the winter and spring months from 1999 through 2002. The bacterial plant pathogen Pseudomonas syringae depends on a type III protein secretion system and the effector proteins that it translocates into plant cells to cause disease and to elicit the defense-associated hypersensitive response on resistant plants. The availability of the P. syringae pv. tomato DC3000 genome sequence has resulted in the identification of many novel effectors. We identified the hopPtoV effector gene on the basis of its location next to a candidate type III chaperone (TTC) gene, shcV, and within a pathogenicity island in the DC3000 chromosome. The extremely skewed female-biased sex ratio in the desert moss Syntrichia caninervis was investigated by assessing the regeneration capacity of detached leaves. Juvenile, green, yellow-green, and brown leaves equating to approximately 0, 2, 6, and 12 yr of age, respectively, were detached from individuals of S. caninervis collected from 10 field populations and grown in a growth chamber for 58 d at a light intensity of 33–128 µmol · m–2 · s–1. Younger leaves (0–2 yr old) tended to have a greater viability, regenerate more quickly, extend their protonemal filaments farther, produce shoots (gametophores) more quickly, produce more shoots, and …

Pseudomonas syringae pv. tomato strain DC3000 is a pathogen of tomato and Arabidopsis. The hrp-hrcencoded type III secretion system (TTSS), which injects bacterial effector proteins (primarily called Hop or Avr proteins) into plant cells, is required for pathogenicity. In addition to being regulated by the HrpL alternative sigma factor, most avr or hop genes encode proteins with N termini that have several characteristic features, including (i) a high percentage of Ser residues, (ii) an aliphatic amino acid (Ile, Leu, or Val) or Pro at the third or fourth position, and (iii) a lack of negatively charged …

A new hierarchy of intellectual property protection for plant innovation is emerging. Utility patent protection is poised to become the dominant intellectual property mechanism for plants in the U.S. and perhaps elsewhere. Plant breeder's rights systems continue to garner a dedicated following, especially in developing countries, as a means for complying with international intellectual property treaty obligations. But while utility patent and plant breeder's rights regimes have come to occupy the first tier of the intellectual property hierarchy for plants, other forms of intellectual property protection remain important, albeit in a supplemental role. Gram-negative bacteria use a variety of virulence factors including phytotoxins, exopolysaccharides, effectors secreted by the type III secretion system, and cell-wall-degrading enzymes to promote parasitism in plants. However, little is known about how these virulence factors alter plant celluar responses to promote disease. In this study, we show that virulent Pseudomonas syringae strains activate the transcription of an Arabidopsis ethylene response factor (ERF) gene, RAP2.6, in a coronatine insensitive 1 (COI1)-dependent manner. A highly sensitive RAP2.6 promoter-firely luciferase (RAP2.6-LUC) reporter line was developed to monitor activities of various bacterial virulence genes. Increasing the stearic acid content to improve soybean [Glycine max (L) Merr] oil quality is a desirable breeding objective for food-processing applications. Although a saturated fatty acid, stearic acid has been shown to reduce total levels of blood cholesterol and offers the potential for the production of solid fat products (such as margarine) without hydrogenation. This would result in the reduction of the level of trans fat in food products and alleviate some current health concerns. This report contains information from a 2001 focus group survey of production practices and marketing decisions of ‘typical’ representative Arkansas farms from selected counties in eastern Arkansas. While the National Agricultural Statistical Service (NASS) publishes similar information through the Arkansas Agricultural Statistics Service, reporting of these statistics is often not differentiated across crops or specific farm type. Therefore, one of the major objectives of this survey was to ascertain production practices for specific crops and farm types typical of row crop farms in eastern Arkansas. Responses were categorized into production regions dominated by either rice or cotton production. The results …

The bacterial plant pathogen Pseudomonas syringae possesses a type III protein secretion system that delivers many virulence proteins into plant cells. A subset of these proteins (called Avr proteins) is recognized by the plant’s innate immune system and triggers defences. One defence-associated response is the hypersensitive response (HR), a programmed cell death (PCD) of plant tissue. We have previously identified HopPtoD2 as a type III secreted protein from P. s. pv. tomato DC3000. Sequence analysis revealed that an N-terminal domain shared homology with Avr- PphD and a C-terminal domain was similar to protein tyrosine phosphatases (PTPs). We demonstrated that purified …
